Type 2 diabetes mellitus is strongly associated with increased risk of cardiovascular disease. Despite availability of several antidiabetic medications, the cardiovascular outcomes remain unchanged. Activation of renin angiotensin system is one of the critical factors in development of diabetic complications. Hyperglycemia causes an increase in tissue and circulating Angiotensin II. Angiotensin converting enzyme (ACE2) and neprilysin (NEP) are angiotensin (1-7) forming enzymes. ACE2 was first cloned from heart failure patients and has cardioprotective and renoprotective properties. Combination of angiotensin receptor antagonism and NEP inhibition is a new therapeutic strategy for treatment of heart failure. The aim of this study was …
Biomarkers Of Sepsis: A Retrospective Approach, Jose M. Roberts
Biomarkers Of Sepsis: A Retrospective Approach, Jose M. Roberts
Williams Honors College, Honors Research Projects
Background: Biomarkers are taking the spotlight in becoming the norm for early diagnoses. Sepsis is an inflammatory disease that increases metabolic rate in children. The first biomarker is hyponatremia. Hyponatremia is a frequent electrolyte imbalance in clinical practice, often observed in children with inflammatory disease and infection. Presence of hyponatremia is associated with electrical signaling imbalances, inflammation and renal dysfunction. The clinical value of hyponatremia in pediatric patients is unknown. The C-reactive protein is a second biomarker. Its presence signifies that necrotic cells and inflammation are present.
Objectives: To evaluate the use of biomarkers in children seen in the Emergency …
Development Of Phenotypic Drug Discovery Models For Tropical Parasitic Diseases, Surendra Kumar Jain
Development Of Phenotypic Drug Discovery Models For Tropical Parasitic Diseases, Surendra Kumar Jain
Electronic Theses and Dissertations
Malaria and visceral leishmaniasis are major killer parasitic diseases. These diseases though occur primarily in tropical countries; economic burden and overall health impacts of malaria and leishmaniasis are global. The emergence of drug-resistant and more-virulent strains of the pathogens has further amplified the problems. New drug discovery approaches primarily rely on in vitro and in vivo models of the disease. The pathogens causing leishmaniasis and malaria are intracellular. Leishmania parasite grows as amastigotes in macrophages cells, and malaria parasite grows within the hepatocytes or erythrocytes. The intracellular forms of the pathogens are responsible for the pathophysiology of the diseases. New …

Phytochemical Investigation Of Psychoactive Medicinal Plants For The Treatment Of Neurological Disorders, Vedanjali Gogineni
Electronic Theses and Dissertations
Neurological disorders include disorders such as migraine, tension-type headaches, Parkinson’s disease (PD), epilepsy, dementia, and Alzheimer’s disease (AD). Globally, PD is the second most comneurodegenerative disorder following AD. In developed countries, PD affects 1% of all population over 60 years of age. The prevalence of PD is increasing over the years and is expected to double by 2030. It is proposed that combination of dopaminergic drugs with monoamine oxidase B (MAO-B) inhibitors, anticholinergics, catechol-O-methyl transferase inhibitors, and other non-dopaminergic drugs can better alleviate levodopa-induced motor complications along with better control of motor symptoms. MAO-B inhibitors play a significant role in …
Impact Of Corporate Social Responsibility On Financial Performance In The Pharmaceutical Industry, Stephen A. Adamu
Impact Of Corporate Social Responsibility On Financial Performance In The Pharmaceutical Industry, Stephen A. Adamu
Theses and Dissertations
Many studies have examined the association between corporate social responsibility (CSR) and corporate financial performance, but scholars argue that the exact relationship between CSR and corporate financial performance remains unclear. This quantitative study examines the impact of CSR on corporate financial performance in the pharmaceutical industry. The study addresses the research question: What is the financial performance in the pharmaceutical industry among companies that have embraced CSR? The alternative hypothesis predicted positive correlations between financial performance and CSR. The related null hypotheses predicted that there would be no correlations between any of 8 dimensions of CSR and corporate financial performance. …

