Traditional x-ray imaging provides only low contrast from low atomic number materials, like soft tissue, due to the small attenuation variations producing very small intensity changes. Higher contrast can be achieved through phase information. The phase change is obtained from the x-ray refracting in a sample, or phase object, due to the difference in refractive indexes. This causes a small angular deviation from the original path. The causative agents of Lyme disease, or Lyme Borreliosis, are spirochetes in the Borrelia burgdorferi sensu lato (s.l.) species complex that are transmitted by ticks in the genus Ixodes. Certain B. burgdorferi s.l. genotypes are more likely to cause invasive, disseminated infection in humans, and the gene, ospC, has been identified as a molecular marker that can be used to predict virulence. ospC is the most polymorphic locus in the B. burgdorferi s.l. genome, and different alleles have been assigned to Major ospC Groups based on sequence dissimilarity. Exposure therapy is effective for addressing social anxiety and public speaking fears, yet there are differences in suggested exposure strategies among empirically supported, manualized treatments. Manualized exposure strategies differ in the relative theoretical emphasis that underpins the specific procedural approach. Some approaches rely more heavily on the principle of habituation and formal cognitive restructuring; alternative approaches emphasize inhibitory learning and the individual's response to his/her own negative emotions, while promoting present-focused awareness. The human response to environmental carcinogens, some of which require metabolic activation, is highly variable. Factors such as environment, lifestyle, and genetics all influence the rates of exposure to and ultimate bioactivation of these compounds. Genetic factors include mutations to cell-cycle regulation, cell proliferation, and DNA repair genes; however, epidemiological studies may lack significance due to inadequate patient numbers. We used budding yeast as a model organism to determine genetic susceptibility to food-associated carcinogens, including aflatoxin (AFB1) and heterocyclic aromatic amines (HAAs). A smart contrast agent is a molecule whose relaxivity changes in response to a biological trigger such as a biomolecule, enzymatic activity, or pH. The focus of my graduate project was to develop a DNA aptamer-based smart MRI contrast agent capable of sensing dopamine. Aptamers are single-stranded oligonucleotides that form stable three-dimensional structures efficient in binding with high affinity and specificity for a range of targets. The DNA aptamer has an attached lanthanide-based probe, consisting of a metal ion Gd(III) and a macrocyclic DOTA ligand. The aptamer was assembled using solid phase oligonucleotide synthesis. Current estimates suggest that approximately 9 million U.S. citizens identify as biracial or multiracial (henceforth “multiracial”) and that the multiracial population grew 32% from 2000 to 2010 (Humes et al., 2011). Despite the growth and increased visibility of this population, the psychological research on this group is limited and further research is needed to address the potential unique needs of multiracial individuals with regard to experiences of racial oppression, racial identity, and connections to health (Choi, Harachi, Gillmore, & Catalano, 2006). RRAM has recently emerged as a strong candidate for non-volatile memory (NVM). Beyond memory applications, RRAM holds promise for use in performing logic functions, mimicking neuromorphic activities, enabling multi-level switching, and as one of the key elements of hardware based encryption or signal processing systems. It has been shown previously that RRAM resistance levels can be changed by adjusting compliance current or voltage level. From the modern protests of the Dakota Access Pipeline to feuding between village dwelling horticulturalists, there is a long history of conflict along the banks of the Missouri River. This dissertation aims to investigate the interplay between the unique landscape of the semi-arid river valley and the continuing influx of culturally diverse people moving to the region, specifically, the semi-sedentary hunting farmers living in the Middle Missouri division of the North American Great Plains, between A.D. 1000 and 1750. In sifting through his letters, famous for his unselfconscious thoughts on the nature of poetry and poetic composition, it is possible to construct the details of his culinary life not only as it pertains to daily eating habits but also in how he uses the language of food to articulate the sensations achieved in the course of writing and reading poetry. Emerging adulthood is a developmental period marked by increased anxiety related to academic, social, and professional stressors. The ability to successfully cope with stress is strongly negatively related to mental health outcomes. Furthermore, recent studies indicate a connection between executive function impairment, anxiety and depression. The current study examines executive functioning (EF), coping, anxiety, and depression in a sample of 210 emerging adults (NMales = 131, Mage = 19.14). Additionally, the relation between EF and coping is explored. Finally, coping’s effect on the relation between EF and outcome (anxiety and depression) is investigated. Molecularly imprinted polymers (MIPs) are a class of materials containing sites capable of selectively binding to the imprinted target molecule. Computational chemistry techniques were used to study the effect of different fabrication parameters (the monomer-to-target ratios, pre-polymerization solvent, temperature, and pH) on the formation of the MIP binding sites. Imprinted binding sites were built in silico for the purposes of better characterizing the receptor – ligand interactions. Chiefly, the sites were characterized with respect to their selectivities and the heterogeneity between sites.

