The Role Of H19, A Long Non-Coding Rna, In Liver Development, Chad Pope Jul 2017

The Role Of H19, A Long Non-Coding Rna, In Liver Development, Chad Pope

Doctoral Dissertations

Liver development at the postnatal age has been understudied despite being a time when the liver is rapidly growing and changing its primary function as an organ involved in hematopoiesis to an organ responsible for metabolism. Long non-coding RNAs (LncRNAs) are important in many biological processes, including organogenesis, normal liver functions, and liver diseases. Expression levels of lncRNAs change in identifiable patterns in liver during development, and H19, due to its expression pattern, may be important for liver development.

H19 RNA is highly expressed at early postnatal ages and precipitously decreases at a specific time corresponding with increases in expression …

Using Mobile Geographic Information Systems To Improve Operational Efficiency, Data Reliability, And Access In Mine Action, Paul Rittenhouse, Lindsay Aldrich Jul 2017

Using Mobile Geographic Information Systems To Improve Operational Efficiency, Data Reliability, And Access In Mine Action, Paul Rittenhouse, Lindsay Aldrich

The Journal of Conventional Weapons Destruction

The inherently complex field of mine action, with its many political, financial, and physical considerations, is also a spatial, data-driven field; and as a result, geographic information systems (GIS) stand to play a major role. Spatial data can help address questions such as: Where are the hazardous areas and what has been cleared or cancelled? Where have teams already surveyed? Where should they go next? How many square meters have been cleared? Due to the complexities surrounding assigning tasks and prioritization, standard operating procedures (SOP), quality assurance/quality control (QA/QC) and database design, GIS often gets limited to high-level planning, database …

Drug Repurposing Using The Anthrax Toxin Internalization Pathway, Leeor Zilbermintz Jul 2017

Drug Repurposing Using The Anthrax Toxin Internalization Pathway, Leeor Zilbermintz

KGI Theses and Dissertations

Pathogens and pathogenic agents such as lethal toxin, exploit host proteins for their success. Antibiotic discovery efforts have been focused on developing drugs that selectively target pathogens, despite the potential for drug resistance and providing no guarantee of fewer side effects for the patient. This dissertation describes the systematic efforts to identify drugs that alter those host proteins that are involved in the anthrax toxin internalization pathway. Anthrax is a rare and lethal disease caused by the toxin producing, gram-positive bacterium, Bacillus anthracis. The lethality of anthrax is largely attributed to the toxins that circulate within the host, destroying host …

Multisequence Algorithm For Coarse-Grained Biomolecular Simulations: Exploring The Sequence-Structure Relationship Of Proteins, Adekunle Aina, Stefan Wallin Jul 2017

Multisequence Algorithm For Coarse-Grained Biomolecular Simulations: Exploring The Sequence-Structure Relationship Of Proteins, Adekunle Aina, Stefan Wallin

Association in Solution IV

Many biologically motivated problems naturally call for the investigation and comparison of molecular variants, such as determining the mechanisms of specificity in biomolecular interactions or the mechanisms of molecular evolution. We consider a generalized ensemble algorithm for coarse-grained simulations of biomolecules which allows the thermodynamic behavior of two or more sequences to be determined in a single multisequence run. By carrying out a random walk in sequence space, the method also enhances conformational sampling. Escape from local energy minima is accelerated by visiting sequences for which the minima are shallower or absent. We test the method on an intermediate-resolution coarse-grained …

Dna Analysis Of Surfactant-Associated Bacteria In A Natural Sea Slick In The Gulf Of Mexico Observed By Terrasar-X, Kathryn Howe

HCNSO Student Theses and Dissertations

Under low wind speed conditions, surfactants accumulate at the air-sea interface, dampen short-gravity capillary (Bragg) waves, and form natural sea slicks that are detectable visually and in synthetic aperture radar (SAR) imagery. Marine organisms, such as phytoplankton, zooplankton, seaweed, and bacteria, produce and degrade surfactants during various life processes. This study coordinates in situ sampling with TerraSAR-X satellite overpasses in order to help guide microbiological analysis of the sea surface microlayer (SML) and associated subsurface water (SSW). Samples were collected in the Gulf of Mexico during a research cruise (LASER) in February 2016 to determine abundance of surfactant associated bacteria …

