In this work, we computationally investigated from quantum chemical calculations (DFT) at the BP86 level with the various basis sets def2-SVP, def2-TZVPP, and TZ2P+, chemical bonding issues of the recently described carbene-analogues gold(I) complexes AuCl-NHEMe (Au1-NHE) with E = C – Pb. The optimized structures and the metal-ligand bond dissociation energy (BDE) were calculated, and the nature of the E→Au bond was studied with charge and energy decomposition methods. The equilibrium structures of the system showed that there were major differences in the bonded orientation from the ligands NHC-NHPb to gold(I) complex between the lighter and the heavier homologues. The …

Self-forgiveness and forgiveness-seeking are important and understudied aspects of forgiveness. We examined the cardiac and emotional patterns of healthy young adults (40 women, 40 men) who recalled an unresolved offense they had caused another person. Participants engaged in four imagery conditions: ruminating about the offense, being humbly repentant and engaging in self-forgiveness, seeking forgiveness from the victim and receiving forgiveness, and seeking forgiveness from the victim and being begrudged. Being repentant and begrudged forgiveness by one’s victim was associated with the same level of guilt as when ruminating, but significantly more negative emotion, less control, and less empathy than when …
Discovering Antibacterial And Anti-Resistance Agents Targeting Multi-Drug Resistant Eskape Pathogens, Renee Fleeman
Discovering Antibacterial And Anti-Resistance Agents Targeting Multi-Drug Resistant Eskape Pathogens, Renee Fleeman
USF Tampa Graduate Theses and Dissertations
Antibiotic resistance has been a developing problem for mankind in recent decades and multi-drug resistant bacteria are now encountered that are resistant to all treatment options available. In 2014, the World Health Organization announced that this problem is driving us towards a “post-antibiotic era” that will change the face of modern medicine as we know it. If lack of novel antibiotic development and FDA approval continues, by the year 2050, 10 million people will die each year to an antimicrobial resistant bacterial infection. With lack of pharmaceutical industry involvement in developing novel antibiotics, the responsibility now lies within the academic …

Periphytic Diatoms In The Polluted Linggi (Sensu Stricto) And Kundor Rivers, Negeri Sembilan, Malaysia, Khan I.S.A. Nather
ASEAN Journal on Science and Technology for Development
The investigation in this paper aimed to describe periphytic diatom assemblage, species composition and distribution in tropical polluted Linggi (sensu stricto or s.s.) and Kundor rivers in Negeri Sembilan, Malaysia. Regardless of ecological and environmental conditions, diatoms were the numerically dominant flora among periphytic algae collected monthly over a period of 13 months at nine stations in the Linggi (s.s.) and Kundor rivers. The freshwater periphytic diatom samples were collected mainly using artificial substrates and supplemented with natural substrates. The periphytic diatoms thus collected from both natural and artificial substrates comprised 86 taxa (82 pennate and 4 centric forms) belonging …

immune response to injury, and are implicated in facilitating neural plasticity. The rodent gustatory system is highly plastic, particularly during development, and outcomes following nerve injury are more severe in developing animals. The mechanisms underlying developmental plasticity in the taste system are largely unknown, making microglia an attractive candidate. To better elucidate microglia’s role in the taste system, we examined these cells in the rostral nucleus of the solitary tract (rNTS) during normal development and following transection of the chorda tympani taste nerve (CTX). Rats aged 5, 10, 25, or 50 days received unilateral CTX or no surgery and were …

Interleukin-15 (IL-15) is a potent cytokine that increases CD8+ T and NK cell numbers and function in experimental models. However, obstacles remain in using IL-15 therapeutically, specifically its low potency and short in vivo half-life. To help overcome this, a new IL-15 superagonist complex comprised of an IL-15N72D mutation and IL-15RαSu/Fc fusion (IL-15SA, also known as ALT-803) was developed. IL-15SA exhibits a significantly longer serum half-life and increased in vivo activity against various tumors. Herein, we evaluated the effects of IL-15SA in recipients of allogeneic hematopoietic stem cell transplantation. Weekly administration of IL-15SA to transplant recipients significantly increased the number …

Scott L. Gardner Publications
An extensive survey of small mammals and ectoparasites along an altitudinal transect in the Manu Biodiversity Reserve in Peru found the sigmodontine rodent genus Nephelomys infested by mites of the genus Gigantolaelaps Fonseca, 1939. Two distinct species co-occurred exclusively in the pelage of Nephelomys keaysi, G. inca Fonseca and G. minima n. sp. Nephelomys levipes, which replaces N. keaysi at higher elevations, was infested exclusively with a single new species, G. nebulosa n. sp. In this paper, we formally describe these new mite species, and provide more information on the morphology of G. inca.

