Cool-season extratropical cyclones near the U.S. East Coast often have significant impacts on the safety, health, environment and economy of this most densely populated region. For example, the “January 2015 blizzard†caused thousands of flights cancellations, travel bans enacted in five states, and two related deaths. Hence it is of vital importance to forecast these high-impact winter storm events as accurately as possible by numerical weather prediction (NWP), including in the medium-range (3-6 days). Ensemble forecasts are appealing to operational forecasters when forecasting such events because they can provide an envelope of likely solutions to serve user communities. However, it …

Science, technology, engineering and mathematics (STEM) education in the United States lags behind that of other industrialized nations. Despite national efforts to enhance the quality of STEM education for students, progress remains elusive. Underperformance is evident in measures of outcomes, participation, and retention. In particular, inequity persists in the attraction and retention of women to STEM fields. Mentoring is heavily cited as a means to improve our national efforts to fortify STEM education. This research explores mentoring styles, gender preferences, and differential impact on outcomes. The results challenge conventional wisdom that women prefer and benefit from a style of mentoring …

An exploration of theatrical history, theory, and technique is a “triumvirate†that can “determine the value of any performance practice for an enlightened theatre†(Gainor 173). My thesis addresses the question of what it means to be a feminist performer, and overall theatre practitioner. Feminist critiques have often attempted to understand a technique best suited for women to practice theatre that is reflective of feminist ideals. My thesis begins by exploring specific examples of female representation in theatre and in society. Signal transduction relies on the proper localization of proteins to properly transfer signals throughout the cell. The cell membrane serves as a vital interface for the proper localization of proteins. Posttranslational modifications cause proteins to become targeted to membranes. One such modification is palmitoylation, the addition of a palmitic acid to a cysteine of a protein. Within the neuron, a large protein complex called the postsynaptic density receives upstream signals at the synapse. Many signaling proteins within the postsynaptic density are known to require palmitoylation for proper trafficking and function. Meiosis is a specialized type of cell division, which produces haploid daughter cells from diploid parental cells. Meiotic recombination is initiated by the introduction of programmed DNA double strand breaks (DSBs), which are catalyzed by an evolutionarily conserved, meiosis-specific endonuclease, Spo11. Meiotic cells monitor the repair of Spo11-induced DSBs via the meiotic recombination checkpoint (MRC), which is activated in response to DSBs made during meiotic prophase. The MRC ensures proper repair of DSBs prior to entry into the first meiotic division. The Mec1 checkpoint kinase is recruited to resected DSBs via the 9-1-1 complex containing Rad17, Mec3 and Ddc1. Yeast sporulation is the process by which yeast cells under limiting nutrient conditions are able to alter their nuclear division cycle from mitotic division to meiotic division, resulting in development of up to 4 spores per diploid cell. Spores are a hardy cell type that can survive until nutrient conditions improve. Under particularly severe conditions, sporulating cells only produce 2 spores, in an arrangement known as a non-sister dyad (NSD). NSDs contain the two spores with the youngest spindle pole bodies (SPBs), which are produced in meiosis II. The SPBs are the central nucleating point of prospore membranes. A scaffolding …

The Drosophila runt gene is the founding member of the RUNX transcription factor family, a group of conserved genes that have vital roles in multiple developmental pathways throughout the animal kingdom. Runt plays important roles in segmentation, as well as in neurogenesis and sex determination in the Drosophila embryo. As a transcription factor, Runt is both an activator and a repressor of its target genes depending on the context. Sloppy-paired (slp1) is a target of runt’s activity as a pair-rule transcription factor in the segmentation pathway that is both activated and repressed by Runt. Tissue necrosis as a consequence of ischemia-reperfusion injury and oxidative damage is a leading cause of permanent disability and death worldwide. The complete mechanism by which cells undergo necrosis upon oxidative stress is not understood. In response to an oxidative insult, wildtype p53 has been implicated as a central regulatory component of the mitochondrial permeability transition (mPT), triggering necrosis. This process is associated with cellular stabilization and translocation of p53 into the mitochondrial matrix. I explore the mechanism by which p53 activates the key mPT regulator cyclophilin D (CypD); how the stability of p53 affects its ability to interact with …