Hydrogen Sulfide Regulation Of Mesenteric Vascular Resistance, Adelaeda Barrera, Laura Gonzalez Bosc, Jay Naik, Perenkita Mendiola, Nancy L. Kanagy Jul 2017

Hydrogen Sulfide Regulation Of Mesenteric Vascular Resistance, Adelaeda Barrera, Laura Gonzalez Bosc, Jay Naik, Perenkita Mendiola, Nancy L. Kanagy

Biomedical Sciences ETDs

Sleep apnea, recurrent obstruction of the upper airway leads to sleep fragmentation and intermittent hypoxia (IH). In animal studies, IH causes wake-time hypertension by elevating vascular resistance due in part to decreased production of the vasodilator, hydrogen sulfide (H2S). However, much is unknown about where and how H2S acts. Inhibiting cystathionine γ-lyase (CSE) to prevent H2S synthesis augments constriction in isolated mesenteric arteries, but H2S regulation of resistance in this vascular bed in vivo is unknown. The goal of this study was to evaluate CSE regulation of blood flow and resistance in …

Articles

Spinach (Spinacia oleracea L.) leaves represent an important dietary source of nutrients, antioxidants, and antimicrobials. As such, spinach leaves play an important role in health and have been used in the treatment of human diseases since ancient times. Here the aims were to optimize the extraction methods for recovering antimicrobial substances of spinach leaves, determine the minimum inhibitory concentrations (MICs) of the antimicrobial substances against Escherichia coli and Staphylococcus aureus and finally, evaluate the effects of spinach leaves’ antimicrobials on bacterial DNA using central composite face centered methods (CCFC). The effect of the extracts on both Gram positive and …

The Role Of Co-Opted Escrt Proteins And Lipid Factors In Protection Of Tombusviral Double-Stranded Rna Replication Intermediate Against Reconstituted Rnai In Yeast, Nikolay Kovalev, Jun-Ichi Inaba, Zhenghe Li, Peter D. Nagy

Plant Pathology Faculty Publications

Reconstituted antiviral defense pathway in surrogate host yeast is used as an intracellular probe to further our understanding of virus-host interactions and the role of co-opted host factors in formation of membrane-bound viral replicase complexes in protection of the viral RNA against ribonucleases. The inhibitory effect of the RNA interference (RNAi) machinery of S. castellii, which only consists of the two-component DCR1 and AGO1 genes, was measured against tomato bushy stunt virus (TBSV) in wild type and mutant yeasts. We show that deletion of the co-opted ESCRT-I (endosomal sorting complexes required for transport I) or ESCRT-III factors makes …

This article contributes to the study of Hispanic-Serving Institutions (HSIs) through a narrative grounded on two points of innovation. It offers frameworks to decenter the conversation on HSIs from normative practices in higher education to focus on pedagogical, cultural, and political relational processes that find greater congruence between nominal HSIs and the Latina/o students, families, and the communities that populate those universities. It looks at points of innovation that emerged in two different parts of the country at different places, spaces, and time. One was initiated at the University of North Florida (UNF) in the early-to-mid-1970s, and the second is …

A Delphi Study: Guidelines For Researchers Working With Teenage Mothers, Alison M. Chamberlain Jul 2017

A Delphi Study: Guidelines For Researchers Working With Teenage Mothers, Alison M. Chamberlain

Nursing Theses and Dissertations

Problem: It is difficult to get teenage mothers to participate in studies, yet some researchers have been successful. Researchers who understand the barriers to conducting studies with a vulnerable population and are guided in ways to overcome these barriers, will experience greater success when doing studies with this group. Improved study methods can lead to larger numbers of teenage mothers who participate in studies. Researchers who follow developmentally appropriate guidelines are more likely to conduct and replicate studies with this population. Access is essential to advance the science related to care for teenage mothers who are at risk for many …

