Immune Modulation In The Treatment Of Amyotrophic Lateral Sclerosis: A Review Of Clinical Trials, Syed I. Khalid, Leonel Ampie, Ryan Kelly, Shafeeq S. Ladha, Christopher Dardis
Neurology
Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease characterized by the degeneration of motor neurons. Though many molecular and genetic causes are thought to serve as predisposing or disease propagating factors, the underlying pathogenesis of the disease is not known. Recent discoveries have demonstrated the presence of inflammation propagating substrates in the central nervous system of patients afflicted with ALS. Over the past decade, this hypothesis has incited an effort to better understand the role of the immune system in ALS and has led to the trial of several potential immune-modulating therapies. Here, we briefly review advances in the …

Neurology
Objective: To systematically review the evidence and make recommendations with regard to diagnostic utility of cervical and ocular vestibular evoked myogenic potentials (cVEMP and oVEMP, respectively). Four questions were asked: Does cVEMP accurately identify superior canal dehiscence syndrome (SCDS)? Does oVEMP accurately identify SCDS? For suspected vestibular symptoms, does cVEMP/oVEMP accurately identify vestibular dysfunction related to the saccule/ utricle? For vestibular symptoms, does cVEMP/oVEMP accurately and substantively aid diagnosis of any specific vestibular disorder besides SCDS? Methods: The guideline panel identified and classified relevant published studies (January 1980- December 2016) according to the 2004 American Academy of Neurology process. Results …

The Impact Of Aging On Brain Pituitary Adenylate Cyclase Activating Polypeptide Pathology And Cognition In Mice And Rhesus Macaques, Pengcheng Han, Megan Nielsen, Melissa Song, Junxiang Yin, Michele R. Permenter, Julie A. Vogt, James R. Engle, Brittany N. Dugger, Thomas G. Beach, Carol A. Barnes, Jiong Shi
Neurology
Pituitary adenylate cyclase activating polypeptide (PACAP) is associated with Alzheimer's disease (AD), but its age-related effects are unknown. We chose the rhesus macaque due to its closeness to human anatomy and physiology. We examined four variables: aging, cognitive performance, amyloid plaques and PACAP. Delayed nonmatching-to-sample recognition memory scores declined with age and correlated with PACAP levels in the striatum, parietal and temporal lobes. Because amyloid plaques were the only AD pathology in the old rhesus macaque, we further studied human amyloid precursor protein (hAPP) transgenic mice. Aging was associated with decreased performance in the Morris Water Maze (MWM). In wild …

Neurology
Seizure-driven brain damage in epilepsy accumulates over time, especially in the hippocampus, which can lead to sclerosis, cognitive decline, and death. Excitotoxicity is the prevalent model to explain ictal neurodegeneration. Current labeling technologies cannot distinguish between excitotoxicity and hypoxia, however, because they share common molecular mechanisms. This leaves open the possibility that undetected ischemic hypoxia, due to ictal blood flow restriction, could contribute to neurodegeneration previously ascribed to excitotoxicity. We tested this possibility with Confocal Laser Endomicroscopy (CLE) and novel stereological analyses in several models of epileptic mice. We found a higher number and magnitude of NG2+ mural-cell mediated capillary …

Objective: We performed a genome-wide brain expression study to reveal the underpinnings of diseases linked to a repeat expansion in chromosome 9 open reading frame 72 (C9ORF72). Methods: The genome-wide expression profile was investigated in brain tissue obtained from C9ORF72 expansion carriers (n = 32), patients without this expansion (n = 30), and controls (n = 20). Using quantitative real-time PCR, findings were confirmed in our entire pathologic cohort of expansion carriers (n = 56) as well as nonexpansion carriers (n = 31) and controls (n = 20). Results: Our findings were most profound in the cerebellum, where we identified …
Regulator Of Cell Cycle (Rgcc) Expression During The Progression Of Alzheimer's Disease, Scott E. Counts, Elliott J. Mufson
Regulator Of Cell Cycle (Rgcc) Expression During The Progression Of Alzheimer's Disease, Scott E. Counts, Elliott J. Mufson
Translational Neuroscience
Unscheduled cell cycle reentry of postmitotic neurons has been described in cases of mild cognitive impairment (MCI) and Alzheimer's disease (AD) and may form a basis for selective neuronal vulnerability during disease progression. In this regard, the multifunctional protein regulator of cell cycle (RGCC) has been implicated in driving G1/S and G2/M phase transitions through its interactions with cdc/cyclin-dependent kinase 1 (cdk1) and is induced by p53, which mediates apoptosis in neurons. We tested whether RGCC levels were dysregulated in frontal cortex samples obtained postmortem from subjects who died with a clinical diagnosis of no cognitive impairment (NCI), MCI, or …
Exploring Alternative Funding Methods For The Lead Remediation Program In Scott County, Iowa, Dr. John Delaney, Brittany A. Poynor
Exploring Alternative Funding Methods For The Lead Remediation Program In Scott County, Iowa, Dr. John Delaney, Brittany A. Poynor
2016-2017: Scott County, Iowa and the Scott County Health Department
Lead poisoning is an environmental as well as social problem. Areas with more older housing and higher rates of poverty are also the areas with the most lead poisoned children. Though the United States banned the use of lead paint in in 1978 and lead poisoning is preventable, lead poisoning remains a public health problem. Homes built before 1978 may pose risk of lead paint exposure, but of special concern to Scott County are the number of homes built before 1950.
Scott County, Iowa is home to some of the oldest housing units in the U.S. Iowa ranks fifth among …
Genetic Basis For Sex Differences In Obesity And Lipid Metabolism, Jenny Link, Karen Reue
Genetic Basis For Sex Differences In Obesity And Lipid Metabolism, Jenny Link, Karen Reue
Biology
Men and women exhibit significant differences in obesity, cardiovascular disease, and diabetes. To provide better diagnosis and treatment for both sexes, it is important to identify factors that underlie the observed sex differences. Traditionally, sex differences have been attributed to the differential effects of male and female gonadal secretions (commonly referred to as sex hormones), which substantially influence many aspects of metabolism and related diseases. Less appreciated as a contributor to sex differences are the fundamental genetic differences between males and females, which are ultimately determined by the presence of an XX or XY sex chromosome complement. Here, we review …
Archelosaurian Color Vision, Parietal Eye Loss, And The Crocodylian Nocturnal Bottleneck, Christopher A. Emerling
Archelosaurian Color Vision, Parietal Eye Loss, And The Crocodylian Nocturnal Bottleneck, Christopher A. Emerling
Biology
Vertebrate color vision has evolved partly through the modification of five ancestral visual opsin proteins via gene duplication, loss, and shifts in spectral sensitivity. While many vertebrates, particularly mammals, birds, and fishes, have had their visual opsin repertoires studied in great detail, testudines (turtles) and crocodylians have largely been neglected. Here I examine the genomic basis for color vision in four species of turtles and four species of crocodylians, and demonstrate that while turtles appear to vary in their number of visual opsins, crocodylians experienced a reduction in their color discrimination capacity after their divergence from Aves. Based on the …

