In this research, atomic force microscope tip-enhanced laser ablation mass spectrometry (AFM TELA-MS), an ambient sub-micrometer scale sampling method for offline MS was developed. AFM TELA was used to transfer molecules from thin films to a suspended silver wire for off-line mass spectrometry using laser desorption ionization (LDI) and matrix-assisted laser desorption ionization (MALDI). An AFM with a 30 nm radius gold-coated silicon tip was used to image the sample and to hold the tip 15 nm from the surface for material removal using a pulsed Nd:YAG laser, which provides output at wavelengths of 532 nm in the visible, 1064 …

The work of this dissertation investigates the chemical processes of novel materials through quantum mechanical calculations and molecular dynamics simulations. The first section of this work focuses on probing copper based metal organic frameworks (MOFs) for their ability to catalytically release nitric oxide from s-nitrosothiol (RSNO) precursors in a slow, controlled and sustained manner and compares these findings to a free copper ion/ethanol solution. The abundance of liquid water found beneath the Antarctic Ice Sheet (AIS) is thought to provide a suitable habitat for microbial life. However, the thick ice sheet has posed a significant obstacle to accessing this isolated system and as a consequence, little data exists to infer ecosystem processes occurring beneath ice sheets. The effects of nitrogen (N) deposition have been studied in many ecosystems, and one general pattern of response that has emerged from these studies is a decline in species richness. Globally, anthropogenic deposition of N has more than doubled the ambient input of N in terrestrial ecosystems, and the rate at which humans produce reactive N continues to increase annually. Hemlock woolly adelgid (HWA) is an exotic insect pest of eastern hemlock. The entomopathogen Lecanicillium muscarium, including the commercially available strain MycotalRTM, is a potential candidate for fungal biocontrol. There are many factors to consider when using a fungal biocontrol such as ecology and genetic variation of candidate strains and interactions with other fungi and life stages of the target insect pest. Use of pesticide has become part of modern day agricultural practice. Some pesticides can remain in the environment for decades and contaminate surface water that is used for irrigation of produce. Two studies were conducted- first to examine possible pesticide residue in surface water and some fruits, vegetables and cereals in Louisiana; and second was to alleviate possible pesticide residues in the water using zeolite filtration. Molecularly Imprinted Polymers (MIPs) encompass a wide range of applications by changing the different components, e.g. the template, crosslinker or functional monomers. Of interest among these different applications are separations and sensors. Separations by MIPs traditionally use a chiral pure template but in some cases that chiral pure template may not be available for imprinting. Using chiral (N-α-bismethacyloyl-L-alanine) and achiral (N,O-bisacrylamide ethanolamine) crosslinkers we investigated imprinting of scalemic and racemic template mixtures of Boc-tyrosine enantiomers. Prickly sida (Sida spinosa L.) is widely distributed in the southern U.S. and is a troublesome weed in many agronomic crops. In northeastern Louisiana on silty clay loam and clay soils, prickly sida seedling emergence began in early March when soil temperature reached 13.8 C and ceased in mid-October. Seasonal peaks in emergence were associated with rainfall events and variability in prickly sida emergence was observed between soil types and years. Total emergence was as high as 3,000 prickly sida plants m-2. In shade studies, prickly sida was able to emerge and persist under a heavy shade environment and to …

Coastal Louisiana is annually threatened by coastal storms. Population grown, wetland loss, and potentially increasing storm frequency are likely to increase coastal vulnerability to these events. Increasingly, coastal management entities are managing land resources to reduce the economic impact of natural disasters with the use of natural infrastructure. This is true in Louisiana where the Louisiana Coastal Master Plan allocates billions of dollars to coastal restoration projects, many of which are intended to mitigate economic damages from tropical storms and hurricanes. The purpose of the research in this dissertation was to elucidate the intrinsic properties of how nanoparticles are different from bulk materials. This was done by mechanical and electronic studies of the properties of designed nanoparticles using advanced modes of atomic force microscopy. Information