Understanding Trait-Turnover Relationships : An Examination Of The Utility Of Holland's Congruence Theory And The Riasec Typology, Sarah Rebecca Strahan Jan 2017

Understanding Trait-Turnover Relationships : An Examination Of The Utility Of Holland's Congruence Theory And The Riasec Typology, Sarah Rebecca Strahan

Legacy Theses & Dissertations (2009 - 2024)

The goal of the current study is to improve our understanding of the relationship between personality and employee turnover. Research is reviewed to demonstrate that after decades of research we still know very little about the role of personality in turnover. As a result, many researchers have called for new approaches to improve our understanding of the relationship between personality and turnover (Griffeth, Hom, & Gaertner, 2000; Hom & Griffeth, 1995; Russell, 2013). One approach that has received little attention is examining whether job context matters –that is, do the traits that predict employee turnover depend on the particular job. …

Analysis Of Candidate Genes For Seizure Susceptibility In Epileptogenesis, Brittany O'Brien Jan 2017

Analysis Of Candidate Genes For Seizure Susceptibility In Epileptogenesis, Brittany O'Brien

Legacy Theses & Dissertations (2009 - 2024)

The occurrence of recurrent spontaneous seizures is the hallmark of epilepsy. The key to understanding how a normal brain can develop epilepsy through a process known as epileptogenesis is identifying susceptibility factors. Previously, various strains of inbred mice from the hybrid mouse diversity panel were subjected to multiple seizures via the repeated flurothyl model (RFM) for epileptogenesis. The results demonstrated differences in kindling among the inbred strains; specifically two main trends in the generalized seizure threshold (GST) data were kindling-sensitive and kindling-resistant strains. In addition, genome wide association studies done from these mice demonstrated a quantitative trait locus (QTL) on …

Child Abuse In The Archaeological Context, Nicole Marie Popielarz Jan 2017

Child Abuse In The Archaeological Context, Nicole Marie Popielarz

Legacy Theses & Dissertations (2009 - 2024)

Children have been a widely ignored subject in archaeology so it follows that child abuse has not been studied in detail in the archaeological context. This paper combines both forensic anthropology studies and Bioarchaeological studies to see if child abuse is a modern phenomenon. Signs of both non-accidental trauma and neglect have been documented by authors in the archaeological record. The purpose of this paper will be to describe signs of both nonaccidental trauma and neglect. Then examples in archaeological studies will be reviewed. The future of the study of abuse in the archaeological context is also discussed at some …

The Role Of N-Myc Downstream Regulated Gene 1 In Breast Cancer Lipid Metabolism, Christopher James Sevinsky Jan 2017

The Role Of N-Myc Downstream Regulated Gene 1 In Breast Cancer Lipid Metabolism, Christopher James Sevinsky

Legacy Theses & Dissertations (2009 - 2024)

Altered carbohydrate and lipid metabolism are increasingly well characterized hallmarks of aggressive breast cancers. While aerobic glycolysis, or “the Warburg effect”, is a well-established metabolic adaptation exploited by tumor cells, the understanding of unique aspects of cancer lipid metabolism lags behind. This is especially true regarding the coordination of complex lipid synthesis and trafficking pathways, which remains poorly understood. N-Myc Downstream Regulated Gene1 (NDRG1) is overexpressed in many solid tumors, but its function is unclear. The importance of NDRG1 is best exemplified by the effect of null mutations on human physiology: inactivating mutations give rise to the severe autosomal recessive …

Female Athlete Body Project Intervention With Professional Ballet Dancers, Sasha Gorrell Jan 2017

Female Athlete Body Project Intervention With Professional Ballet Dancers, Sasha Gorrell

Legacy Theses & Dissertations (2009 - 2024)

Professional dancers have increased vulnerability for eating disorders (EDs), with three times higher risk than non-dancers. Among ballet dancers, generalized risk for EDs associated with internalization of female beauty ideals (i.e., the thin-ideal) in western culture is compounded by idealization of a ballet-specific body ideal, a combination that confers unique vulnerability for eating pathology. Empirical support has been established for a healthy weight intervention (HWI) that promotes body acceptance and reducing eating pathology; the current study adapted a HWI specifically tailored to female athletes for implementation among professional ballet dancers. Participants from two elite ballet companies were randomized to a …

