Antidepressant-Like Effects Of Amisulpride, Ketamine, And Their Enantiomers On Differential-Reinforcement-Of-Low-Rate (Drl) Operant Responding In Male C57/Bl/6 Mice, Doug Smith

Theses and Dissertations

Major depressive disorder (MDD) is a widespread psychiatric disorder that affects millions of people worldwide and is hypothesized to occur due to impairments in several neurotransmitter systems, including the monoaminergic and glutamatergic neurotransmitter systems. Antidepressant medications targeting multiple monoamine neurotransmitters have been shown to be effective for the treatment of depression. Racemic amisulpride is an atypical antipsychotic that has been used at low doses to treat dysthymia, a mild form of depression, and functions as an antagonist at DA2/3, 5-HT2B, and 5-HT7 receptors. Recent preclinical studies have suggested that the S(+) isomer may be more …

Role Of Translation Initiation In Regulation Of Epithelial Junctions And Cell Motility, Fahda Fawaz Alsharief Jan 2017

Role Of Translation Initiation In Regulation Of Epithelial Junctions And Cell Motility, Fahda Fawaz Alsharief

Theses and Dissertations

The integrity and barrier properties of intestinal epithelium are determined by specialized adhesive structures known as intercellular junctions; composed of adherens junctions (AJs), tight junctions (TJs) and focal adhesions that mediate cell-cell and cell matrix interactions, respectively. These two types of epithelial cell adhesions regulate each other during disruption and restitution of the epithelial barrier. Inflammatory cytokines such as interferon gamma (IFNγ) and tumor necrosis factor alpha (TNFα) are elevated during intestinal inflammation. The most notable effects of IFNγ and TNFα on intestinal epithelial homeostasis involve disruption of apical junctions and attenuation of cell migration. Although molecular mechanisms underlying these …

Our recent discovery of sulfated non-saccharide glycosaminoglycan …

Expanding Monoamine Transporters Pharmacology Using Calcium Channels, Iwona Ruchala Jan 2017

Expanding Monoamine Transporters Pharmacology Using Calcium Channels, Iwona Ruchala

Theses and Dissertations

Research in drug development meets many challenges including lengthy, complex and costly procedures to identify novel pharmacotherapies. In our lab, we developed a method for fast screening of small molecules that interact with monoamine transports – dopamine and serotonin (DAT, SERT). These membrane proteins play important roles in brain neurotransmission responsible for cognition, motion and pleasure. Dysfunction in dopaminergic and serotonergic systems result in neurological disorders such as depression, Attention Deficit Hyperactivity Disorder (ADHD), schizophrenia and addiction.

DAT and SERT are responsible for uptake of dopamine (DA) or serotonin (5HT) into the synapse and they limit neurotransmitter signaling. Drugs that …

Drinking Motives Underlying Internalizing And Externalizing Pathways To Alcohol Misuse In College Students, Jeanne Savage Jan 2017

Drinking Motives Underlying Internalizing And Externalizing Pathways To Alcohol Misuse In College Students, Jeanne Savage

Theses and Dissertations

Alcohol misuse, including heavy episodic use and negative consequences, is a major public health concern and a particular problem among college students. The etiology of alcohol misuse is not well resolved, with multiple and often contradictory factors implicated in its development. Genetic factors influence alcohol misuse but few specific genes have been identified. A potential reason for these challenges is that alcohol misuse is phenotypically and genetically heterogeneous; that is, there are multiple causal pathways underlying its development. Previous typologies have suggested that distinct internalizing and externalizing pathways are involved, with corresponding differences in profiles of personality, temperament, and comorbid …

How Does Alcoholics Anonymous Affect Drinking Outcomes? A Grounded Theory Perspective, Bryan Boyles Jan 2017

How Does Alcoholics Anonymous Affect Drinking Outcomes? A Grounded Theory Perspective, Bryan Boyles

Theses and Dissertations

The mutual aid of Alcoholics Anonymous helps more people with alcohol-related problems than all forms of clinical treatment combined. Yet, efforts to understand AA’s action have been conducted, almost exclusively, from a clinical perspective. Thus far, the literature representing Alcoholics Anonymous from the perspective of its members is very limited. This study uses qualitative content analysis of the AA literature and a grounded theory approach to AA’s fellowship to provide an insider’s perspective of change processes associated with AA involvement. This understanding of the AA literature posits that Alcoholics Anonymous involvement results in life improvement, changes in spirituality, improved well-being, …