A Case Study Of Upper-Room Uvgi In Densely-Occupied Elementary Classrooms By Real-Time Fluorescent Bioaerosol Measurements, Chunxiao Su, Josephine Lau, Fang Yu
Durham School of Architectural Engineering and Construction: Faculty Publications
Recently, the requirement to continuously collect bioaerosol samples using shorter response times has called for the use of real-time detection. The decreased cost of this technology makes it available for a wider application than military use, and makes it accessible to pharmaceutical and academic research. In this case study, real-time bioaerosol monitors (RBMs) were applied in elementary school classrooms—a densely occupied environment—along with upper-room ultraviolet germicidal irradiation (UVGI) devices. The classrooms were separated into a UVGI group and a non-UVGI control group. Fluorescent bioaerosol counts (FBCs) were monitored on 20 visiting days over a four-month period. The classroom with upper-room …

The prevention and cure of malaria is difficult as it thrives under poor socio- and pharmacoeconomic conditions in third world, tropical and subtropical regions. It is a parasitic disease spread through the bite of an infected Anopheles mosquito. In humans, malaria can be caused by several Plasmodium species that include: Plasmodium falciparum, Plasmodium vivax, Plasmodium ovale, Plasmodium malariae and Plasmodium knowlesi. The natural product artemisinin is a sesquiterpene lactone with an endoperoxide bridge isolated from the Chinese annual herb qinghao (Artemisia annua L or sweet wormwood). The relatively stable endoperoxide bridge is essential for their antimalarial activity. The synthesis of …

Background: Biting midges in the genus Culicoides (Diptera; Ceratopogonidae) have been implicated in the transmission of a number of parasites and highly pathogenic viruses. In North America, the complete transmission cycles of many of these pathogens need further elucidation. One way to increase our knowledge about the evolution and ecology of Culicoides species and the pathogens they transmit is to document the diversity of vertebrate hosts that Culicoides feed upon. Our objective was to identify the diversity of Culicoides hosts in the United States.
Results: We sequenced two vertebrate mitochondrial genes (cytochrome c oxidase subunit 1 and cytochrome b) from …

The common vampire bat, Desmodus rotundus, ranges from South America into northern Mexico in North America. This sanguivorous species of bat feeds primarily on medium to large-sized mammals and is known to rely on livestock as primary prey. Each year, there are hotspot areas of D. rotundus-specific rabies virus outbreaks that lead to the deaths of livestock and economic losses. Based on incidental captures in our study area, which is an area of high cattle mortality from D. rotundus transmitted rabies, it appears that D. rotundus are being caught regularly in areas and elevations where they previously were …

Optimization Of Human, Animal, And Environmental Health By Using The One Health Approach, Jonathan M. Sleeman, Thomas J. Deliberto, Natalie Nguyen
United States Department of Agriculture Wildlife Services: Staff Publications
Emerging diseases are increasing burdens on public health, negatively affecting the world economy, causing extinction of species, and disrupting ecological integrity. One Health recognizes that human, domestic animal, and wildlife health are interconnected within ecosystem health and provides a framework for the development of multidisciplinary solutions to global health challenges. To date, most health-promoting interventions have focused largely on single-sector outcomes. For example, risk for transmission of zoonotic pathogens from bush-meat hunting is primarily focused on human hygiene and personal protection. However, bush-meat hunting is a complex issue promoting the need for holistic strategies to reduce transmission of zoonotic disease …

Antibiotic resistance, particularly to fluoroquinolones and macrolides, in the major foodborne pathogen Campylobacter is considered a serious threat to public health. Although ruminant animals serve as a significant reservoir for Campylobacter, limited information is available on antibiotic-resistant Campylobacter of bovine origin. Here, we analyzed the antimicrobial susceptibilities of 320 C. jejuni and 115 C. coli isolates obtained from feedlot cattle farms in multiple states in the U.S. The results indicate that fluoroquinolone resistance reached to 35.4% in C. jejuni and 74.4% in C. coli, which are significantly higher than those previously reported in the U.S. While all fluoroquinolone resistant …