Impedance Biosensors And Deep Crater Salivary Gland Scaffolds For Tissue Engineering, Robert Arthur Schramm Jan 2017

Impedance Biosensors And Deep Crater Salivary Gland Scaffolds For Tissue Engineering, Robert Arthur Schramm

Legacy Theses & Dissertations (2009 - 2024)

The salivary gland is a complex, branching organ whose primary biological function is the production of the fluid critical to alimentary function and the lubrication and maintenance of the oral cavity, saliva. The most frequent disruption of the salivary organ system is one in which the rate of supply of saliva into the oral cavity is diminished, and this may vary from a minor reduction, to near cessation. Regenerative medicine is a field which seeks to find ways to overcome the symptoms of organ malfunction or damage by inducing regrowth, repair and replacement of partial or whole organ function. Historically, …

Mass Spectrometric Analysis And Machine Learning Enable Microorganism Classification Based On Rna Posttranscriptional Modifications, Colin Christopher Aldrich

Legacy Theses & Dissertations (2009 - 2024)

RNA post-transcriptional modifications (PTMs) are dynamic features that can be up- or down-regulated by the health and metabolic state of a cell. These covalent modifications are installed and removed on RNA nucleosides by enzymes controlled by the activation and deactivation of specific genes. The goal of this research was to demonstrate that RNA PTMs can serve as a unique feature for the classification/identification of microorganisms. We utilized a scheme based on electrospray ionization mass spectrometry (ESI-MS) to obtain global PTM profiles from total RNA extracted from various microorganisms in optimal growth conditions as well as Salmonella typhimurium (S. typhimurium) spiked …

Biomarkers And Metabolic Mechanisms Of Tobacco Smoke-Induced Lung Disease, Matthew Alan Hartog Jan 2017

Biomarkers And Metabolic Mechanisms Of Tobacco Smoke-Induced Lung Disease, Matthew Alan Hartog

Legacy Theses & Dissertations (2009 - 2024)

The overall goal of the research conducted in this dissertation is to determine the role of cytochrome (CYP) P450-mediated xenobiotic bioactivation in the development of tobacco smoke-induced lung toxicity. The central hypothesis of this work is that CYP-mediated bioactivation of specific tobacco smoke constituents is an important contributor to the generation of tobacco smoke-induced lung inflammation and injury. The specific aims are 1) to demonstrate that CYP-mediated metabolism contributes to the onset of lung inflammation following tobacco smoke inhalation; 2) to demonstrate that specific tobacco smoke constituents undergo CYP-mediated bioactivation in vivo; 3) to demonstrate that CYP-mediated bioactivation of tobacco …

Factors Associated With Successful Completion Of A Transitional Living Program For Homeless And Former Foster Youth, Megan Brophy-Mclean Jan 2017

Factors Associated With Successful Completion Of A Transitional Living Program For Homeless And Former Foster Youth, Megan Brophy-Mclean

Legacy Theses & Dissertations (2009 - 2024)

Transitional and Independent Living programs exist to help prepare homeless, at-risk and foster youth to live independently in the community as adults at an age earlier than expected from more privileged youth. The challenges they face often result in poor outcomes such as ongoing homelessness, substance abuse problems, low levels of education, and high levels of unemployment. Because the stakes are so high for these youth, it is imperative that the programs designed to help them are able to meet their needs by having them remain in the program until they are competent to live independently. This study attempts to …

Mediating And Moderating Factors Of The Relationship Between Sexual Orientation And Eating Pathology And Body Satisfaction In Sexual Minority And Heterosexual Men, Andrea Denae Murray Jan 2017

Mediating And Moderating Factors Of The Relationship Between Sexual Orientation And Eating Pathology And Body Satisfaction In Sexual Minority And Heterosexual Men, Andrea Denae Murray

Legacy Theses & Dissertations (2009 - 2024)

Most research on body image and disordered eating has focused largely on women, as women are at higher risk than men for eating disorders. In recent years research has revealed that men are at increasing risk for these outcomes, especially as the ideal male body represented in media images and therefore frequently internalized among men is becoming so lean and muscular as to make it very difficult for most men to realistically achieve. Sexual minority men in particular have been found to be at increased risk for body dissatisfaction, body shame, and disordered eating than their heterosexual counterparts. The research …

