Purpose:
The tumor growth kinetics of the human LoVo colorectal xenograft model was assessed in response to vandetanib, an orally available receptor tyrosine kinase inhibitor, radiotherapy (RT), or irinotecan (CPT-11), as single therapies and in combination.
Methods and Materials:
LoVo cells were injected subcutaneously into the right hind limb (5x106 cells in 100μL phosphate-buffered saline) of athymic NCR NUM mice and tumors were grown to a volume of 200–300mm3 before treatment. Vandetanib was administered at 50 mg/kg daily orally for 14 days starting on Day 1. RT was given as three fractions (3x3 Gy) on Days 1, 2, and 3. …

Inflammatory changes are a major component of the normal tissue response to ionizing radiation, and increased nuclear factor κB (NF-κB) activity is an important mediator of inflammatory responses. Here, we used zebrafish embryos to assess the capacity of two different classes of pharmacologic agents known to target NF-κB to modify radiation toxicity in the vertebrate organism. These were proteasome inhibitors, including lactacystin, MG132, and PS-341 (Bortezomib/VELCADE), and direct inhibitors of NF-κB activity, including ethyl pyruvate (EP) and the synthetic triterpenoid CDDO-TFEA (RTA401), among others. Peridynamic Modeling Of Ruptures In Biomembranes, Michael Taylor, Irep Gözen, Samir Patel, Aldo Jesorka, Katia Bertoldi

We simulate the formation of spontaneous ruptures in supported phospholipid double bilayer membranes, using peridynamic modeling. Experiments performed on spreading double bilayers typically show two distinct kinds of ruptures, floral and fractal, which form spontaneously in the distal (upper) bilayer at late stages of double bilayer formation on high energy substrates. It is, however, currently unresolved which factors govern the occurrence of either rupture type. Variations in the distance between the two bilayers, and the occurrence of interconnections (“pinning sites”) are suspected of contributing to the process. Confidence Intervals For Heritability Via Haseman-Elston Regression, Tamar Sofer

Heritability is the proportion of phenotypic variance in a population that is attributable to individual genotypes. Heritability is considered an important measure in both evolutionary biology and in medicine, and is routinely estimated and reported in genetic epidemiology studies. In population-based genome-wide association studies (GWAS), mixed models are used to estimate variance components, from which a heritability estimate is obtained. The estimated heritability is the proportion of the model's total variance that is due to the genetic relatedness matrix (kinship measured from genotypes). Current practice is to use bootstrapping, which is slow, or normal asymptotic approximation to estimate the precision …

Cervical and high thoracic spinal cord injury (SCI) drastically impairs autonomic nervous system function. Individuals with SCI at thoracic spinal-level 5 (T5) or higher often present cardiovascular disorders that include resting systemic arterial hypotension. Gastrointestinal (GI) tissues are critically dependent upon adequate blood flow and even brief periods of visceral hypoxia triggers GI dysmotility. The aim of this study was to test the hypothesis that T3-SCI induces visceral hypoperfusion, diminished postprandial vascular reflexes and concomitant visceral inflammation. Previous studies have shown that the anterior thalamic nuclei (ATN) contain a large population of head direction cells, which fire as a function of an animal’s directional orientation in an environment, thereby providing a compass-like representation guiding navigation. Recent work has suggested that directional orientation information stemming from the ATN is critical for the generation of hippocampal and parahippocampal spatial representations, and may contribute to the establishment of unique spatial representations in radially oriented tasks such as the radial arm maze. Structural changes induced by radiation damage in X-ray crystallography hinder the ability to understand the structure/function relationship in chemical reactions. Serial femtosecond crystallography overcomes this problem by exposing the sample to very short and intense laser pulse leading to measurement before destruction. Here we use molecular modeling to map the radiation damage during the 10–50 fs to the intensity, the energy and the time duration of the laser pulse on the oxygen-evolving complex (OEC) of photosystem II. Association between histones and DNA is crucial for many cellular functions such as gene transcription and epigenetic silencing. Changes to chromatin structure influence gene expression via histone modifications. Chromatin immunoprecipitation (ChIP) is an experimental technique used to investigate the interactions between histones and DNA. ChIP determines the specific location in the genome that histone modifications are associated with, indicating the target of the histone modifiers. Despite the appeal of ChIP as an in vitro technique, there are limitations to its use. There is variability from one preparation to the next, given the many steps and reagents used throughout the technique. …