Mast Cell Activation By Diverse Stimuli Can Be Suppressed By Steroid Therapy And Targeting The Fyn-Stat5b Cascade, Anuya Paranjape Jan 2017

Mast Cell Activation By Diverse Stimuli Can Be Suppressed By Steroid Therapy And Targeting The Fyn-Stat5b Cascade, Anuya Paranjape

Theses and Dissertations

Mast cells are critical effectors of allergic disease that can be activated by numerous stimuli. We have examined mast cell control by the inflammatory cytokine, IL-33, as well as IgG. In the first study reported here, we found that the synthetic glucocorticoid, dexamethasone, potently and rapidly suppressed IL-33-induced cytokine production from murine bone marrow–derived and peritoneal mast cells, as well as human mast cells. Dexamethasone also antagonized IL-33-mediated enhancement of IgE-induced cytokine production and migration. Although dexamethasone had no effect on IL-33-induced phosphorylation of MAP kinases or NFκB p65 subunit, it antagonized AP-1 and NFκB-mediated transcriptional activity. Finally, intraperitoneal administration …

Contribution Of Asc-Inflammasome To Vascular Endothelial Dysfunction: Role Of Rna Receptor Rig-I, Ashley Pitzer Jan 2017

Contribution Of Asc-Inflammasome To Vascular Endothelial Dysfunction: Role Of Rna Receptor Rig-I, Ashley Pitzer

Theses and Dissertations

Retinoic acid-inducible gene-I (RIG-I) is a putative RNA helicase and recently identified as a cytosolic RNA receptor in mammalian cells. The role of RIG-I in the regulation of vascular function under physiological and pathological conditions is unknown. Recent studies have shown that the inflammasome serves as a crucial initiator of cytokine-mediated inflammation mediating the pathogenesis of cardiovascular disease. The present study tested whether RIG-I activation triggers inflammasome formation and subsequent cytokine-mediated inflammation in the endothelium of mice coronary arteries. Using both genetic and pharmacological interventions of the RIG-I inflammasome, we first characterized whether specific activation of RIG-I via 3pRNA transfection …

A Hierarchical Architectural Framework For Securing Unmanned Aerial Systems, Matthew Leccadito Jan 2017

A Hierarchical Architectural Framework For Securing Unmanned Aerial Systems, Matthew Leccadito

Theses and Dissertations

Unmanned Aerial Systems (UAS) are becoming more widely used in the new era of evolving technology; increasing performance while decreasing size, weight, and cost. A UAS equipped with a Flight Control System (FCS) that can be used to fly semi- or fully-autonomous is a prime example of a Cyber Physical and Safety Critical system. Current Cyber-Physical defenses against malicious attacks are structured around security standards for best practices involving the development of protocols and the digital software implementation. Thus far, few attempts have been made to embed security into the architecture of the system considering security as a holistic problem. …

Theses and Dissertations

This study investigated validity evidence for the Science Literacy Assessment, an instrument designed to assess middle school students’ ability to think scientifically as well as their motivation and beliefs about science (Fives, Huebner, Birnbaum, & Nicolich, 2014). Specifically, three sources of evidence were considered; internal structure, concurrent criterion-related, and predictive criterion-related. Exploratory factor analysis was utilized to examine the underlying factor structure of each of the instrument’s two components, motivation and beliefs related to science and demonstrated scientific literacy. Pearson product-moment correlations were calculated to determine the relationship between scores on the motivation and belief component of the Science Literacy …

Predictors Of Vicarious Traumatization Among Trauma Clinicians And General Mental Health Providers: A Comparison, Shaina Gulin Jan 2017

Predictors Of Vicarious Traumatization Among Trauma Clinicians And General Mental Health Providers: A Comparison, Shaina Gulin

Theses and Dissertations

Vicarious traumatization (VT) describes the gradual, transformative shifts in internal experience that occur as a result of cumulative exposure to clients’ trauma material. VT is thought to develop in the therapist due to empathic engagement with clients, resulting in profound disruptions in frame of reference. Because VT is conceptualized as a condition that develops due to frequent exposure to clients’ traumatic material, a rapidly emerging body of theoretical literature suggests that clinicians can safeguard against VT by maintaining a more balanced workload (i.e., a caseload of clients with a variety of presenting problems) and limiting the number of trauma cases. …