Homo- And Heterometallic Bis(Pentafluorobenzoyl)Methanide Complexes Of Copper(Ii) And Cobalt(Ii), Janell Crowder Jan 2017

Homo- And Heterometallic Bis(Pentafluorobenzoyl)Methanide Complexes Of Copper(Ii) And Cobalt(Ii), Janell Crowder

Legacy Theses & Dissertations (2009 - 2024)

β-Diketones are well known to form metal complexes with practically every known metal and metalloid. Metal complexes of fluorinated β-diketones generally exhibit increased volatility and thermal stability compared to the non-fluorinated analogues, and thus are used extensively in various chemical vapor deposition (CVD) processes for the deposition of metal, simple or mixed metal oxides, and fluorine-doped metal oxide thin films. Furthermore, the electron-withdrawing nature of the fluorinated ligand enhances the Lewis acidity of a coordinatively unsaturated metal center which facilitates additional coordination reactions. The physical and structural properties of fluorinated β-diketonate complexes are discussed in Chapter 1 …

A Kinetic Study Of Regulation Of Glua1 Homomeric And Heteromeric Ampa Receptors By 2,3-Benzodiazepines, Yin-Shuo (Andrew) Wu

Legacy Theses & Dissertations (2009 - 2024)

AMPA receptors are a subtype of the ionotropic glutamate receptor family. They are ligand-gated or glutamate-activated, transmembrane ion channels that mediate the majority of excitatory synaptic transmission in the central nervous system. AMPA receptors are essential for neuronal development and brain functions, including learning and memory. Over-activation of calcium permeable AMPA receptors has been linked to some neurological diseases such as stroke, Alzheimer’s disease and amyotrophic lateral sclerosis. Inhibitors of glutamate receptors are therefore drug candidates for the potential treatment of these neurological diseases. One of the most promising classes of AMPA receptor antagonists is 2,3-benzodiazepine derivatives, thought to be …

Evolution Of Lemur Pelage Color Variation, Amanda Nicole Spriggs Jan 2017

Evolution Of Lemur Pelage Color Variation, Amanda Nicole Spriggs

Legacy Theses & Dissertations (2009 - 2024)

This dissertation investigates the selective pressures that may be driving variation in pelage color patterns in Eulemur, Varecia, and Propithecus. This is addressed by exploring the relationship between the lemur pelage colors and ecological variables, such as habitat type, predation pressures, visibility variables, and level of sympatry. The following model is presented here to describe the evolutionary mechanisms that are thought to be maintaining pelage coloration in lemurs: Pelage color and pattern are potentially used to (1) signal to conspecifics information such as mate quality, (2) signal to congenerics species identity, and (3) avoid signaling the individual’s presence to predators. …

Detecting And Analyzing Trna Modification Systems And Homologs Using In Silico And Colorectal Cancer Models, Khadijah Onanuga

Legacy Theses & Dissertations (2009 - 2024)

tRNA modifications can be considered epitranscriptomic signaling components that regulate translation and play integral roles in stress response pathways. As such, tRNA modification enzymes have roles in cancer etiology and potential utility as biomarkers of pathological states. For my thesis project I have used computational and wet bench approaches to study tRNA modification systems. Chapter two of my thesis deals with tRNA modification detection, as current methods are costly, time consuming, and require RNA fragmentation. I present a single-molecule-based approach for RNA modification detection, which involves in slico studies using a 5-layered graphene nanopore. Our simulations using a 1.5 nm …

Coupling Of Sterically Demanding Peptides By Strain Driven Beta-Thiolactone Mediated Native Chemical Ligation, Yunxian Xiao Jan 2017

Coupling Of Sterically Demanding Peptides By Strain Driven Beta-Thiolactone Mediated Native Chemical Ligation, Yunxian Xiao

Legacy Theses & Dissertations (2009 - 2024)

