The Soybean Rfg1 Gene Restricts Nodulation By Sinorhizobium Fredii Usda193, Yinglun Fan, Jinge Liu, Shanhua Lyu, Qi Wang, Shengming Yang, Hongyan Zhu

Plant and Soil Sciences Faculty Publications

Sinorhizobium fredii is a fast-growing rhizobial species that can establish a nitrogen-fixing symbiosis with a wide range of legume species including soybeans (Glycine max). In soybeans, this interaction shows a high level of specificity such that particular S. fredii strains nodulate only a limited set of plant genotypes. Here we report the identification of a dominant gene in soybeans that restricts nodulation with S. fredii USDA193. Genetic mapping in an F2 population revealed co-segregation of the underlying locus with the previously cloned Rfg1 gene. The Rfg1 allele encodes a member of the Toll-interleukin receptor/nucleotide-binding site/leucine-rich repeat class of …

Genetically Engineering Of Synechocystis Pcc 6803 For Sustainable Production Of Cinnamic Acid, Shahrah Saad Alqahtani

Theses and Dissertations

Cinnamic acid is the precursor for phenylpropanoids, which play important roles in plant growth, development, and stress responses. It is highly beneficial to human health for its anticancer, antitoxic and anti-inflammatory functions. Currently, the major methods of industrial production of cinnamic acid are chemical synthesis and extraction from plants. However, these methods are often associated with low production yields and high costs. Cyanobacterial platforms could be highly suitable for phenylpropanoid production due to potential high yields and eco- friendly, cost- effective production processes. The primary purpose of this study is to explore the possibility of using the cyanobacterium Synechocystis PCC …

SEM study clearly revealed its 3-dimentional pore structure, while …

Cgg Repeat Rnas Regulate Granule Translation, Rene Rovozzo Sep 2017

Cgg Repeat Rnas Regulate Granule Translation, Rene Rovozzo

Doctoral Dissertations

Fragile X tremor ataxia syndrome (FXTAS) and fragile X primary ovarian insufficiency (FXPOI) are associated with CGG repeat expansions in the 5’UTR of the FMR1 gene. RNAs containing an hnRNP A2 response element (A2RE) are localized and transported in hnRNP A2 granules that are organized by multivalent interactions. We show that RNAs containing CGG repeats are also localized in hnRNP A2 granules. Exogenous expanded CGG repeat RNA microinjected in mouse hippocampal neurons or oocytes inhibits granule RNA translation. Endogenous premutation CGG repeats in FMR1 in human fibroblast cells from females at risk for FXPOI also inhibit granule RNA translation. We …

The Importance Of Saperda Inornata And Oberea Schaumii (Coleoptera: Cerambycidae) Galleries As Infection Courts Of Hypoxylon Pruinatum In Trembling Aspen, Populus Tremuloides, John C. Nord, Fred B. Knight

The Great Lakes Entomologist

(excerpt) Trembling aspen, Populus tremuloides Michaux, and bigtooth aspen, P. grandidentata Michaux, are hosts of numerous species of injurious insects and microorganisms (Harrison 1959). Only a few of those organisms, however, are directly responsible for mortality of healthy trees. The fungus Hypoxylon pruinatum (Klotzsche) Cke. is most important in that respect, killing 1-2%o f the standing volume annually in the Lake States (Anderson 1964). It invades and spreads in cambial tissue, killing it and eventually the branch or stem by girdling. Initially, a canker appears as a sunken, yellowish-orange area in the bark (Anderson 1956). In a later stage the …

Neuroprotective Mechanisms Of Ischemic Tolerance Induced By Resveratrol Preconditioning, Kevin B. Koronowski Sep 2017

Neuroprotective Mechanisms Of Ischemic Tolerance Induced By Resveratrol Preconditioning, Kevin B. Koronowski

Open Access Dissertations

Ischemic stroke is a devastating neurological disorder that currently has no clinically approved neuroprotective treatments. Resveratrol is a naturally occurring polyphenol that protects against cerebral ischemic injuries when administered in a prophylactic manner. Yet, the molecular underpinnings of resveratrol-induced neuroprotection are still unclear. This is partly because the metabolic adaptations induced by a major target of resveratrol, the NAD+-dependent deacetylase Sirt1, have not been characterized in the brain. The goal of this study was to better understand the efficacy of resveratrol as a therapeutic agent in ischemic stroke and the metabolic pathways regulated by brain Sirt1 that promote ischemic tolerance. …