Various tissues, nasal swabs, urine and blood samples were collected from 376 feral swine at two federally inspected abattoirs in Texas during six separate sampling periods in 2015. Samples were tested for Brucella spp. by culture and serology. Brucella spp. were cultured from 13.0% of feral swine, and antibodies were detected in 9.8%. Only 32.7% of culture-positive feral swine were also antibody positive, and 43.2% of antibody-positive feral swine were culture positive. Approximately, the same number of males (14.0%) and females (12.1%) were culture positive, and slightly more males (10.5%) than females (8.7%) were antibody positive. Our results indicate that …
A Diagnostic And Control Programme For Johne’S Disease On Irish Dairy Farms, Aideen Kennedy
A Diagnostic And Control Programme For Johne’S Disease On Irish Dairy Farms, Aideen Kennedy
Theses
Johne’s disease (JD) is an enteritis of ruminants with potential zoonotic implications. JD diagnosis and control present difficulties given JDs prolonged subclinical nature. In order to improve JD control in Ireland, PhD aims included conducting a national survey documenting high-risk husbandry practices employed on Irish farms, thereby targeting areas for improved management. Furthermore as JD has been associated with on-farm economic losses internationally, this research programme aimed to analyse production losses associated with testing JD ELISA positive on Irish dairy farms. A longitudinal study was also conducted to assess if successful JD control can be achieved using gold-standard protocols. Due …

An Observational Study Of Ticks On The 30 Acre Lake Trail At Turnbull National Wildlife Refuge, Robert E. Shadix Jr.
EWU Masters Thesis Collection
Ticks are known carriers of bacterial pathogens that cause diseases in humans and other mammals. Hosts (mice, chipmunks, rabbits, deer, and elk) must be in the tick’s questing range to fulfill the life cycle. The questing range depends on the life stage of the tick, vegetation, and host accessibility. Mammal densities directly affect the number of questing ticks observed in the environment. The 30 Acre Lake Trail site was selected for this study due to the high density of Dermacentor species ticks observed in past studies and the only known site of Rickettsia rickettsii pathogen isolated from ticks on the …

Background: A statistical methodology is available to estimate the proportion of cell types (cellular heterogeneity) in adult whole blood specimens used in epigenome-wide association studies (EWAS). However, there is no methodology to estimate the proportion of cell types in umbilical cord blood (also a heterogeneous tissue) used in EWAS.
Objectives: The objectives of this study were to determine whether differences in DNA methylation (DNAm) patterns in umbilical cord blood are the result of blood cell type proportion changes that typically occur across gestational age and to demonstrate the effect of cell type proportion confounding by comparing preterm infants exposed and …
Longitudinal Assessment Of Blood Brain Barrier Disruption In Primary Hiv Infection And Effect Of Cart Therapy, Elham Rahimy
Longitudinal Assessment Of Blood Brain Barrier Disruption In Primary Hiv Infection And Effect Of Cart Therapy, Elham Rahimy
Yale Medicine Thesis Digital Library
Abnormal blood brain barrier (BBB) permeability has been implicated in the neuropathogenesis of chronic HIV infection. As neurocognitive impairment can persist despite effective combination antiretroviral therapy (cART), it is possible that irreversible central nervous system (CNS) processes are initiated in early infection, before cART is typically initiated. We analyzed the natural history of BBB permeability in primary HIV infection (PHI), and the effects of cART initiated during this period. CSF:Serum albumin quotient (QAlb), a marker of BBB permeability, was measured in longitudinal observational studies of PHI. We analyzed trajectories of QAlb pre- and post-cART using mixed-effects models, and associations between …