The association between sterically requesting peptidyl destinations is hard to accomplish. The ligation between amino acids, for example, Val-Val has been ended up being amazingly testing with ordinary NCL strategies that are depending on exogenous thiol added substances. Besides, the extra of overabundance measures of outer thiol activators, for example, a 4-mercaptophenylacetic corrosive (MPAA) impede the consequent desulfurization, and it needs the additional progression to be expelled. Although various methodology has been produced to encourage MPAA expulsion, reverse phase preparative HPLC purification remains the best. Nevertheless, the additional purification decreases response proficiency and yield. Thus, we report a β-thiolactone-mediated additive-free …

Journal Articles: Pulmonary & Critical Care Med

BACKGROUND: Chronic obstructive pulmonary disease (COPD) is currently the third leading cause of death and there is a huge unmet clinical need to identify disease biomarkers in peripheral blood. Compared to gene level differential expression approaches to identify gene signatures, network analyses provide a biologically intuitive approach which leverages the co-expression patterns in the transcriptome to identify modules of co-expressed genes.

METHODS: A weighted gene co-expression network analysis (WGCNA) was applied to peripheral blood transcriptome from 238 COPD subjects to discover co-expressed gene modules. We then determined the relationship between these modules and forced expiratory volume in 1 s (FEV …

The Impact Of Stress On Steroid Hormones And Cue Reactivity In Smokers And Gamblers, Stephanie Elaine Wemm Jan 2017

The Impact Of Stress On Steroid Hormones And Cue Reactivity In Smokers And Gamblers, Stephanie Elaine Wemm

Legacy Theses & Dissertations (2009 - 2024)

Addictions, both substance and behavioral, have been conceptualized as having similar biopsychosocial processes with different opportunistic expressions (Shaffer et al., 2004). Biological processes such as the hormonal response to stress as measured by cortisol, dehydroepiandrosterone (DHEA), and the ratio of the DHEA/cortisol may be among the variables underlying the disposition to develop an addictive disorder, regardless of whether it is a substance-based or a so-called behavioral addiction. The current study aimed to examine whether physiological and psychological reactions to stress are similar in high-frequency smokers and gamblers. The subjective (urges, cravings) and physiological responses (skin conductance and heart rate) of …

Investigating The Role Of Epitranscriptomics In Viral Immunity, Kaycie S. Lawson Jan 2017

Investigating The Role Of Epitranscriptomics In Viral Immunity, Kaycie S. Lawson

Legacy Theses & Dissertations (2009 - 2024)

The characterization of host-virus relationships is essential for understanding the process of viral replication and driving the development of effective viral targeting strategies. In this work, I tested the hypothesis that ribonucleic acid (RNA) post-transcriptional modifications (PTMs) may play a significant role in defining host-virus interactions. The model system consisted of a yeast strain capable of harboring the endogenous L-A virus, which allowed me to evaluate the possible involvement of PTMs in the process of cellular immunity to virus-borne toxicity. A mass spectrometry-based platform was utilized to compare the global PTM profile of wild-type yeast with those of the same …

Understanding The Genotoxicity Of Silver Nanoparticles And The Chemoprevention Of Pomegranate Extract, Sameera Nallanthighal Jan 2017

Understanding The Genotoxicity Of Silver Nanoparticles And The Chemoprevention Of Pomegranate Extract, Sameera Nallanthighal

Legacy Theses & Dissertations (2009 - 2024)

The use of silver nanoparticles (AgNPs) in a wide variety of consumer products (i.e. toothpastes, food containers, dietary supplements and garments) for their antimicrobial properties can lead to potential oral exposure in humans. To enhance their stability, AgNPs are coated with capping agents such as citrate and polyvinylpyrrolidone (PVP). Despite the lack of significant general toxicity based on hematology, blood chemistry and histology evaluations, the potential genotoxic effects of AgNPs cannot be ruled out and have to be addressed. Studies examining the genotoxic risks of AgNPs are needed because genotoxicity is a strong indicator of cancer risk. Here we examined …

Retinal Disease Modeling And Drug Screening Using Stem Cell Derived Retinal Pigment Epithelium, Janmeet Singh Saini Jan 2017

Retinal Disease Modeling And Drug Screening Using Stem Cell Derived Retinal Pigment Epithelium, Janmeet Singh Saini

Legacy Theses & Dissertations (2009 - 2024)