relating to the work functions, contact potential difference, Young’s Moduli, elasticity, and viscoelasticity can be investigated using state-of-the-art atomic force microscope (AFM) experiments. Helicoidea is a diverse group of land snails with a global distribution. While much is known regarding the relationships of helicoid taxa, comparatively little is known about the evolution of the mitochondrial genome in the superfamily. We sequenced two complete mitochondrial genomes from Praticolella mexicana Perez, 2011 representing the first such data from the helicoid family Polygyridae, and used them in an evolutionary analysis of mitogenomic gene order. We found the mitochondrial genome of P. mexicana to be 14,008 bp in size, possessing the typical 37 metazoan genes. Multiple alternate stop codons are used, as are incomplete stop codons. We have previously reported the use of one-bead-one-compound (OBOC) combinatorial technology to develop a disulfide cyclic, Arg-Gly-Asp containing octapeptide LXW7 (cGRGDdvc), that targets αvβ3 integrin with high affinity and specificity (Mol Cancer Ther, 9:2714–23, 2010). αvβ3 integrin is known to be over-expressed in many cancers and in tumor vasculature, and it has been established as a cancer therapeutic target. To further optimize LXW7, we have performed systematic structure activity relationship (SAR) studies. Cultural diversity features prominently in Canadian social and political life. Canadian social psychologists, for their part, have led a rich tradition of research on intercultural relations. We identified the undergraduate social psychology textbooks used to introduce students of psychology in Canada to the field and examined the extent that culture is incorporated into the teaching of introductory social psychology. Through a content analysis of Canadian-published introductory social psychology texts, we found that these textbooks are inclusive of cultural diversity, although not all textbooks covered culture with the same depth. The objective of this dissertation is to understand the functions of Escherichia coli YqjA and YghB, which are members of the conserved DedA/Tvp38 membrane protein family. YqjA and YghB are inner membrane (IM) proteins with multiple predicted membrane–spanning domain, sharing 62% amino acid identity and partly overlapping functions. Simultaneous in-frame deletions of these two genes in a strain named BC202 results in various phenotypes including cell division defects, temperature sensitivity, and sensitivity to drugs and alkaline pH. This quantitative, non-experimental, correlational study examined the relationship between secondary school staff perceptions of their school’s effectiveness and the change in student literacy over a one year period. The staff perception data was elicited through an anonymous, electronically administered survey, the SEDL’s School Professional Staff as Learning Community Questionnaire (SPSLCQ) (Hord, 1996). VGSCs participate in the acidification of the perimembrane, activating the cysteine proteases, cathepsin B and S, thereby triggering the proteolysis of the extracellular matrix and the basement membrane, thereby promoting cell invasion. Evidence suggests that serum 25(OH)D (serum vitamin D), generally low in African-Americans, may play a role in glucose control. Selective serotonin reuptake inhibitors (SSRIs) have been a mainstay pharmacological treatment for women experiencing depression during pregnancy and postpartum for nearly three decades. Recently, though, growing evidence indicates that early-life SSRI exposure triggers long-lasting behavioral abnormalities. Clinically, children exposed to SSRIs in early life exhibit increased internalizing behavior, reduced social behavior, and increased risk for depression in adolescence. Similarly, in rodents, perinatal SSRI exposure leads to increased traits of anxiety- and depression-like behavior. This dissertation was a qualitative study of school social workers who worked with special needs adolescent youth with emotional and behavioral disabilities in a public high school. This study centered on the experiences of 10 social workers in a public high school in New Jersey. This study investigates whether or not parental acculturation is associated with depression levels among youth in US-based immigrant families. Previous research has shown that even though individual acculturation is negatively correlated with adolescents’ good mental health, parental acculturation is associated with higher levels of mental health among youth. This study elaborates on these prior findings by using a large national sample of immigrant children ages 13-14 and considering the mediating role of social ties and social support.
