Journal Articles: Pathology and Microbiology

Menaquinone (MK) biosynthesis pathway is a potential target for evaluating antimicrobials in gram-positive bacteria. Here, 1,4-dihydroxy-2-naphthoate prenyltransferase (MenA) was targeted to reduce methicillin-resistant Staphylococcus aureus (MRSA) growth. MenA inhibiting, long chain-based compounds were designed, synthesized and evaluated against MRSA and menaquinone utilizing bacteria in aerobic conditions. The results showed that these bacteria were susceptible to most of the compounds. Menaquinone (MK-4) supplementation rescued MRSA growth, suggesting these compounds inhibit MK biosynthesis. 3a and 7c exhibited promising inhibitory activities with MICs ranging 1-8 μg/mL against MRSA strains. The compounds did not facilitate small colony variant formation. These compounds also inhibited the …

Neisseria gonorrhoeae is the causative agent of one of the most common sexually transmitted diseases, gonorrhea. Over the past two decades there has been an alarming increase of reported gonorrhea cases where the bacteria were resistant to the most commonly used antibiotics thus prompting for alternative antimicrobial treatment strategies. The crucial step in this and many other bacterial infections is the formation of microcolonies, agglomerates consisting of up to several thousands of cells. The attachment and motility of cells on solid substrates as well as the cell–cell interactions are primarily mediated by type IV pili, long polymeric filaments protruding from …

Putting Resilience Theory Into Practice: The Example Of Fisheries Management, Robin Kundis Craig Jan 2017

Putting Resilience Theory Into Practice: The Example Of Fisheries Management, Robin Kundis Craig

Utah Law Faculty Scholarship

By acknowledging a world of continuous change and reduced human control over nature, resilience theory thus suggests a wide range of potential changes to marine fisheries management for a changing ocean. Even the most modest of these, however, should inspire comprehensive amendments to both domestic and international fisheries law, particularly to their emphases on MSY. Full incorporation of resilience thinking, in turn, demands a longer-term and system-based perspective on marine management, empowering humans to make choices now to strengthen the ecological resilience of marine ecosystems to the changes that are still coming, increasing the chances that the ocean will remain …

Structure-Function Investigation Of Membrane Proteins Using Nanodiscs, Robbins J. Puthenveetil Jan 2017

Structure-Function Investigation Of Membrane Proteins Using Nanodiscs, Robbins J. Puthenveetil

Doctoral Dissertations

Several membrane proteins, comprising of either α-helical or β-barrel structures have been successfully studied in vitro using several membrane mimetic systems. Nanodiscs, a new class of discoidal nanoscale lipoprotein complex have been used extensively in the present study to investigate different membrane proteins. We begin by delineating the development of small (D7) nanodiscs through its rigorous characterization and demonstrate advantages in solution NMR applications. In addition, we show the development of an on-column method to generate nanodiscs through the selective use of variable protein tags. D7 discs have been further used to visualize the downstream phosphorylation events of activated integrin …

Δnp63Α And Microrna: Leveraging The Epithelial-Mesenchymal Transition, Andrew J. Stacy, Michael P. Craig, Suraj Sakaram, Madhavi Kadakia

Biochemistry and Molecular Biology Faculty Publications

The epithelial-mesenchymal transition (EMT) is a cellular reprogramming mechanism that is an underlying cause of cancer metastasis. Recent investigations have uncovered an intricate network of regulation involving the TGFβ Wnt, and Notch signaling pathways and small regulatory RNA species called microRNAs (miRNAs). The activity of a transcription factor vital to the maintenance of epithelial stemness, ?Np63a, has been shown to modulate the activity of these EMT pathways to either repress or promote EMT. Furthermore, ?Np63a is a known regulator of miRNA, including those directly involved in EMT. This review discusses the evidence of ?Np63a as a master regulator of EMT …

Effect Of Seed Morph And Light Level On Growth And Reproduction Of The Amphicarpic Plant Amphicarpaea Edgeworthii (Fabaceae), Keliang Zhang, Jerry M. Baskin, Carol C. Baskin, Xuejun Yang, Zhenying Huang

