Transcriptional And Post-Transcriptional Regulation Of Nrf2 In The Heart By The Deubiquitinase Cyld, Bryan J. Mathis Jun 2016

Transcriptional And Post-Transcriptional Regulation Of Nrf2 In The Heart By The Deubiquitinase Cyld, Bryan J. Mathis

Theses and Dissertations

The cylindromatosis (CYLD) is a K63-linked deubiquitinase (DUB) that has been linked to the regulation of multiple physiological or pathological processes, such as neural development, inflammation and fibrosis. However, a novel paradigm for CYLD has been recently postulated; namely that of CYLD as a mediator of cardiac disease. Nuclear factor, erythroid-2 related factor 2 (Nrf2), a master antioxidant transcription factor, has been shown to suppress cardiac pathological remodeling and dysfunction via downregulation of reactive oxygen species formation (ROS). It is normally regulated by Kelch-like ECH associated protein 1 (Keap1). However, the regulatory link between CYLD and Nrf2 in the diseased …

The Undiagnosed Patient And The Diagnostic Odyssey: Current Genetic Counseling Practices And Perspectives, Amelia Cordell Wardyn Jun 2016

The Undiagnosed Patient And The Diagnostic Odyssey: Current Genetic Counseling Practices And Perspectives, Amelia Cordell Wardyn

Theses and Dissertations

Patients seen in genetics clinics often endure a diagnostic odyssey in their search for answers for their medical symptoms. This time is not only challenging for patients and their families, but also for the genetic counselors who are trying to help the patients. Previous research has shown that parents of children with undiagnosed medical disorders have specific goals and reasons for wanting to find a diagnosis, and there are many difficulties faced by these parents. Genetic counselors often serve as a prominent figure during the diagnostic odyssey, but little known research has assessed the current practices of and impact that …

An Investigation Of The Succination Of The Selenoproteins Thioredoxin Reductase And Glutathione Peroxidase, Scott Lanci Jun 2016

An Investigation Of The Succination Of The Selenoproteins Thioredoxin Reductase And Glutathione Peroxidase, Scott Lanci

Theses and Dissertations

We have identified the irreversible protein modification S-(2-succino)cysteine, a product of the Kreb’s cycle intermediate fumarate reacting with the thiol group of cysteine (Cys) residues, also termed succination. Succination is increased in adipose tissue of diabetic mice in vivo and in adipocytes matured in high glucose medium (25 mM) in vitro. Selenocysteine (Sec) is a less common but highly conserved amino acid that has major functions in redox chemistry. Sec possesses a lower pKa (~5.4) and is much more reactive in comparison to Cys. This project aimed to determine if succination occurred on Sec residues in two selenoproteins that play …

Novel Methods For Analyzing Longitudinal Data With Measurement Error In The Time Variable, Caroline Munindi Mulatya Jun 2016

Novel Methods For Analyzing Longitudinal Data With Measurement Error In The Time Variable, Caroline Munindi Mulatya

Theses and Dissertations

In some longitudinal studies, the observed time points are often confounded with measurement error due to the sampling conditions, resulting into data with measurement error in the time variable. This type of data occurs mainly in observational studies when the onset of a longitudinal process is unknown or in clinical trials when individual visits do not take place as specified by the study protocol, but are often rounded to coincide with the study protocol. Methodological and inferential implications of error in time varying covariates for both linear and nonlinear models have been studied widely. In this dissertation, we shift attention …

Assessing The Effects Of The Mud Snail, Ilyanassa Obsoleta, On The Benthic Microalgal Community In A Pristine Saltmarsh, Miranda Gore

Theses and Dissertations

Saltmarshes are among the most productive ecosystems globally. At North Inlet estuary, South Carolina, about one third of the primary production comes from benthic microalgae. During the tidal cycle, mobile microalgae vertically migrate through the upper 3 mm of sediment. At low tide, algae are vulnerable to a variety of grazers, including the mud snail, Ilyanassa obsoleta, which is abundant in tidal creeks. Many species of intertidal snails have been shown to significantly alter the community structure and density of microalgae within the sediment. The purpose of this study was to determine how I. obsoleta affects the benthic microalgae community …

Conversations About Sexuality On A Public University Campus: Exploring Perspectives From Campus Ministry Leaders And Students, Charis Rebecca Davidson

Theses and Dissertations

Since the 1800s, religious organizations have been a presence on public universities. However, there is limited research on the influence these organizations may have on student health communication and behaviors. Prior research on campus ministries has focused primarily on the culture of individual organizations. In this study, researchers explored the goals and experiences of USC leaders related to communication around sexuality with their students, as well as student perceptions of these messages, and how students incorporate religious messages about sexuality into their lives. The present study had four aims:

