Development, Manufacturing, And Supply Of Msd’S Ebola Vaccine, Jeffrey T. Blue
Development, Manufacturing, And Supply Of Msd’S Ebola Vaccine, Jeffrey T. Blue
Vaccine Technology VI
Ebola was first discovered in 1976 and is a member of the filoviridae family of viruses. There are multiple strains of Ebola and infection can lead to hemorrhagic fever and death. In March 2014, a historic Ebola outbreak occurred in three Western African countries, Guinea, Liberia, and Sierra Leone. Over 28,000 cases were reported and led to more than 11,000 deaths, more than ten times the amount of cases compared to all past outbreaks combined. On August 8th 2014, the World Health Organization declared a Public Health Emergency of International Concern (PHEIC). Merck Sharp & Dohme (MSD) partnering with …

Department of Plant Pathology: Faculty Publications
A new species of Mesocriconema and a unique assemblage of plant-parasitic nematodes was discovered in a heath bald atop Brushy Mountain in Great Smoky Mountains National Park. Mesocriconema ericaceum n. sp., a species with males, superficially resembles M. xenoplax. DNA barcoding with the mitochondrial COI gene provided evidence of the new species as a distinct lineage. SEM revealed significant variability in arrangement of labial submedian lobes, plates, and anterior and posterior annuli. Three other nematodes in the family Criconematidae were characterized from the heath bald. Ogma seymouri, when analyzed by statistical parsimony, established connections with isolates from north-eastern Atlantic …
Improving Global Human Health Through Norovirus Virus-Like Particle Manufacturing, Scot Shepard
Improving Global Human Health Through Norovirus Virus-Like Particle Manufacturing, Scot Shepard
Vaccine Technology VI
Norovirus infection is the most common cause of acute gastroenteritis in the U.S., estimated to afflict 21 million people per year. For scale-up manufacturing of a norovirus vaccine candidate, the baculovirus expression system has been used in 1000 L working volume stirred-tank bioreactors for production of two distinct norovirus virus-like particles (VLP). Downstream processing using methods for enveloped virus inactivation and a series of orthogonal chromatography steps results in norovirus VLP that contain low residual levels of host-cell protein, host cell and baculovirus DNA, and are free of replicating baculovirus. Emphasis has been placed on employing single-use technologies including disposable …

Abiotic stress tolerance in dry bean (Phaseolus vulgaris L.) is complex. Increased population sizes are contributing to finding QTL conditioning stress response but phenotyping has not kept pace with high throughput genotyping for such studies. Our objectives were to determine effectiveness of 20 most tolerant and 20 most susceptible lines representing phenotypic extremes from a RIL population (‘Buster’ x 'Roza’ [BR]) to facilitate examination of 19 traits for relevance to stress response and to validate existing QTL conditioning stress response. Using phenotypic extremes tested across multiple trials, eight of the 19 traits were clearly associated with drought stress. Pod …

Nfat5/Stat3 Interaction Mediates Synergism Of High Salt With Il-17 Towards Induction Of Vegf-A Expression In Breast Cancer Cells, Suneetha Amara, Dalal Alotaibi, Venkataswarup Tiriveedhi

Chronic inflammation has been considered an important player in cancer proliferation and progression. High salt (sodium chloride) levels have been considered a potent inducer of chronic inflammation. In the present study, the synergistic role of high salt with interleukin (IL)‑17 towards induction of the inflammatory and angiogenic stress factor vascular endothelial growth factor (VEGF)‑A was investigated. Stimulation of MCF-7 breast cancer cells with high salt (0.2 M NaCl) and sub‑minimal IL‑17 (1 ng/ml) enhanced the expression of VEGF-A (2.9 and 2.6-fold, respectively, P<0.05) compared with untreated cells. Furthermore, co‑treatment with both high salt and sub‑minimal IL‑17 led to a 5.9‑fold increase in VEGF‑A expression (P<0.01), thus suggesting a synergistic role of these factors. VEGF‑A promoter analysis and specific small interfering RNA knock‑down of transcription factors revealed that high salt induced VEGF‑A expression through nuclear factor of activated T‑cells (NFAT)5, while IL‑17 induced VEGF‑A expression via signal transducer and activator of transcription (STAT)3 signaling mechanisms. Treatment of normal human aortic endothelial cells with the supernatant of activated MCF‑7 cells enhanced cell migration and induced expression of migration‑specific factors, including vascular cell adhesion protein, β1 integrin and cluster of differentiation 31. These data suggest that high salt levels synergize with pro‑inflammatory IL‑17 to potentially induce cancer progression and metastasis through VEGF‑A expression. Therefore, low‑salt diet, anti‑NFAT5 and anti‑STAT3 therapies may provide novel avenues for enhanced efficiency of the current cancer therapy.