Phylogeography And Population Dynamics Of North American Wolverines (Gulo Gulo Luscus) In Alaska And Western Canada, Dianna M. Krejsa
Biology ETDs
The circumboreal wolverine (Gulo gulo sp.) is ideal for studying responses to environmental perturbation in the North due to a history of persistence in glacial refugia and subsequent glacial recolonization. Wolverines are also excellent indicators of human influence on the environment due to their close association with remote areas and cold, snowy climes. Through the use of genetic tools (i.e., nuclear microsatellite loci and mitochondrial sequences), I examined population structure of wolverines in Alaska and western Canada to identify signatures of glacial refugia, bottlenecks, and distinctive populations, sex-biased dispersal, gene flow, and source and sink population dynamics. I identified genetic …

The olivary pretectal nucleus (OPT) is a midbrain structure that receives reciprocal bilateral retinal projections, is involved in the pupillary light reflex, and connects reciprocally with the intergeniculate leaflet (IGL), a retinorecipient brain region that mediates behavioral responses to light pulses (i.e., masking) in diurnal Nile grass rats. Here, we lesioned the OPT and evaluated behavioral responses in grass rats to various lighting conditions, as well as their anxiety-like responses to light exposure. While control grass rats remained diurnal, grass rats with OPT lesions exhibited a more night-active pattern under 12h:12h light-dark (LD) conditions. However, when placed in constant darkness, …

BACKGROUND:
The Family Smoking Prevention and Tobacco Control Act gave the Food and Drug Administration jurisdiction over the regulation of all tobacco products, including their nicotine content. Under this act, a major strategy to reduce harm from cigarette tobacco is lowering the nicotine content without causing unintended adverse consequences. Initial research on reduced nicotine content (RNC) cigarettes has shown that smokers of these cigarettes gradually decrease their smoking frequency and biomarkers of exposure. The effectiveness of this strategy needs to be demonstrated in different populations whose response to RNC cigarettes might be substantially mediated by personal or environmental factors, such …

Growth phenotypes of microorganisms are a strong indicator of their underlying genetic fitness and can be segregated into 3 growth regimes: lag-phase, log-phase, and stationary-phase. Each growth phase can reveal different aspects of fitness that are related to various environmental and genetic conditions. High-resolution and quantitative measurements of all 3 phases of growth are generally difficult to obtain. Here we present a detailed method to characterize all 3 growth phases on solid media using an assay called One-cell Doubling Evaluation of Living Arrays of Yeast (ODELAY). ODELAY quantifies growth phenotypes of individual cells growing into colonies on solid media using …

The Role Of Lkb1 In Obesity-Induced Skeletal Muscle Inflammation And Insulin Resistance, David Thomson
Journal of Undergraduate Research
The incidence of obesity and the health consequences that accompany it have increased rapidly over the past decade. Understanding the molecular pathways involved in its development will be crucial in developing novel treatment strategies for obesity and associated disorders. Low-grade inflammation likely plays a major role in many of the complications of obesity. We proposed to study the role of liver kinase B1 (LKB1) in the development of inflammation and insulin resistance in obese mice and saturated fatty acid-induced insulin-resistant cells.

On Axon-Axon Interaction Via Currents And Fields, Aman Chawla
USF Tampa Graduate Theses and Dissertations
In this dissertation, we investigate coupling between axons in a tract, when the tract has an arbitrary cross-section, with the coupling being mediated by currents as well as electric fields. Under the current mediated setting, we develop a new master equation which captures the relative axonal geometry, specifically, the axon inclinations θi and the inter-axon distances Wip and perform a number of simulations. We observe synchronization in our simulations of axons with differing diameters and separation-dependent coupling delays in the case of non-trivial tract geometries. For the field-mediated interaction setting, we determine the electric near-field of a firing …

© FEMS 2017. All rights reserved. Under alkaline pH conditions, Escherichia coli must maintain a stable cytoplasmic pH of about 7.6 that is acidic relative to the environment. Bacteria employ various mechanisms to survive alkaline pH; however, membrane cation/H+ antiporters play a primary role by facilitating inward transport of protons. Escherichia coli YqjA belongs to the DedA/Tvp38 membrane protein family and, along with its paralog YghB, is required for growth at 42°C, proper cell division and antibiotic resistance. YqjA is required for viability at alkaline pH, requiring cations sodium or potassium to support growth under these conditions, suggesting it may …