Misophonia: An Investigation Of The Lesser-Known Decreased Sound Tolerance Condition, Shannon Cusack Jan 2017

Misophonia: An Investigation Of The Lesser-Known Decreased Sound Tolerance Condition, Shannon Cusack

Theses and Dissertations

Misophonia is a decreased sound tolerance condition (DST) that is not yet well-established in the literature. However, the existing research on misophonia shows that it is occurring at substantial levels in the population. The majority of the existing literature has focused on the clinical correlates of misophonia. Although the correlates have been investigated, there is no accepted mechanism behind misophonia etiology or maintenance. The present study examined three hypotheses in order to start to identify potential mechanisms behind misophonia: how emotional predilections are related to the emotional response, how obsessive-compulsive disorder (OCD) symptoms and misophonia are related, and the possibility …

Understanding Over-The-Counter Medication Use And Decision-Making Among Community-Dwelling Us Older Adults: A Mixed-Methods Approach, Yoshita Paliwal Jan 2017

Understanding Over-The-Counter Medication Use And Decision-Making Among Community-Dwelling Us Older Adults: A Mixed-Methods Approach, Yoshita Paliwal

Theses and Dissertations

Introduction

Older adults are regular consumers of over-the-counter (OTC) medications. OTC medications are generally considered safe, and convenient to use without requiring a prescription. However, the safety of an OTC medication and the final health outcome depends in part upon consumers’ perceptions, beliefs, and their decision-making about OTC medication use. The main objectives of this study were: 1) to examine the prevalence and characteristics of OTC medication use among community-dwelling US older adults (65 years and older), 2) to explore older adults’ knowledge, attitudes, beliefs and experiences about OTC medications, and 3) to elicit the OTC medication decision-making process in …

Developmental Trajectories Of Alcohol Use And Alcohol Use Disorder, Elizabeth C. Long Jan 2017

Developmental Trajectories Of Alcohol Use And Alcohol Use Disorder, Elizabeth C. Long

Theses and Dissertations

Alcohol use (AU) and alcohol use disorder (AUD) are leading causes of morbidity, premature death, and economic burden. They are also associated with high levels of disability and many other negative outcomes. Twin and family studies have consistently shown that AU and AUD are complex traits influenced by both genetic and environmental factors. Although much has been learned about the genetic and environmental etiology of AU and AUD, significant gaps remain. These include the need for a more comprehensive understanding of the roles of risk and protective factors, and the nature of developmental trajectories underpinning the progression from AU to …

The Role Of Afrocentric Features In Mental Healthcare Utilization And Counselor Preferences In Black College Students, Randl B. Dent Jan 2017

The Role Of Afrocentric Features In Mental Healthcare Utilization And Counselor Preferences In Black College Students, Randl B. Dent

Theses and Dissertations

Though mental health issues are prevalent in Black young adults, they underutilize mental healthcare services. This research examined the role of feature-based discrimination in mental healthcare (under)utilization. Study 1, a secondary analysis of the National Longitudinal Study of Adolescent to Adult Health, provided no evidence supporting a link between skin tone and mental healthcare utilization, when controlling for depression diagnosis. However, when controlling for depression symptoms, there was a trend such that Black young adults with darker, as opposed to lighter, skin tone utilized healthcare less. Study 2, an experimental study with 33 Black college students, showed 73% of the …

Ain’T I Black Enough?: Anxiety Related To The Questioning Of One’S Group Membership, Michelle Stith Jan 2017

Ain’T I Black Enough?: Anxiety Related To The Questioning Of One’S Group Membership, Michelle Stith

Theses and Dissertations

During late adolescence and emerging adulthood, individuals undergo identity formation (Erikson, 1956). During this period, individuals may be insecure about their group memberships (Myers & DeWall, 2015). Subcategorizations have developed among Black people in America resulting from their traumatic history (Blair, Judd, Sadler, & Jenkins, 2002; Levingston & Brewer, 2002). This subcategorization creates in-group/out-group dichotomies. In order to increase one’s self-image, Social Identity Theory (Tajfel & Turner, 1979) poses that individuals attempt to enhance the image of their in-group. This study aimed to determine whether a person feels anxiety due to fear of rejection from the in-group when challenged on …

