Background: Non-alcoholic and alcoholic fatty liver disease (NAFLD and AFLD, respectively) are major health problems, as patients with either condition can progress to hepatitis, fibrosis, and cirrhosis. Although histologically similar, key differences likely exist in these two models. For example, altered content of several vesicle trafficking proteins have been identified in AFLD, but their content in NAFLD is unknown. In this study, we compared select parameters in NAFLD and AFLD in a rat model.
Methods: We fed either Lieber- DeCarli liquid control or alcohol-containing (35 % as calories) diet (AFLD model) or lean or high-fat (12 or 60 % derived …

Calmodulin (CaM) is the primary calcium signaling protein in eukaryotes and has been extensively studied using various biophysical techniques. Prior crystal structures have noted the presence of ambiguous electron density in both hydrophobic binding pockets of Ca2+-CaM, but no assignment of these features has been made. In addition, Ca2+-CaM samples many conformational substates in the crystal and accurately modeling the full range of this functionally important disorder is challenging. In order to characterize these features in a minimally biased manner, a 1.0 A resolution single-wavelength anomalous diffraction data set was measured for selenomethionine-substituted Ca2+-CaM. …

Stable transformation of plants is a powerful tool for hypothesis testing.Arapid and reliable evaluation method of the transgenic allele for copy number and homozygosity is vital in analysing these transformations. Here the suitability of Southern blot analysis, thermal asymmetric interlaced (TAIL-)PCR, quantitative (q)PCR and digital droplet (dd)PCR to estimate T-DNA copy number, locus complexity and homozygosity were compared in transgenic tobacco. Southern blot analysis and ddPCR on three generations of transgenic offspring with contrasting zygosity and copy number were entirely consistent, whereas TAIL-PCR often underestimated copy number. qPCR deviated considerably from the Southern blot results and had lower precision and higher …

Parasites and pathogens are increasingly recognized as significant drivers of ecological and evolutionary change in natural ecosystems. Concurrently, transmission of infectious agents among human, livestock, and wildlife populations represents a growing threat to veterinary and human health. In light of these trends and the scarcity of long-term time series data on infection rates among vectors and reservoirs, the National Ecological Observatory Network (NEON) will collect measurements and samples of a suite of tick-, mosquito-, and rodent-borne parasites through a continental-scale surveillance program. Here, we describe the sampling designs for these efforts, highlighting sampling priorities, field and analytical methods, and the …

Human cytomegalovirus (HCMV) is a large double-stranded DNA virus that causes severe disease in newborns and immunocompromised patients. During infection, HCMV is able to reconfigure the host cell machinery to establish a virus producing factory, termed the cytoplasmic virion assembly complex (cVAC). Generating drugs that affect cVAC development or function provides an alternative mode of action for HCMV antivirals that can essentially eliminate virion production. The objective of …
Children Once, Not Forever: Harper Lee’S Go Set A Watchman And Growing Up, Allen Mendenhall
Children Once, Not Forever: Harper Lee’S Go Set A Watchman And Growing Up, Allen Mendenhall
Indiana Law Journal
The narratives of Jean Louise in To Kill a Mockingbird and Go Set a Watchman are as consistent as lived experience, which is marked by disruption and contingency, ambiguity and rupture, fragmentation and complexity. Only the careless would have accepted Jean Louise and Atticus as one-dimensional, self-contained figures unspoiled by the mores, customs, and vocabularies of their white discursive community. Such a sanitized view of Jean Louise and Atticus erases and rewrites rather than represents history in its disturbing, enlightening variety and complexity. Jean Louise and Atticus are not stock character types; their thoughts and behaviors are irreducible and inexhaustible.

Crossing The Troll Bridge - The Framing Of Fat Bodies On Social Media, Amanda Levitt
Wayne State University Theses
As the use of social media grows, cyber bullying and trolling plague the way that people interact online. Even within spaces specifically dedicated to deconstructing dominant discourse around fatness, cyber bullying is an everyday occurrence. Data for this research came from a randomly selected sample of 500 messages sent to moderators of This is Thin Privilege, a Tumblr blog that explores and critiques the experiences of fat people in society. This study contributes to the growing body of literature on online communication by analyzing how commenters’ messages construct fatness and reify dominant narratives, the common tactics commenter’s use, and the …

Analyzing Sumo-2/3 Modification In Regulation Of Breast Cancer Progression And Mitotic Chromosome Segregation, Divya Subramonian
Wayne State University Dissertations
Roles of SUMOylation in cancer metastasis and mitotic progression has been elucidated in this thesis. This vital and dynamic post-translational modification has been known to be dysregulated in several kinds of cancer. Global SUMO-2/3 conjugation is upregulated in a mouse metastatic breast cancer cell line, 66cl4, compared to a non-metastatic cell line 168FARN from the same genetic background. In order to identify the SUMO-2/3 substrates, SILAC labeled lysates from the two cell lines were immunopurified using SUMO-2/3 monoclonal antibodies. By using SUMO-2/3 epitope peptide elution (SUMO-2/3-ePIPE) and quantitative mass spectrometry, 66 SUMO-2/3 targets were identified, of which 15 targets are …

Variables That Impact The Implementation Of Project-Based Learning In High School Science, Kellie Renee Cunningham
Wayne State University Dissertations
Wagner and colleagues (2006) state the mediocrity of teaching and instructional leadership is the central problem that must be addressed if we are to improve student achievement. Educational reform efforts have been initiated to improve student performance and to hold teachers and school leaders accountable for student achievement (Wagner et al., 2006). Specifically, in the area of science, goals for improving student learning have led reformers to establish standards for what students should know and be able to do, as well as what instructional methods should be used. Key concepts and principles have been identified for student learning. Additionally, reformers …