Four groups (n=10) of one year old Labeo rohita were exposed to 96-hr LC50 and sub-lethal (2/3rd, 1/4th and 1/5th) concentrations of MnCl2 for a duration of 30 days in the glass aquaria of 50L water capacity. After an exposure period of 30 days, the activity of peroxidase enzyme in the liver and brain of MnCl2 exposed fish was measured and compared with the control group. This paper characterizes three novel microwave applicators based on zero-order mode resonators for use in hyperthermia treatment of cancer. The radiation patterns are studied with numerical simulations in muscle tissue-equivalent model at 434 MHz. The relative performance of the applicators is compared in terms of reflection coefficient, current distribution, power deposition (SAR) pattern, effective field size in 2D and 3D tissue volumes, and penetration depth. One particular configuration generated the most uniform SAR pattern, with 25% SAR covering 84 % of the treatment volume extending to 1 cm depth under the aperture, while remaining above 58% coverage as deep as …

Background
Streptococcus pneumoniae is one of the leading causes of community acquired pneumonia and acute otitis media. Certain aspects of S. pneumoniae’s virulence are dependent upon expression and release of the protein toxin pneumolysin (PLY) and upon the activity of the peroxide-producing enzyme, pyruvate oxidase (SpxB). We investigated the possible synergy of these two proteins and identified that release of PLY is enhanced by expression of SpxB prior to stationary phase growth.
Results
Mutants lacking the <>spxB gene were defective in PLY release and complementation of spxB restored PLY release. This was demonstrated by cytotoxic effects of sterile filtered …

Alcohol use disorders (AUD) exacerbate neurocognitive dysfunction in Human Immunodeficiency Virus (HIV+) patients. We have shown that chronic binge alcohol (CBA) administration (13-14 g EtOH/kg/wk) prior to and during simian immunodeficiency virus (SIV) infection in rhesus macaques unmasks learning deficits in operant learning and memory tasks. The underlying mechanisms of neurocognitive alterations due to alcohol and SIV are not known. This exploratory study examined the CBA-induced differential expression of hippocampal genes in SIV-infected (CBA/SIV+; = 2) macaques in contrast to those of sucrose administered, SIV-infected (SUC/SIV+; = 2) macaques. Within the last century, the floristic composition of riparian communities in the Southwest has drastically changed following introduction of the exotic tree Tamarix ramosissima. In an attempt to control Tamarix populations, the tamarisk leaf beetle (Diorhabda carinulata) has been utilized as a biological control agent. Three years of data collection at our study sites along Fountain Creek (Fountain, CO) allowed us to characterize the response of Tamarix to invasion by the biological control agent. The NASA Balloon Satellite, which is a balloon that is sent 100,000 feet into the upper atmosphere to collect data, and stream that data live to NASA webpage, will carry experiments that are part of a payload. My experiment will be a Neulog Sensor Logger that will collect data on the UVA and UVB exposure rate of light from the upper stratosphere and on the ground. Responding To Modern Flooding: Old English Place-Names As A Repository Of Traditional Ecological Knowledge, Richard L.C. Jones
Journal of Ecological Anthropology
Place-names are used to communicate Traditional Ecological Knowledge (TEK) by all indigenous, aboriginal and First Nations people. Here and for the first time, English place-names are examined through a TEK lens. Specifically, place-names formed in Old English—the language of the Anglo-Saxon—and coined between c. 550 and c. 1100 A.D., are explored. This naming horizon provides the basic name stock for the majority of English towns and villages still occupied today. While modern English place-names now simply function as convenient geographical tags Old English toponymy is shown here to exhibit close semantic parallels with many other indigenous place-names around the world. …

Smooth muscle presents several unique contractile properties when compared to striated muscle. Data suggests that differential remodeling of the alpha-actin and beta-actin and the regulation of such remodeling may play an essential role in smooth muscle contraction. At rest both alpha- and beta-actin both exhibit a cable-like appearance. During contraction, alpha-actin appears to undergo significant remodeling with dissolution on the majority of the cables and the formation of podosomes where it demonstrates co-localization with myosin while beta-actin retains a cable-like appearance. While the remodeling of the two actin domains has been characterized under normal gravity little is known regarding cytoskeletal …