In excess, adipogenesis and adiposity can compromise the quality of the musculoskeletal system, disrupt progenitor cell populations, and increase susceptibility to diabetes. The extent to which muscle and bone tissue are impaired by obesity-related pathologies is not clear. Additionally, while fat, bone, and muscle are distinct tissue systems, the degree to which bone marrow cells contribute to the fat/muscle phenotype in the pre-obese state is yet to be elucidated. Limitations associated with current bone grafting techniques utilizing autologous or allogeneic bone have led to an increase in bone tissue engineering strategies. The ideal scheme of repairing a bone defect is to employ a material that can act as a scaffold and assist regeneration of native tissue in the area. Although biodegradable polymers fulfill many of the requirements for synthetic bone grafts, a major limitation is poor integrity under load bearing conditions prompting investigation of nanoparticle-reinforced biodegradable polymer nanocomposites. Red Blood Cell (RBC) aggregation is an important hemorheological phenomenon especially in microcirculation. In healthy individuals, RBCs are known to aggregate and gravitate toward the faster flow in the center of vessels to increase their throughput for more efficient oxygen delivery. Their aggregation is known to occur during a variety of environmental, pathological, and physiological conditions and is reversible when aggregates are subject to the relatively high shear forces in the circulation. Mycobacterium tuberculosis (Mtb) is the bacterium that causes tuberculosis (TB) in humans. TB is one of the deadliest infectious diseases in the world. Mtb resides in the lipid- enriched environment of the host and metabolizes cholesterol as a carbon source using its own enzymes. Among the total 4000 genes of Mtb, 250 genes correlate to the enzymatic reactions of lipid utilization. The degradation of cholesterol side chain is characterized by three cycles of β-oxidation. Each of the β-oxidation cycles involves hydration of the dehydrogenated cholesterol intermediate substrate. Rv3538 and EchA19 are expected to function as an enoyl-CoA hydratase in the …

In order to bridge the computational gap between DOCK and MD-based methods, a pipeline was designed to combine external information derived from DOCK with replica exchange molecular dynamics using MELD, an algorithm recently developed by our lab. Our goal was to predict the lowest free energy binding pose within the DOCK ensemble, therefore enabling us to refine DOCK results in cases where the correct structure is incorrectly scored. Our method was tested on 8 such ‘scoring failures’ found in the DOCK database SB2012. Three protocols of extracting structural information from DOCK ensembles were designed and tested. With the growing need to find alternative clean energy sources to fossil fuels, research into developing efficient fuel cells and batteries stands at the forefront of this grand effort. In these days, high frequency hedge funds have developed as a new and successful category of hedge funds. Accordingly, risk management is now obliged to keep pace with this market and takes intraday-risk management into consideration. To aim to contribute on answering questions on intraday risk management, the dissertation consists of three parts. In first part, an intraday risk assessment model incorporating long-range dependence and heavy-tailness is suggested. Since the publication of Hamilton’s (1989) seminal work on Markov switching model, a large number of applications have been found in economics and finance. The classical Markov switching models characterize the estimation of parameters in finite state, limited by the pre-specified number of regimes, thus it is restrictive in empirical studies. In this thesis, we develop a stochastic regime switching model, where the model parameters are both categorical and continuous. By assuming conjugate priors and defining stochastic regime switching variables, we derive recursive filtering and smoothing algorithms to estimate the regimes and develop closed-form recursive Bayes estimates of the regression …