The eye is an intricate organ, possessing sensory receptors in the retina that are involved in visual perception. The retina consists primarily of the neural layer, referred to as the neural retina, and the pigmented layer, referred to as the retinal pigment epithelium (RPE). The neural retina and RPE develop and mature in a coordinated manner, and depend on each other for survival. In a mature healthy retina, the retinal photoreceptor outer segments interdigitate with the RPE monolayer, and they co-exchange key supportive factors. RPE cells are indispensable for the health of the retina as they are involved in a …

The Right Approach To Success : Approach And Avoidance Motivation In Anticipatory Self-Control, Ellen O'Malley Jan 2017

The Right Approach To Success : Approach And Avoidance Motivation In Anticipatory Self-Control, Ellen O'Malley

Legacy Theses & Dissertations (2009 - 2024)

Self-control success often requires considerable effort in moments of temptation. The current research examines the effectiveness of self-control strategies based on individual differences in self-regulatory motivation. Although self-control appears to come easier to those who are avoidance-motivated rather than approach-motivated, this may be due to the conditions under which self-control is needed and tested. Specifically, the temporal context of an anticipated selfcontrol conflict (i.e., if it is near or distant) may be more instrumental for approach-motivated self-regulation relative to avoidance-motivated self-regulation. Three studies test the temporal and motivational conditions under which participants successfully pursue self-control goals. Results suggest that anticipatory …

Influence Of The Pre-Initiation Complex On Mediator Recruitment In Saccharomyces Cerevisiae, Elisabeth Rose Knoll Jan 2017

Influence Of The Pre-Initiation Complex On Mediator Recruitment In Saccharomyces Cerevisiae, Elisabeth Rose Knoll

Legacy Theses & Dissertations (2009 - 2024)

The Mediator complex plays a central, highly conserved role in eukaryotic transcription by RNA Polymerase II (Pol II) by stimulating the cooperative assembly of a pre-initiation complex (PIC) and recruitment of Pol II for gene activation. Mediator recruitment has generally been ascribed to sequence-specific activators engaging subunits from the tail module which in turn function to recruit the middle and head for complete assembly at the UAS. Mediator subunits of the middle and head then bridge the enhancer to connect with the PIC at the core promoter. It is reported that Mediator recruitment at the UAS preferentially occurs at SAGA-dependent, …

Examining Variability In Model Skill During The 7 January 2013 Sudden Stratospheric Warming Event, Jonathan Lee Blufer Jan 2017

Examining Variability In Model Skill During The 7 January 2013 Sudden Stratospheric Warming Event, Jonathan Lee Blufer

Legacy Theses & Dissertations (2009 - 2024)

Recent analyses of numerical weather prediction models have shown that stratospheric regime changes (e.g. strong and weak vortex events) are not skillfully predicted at medium-range lead times. Motivated by these recent analyses, this thesis investigates the sources of variability in stratospheric forecast skill amongst several operational models initiated at different lead times prior to the 7 January 2013 sudden stratospheric warming (SSW). This study extends upon a previous analysis by the Stratospheric Network for the Assessment of Predictability (SNAP), which concluded that a change in forecast lead-time from 15 to 10 days increased model skill in predicting the 2013 SSW …

The Role Of The Mediator Transcriptional Co-Activator Complex And Promoter Dependence In Ty1 Retrotransposition In Saccharomyces Cerevisiae, Alicia Salinero Jan 2017

The Role Of The Mediator Transcriptional Co-Activator Complex And Promoter Dependence In Ty1 Retrotransposition In Saccharomyces Cerevisiae, Alicia Salinero

Legacy Theses & Dissertations (2009 - 2024)

Retrotransposons are mobile genetic elements that replicate via an RNA intermediary and constitute a significant portion of most eukaryotic genomes. Saccharomyces cerevisiae has been invaluable to retrotransposon research due to the presence of an active retroelement known as Ty1. The mobility of Ty1 is regulated both positively and negatively by numerous host factors, including several subunits of the Mediator transcriptional co-activator complex. The Mediator core complex is organized into genetically and structurally defined head, middle, and tail modules, along with a transiently associated kinase module. We show that with the exception of the kinase module, deletion of non-essential subunits from …