One In Eight: Deciding To Pursue A College-Going Possible Self In A High-Poverty High School, David B. Naff Jan 2017

One In Eight: Deciding To Pursue A College-Going Possible Self In A High-Poverty High School, David B. Naff

Theses and Dissertations

There is considerable research evidence suggesting that low-income, racial minority students value education and aspire for postsecondary educational attainment (Bloom, 2007; Destin & Oyserman, 2009; Wigfield & Eccles, 2002). However, their performance in school often does not align with those values and ambitions, as these students tend to underachieve in comparison with their higher-income, non-minority peers (Reardon, 2011), with particular gaps found in those attending schools of concentrated poverty (Rowan, 2011). This gap between educational ambition and attainment suggests that the experience of living and going to school in a high-poverty context could be related to the motivational processes driving …

Pathways Linking Clinician Demographics To Mental Health Diagnostic Accuracy: An International Perspective, Julia Brechbiel Jan 2017

Pathways Linking Clinician Demographics To Mental Health Diagnostic Accuracy: An International Perspective, Julia Brechbiel

Theses and Dissertations

Significant research efforts have focused on examining the effect of patient factors on providing diagnoses across clinical settings; however, the influence of clinician demographics have received less attention. This study aimed to understand the impact of nonclinical factors such as clinician characteristics and response time on diagnostic accuracy. The study used data from a WHO field study of the ICD-11 development (n = 1822) that required clinicians to diagnose two case vignettes. Clinicians’ slower response times had a significant positive impact on their rates of diagnostic accuracy. However, there was no evidence that clinicians’ demographic features were directly related to …

Modeling Psychologists’ Openness To Performing Clinical Work With Telepsychology, Bradford S. Pierce Jan 2017

Modeling Psychologists’ Openness To Performing Clinical Work With Telepsychology, Bradford S. Pierce

Theses and Dissertations

This cross-sectional study examined whether the Theory of Reasoned Action (TRA) and Technology Acceptance Model (TAM) apply to psychologists’ openness to using telepsychology, and to identify significant personal or environmental predictors of telepsychology adoption. A total of 1,791 licensed psychologists currently practicing psychotherapy in the United States were recruited to complete a previously developed 21-item pool. Personal and environmental characteristics were also collected. Path models representing the TRA, TAM, and a hybrid of both were tested. Also, a logistic regression was used to identify personal and environmental predictors of current use of telepsychology. Results indicated the TRA and TAM were …

A Biobehavioral Approach To Examining Moral Distress In Critical Care Nurses, Marian Altman Jan 2017

A Biobehavioral Approach To Examining Moral Distress In Critical Care Nurses, Marian Altman

Theses and Dissertations

Moral distress is a complex and challenging problem that may cause negative biopsycohosical and professional outcomes for critical care nurses. The purpose of this work was to explore the relationship between the ethical climate of the work environment and moral distress as experienced by critical care nurses; and to explore relationships among mediators of stress (nurse characteristics e.g. education (BSN, nonBSN), years certified as a critical care nurse, and tolerance of ambiguity) and their relationship with perceived stress, moral distress, health status and salivary alpha amylase. A descriptive correlational cross-sectional design was used for this pilot study of 100 critical …

Influence Of Contextual Factors And Self Efficacy On Self- Management In Parents Of Children With Cystic Fibrosis, Erin B. Booth

Theses and Dissertations

Cystic Fibrosis, a life threatening autosomal recessive genetic disease, is characterized by a defective gene resulting in the production of thick mucus that obstructs the lungs and pancreas. CF requires intensive management performed at the home. An initial pilot study was performed to describe knowledge of CF related diabetes (CFRD) in adults with CF. The findings of this study, which demonstrated that adults with CF lacked sufficient knowledge about CFRD confirmed the need to explore additional factors of self-management guided by a theoretical framework. The second study presented in this dissertation used the Individual and Family Self-Management Theory (IFSMT) to …

Using Restoration-Oriented Coping And The Dual Process Model With Bereaved Undergraduates, Farah J. Aslanzadeh Jan 2017