This dissertation examines the ways in which Ahmet Hamdi Tanpınar’s novel, Huzur (1949), Mahmoud Darwish’s prose-poem Memory for Forgetfulness: August, Beirut, 1982 (1995), and Virginia Woolf’s Mrs. Dalloway (1925) manifest a cognitive landscape of crisis in discursive socio-cultural contexts that provide the reader with a critical awareness of worldliness. It argues that the crisis serves as an overarching theme and a structural and thematic principle in determining the conceptual frameworks of the critical thresholds that these works address. Accurate prediction of solar energy in short-term and mid-term horizons becomes increasingly important for harvesting solar energy and improving its viability in comparing with fossil fuels. Traffic differentiation---giving better or worse performance to certain classes of Internet traffic---is a well-known but poorly understood traffic management policy. There is active discussion on whether and how ISPs should be allowed to differentiate Internet traffic, but little data about current practices to inform this discussion. This dissertation is an excursion into computer-discovered algorithms and computer-aided algorithm design. In our vision of the not-so-distant future of computing, machines will take the responsibility of designing and implementing most, if not all, machine-specific highly efficient nontrivial algorithms with provable correctness and performance guarantees. Indeed, as the complexity of computing hardware grows and their basic architectures keep changing rapidly, manually redesigning and reimplementing key algorithms for high performance on every new architecture is quickly becoming an impossible task. Over the past decade, web application vulnerabilities have become far more common than vulnerabilities in conventional applications. To mitigate them, we approach the problem from two extremes: one that requires no changes to existing applications but is limited to a few well-defined vulnerability classes, and the second that provides a comprehensive solution but requires a re-thinking of web applications. Our first approach mitigates specific vulnerabilities using policies that do not depend on the application logic, and thus require no developer involvement or effort. In Provision-after-Wait in Healthcare (PaW), a social planner operating on a constrained budget is required to sponsor medical treatments to a population of patients. Specifically, each patient is allocated to any of the k hospitals to receive a single unit of medical treatment. The cost of treatment depends upon the hospital and is paid for by the planner. Associated with every patient is a set of k non-negative numbers which denote the patients’ preference or intrinsic value towards getting treated in each of the k hospitals. In this dissertation, we focus on extending classical spectral shape analysis by incorporating spectral graph wavelets and sparsity-seeking algorithms. Defined with the graph Laplacian eigenbasis, the spectral graph wavelets are localized both in the vertex domain and graph spectral domain, and thus are very effective in describing local geometry. With a rich dictionary of elementary vectors and forcing certain sparsity constraints, a real life signal can often be well approximated by a very sparse coefficient representation. The many successful applications of sparse signal representation in computer vision and image processing inspire us to explore the idea of employing sparse modeling …

Self-control failure is a potent predictor of unhealthful behaviors such as drinking and smoking and a risk factor for addiction. Although research examining self-control failure among addicted individuals is growing, less is known about non-addicted people who fail at controlling health behavior. Social-cognitive models are often used to examine health behaviors among non-addicted individuals. However, health behavior theories tend to be trait-based and underemphasize the role of context. It is important to take into account the influence of situational factors (e.g. | stress, fatigue, and negative mood) when understanding the association of dispositional self-control and health-impairing behaviors. The current research explored potential predictors of romantic relationship formation with a particular target of romantic interest. Possible predictors were derived from a variety of close relationship theories, including Social Penetration Theory, the Investment Model, and the Risk Regulation System. In the main study, single participants who were romantically interested in someone reported on their experiences with the target of interest and were followed-up with for 6 months to determine if they formed a relationship with the target. A factor analysis of the possible predictors yielded three factors – a Closeness/Low Risk factor that included reciprocal liking, mutual self-disclosure, and …

For the past three years, while an MFA student at Stony Brook University, I have pursued an intense study of sculptural work in the medium of ceramic. My goal was to create large-scale ceramic installations. Theses installations comprised of many ceramic pieces I meticulously sculpted and carved to reference the fragility, beauty and finite existence of the human body as it relates to earthly landscapes. My aim has been to imbue an exhibition space with a sense of reference and ambiguity, allowing the viewer of the large scaled installed work to experience the work independent of direction.