A Multicriteria Decision Analysis For Identifying Priority Conservation Areas For Grassland Birds, Flavio Sutti, Allan Strong, Noah G. Perlut
Environmental Studies Faculty Publications
Biodiversity conservation frequently competes with the needs of society for agricultural production and development. However, properly designed and efficiently implemented conservation programs can be used to integrate wildlife and human needs. We tested the efficacy of multicriteria decision analysis as a tool to select priority areas for conservation in human-dominated landscapes using grassland birds in the northeastern US as a test case. We created detailed GIS layers including landscape- (forest, grassland, development, and roads within a 3000-m buffer around each grassland patch) and patch-level (size, management, and conservation status) criteria important in grassland bird habitat selection and conservation. We developed …

Angular leaf spot (ALS) and powdery mildew (PWM) are two important fungi diseases causing significant yield losses in common beans. In this study, a new genetic linkage map was constructed using single sequence repeats (SSRs) and single nucleotide polymorphisms (SNPs), in a segregating population derived from the AND 277 x SEA 5 cross, with 105 recombinant inbred lines. Phenotypic evaluations were performed in the greenhouse to identify quantitative trait loci (QTLs) associated with resistance by means of the composite interval mapping analysis. Four QTLs were identified for ALS resistance. The QTL ALS11AS, linked on the SNP BAR 5054, mapped on …

The common bean is characterized by high sensitivity to drought and low productivity. Breeding for drought resistance in this species involves genes of different genetic groups. In this work, we used a SEA 5 x AND 277 cross to map quantitative trait loci associated with drought tolerance in order to assess the factors that determine the magnitude of drought response in common beans. A total of 438 polymorphic markers were used to genotype the F8 mapping population. Phenotyping was done in two greenhouses, one used to simulate drought and the other to simulate irrigated conditions. Fourteen traits associated with drought …

The metabolic demand of walking generally increases following lower extremity amputation. This study used real-time visual feedback to modify biomechanical factors linked to an elevated metabolic demand of walking in individuals with transtibial amputation. Eight persons with unilateral, traumatic transtibial amputation and 8 uninjured controls participated. Two separate bouts of real-time visual feedback were provided during a single session of gait retraining to reduce 1) center of mass sway and 2) thigh muscle activation magnitudes and duration. Baseline and post-intervention data were collected. Metabolic rate, heart rate, frontal plane center of mass sway, quadriceps and hamstrings muscle activity, and co-contraction …

Race, Culture & Abuse Of Persons With Disabilities, Elizabeth P. Cramer, Y. Joon Choi, Avina I. Ross
Social Work Publications
This chapter will explore how race and culture influence the lives of persons with disabilities who are experiencing abuse. The discussion will be framed by an intersectional lens and will be informed by cultural humility and critical race theory. Practitioners need to remain open to the idea that they cannot and will not know all there is to know about any given culture, and they should be open to hearing about their clients’ understanding and experiences of culture. Rather than knowing certain pieces of “knowledge” about a cultural group, it is more important to understand what pieces of culture the …

Noise-Enhanced Coding In Phasic Neuron Spike Trains, Cheng Ly, Brent Doiron
Statistical Sciences and Operations Research Publications
The stochastic nature of neuronal response has lead to conjectures about the impact of input fluctuations on the neural coding. For the most part, low pass membrane integration and spike threshold dynamics have been the primary features assumed in the transfer from synaptic input to output spiking. Phasic neurons are a common, but understudied, neuron class that are characterized by a subthreshold negative feedback that suppresses spike train responses to low frequency signals. Past work has shown that when a low frequency signal is accompanied by moderate intensity broadband noise, phasic neurons spike trains are well locked to the signal. …

Quantitative relationship between the magnitude of variation in minor histocompatibility antigens (mHA) and graft versus host disease (GVHD) pathophysiology in stem cell transplant (SCT) donor-recipient pairs (DRP) is not established. In order to elucidate this relationship, whole exome sequencing (WES) was performed on 27 HLA matched related (MRD), & 50 unrelated donors (URD), to identify nonsynonymous single nucleotide polymorphisms (SNPs). An average 2,463 SNPs were identified in MRD, and 4,287 in URD DRP (p