Fibroblast Growth Factor 21 Induces Cardiac Hypertrophy, Karla Schramm Sep 2017

Fibroblast Growth Factor 21 Induces Cardiac Hypertrophy, Karla Schramm

Open Access Dissertations

Diabetic cardiomyopathy is a diabetes-associated cardiovascular disease (CVD), which causes heart dysfunction. Left ventricular hypertrophy (LVH) is a key feature of diabetic cardiomyopathy. Molecular mechanisms underlying this condition are largely unknown. Fibroblast growth factor (FGF) 21, an adipokine that regulates metabolism, has been found to be elevated in the circulation of type 2 diabetic patients. This thesis evaluates the long-term effects of increased serum FGF21 levels on the heart. It reveals that FGF21 induces hypertrophy of cardiac myocytes via FGF receptor (FGFR) 4-dependent activation of the pathological and pro-hypertrophic PLCγ/calcineurin/NFAT pathway, but independently of β-klotho, the canonical co-receptor for FGF21 …

Endophytic actinomycetes from medicinal plants produce a wide diversity of secondary metabolites (SM). However, to date, the knowledge about endophytes from Brazil remains scarce. Thus, we analyzed the antimicrobial potential of 10 actinomycetes isolated from the medicinal plant Vochysia divergens located in the Pantanal sul-mato-grossense, an unexplored wetland in Brazil. Strains were classified as belonging to the Aeromicrobium, Actinomadura, Microbacterium, Microbispora, Micrococcus, Sphaerisporangium, Streptomyces, and Williamsia genera, through morphological and 16S rRNA phylogenetic analyzes. A susceptibility analysis demonstrated that the strains were largely resistant to the antibiotics oxacillin and nalidixic acid. Additionally, different culture media (SG and R5A), and …

Climate change is a well-documented driver of both wildlife extinction and disease emergence, but the negative impacts of climate change on parasite diversity are undocumented. We compiled the most comprehensive spatially explicit data set available for parasites, projected range shifts in a changing climate, and estimated extinction rates for eight major parasite clades. On the basis of 53,133 occurrences capturing the geographic ranges of 457 parasite species, conservative model projections suggest that 5 to 10% of these species are committed to extinction by 2070 from climate-driven habitat loss alone. We find no evidence that parasites with zoonotic potential have a …

Transfer RNAs (tRNAs) function in translational machinery and further serves as a source of short non-coding RNAs (ncRNAs). tRNA-derived ncRNAs show differential expression profiles and play roles in many biological processes beyond translation. Molecular mechanisms that shape and regulate their expression profiles are largely unknown. Here, we report the mechanism of biogenesis for tRNA-derived Piwi-interacting RNAs (td-piRNAs) expressed in Bombyx BmN4 cells. In the cells, two cytoplasmic tRNA species, tRNAAspGUC and tRNAHisGUG, served as major sources for td-piRNAs, which were derived from the 5'-part of the respective tRNAs. cP-RNA-seq identified the two tRNAs as major substrates for the 5'-tRNA halves …

Micro-Contacts Testing Using A Micro-Force Sensor Compatible With Biological Systems, Ronald A. Coutu Jr., Dushyant Tomer Sep 2017

Micro-Contacts Testing Using A Micro-Force Sensor Compatible With Biological Systems, Ronald A. Coutu Jr., Dushyant Tomer

Electrical and Computer Engineering Faculty Research and Publications

This paper presents the performance and reliability testing of microelectromechanical systems (MEMS) switches by using a micro-force sensor which was originally designed/used to conduct mechanical testing of biological cells. MEMS switches are key components for radio frequency (RF) applications due to their extremely low power consumption and small geometries over conventional technologies. However, unstable electrical contact resistance severely degrades the performance and reliability of such micro-switches. Therefore, our focus is to improve the performance and reliability of “cold” switched micro-contacts by using novel contact materials and engineered micro-contact surfaces. The contact metallurgies considered in this work are “similar” thin film …

ABSTRACT The scavenger receptor MARCO is expressed in several subsets of naive tissue-resident macrophages and has been shown to participate in the recognition of various bacterial pathogens. However, the role of MARCO in antiviral defense is largely unexplored. Here, we investigated whether MARCO might be involved in the innate sensing of infection with adenovirus and recombinant adenoviral vectors by macrophages, which elicit vigorous immune responses in vivo. Using cells derived from mice, we show that adenovirus infection is significantly more efficient in MARCO-positive alveolar macrophages (AMs) and in AM-like primary macrophage lines (Max Planck Institute cells) than in MARCO-negative bone …