Biology Faculty Publications

Amphicarpic plants produce aerial and subterranean fruits on an individual plant, and these heteromorphic diaspores give rise to plants that differ in growth and ecology. Amphicarpaea edgeworthii is a summer annual amphicarpic species that grows over a range of light levels. We aimed to compare the response to shading intensity of plants of A. edgeworthii grown throughout their life cycle from aerial seeds (ASP) and from subterranean seeds (SSP). We hypothesized that vegetative and reproductive growth of plants from ASP and SSP respond differently to light. Plants were grown from ASP and SSP under 0, 46, 71 and 90% shading …

A Cascade Of Multiple Proteins And Lipids Catalyzes Membrane Fusion, William Wickner, Josep Rizo Jan 2017

A Cascade Of Multiple Proteins And Lipids Catalyzes Membrane Fusion, William Wickner, Josep Rizo

Dartmouth Scholarship

Recent studies suggest revisions to the SNARE paradigm of membrane fusion. Membrane tethers and/or SNAREs recruit proteins of the Sec 1/Munc18 family to catalyze SNARE assembly into trans-complexes. SNARE-domain zippering draws the bilayers into immediate apposition and provides a platform to position fusion triggers such as Sec 17/α-SNAP and/or synaptotagmin, which insert their apolar "wedge" domains into the bilayers, initiating the lipid rearrangements of fusion.

The induction of tumor suppressor proteins capable of cancer cell apoptosis represents an attractive option for the re-purposing of existing drugs. We report that the anti-malarial drug, chloroquine (CQ), is a robust inducer of Par-4 secretion from normal cells in mice and cancer patients in a clinical trial. CQ-inducible Par-4 secretion triggers paracrine apoptosis of cancer cells and also inhibits metastatic tumor growth. CQ induces Par-4 secretion via the classical secretory pathway that requires the activation of p53. Mechanistically, p53 directly induces Rab8b, a GTPase essential for vesicle transport of Par-4 to the plasma membrane prior to secretion. Our findings …

Androgen Receptor-Dependent And -Independent Mechanisms Driving Prostate Cancer Progression: Opportunities For Therapeutic Targeting From Multiple Angles., David T. Hoang, Kenneth A. Iczkowski, Deepak Kilari, William See, Marja T. Nevalainen

Student Papers, Posters & Projects

Despite aggressive treatment for localized cancer, prostate cancer (PC) remains a leading cause of cancer-related death for American men due to a subset of patients progressing to lethal and incurable metastatic castrate-resistant prostate cancer (CRPC). Organ-confined PC is treated by surgery or radiation with or without androgen deprivation therapy (ADT), while options for locally advanced and disseminated PC include radiation combined with ADT, or systemic treatments including chemotherapy. Progression to CRPC results from failure of ADT, which targets the androgen receptor (AR) signaling axis and inhibits AR-driven proliferation and survival pathways. The exact mechanisms underlying the transition from androgen-dependent PC …

Cultivating Relationships From A Common Cor*: What Teachers Do To Relate To Othered Students, Lucia Desantis-Heddleson Jan 2017

Cultivating Relationships From A Common Cor*: What Teachers Do To Relate To Othered Students, Lucia Desantis-Heddleson

Student Scholarship

This research is focused on the teacher's perspective of the dyadic relationship between teachers and othered students--students who are traditionally disregarded and/or are considered outside of the norm (Borrero et al., 2012). The study at hand is limited to this dyad, but is crucial for the understanding of what behaviors, social and emotional intelligence competencies, and practices make a teacher good at reaching othered students beyond the mere academics of doing school. The role of a teacher within the confines of schooling is changing because schools have become institutions involved in socialization, motivation, and adapting to change (i.e., preparing students …

Inferring Condition-Specific Targets Of Human Tf-Tf Complexes Using Chip-Seq Data, Chia-Chun Yang, Min-Hsuan Chen, Sheng-Yi Lin, Erik H. Andrews, Chao Cheng, Jeremy J.W Chen

Dartmouth Scholarship

Background:

Transcription factors (TFs) often interact with one another to form TF complexes that bind DNA and regulate gene expression. Many databases are created to describe known TF complexes identified by either mammalian two-hybrid experiments or data mining. Lately, a wealth of ChIP-seq data on human TFs under different experiment conditions are available, making it possible to investigate condition-specific (cell type and/or physiologic state) TF complexes and their target genes.