Novel Design Strategies For Platinum-Containing Conjugated Polymers And Small Molecules For Organic Solar Cells, Wenhan He Jul 2017

Novel Design Strategies For Platinum-Containing Conjugated Polymers And Small Molecules For Organic Solar Cells, Wenhan He

Chemistry and Chemical Biology ETDs

Current state-of-the-art organic solar cells (OSCs) adopt the strategy of using conjugated polymers or small molecules as donors and fullerene derivatives as acceptors in their active layers. Regarding to the donors of interest, the conjugated polymers and small molecules coupled with heavy metals have been less explored compared to their counterparts. Among various transition metal complexes applied, Pt(II) complexes are unique because of their intrinsic square planar geometries and ability to serve as building blocks for conjugated systems. Furthermore, the heavy metal Pt facilitates the formation of triplet excitons with longer life times through spin-orbital coupling which are of benefit …

Modern metagenomic environmental DNA studies are almost completely reliant on next-generation sequencing, making evaluations of these methods critical. We compare two next-generation sequencing techniques – amplicon and shotgun – on water samples across four of Brazil’s major river floodplain systems (Amazon, Araguaia, Paraná, and Pantanal). Less than 50% of phyla identified via amplicon sequencing were recovered from shotgun sequencing, clearly challenging the dogma that mid-depth shotgun recovers more diversity than amplicon-based approaches. Amplicon sequencing also revealed ~27% more families. Overall the amplicon data were more robust across both biodiversity and community ecology analyses at different taxonomic scales. Our work doubles …

Neural Coding And Models For Natural Sounds Recognition: Effects Of Temporal And Spectral Features, Seyedeh Fatemeh Khatami Firoozabadi Jul 2017

Neural Coding And Models For Natural Sounds Recognition: Effects Of Temporal And Spectral Features, Seyedeh Fatemeh Khatami Firoozabadi

Doctoral Dissertations

The mammalian brain is able to recognize natural sounds in the presence of acoustic uncertainties such as background noise. A prevailing theory of neural coding suggest that neural systems are optimized for natural environment signals and sensory inputs that are biologically relevant. The optimal coding hypothesis thus suggests that neural populations should encode sensory information so as to maximize efficient utilization of environmental inputs. In the first part of my thesis, I will explore the origins of scale invariance phenomena which has been previously described for natural sounds and has been observed in a variety of natural sensory signals including …

Behavioral And Neuroanatomical Assessment Of Transgenic Mouse Models Of Language-Related Developmental Disorders, Amanda R. Rendall Jul 2017

Behavioral And Neuroanatomical Assessment Of Transgenic Mouse Models Of Language-Related Developmental Disorders, Amanda R. Rendall

Doctoral Dissertations

Language encompasses an ability to acquire and integrate complex cognitive systems in order to communicate with others, and inherited factors are thought to play a key role in modulating these emergent skills. In recent years, the role of genetics in language has gained focus and attention, based on accumulating empirical knowledge about the genes, proteins, and cellular machinery involved. In particular, disruption to various mechanisms has been shown to relate to impairments in language -- as seen in neurodevelopmental disorders such as autism, dyslexia and SLI. Mouse models can serve as a useful tool in studying the genetic modulators of …

Advanced Science Research Center

Differentiation of stem cells to chondrocytes in vitro usually results in a heterogeneous phenotype. This is evident in the often detected over expression of type X collagen which, in hyaline cartilage structure is not characteristic of the mid-zone but of the deep-zone ossifying tissue. Methods to better match cartilage developed in vitro to characteristic in vivo features are therefore highly desirable in regenerative medicine. This study compares phenotype characteristics between pericytes, obtained from human adipose tissue, differentiated using diphenylalanine/serine (F2/S) peptide hydrogels with the more widely used chemical induced method for chondrogenesis. Significantly higher levels of type II collagen were …
