The developmental origins of health and disease (DOHaD) theory and life course theory (LCT) are emerging fields of research that have significant implications for the public health and health promotion professions. Using a DOHaD/LCT perspective, social determinants of health (SDH) take on new critical meaning by which health promotion professionals can implement DOHaD/LCT guided interventions, including recommended policies. Through these interventions, public health could further address the sources of worldwide chronic disease epidemics and reduce such disease rates substantially if related policy, programs, and interdisciplinary and multi-sector collaboration are emphasized. We study a hydrodynamic limit for a system of N diffusions moving in an open domain D ⊆ Rd undergoing branching when one particle reaches a certain subset of the boundary. The particle at the boundary and another random neighbor are eliminated and replaced with two new particles created instantaneously at a random point with distribution γ(dx) in D. This thesis proves the d = 1 case with D = (0,1), γ(dx) = δc(dx), c ∈ (0,1) while the general case is done in an upcoming paper. The Physiological Effects Of The Guanylin Peptides In The Intestine Of The Gulf Toadfish (Opsanus Beta), Ilan M. Ruhr

The findings in the present dissertation describe the regulatory role the guanylin peptides play in seawater teleost intestinal physiology through the use of the model organism, the Gulf toadfish (Opsanus beta). For the majority of the findings, renoguanylin (a guanylin peptide exclusive to teleost fish) is used as an experimental tool and elicits responses in the posterior intestine and rectum of the Gulf toadfish, but does not affect the anterior intestine. Both the posterior intestine and rectum display secretory short-circuit currents, reversed transepithelial potentials, and increased conductance in response to renoguanylin stimulation. These changes are driven primarily by chloride secretion …

The adoption of genetically engineered (GE) crops has created a demand for practical methods to mitigate pollen dispersal and gene flow. The goal of this project was to measure the ability of a narrow forest windbreak to reduce downwind pollen fluxes from switchgrass (Panicum virgatum L.), a North American grass and model biofuels feedstock. Switchgrass fields were established in two identical plots where one had a forest windbreak and the other was in an open (control) site. Switchgrass reproduction, pollen dispersal, wind speed, and wind direction were measured over two years. Daily release of switchgrass pollen peaked at 11:00-13:30 …

The effect of grazers on the diversity, distribution, and composition of their principal food source has rarely been described for the high intertidal zone of rocky shores, a model system for studying the potential effects of climate change. Along rocky, wave-swept shores in central California, the microphytobenthos (MPB) supports diverse assemblages of limpets and littorine snails, which, at current benign temperatures, could potentially partition food resources in a complementary fashion, thereby enhancing secondary productivity. Two limpet species in particular, Lottia scabra and L. austrodigitalis, may partition components of the MPB, and are likely to affect the composition of the …

Some introduced species spread rapidly beyond their native range and into novel habitats mediated by a high degree of phenotypic plasticity and/or rapid evolutionary responses. In this context, clonality has been described as a significant factor contributing to invasiveness. We studied the abiotic environment and the responses of different tussock architecture traits of the invasive cordgrass Spartina densiflora Brongn. (Poaceae). Biofouling of experimental cages and other field apparatuses can be problematic for scientists and has traditionally been addressed using frequent manual removal (e.g., scraping, scrubbing). Recent environmental restrictions and legislative changes have driven the development of less hazardous antifouling products, making antifouling paint a potential alternative option to manual removal. Consequently, the viability of using these newly developed products as a replacement for the manual cleaning of exclusion cages was experimentally investigated. Understanding the relationship between species distributions and their underlying mechanisms is a major question in ecology and evolutionary biology. The mechanisms involved and the degree to which they affect distributions vary among and within species. Examining the diversity of these mechanisms through behavioral, physiological, and integrative approaches can further our knowledge of the evolution of such traits and the maintenance of biodiversity and geographic distributions. Many of these traits may be conserved after speciation, therefore examination should be thorough, involve many species, and incorporate a phylogenetic approach. Single-stranded DNA binding (SSB) proteins coordinate DNA replication, repair, and recombination and are critical for maintaining genomic integrity. SSB binds to single-stranded DNA (ssDNA) rapidly and with very high affinity making it a useful molecular tool to detect free ssDNA in solution. We have labeled SSB from Plasmodium falciparum (Pf-SSB) with the MDCC (7-diethylamino-3-((((2-maleimidyl)ethyl)amino)-carbonyl)coumarin) fluorophore which yields a four-fold increase in fluorescence upon binding to ssDNA. Pf-SSBMDCC binding to DNA is unaffected by NaCl or Mg2+ concentration and does not display salt-dependent changes in DNA binding modes or cooperative binding on long DNA substrates. These …

Boolean networks have been widely used as models for gene regulatory networks, signal transduction networks, or neural networks, among many others. One of the main difficulties in analyzing the dynamics of a Boolean network and its sensitivity to perturbations or mutations is the fact that it grows exponentially with the number of nodes. Therefore, various approaches for simplifying the computations and reducing the network to a subset of relevant nodes have been proposed in the past few years. We consider a recently introduced method for reducing a Boolean network to its most determinative nodes that yield the highest information gain. …

Plasminogen activator inhibitor type-1 (PAI-1) is a member of the serine protease inhibitor (serpin) family. PAI-1 is involved in the regulation of fibrinolysis, which is the breakdown of blood clots. PAI-1 inhibits serine proteases tissue-type plasminogen activator (tPA) and urokinase-type plasminogen activator (uPA), which are key factors in fibrinolysis. Excess levels of PAI-1 have also been shown to increase the risk of diabetes, stroke, and atherosclerosis, as well as tumor development. Previous attempts at synthesizing a practical drug for the regulation of PAI-1 have not been successful; therefore, it has been the purpose of this study to further advance progress …

We have developed a Lewis-acid catalyzed, diastereoselective aza-Cope rearrangement— Mannich cyclization (ACM) to form acyl pyrrolidines from conformationally mobile substrates. Earlier studies from our lab have shown that substituted acyl pyrrolidines can be synthesized with diastereoselectivity of 50:1 trans to cis, and that unsubstituted pyrrolidines can be synthesized with 13:1 enantioselectivity. This study attempted to observe the stereochemical outcomes of the ACM for disubstituted pyrrolidine systems using oxazolidine starting materials. A proof of concept dimethyl-substituted oxazolidine was subjected to ACM conditions, producing the pyrrolidine with good yields. Widespread use of oxidized graphene nanomaterials in industry, medicine, and consumer products raises concern about potential adverse impacts on human health. The p53 tumor suppressor protein is crucial to maintaining cellular and genetic stability to prevent carcinogenesis. Here, we show that oxygen functionalized graphene (f-G) absorption and p53 functional status correlate with cytotoxicity and genotoxicity in human B-lymphoblastoid cells. Trends in f G absorption by were dose-dependent. Cells with functional p53 exposed to f-G arrested in G0/G1 phase of the cell cycle, suppressed f-G induced reactive oxygen species (ROS), and had elevated apoptosis.
