Retrotransposons are mobile genetic elements that replicate via an RNA intermediary and constitute a significant portion of most eukaryotic genomes. Saccharomyces cerevisiae has been invaluable to retrotransposon research due to the presence of an active retroelement known as Ty1. The mobility of Ty1 is regulated both positively and negatively by numerous host factors, including several subunits of the Mediator transcriptional co-activator complex. The Mediator core complex is organized into genetically and structurally defined head, middle, and tail modules, along with a transiently associated kinase module. Introduction: Health-related Quality of life (HRQoL) is recognized as a significant medical outcome measure for persons living with HIV (PLWH). Identification of potentially modifiable determinants of HRQoL could help target people in need of additional health care and social services in order to improve their HRQoL. This dissertation delineates the process and advancements in professionalizing the police departments and the development of forensic science in New York. Furthermore, as a preliminary and exploratory study it examines the evolution and changes of forensic science within the growth and expansion of a New York county police department. The relationship between police and forensic science was complex; for example, the impetus for the police to adopt a particular forensic methodology varied. At times, the development of forensic methodologies necessitated a further step in professionalizing the police departments. Expression of metabolic enzymes Cytochrome P450 (CYP) is highly affected by drugs, diseases, age and gender. The Intestinal microflora has been suggested to play a role in regulating CYP expression and antibiotic treatments would lead to changes in level and composition of microbiota in mouse gut. This study aims to analyze CYP expression changes in the liver and small intestine resulted from the reduction of intestinal microflora by antibiotics. Neurodegenerative diseases like Alzheimer’s, Parkinson’s, Huntington’s, Amyotrophic Lateral Sclerosis and Prion Diseases tend to have similar cellular and molecular mechanisms of protein aggregation and inclusion body formation. These processes suggest common mechanisms of pathogenesis. Similarities in structure, mechanism of formation and physiological effects of protein aggregates in neurodegenerative diseases may lead to development of common preventative or therapeutic strategies.

The challenges to forensic body fluid analysis have placed limitations on the type of information that investigators can acquire and how that information can be collected. In recent years, Raman spectroscopy has proven itself useful for characterizing body fluids. In 2008, a large-scale investigation was undertaken to explore the use of Raman spectroscopy as a means of identifying body fluids. This work resulted in multidimensional Raman spectroscopic signatures for the five main body fluids: semen, peripheral blood, saliva, vaginal fluid, and sweat. The allosteric regulation of SIRT6 by nicotinamide (NAM), along with the growing evidence of this enzyme's key role in the immune response, prompted the mechanistic study of SIRT6 inhibition by pyrazinamide (PZA) and analogs. In our current study, PZA, an analog of NAM, was revealed to have a modest modulatory effect on SIRT6, an enzyme that regulates the NF-κB signaling pathway at the transcriptional level (a relevant pathway to inflammation). Similarly, the analogs of PZA, 5-Cl PZA, 5-MeO PZA, and POA exhibited a modulatory effect against SIRT6 in our in vitro studies, enabling identification of a potential new target for …

HIV contributes to the highest number of deaths from infectious diseases in low-income countries. In this three-paper dissertation, using the case of Pakistan, I focus on understanding how the exercise of power by actors at three different levels of the health system, individual, program and societal, influences policy implementation and shapes HIV program outcomes. The objective is to improve HIV service provision thereby achieving improved health policy outcomes. I adopt a mixed-methods approach and use quantitative (logit regression) and qualitative techniques (content analysis of newspapers and semi-structured interviews). BADGE (bisphenol A diglycidyl ether) based epoxy resins have been used in a variety of applications since the 1940s. However, limited studies are available about the occurrence of BADGE and its derivatives (BADGEs) in the environment. The aims of this thesis were to determine the concentrations of BADGE and its derivatives in dental sealants and textile products from the U.S. to characterize the sources of these substances. Recent avian and swine-origin influenza virus outbreaks illustrate the ongoing threat of influenza pandemics. We investigated immunogenicity and protective efficacy of a multi-antigen (MA) universal influenza DNA vaccine consisting of HA, M2, and NP antigens in cynomolgus macaques. Following challenge with a heterologous pandemic H1N1 strain, vaccinated animals exhibited significantly lower viral loads and more rapid viral clearance when compared to unvaccinated controls. The MA DNA vaccine induced robust serum and mucosal antibody responses but these high antibody titers were not broadly neutralizing. In contrast, the vaccine induced broadly-reactive NP specific T cell responses that cross-reacted with the challenge virus …