Results:

Here, we developed a systematic pipeline to infer Condition-Specific Targets of human TF-TF complexes (called the CST pipeline) by integrating ChIP-seq data and TF motifs. In total, we predicted …

Alzheimer's disease (AD) is a slow, progressive neurodegenerative disease and the most common type of dementia in the elderly. The etiology of AD and its underlying mechanism are still not clear. In a previous study, we found that an ethyl acetate extract of Centipedegrass (CG) (i.e., EA-CG) contained 4 types of Maysin derivatives, including Luteolin, Isoorientin, Rhamnosylisoorientin, and Derhamnosylmaysin, and showed protective effects against Amyloid beta (Aβ) by inhibiting oligomeric Aβ in cellular and in vitro models. Here, we examined the preventative effects of EA-CG treatment on the Aβ burden in the Tg (Mo/Hu APPswe PS1dE9) AD mouse model. We …

Fröhlich Effect And Delays Of Visual Attention, Nika Adamian, Patrick Cavanagh Jan 2017

Fröhlich Effect And Delays Of Visual Attention, Nika Adamian, Patrick Cavanagh

Dartmouth Scholarship

In the Fröhlich effect, the initial position of an object that suddenly appears in motion is perceived as being shifted in the direction of its motion. Here we establish that this shift is not an obligatory consequence of motion, but it is driven by focused attention. In Experiment 1 using different cueing conditions, we found that invalid cues produced larger perceptual shifts, although the Fröhlich effect was still present for valid and neutral cues. These results support Müsseler and Aschersleben's (1998) proposal that the Fröhlich effect is the result of the time it takes to shift focal attention to the …

Evidence For Genetic Heterogeneity Between Clinical Subtypes Of Bipolar Disorder, Aw Charney, Dm Ruderfer, Ea Stahl, Et Al

School of Biomedical Sciences

We performed a genome-wide association study of 6447 bipolar disorder (BD) cases and 12 639 controls from the International Cohort Collection for Bipolar Disorder (ICCBD). Meta-analysis was performed with prior results from the Psychiatric Genomics Consortium Bipolar Disorder Working Group for a combined sample of 13 902 cases and 19 279 controls. We identified eight genome-wide significant, associated regions, including a novel associated region on chromosome 10 (rs10884920; P=3.28 × 10−8) that includes the brain-enriched cytoskeleton protein adducin 3 (ADD3), a non-coding RNA, and a neuropeptide-specific aminopeptidase P (XPNPEP1). Our large sample size allowed us to test the heritability and …

Can Scientists And Their Institutions Become Their Own Open Access Publishers?, Karen Shashok

Copyright, Fair Use, Scholarly Communication, etc.

This article offers a personal perspective on the current state of academic publishing, and posits that the scientific community is beset with journals that contribute little valuable knowledge, overload the community’s capacity for high-quality peer review, and waste resources. Open access publishing can offer solutions that benefit researchers and other information users, as well as institutions and funders, but commercial journal publishers have influenced open access policies and practices in ways that favor their economic interests over those of other stakeholders in knowledge creation and sharing. One way to free research from constraints on access is the diamond route of …

Helping Hand Tools V. United States Environmental Protection Agency, Emily A. Slike

Public Land & Resources Law Review

When the EPA decided to treat biomass fuel sources differently within the BACT analysis, the Ninth Circuit continued Chevron’s legacy and granted the agency deference. The Bioenergy BACT may develop as science continues to evolve, but because the EPA took a “hard look” during a thorough permit review, the court held that agency issuance of new BACT guidelines was reasonable.

Quantifying The Totality Of Treatment Effect With Multiple Event-Time Observations In The Presence Of A Terminal Event From A Comparative Clinical Study, Brian Claggett, Lu Tian, Haoda Fu, Scott D. Solomon, L. J. Wei Jan 2017

Quantifying The Totality Of Treatment Effect With Multiple Event-Time Observations In The Presence Of A Terminal Event From A Comparative Clinical Study, Brian Claggett, Lu Tian, Haoda Fu, Scott D. Solomon, L. J. Wei

Harvard University Biostatistics Working Paper Series

To evaluate the totality of one treatment's benefit/risk profile relative to an alternative treatment via a longitudinal comparative clinical study, the timing and occurrence of multiple clinical events are typically collected during the patient's followup. These multiple observations reflect the patient's disease progression/burden over time. The standard practice is to create a composite endpoint from the multiple outcomes, the timing of the occurrence of the first clinical event, to evaluate the treatment via the standard survival analysis techniques. By ignoring all events after the composite outcome, this type of assessment may not be ideal. Various parametric or semi-parametric procedures have …