Regional species diversity generally increases with primary productivity whereas local diversity–productivity relationships are highly variable. This scale-dependence of the biodiversity–productivity relationship highlights the importance of understanding the mechanisms that govern variation in species composition among local communities, which is known as β-diversity. Hypotheses to explain changes in β-diversity with productivity invoke multiple mechanisms operating at local and regional scales, but the relative importance of these mechanisms is unknown. Here we show that changes in the strength of local density-dependent interactions within and among tree species explain changes in β-diversity across a subcontinental-productivity gradient. Stronger conspecific relative to …

Coulombic forces are vital in modulating the electron transfer dynamics in both synthetic and biological polychromophoric assemblies, yet quantitative studies of the impact of such forces are rare, as it is difficult to disentangle electrostatic forces from simple electronic coupling. To address this problem, the impact of Coulombic interactions in the successive removal of two electrons from a model set of spirobifluorenes, where the interchromophoric electronic coupling is nonexistent, is quantitatively assessed. By systematically varying the separation of the bifluorene moieties using model compounds, ion pairing, and solvation, these interactions, with energies up to about 0.4 V, are absent at …

Crisis Leadership And Decision-Making: Hospital Administration And Nurse Leaders' Concerns For Disaster Response, Tener Goodwin Veenema, Katherine Deruggiero, Sarah Lynn Losinski, Daniel Barnett
International Nursing Research Congress (INRC)
Purpose: The purpose of this study was to characterize and explore the essential elements of effective hospital and nursing leadership during disasters and MCIs and to identify those events of greatest concern for response. Disaster and mass casualty incidents (MCIs) impose a huge burden on health care leaders. These events create a sudden, unanticipated demand for care referred to as ‘patient surge" that has the potential to quickly overwhelm a hospital"s capacity to function properly. The demand for health care services exceeds capacity to respond and the institution"s normally acceptable patterns of operations and logistics are disrupted. Strong crisis leadership …

Lack Of Evidence For Involvement Of Tonebp And Hyperosmotic Stimulus In Induction Of Autophagy In The Nucleus Pulposus., Chao Liu, Hyowon Choi, Zariel I. Johnson, Jiwei Tian, Irving Shapiro, Makarand V Risbud
Department of Orthopaedic Surgery Faculty Papers
Nucleus pulposus (NP) cells reside in a physiologically hyperosmotic environment within the intervertebral disc. TonEBP/NFAT5 is an osmo-sensitive transcription factor that controls expression of genes critical for cell survival under hyperosmotic conditions. A recent report on NP and studies of other cell types have shown that hyperosmolarity triggers autophagy. However, little is known whether such autophagy induction occurs through TonEBP. The goal of this study was to investigate the role of TonEBP in hyperosmolarity-dependent autophagy in NP. Loss-of-function studies showed that autophagy in NP cells was not TonEBP-dependent; hyperosmolarity did not upregulate autophagy as previously reported. NP tissue of haploinsufficient …

Metastasis accounts for a majority of cancer death. One key feature during metastasis is epithelial-mesenchymal transition (EMT), which is regulated by transcription factors such as Snail and Twist. In non-malignant cells, Snail has a short half-life and is degraded via ubiquitination, but its stability is increased in cancer cell. However, the mechanism by which Snail escapes ubiquitination and degradation remains unknown. Recently, we found that Dub3 is a deubiquinase of Snail. Most importantly, we determined that Dub3 responded to extracellular signals such as IL-6, and that the resultant signaling prevented Snail degradation, and promoted cancer growth, invasion, and migration. In …

Understanding The Experience Of Early-Onset Bipolar Disorder: A Phenomenological Study Of Emerging Adults, Kristin M. Smyth
USF Tampa Graduate Theses and Dissertations
While early-onset bipolar disorder (EOBD) has increased in prevalence, much remains to be understood about its phenomenology. Research and treatment models remain rooted in neurobiological conceptualizations of the illness that borrow heavily from models for the traditional adult-onset form of bipolar disorder. This study utilized a transcendental phenomenological design as a first step in obtaining an understanding of the lived experience of EOBD. A purposive sample of eight participants ages 18-25 participated in semi-structured interviews that elicited information on experiences of EOBD symptomatology and course of illness, stigma, experience with healthcare and treatment, and impact on identity, interpersonal relationships, and …