Opioid analgesic tolerance remains a considerable drawback to chronic pain management. The finding that concomitant administration of delta opioid receptor (DOR) antagonists attenuates the development of tolerance to mu opioid receptor (MOR) agonists has led to interest in producing bifunctional MOR agonist/DOR antagonist ligands. Herein, we present 7-benzylideneoxymorphone (6, UMB 246) displaying MOR partial agonist/DOR antagonist activity, representing a new lead for designing bifunctional MOR/DOR ligands.

Diet can influence the composition of the human microbiome, and yet relatively few dietary ingredients have been systematically investigated with respect to their impact on the functional potential of the microbiome. Dietary resistant starch (RS) has been shown to have health benefits, but we lack a mechanistic understanding of the metabolic processes that occur in the gut during digestion of RS. Here, we collected samples during a dietary crossover study with diets containing large or small amounts of RS. Assessing the risk of transgenic crop products is essential when determining the safety of a crop for deregulation and commercialization. The Organization of Economic and Cooperative Development (OECD), International Standards Organization (ISO), and governmental regulatory agencies require a battery of tests to demonstrate the safety of a GM product against several surrogate species of organisms that perform various ecosystem services. Assays are performed using toxicology methods established for pesticides. Lytopylus is species-rich genus of Agathidinae (Hymenoptera: Braconidae: Agathidinae), but limited information of the genus has been published. Morphological, molecular, and ecological data were compared to resolve the species limits of Lytopylus reared from caterpillars collected the Area de Conservación Guanacaste (ACG) in Costa Rica. Molecular data were initially used to hypothesize species limits, and morphological and host use data were employed to make a final decision when molecular data was indecisive, e.g., when the genetic difference between species was slight. Throughout Peirce’s writing, we witness his developing vision of a machine that scientists will eventually be able to create. Nadin (2010) raised the question: Why do computer scientists continue to ignore Peirce’s sign theory? A review of the literature on Peirce’s theory and the semiotics machine reveals that many authors discussed the machine; however, they do not differentiate between a physical computer machine and its software. This paper discusses the problematic issues involved in converting Peirce’s theory into a programming language, machine and software application. Prostate cancer is the most common noncutaneous cancer in males. Men of African origin are at a significantly higher risk as reflected in the higher incidence and mortality rates in this racial group. Metastases incidence increases parallel to serum levels of prostate-specific antigen (PSA), contributing significantly to morbidity and mortality. Staging of the disease involves bone scans, which are sensitive in detecting skeletal metastases. PURPOSE In California, the passage of SB493 in July of 2013 was a milestone in advancing pharmacy practice. Among other things, the new legislation allows pharmacists to provide routine immunizations without a protocol and furnish medications for international travelers for conditions not requiring a diagnosis. When developing a pharmacist-run travel health service, consideration must be given to multiple important factors, including pharmacist training, physician partnership, logistics, from scheduling to documentation, and the resources necessary to provide a travel health service.5 This article sets out to provide guidance and insight to pharmacists seeking to implement a travel health service.

Biomass is considered a renewable source of energy with minimum carbon foot print if managed sustainably. The majority of the worlds energy is spend on transportation, and fast pyrolysis of biomass could be a potential route for production of a sustainable liquid transportation fuel. However, there are several hurdles in the conversion process. This work addresses these hurdles by investigating the impact of several pretreatment methods on fast pyrolysis including thermal pretreatment (torrefaction), comminution/grinding, mineral reduction. The impact of important parameters like heat transfer medium, conversion temperature and particle size were also investigated.

The invasion of Eurasian watermilfoil (Myriophyllum spicatum) can influence littoral zone communities within lakes. Its formation of dense mats at the water surface can suppress native macrophyte growth and impact fish diets and community structure, as well as invertebrate assemblages. However, in the colder waters of the upper Great Lakes region, Eurasian watermilfoil is patchily distributed and integrates more with the native macrophyte community.