A Generalized Computer Vision Approach To Mapping Crop Fields In Heterogeneous Agricultural Landscapes, Stephanie R. Debats, Dee Luo, Lyndon Estes, Thomas J. Fuchs, Kelly K. Caylor
Geography
Smallholder farms dominate in many parts of the world, particularly Sub-Saharan Africa. These systems are characterized by small, heterogeneous, and often indistinct field patterns, requiring a specialized methodology to map agricultural land cover. Using a variety of sites in South Africa, we present a new approach to mapping agricultural fields, based on efficient extraction of a vast set of simple, highly correlated, and interdependent features, followed by a random forest classifier. We achieved similar high performance across agricultural types, including the spectrally indistinct smallholder fields as well as the more easily distinguishable commercial fields, and demonstrated the ability to generalize …

Temperature-Dependent Pathogenicity Of Grouper Iridovirus Of Taiwan (Tgiv), Hsin-I Liu, Ying-Chu Lin, Pinwen Peter Chiou, Hsin-Yiu Chou
Journal of Marine Science and Technology–Taiwan
Grouper iridovirus of Taiwan (TGIV) is one of the most devastating infectious pathogens of grouper fry in Taiwan. As environmental temperature often plays an important role in the outbreak of diseases, we assayed the impact of different temperatures (18, 25 and 32℃) on TGIV infection both in vitro and in vivo. Cytopathic effect (CPE) characterized as cell rounding and lysis were observed in TGIV-infected grouper SB (swim bladder) cells at 25℃ and 32℃, but was absent at 18℃. With temperature rising to 32℃, the presence of pycnotic nuclei and chromatin margination became prominent in the infected cells, indicating an apoptotic …

Journal of Marine Science and Technology–Taiwan
For facilitating the release of bionutrients from noni, 6 h of hydrolysis with 100 U/mL cellulase at 50℃ and an additional 48 h of fermentation with Pediococcus pentosaceus BCRC 14053 at 37℃ were performed. Increases in the extraction yields (87.8-607.8 mg/g) and the total phenolic content (2.02-7.45 mg/g) of various extracts were obtained, suggesting that bionutrients were released from noni during hydrolysis or fermentation. In hydrolyzed and/or additionally fermented samples, significant decreases in the half maximal inhibitory concentration (IC50) against α-amylase (10.28-1.61 mg/mL) and α-glucosidase (46.47-3.73 mg/mL) were observed. Gas chromatography–mass spectrometry results revealed that the major components with these …

© 2016 The Author 2016. Published by Oxford University Press. Motivation: A growing number of studies have explored the process of pre-implantation embryonic development of multiple mammalian species. However, the conservation and variation among different species in their developmental programming are poorly defined due to the lack of effective computational methods for detecting co-regularized genes that are conserved across species. The most sophisticated method to date for identifying conserved co-regulated genes is a two-step approach. This approach first identifies gene clusters for each species by a cluster analysis of gene expression data, and subsequently computes the overlaps of clusters identified …

Transgenic crops producing Bacillus thuringiensis (Bt) proteins have become a primary tool in pest management. Due to the intensive use of Bt crops, resistance of the fall armyworm, Spodoptera frugiperda, to Cry1F maize has occurred in Puerto Rico, Brazil, and some areas of the southeastern U.S. The sustainability of Bt crops faces a great challenge because the Cry1F-maize resistant S. frugiperda may also infest other Bt crops in multiple cropping ecosystems. Here we examined the survival and plant injury of a S. frugiperda population selected with Cry1F maize on three single-gene and five pyramided Bt cotton products. Larvae of Cry1F-susceptible …

The ENaC/degenerin ion channel superfamily includes the amiloride-sensitive epithelial sodium channel (ENaC) and acid sensitive ionic channel (ASIC). ENaC is a multimeric ion channel formed by heteromultimeric membrane glycoproteins, which participate in a multitude of biological processes by mediating the transport of sodium (Na+) across epithelial tissues such as the kidney, lungs, bladder, and gut. Aberrant ENaC functions contribute to several human disease states including pseudohypoaldosteronism, Liddle syndrome, cystic fibrosis, and salt-sensitive hypertension. Increasing evidence suggests that ion channels not only regulate ion homeostasis and electric signaling in excitable cells but also play important roles in cancer cell behaviors such …