Bartonella bacilliformis is a gram negative alpha-proteobacterium native to the Andes of South America. Bartonella causes Carrion’s disease, a potentially life threatening disease transmitted by the sand fly. It has a mortality rate of 88% when untreated and 10% when treated. Diagnostics and control measures for the disease are underdeveloped and no vaccine is available. Recent outbreaks indicate that the range of the pathogen is increasing with nearly 1.7 million people in western South America at risk. Little is known about the epidemiology and pathogenesis of B. bacilliformis. Recent research done by UC San Diego found a paralogous gene …

Given that neurocognitive impairment is a frequent complication of HIV-1 infection in Spanish-speaking adults, the limited number of studies assessing HIV-associated neurocognitive disorders (HAND) in this population raises serious clinical concern. In addition to being appropriately translated, instruments need to be modified, normed, and validated accordingly. The purpose of the current study was to examine the diagnostic utility of the HIV Dementia Scale (HDS) and International HIV Dementia Scale (IHDS) to screen for HAND in Spanish-speaking adults living with HIV infection. Participants were classified as either HAND (N = 47) or No-HAND (N = 53) after completing a comprehensive neuropsychological …

Opioid Abuse In Pregnancy: A Systematic Review, Emily Sebunia, Colleen Fearon
Williams Honors College, Honors Research Projects
Prenatal opioid abuse is a serious public health concern and linked with numerous health consequences for both mothers and children. Despite research describing negative outcomes of drug use during pregnancy, it is still a prevalent problem in the United States, with researchers finding that women are at highest risk for substance abuse during their reproductive years. Opioid abuse during pregnancy is common, including opioids such as hydrocodone, oxycodone, morphine, and heroin. Use of these drugs results in both short and long term side effects for the mother and child. Therefore, the prevalence of this problem should not be underestimated. …

Not all women infected with chlamydiae develop upper genital tract disease, but the reason(s) for this remains undefined. Host genetics and hormonal changes associated with the menstrual cycle are possible explanations for variable infection outcomes. It is also possible that disease severity depends on the virulence of the chlamydial inoculum. It is likely that the inoculum contains multiple genetic variants, differing in virulence. If the virulent variants dominate, then the individual is more likely to develop severe disease. Based on our previous studies, we hypothesized that the relative degree of virulence of a chlamydial population dictates the microRNA (miRNA) expression …

Walden Dissertations and Doctoral Studies
Although evidence is currently available for population-based genetic screening and testing of individuals and their family members for certain hereditary chronic disease conditions (Tier 1), few states have integrated these genomic applications into chronic disease prevention programs. State and territorial chronic disease directors (CDDs) could provide the leadership needed to deliver these applications in more states. The purpose of this study was to determine whether an association exists between current chronic disease genomics funding or specific state genomic activities and the level of knowledge and interests in genomics by these directors. Rogers's diffusion of innovations (DIT) theory was used to …

More U.S. men and women are returning from military service obligations with physical and mental disabilities which complicate their transition to civilian life. Few researchers, however, have examined the post service experiences of blind veterans and whether they are affected by societal expectations of people living with disabilities. The purpose of this qualitative study was to gain knowledge about the experiences of U.S. veterans who suffered vision loss, and the influence of societal expectations on their lives. Hermeneutic phenomenological methodology was used to explore the personal meanings 8 veterans, who lost their vision during active service, attached to their experiences …
Effect Of Therapeutic Alliance Of Clients On Methadone Maintenance Treatment Outcomes, Teresa Lyn Fresquez
Effect Of Therapeutic Alliance Of Clients On Methadone Maintenance Treatment Outcomes, Teresa Lyn Fresquez
Walden Dissertations and Doctoral Studies
Opioid abuse costs affect the majority of the adult population in our society directly or indirectly. The current prevailing medical treatment for opioid addiction is methadone maintenance treatment (MMT). MMT reduces infectious disease spread, illicit drug use, criminal activity, and overdose potential. MMT is only as effective as the length of time a client remains active and compliant with the program. In previous studies, therapeutic alliance (TA) has been shown to positively influence the effectiveness of substance abuse treatment. However, a gap exists in research in regards to the impact of TA on the effectiveness of MMT outcomes. The theoretical …