Using Restoration-Oriented Coping And The Dual Process Model With Bereaved Undergraduates, Farah J. Aslanzadeh

Theses and Dissertations

Approximately 60% of college seniors lost at least one family member or friend since beginning college (Cox, Dean, & Kowalski, 2015). Research reveals that bereaved students are more likely than their nonbereaved peers to struggle with academic problems and attrition (Cousins, Servaty-Seib & Lockman, 2017), highlighting the importance of identifying protective factors for this group of individuals. Researchers have identified restoration-oriented coping as a helpful coping mechanism in other samples (Caserta & Lund, 2007; Caserta, Lund, Utza, & de Vries, 2009). Despite qualitative evidence suggesting bereaved undergraduates often employ restoration-oriented coping, no research has formally assessed the effects of restoration-oriented …

The Justification Of Prejudice Toward Childfree Women, Annalucia Bays Jan 2017

The Justification Of Prejudice Toward Childfree Women, Annalucia Bays

Theses and Dissertations

Previous research suggests that women without children are perceived negatively by others and experience adverse outcomes in various settings. This study investigated psychosocial justifications of prejudice toward childfree women. Participants (N = 891) completed measures of right-wing authoritarianism (RWA), the internal (IMS-S) and external (EMS-S) motivations to respond without sexism, and prejudice toward childfree women (i.e., perceived disadvantages of being childfree, evaluations of childfree women, and perceptions of childfree women’s warmth). Most participants also completed one or more justification measures of hostile sexism (HS), benevolent sexism (BS), gender-specific system justification (GSSJ), and femininity ideology. Results indicated that greater RWA …

Identity Politics, State Standards, And On The Ground Realities: A Critical Policy Analysis Of Teaching/Learning Gender/Sexuality In A Virginia Elementary School, Stefanie Hudson Jan 2017

Identity Politics, State Standards, And On The Ground Realities: A Critical Policy Analysis Of Teaching/Learning Gender/Sexuality In A Virginia Elementary School, Stefanie Hudson

Theses and Dissertations

Even though America has seen an increase in the level of acceptance for people who identify as LGBTQ and/or gender non-conforming, certain aspects within society continue to hinder their rights, especially within public education. Specifically, there are insufficiencies regarding content of and attitudes toward including LGBTQ issues in teaching and leadership training programs. Add to that, the deficits in most Family Life Education (FLE) programs in elementary schools as they lack adequate coverage of developmentally appropriate teaching and learning about gender and sexuality. Taken together, it remains questionable whether the needs of LGBTQ and/or gender non-conforming children can truly be …

The Status Of Snapping Turtles (Chelydra Serpentina) In Virginia: Population Viability, Demography, Regulatory Analysis, And Conservation, Benjamin C. Colteaux

Theses and Dissertations

Snapping turtles (Chelydra serpentina) are being harvested in unprecedented numbers in the United States (US) to meet the needs of international markets. Over three million live snapping turtles from farm and wild caught stock were exported from the US to Asia in 2012-14 alone. In the Commonwealth of Virginia, records indicate that 29,860 snapping turtles were commercially harvested between 2000 and 2015. Size limits are often used to regulate harvest pressure in snapping turtles and other game species. I analyzed the historic harvest of eleven US states to test the efficacy of minimum-size limit regulations at reducing commercial …

"C-Terminal Binding Protein 2" An Emerging Oncogene In Colon And Pancreatic Cancers, Ayesha Chawla Jan 2017

"C-Terminal Binding Protein 2" An Emerging Oncogene In Colon And Pancreatic Cancers, Ayesha Chawla

Theses and Dissertations

The C terminal binding proteins (CtBP) 1 and 2 are a family of transcriptional co-repressors overexpressed in a variety of cancers, and are frequently associated with poor prognosis and chemoresistance. CtBP has also been characterized in cell culture models as drivers of migration/invasion and epithelial-mesenchymal transition. CtBP mediates its transcriptional corepressor activity via its dehydrogenase domain, and inhibition of this domain interferes with CtBP oncogenic functions. The role of CtBP in APC mutant neoplasia remains obscure even though APC is responsible for degradation of both β-catenin and CtBP in suppressing colorectal tumorigenesis. Our prior work demonstrated that CtBP proteins can …