In this dissertation, I explore the gendered, sexual, and embodied negotiations of the social world by thirty-five partners of transgender people as their significant other experienced various aspects of gender transition. This research shows the ways that people peripheral to a gender transition can be intimately impacted by it. Overexpression of HER2 (ERBB2/neu) in breast cancer is an established clinical marker for aggressive disease and increased mortality. HER2+ breast cancers have increased protein levels of Human Epidermal Growth Factor Receptor 2 and overexpression of its coding gene, ERBB2. Many HER2+ tumors feature concomitant co-expression of the Nuclear Receptor subfamily 1, Group D, Member 1 (NR1D1/RevERBα) which regulates adipogenesis and circadian rhythm; the dysregulation of these two processes are known risk factors for breast cancer. Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis in humans, is an intracellular pathogen that infects millions of people every year. Mtb can survive inside the host for extended periods of time by sensing and adapting to the host environmental stressors. Transcriptional gene regulation plays a critical role in this adaptation. The technology development of two centrifuge-based instruments gave rise to an Internet-of-Things (IOT) biotech startup. The first instrument: the centrifuge force microscope (CFM) was recently introduced as a platform for massively parallel single-molecule manipulation and analysis. Here we developed a low-cost and self-contained CFM module that works directly within a commercial centrifuge, greatly improving accessibility and ease of use. Our instrument incorporates research grade video microscopy, a power source, a computer, and wireless transmission capability to simultaneously monitor many individually tethered microspheres. Intrinsically disordered proteins or unstructured segments within proteins play an important role in cellular physiology and pathology. A combination of peptide aptamers selected by using the yeast-two-hybrid scheme, and in-cell NMR identified high affinity binders to a transiently structured intrinsically disordered proteins (IDP). This method was validated using the prokaryotic ubiquitin-like protein, Pup, of the Mycobacterium proteasome. We discover two peptide aptamers that bind to opposite sites of a transient helix in Pup, an intrinsically disordered protein, that have vastly different effects on the survival of Mycobacterium bovis BCG.

Development of electrospun nanofibers that mimic the structural, mechanical and biochemical properties of natural extracellular matrices (ECMs) and the basement membranes is a promising approach for tissue regeneration. Electrospun fibers of synthetic polymers partially mimic the topography of the ECM, however, their high stiffness, poor hydrophilicity and lack of in vivo-like biochemical cues is not optimal for epithelial cell self-organization and function. In search of a biomimetic scaffold for salivary gland tissue regeneration, we utilized proteins and peptides to induce biomechanical and biochemical cues that resembles the native tissue environment. DNA nanotechnology has shown great promise in molecular diagnostic, bioanalytical and biomedical applications. The great challenge of detecting target analytes, biomarkers and small molecules, in molecular diagnostics is low yield sensitivity. The New Right is the fastest growing party family in Europe. Factors contributing to its success have been grouped broadly into two types: demand-side factors and supply-side factors. Demand-side factors comprise socio-economic developments such as unemployment, public mistrust in the political establishment, and levels of immigration. Supply-side factors relate to the mechanics of the party system, the type of the electoral system, and endogenous features of parties such as ideology, leadership and organization. In recent decades, scholarly interest on immigration and crime has been on the rise. This increase in interest has even recently produced an academic consensus on the structural-level relationship between immigration and crime: on average, changes in immigration levels do not disproportionately increase crime rates. However, what is less explored in the literature is the individual-level relationship between immigration and criminal victimization, and specifically, the role played by criminological variables among generations of immigrant ethnic groups and their victimization experiences.