1. Assess whether students participating in campus ministries at USC differ …

The Influence Of Motivational Salience On Attention Selection: An Erp Investigation, Constanza De Dios Jun 2016

The Influence Of Motivational Salience On Attention Selection: An Erp Investigation, Constanza De Dios

USF Tampa Graduate Theses and Dissertations

The current study used event-related potentials (ERPs) to investigate how motivational salience in the form of expectation violation influences spatial attention. The medial frontal negativity (MFN) ERP indexes expected value, being negative to unexpected punishments and positive to unexpected rewards. The P1 and N1 ERPs index spatial attention, being larger to stimuli in attended locations. This design attached motivational value to locations by making one visual hemifield economically rewarding (greater probability of a rewarding outcome) and the other punishing (greater probability of a punishing outcome). Keypresses to a dot probe following a reward-signifying stimulus were awarded money if correct, and …

A New Method For Ecoacoustics? Toward The Extraction And Evaluation Of Ecologically-Meaningful Soundscape Components Using Sparse Coding Methods, Alice Eldridge, Michael Casey, Paola Moscoso, Mika Peck

Dartmouth Scholarship

Passive acoustic monitoring is emerging as a promising non-invasive proxy for ecological complexity with potential as a tool for remote assessment and monitoring (Sueur & Farina, 2015). Rather than attempting to recognise species-specific calls, either manually or automatically, there is a growing interest in evaluating the global acoustic environment. Positioned within the conceptual framework of ecoacoustics, a growing number of indices have been proposed which aim to capture community- level dynamics by (e.g., Pieretti, Farina & Morri, 2011; Farina, 2014; Sueur et al.,2008b) by providing statistical summaries of the frequency or time domain signal. Although promising, the ecological relevance and …

Additionally, severe RSV infection early in life is associated with an increased risk of wheeze and other airway disorders later in life. Despite this, there is currently no vaccine or economically reasonable prophylactic regimen to prevent infection. While disease is typically more severe in infancy RSV can infect throughout the lifespan repeatedly as the …

Assessment Of Prolonged Occupational Exposure To Heat Stress, Ximena Garzon-Villalba Garzon-Villalba Jun 2016

Assessment Of Prolonged Occupational Exposure To Heat Stress, Ximena Garzon-Villalba Garzon-Villalba

USF Tampa Graduate Theses and Dissertations

Heat stress is a recognized occupational hazard present in many work environments. Its effects increase with increasing environmental heat loads. There is good evidence that exertional heat illness is associated with ambient thermal conditions in outdoor environments. Further, there is reason to believe that risk of acute injury may also increase with the ambient environment. For these reasons, the assessment of heat stress, which can be done through the characterization of the wet bulb globe temperature (WBGT), is designed to limit exposures to those that could be sustained for an 8-h day. The ACGIH Threshold Limit Value (TLV) for heat …

Applications Of Molecular Modelling And Structure Based Drug Design In Drug Discovery, Sreya Mukherjee Jun 2016

Applications Of Molecular Modelling And Structure Based Drug Design In Drug Discovery, Sreya Mukherjee

USF Tampa Graduate Theses and Dissertations

Calcium ions have important roles in cellular processes including intracellular signaling, protein folding, enzyme activation and initiation of programmed cell death. Cells maintain low levels of calcium in their cytosol in order to regulate these processes. When activation of calcium-dependent processes is needed, cells can release calcium stored in the endoplasmic reticulum (ER) into the cytosol to initiate the processes. This can also initiate formation of plasma membrane channels that allow entry of additional calcium from the extracellular milieu. The change in calcium levels is referred to as calcium flux. A key protein involved in initiation of calcium flux is …

Cell Cycle Arrest By Tgfß1 Is Dependent On The Inhibition Of Cmg Helicase Assembly And Activation, Brook Samuel Nepon-Sixt

USF Tampa Graduate Theses and Dissertations

Tumorigenesis is a multifaceted set of events consisting of the deregulation of several cell-autonomous and tissue microenvironmental processes that ultimately leads to the acquisition of malignant disease. Transforming growth factor beta (TGFß) and its family members are regulatory cytokines that function to ensure proper organismal development and the maintenance of homeostasis by controlling cellular differentiation, proliferation, adhesion, and survival, as well as by modulating components of the cellular microenvironment and immune system. The pleiotropic control by TGFß of these cell intrinsic and extrinsic factors is intimately linked to the prevention of tumor formation, the specifics of which are dependent on …