Big Data Management Using Scientific Workflows, Andrii Kashliev
Wayne State University Dissertations
Humanity is rapidly approaching a new era, where every sphere of activity will be informed by the ever-increasing amount of data. Making use of big data has the potential to improve numerous avenues of human activity, including scientific research, healthcare, energy, education, transportation, environmental science, and urban planning, just to name a few. However, making such progress requires managing terabytes and even petabytes of data, generated by billions of devices, products, and events, often in real time, in different protocols, formats and types. The volume, velocity, and variety of big data, known as the "3 Vs", present formidable challenges, unmet …

Physical And Electrochemical Area Determination Of Electrodeposited Ni, Co, And Nico Thin Films, Matthew Gira, Kevin Tkacz, Jennifer R. Hampton
Faculty Publications
The surface area of electrodeposited thin films of Ni, Co, and NiCo was evaluated using electrochemical double-layer capacitance, electrochemical area measurements using the [Ru(NH3)6]3+/[Ru(NH3)6]2+ redox couple, and topographic atomic force microscopy (AFM) imaging. These three methods were compared to each other for each composition separately and for the entire set of samples regardless of composition. Double-layer capacitance measurements were found to be positively correlated to the roughness factors determined by AFM topography. Electrochemical area measurements were found to be less correlated with measured roughness factors as well as applicable …

Abstract: Children affected by HIV are at increased risk of developmental and neuropsychological disturbances due to direct effects of HIV on the brain and direct effects associated with living in poverty. Caregivers can play an important role, through quality caregiving, in mitigating the negative effect of these stressors. This study used baseline data from an ongoing caregiver training intervention trial to evaluate the association between quality of caregiver-child interactions and neurocognitive outcomes in rural HIV-infected and HIV-exposed but uninfected children in Uganda. We also assessed the extent to which caregiver distress moderated this relationship. Data on 329 caregiver-child dyads were …
Variations In Adolescent Disclosure Among Latinas, Eve Veliz-Moran
Variations In Adolescent Disclosure Among Latinas, Eve Veliz-Moran
Journal of Latino/Latin American Studies (JOLLAS)
This study examines attitudes toward adolescent disclosure among a diverse sample of Latina adolescents. Using ecological and acculturation frameworks, this qualitative, exploratory study analyzes responses from 18 Latina adolescents to open-ended, semi-structured questions from the National Study of Youth and Religion on family relationships, morality, and religion, to explore disclosure attitudes. Results suggest high disclosure to mothers, particularly by Latinas from low-income and religiously congruent families. Fathers’ long work hours and singular religiosity thwart disclosure to them, and family structure, migration, and nativity influence whether adolescent Latinas disclose information to parents. Latino cultural values of respeto and marianismo did not …

Albomycin (ABM), also known as grisein, is a sulfur-containing metabolite produced by Streptomyces griseus ATCC 700974. Genes predicted to be involved in the biosynthesis of ABM and ABM-like molecules are found in the genomes of other actinomycetes. ABM has potent antibacterial activity, and as a result, many attempts have been made to develop ABM into a drug since the last century. Although the productivity of S. griseus can be increased with random mutagenesis methods, understanding of Streptomyces sulfur amino acid (SAA) metabolism, which supplies a precursor for ABM biosynthesis, could lead to improved and stable production. We previously characterized the …
Abortion And The Right To Not Be Pregnant, James E. Mahon
Abortion And The Right To Not Be Pregnant, James E. Mahon
Publications and Research
In this paper I defend Judith Jarvis Thomson's 'Good Samaritan Argument' (otherwise known as the 'feminist argument') for the permissibility of abortion, first advanced in her important, ground-breaking article 'A Defense of Abortion' (1971), against objections from Joseph Mahon (1979, 1984). I also highlight two problems with Thomson's argument as presented, and offer remedies for both of these problems. The article begins with a short history of the importance of the article to the development of practical ethics. Not alone did this article put the topic of the abortion on the philosophical map, but it made 'practical ethics' in the …

Predicting long-term trends in forest growth requires accurate characterisation of how the relationship between forest productivity and climatic stress varies across climatic regimes. Using a network of over two million tree-ring observations spanning North America and a space-for-time substitution methodology, we forecast climate impacts on future forest growth. We explored differing scenarios of increased water-use efficiency (WUE) due to CO2-fertilisation, which we simulated as increased effective precipitation. In our forecasts: (1) climate change negatively impacted forest growth rates in the interior west and positively impacted forest growth along the western, southeastern and northeastern coasts; (2) shifting climate sensitivities offset positive …

Indoleamine 2,3-dioxygenase-1 (IDO1) is a promising therapeutic target for the treatment of cancer, chronic viral infections, and other diseases characterized by pathological immune suppression. Recently important advances have been made in understanding IDO1’s catalytic mechanism. Although much remains to be discovered, there is strong evidence that the mechanism proceeds through a heme-iron bound alkylperoxy transition or intermediate state. Accordingly, we explored stable structural mimics of the alkylperoxy species and provide evidence that such structures do mimic the alkylperoxy transition or intermediate state. We discovered that O-benzylhydroxylamine, a commercially available compound, is a potent sub-micromolar inhibitor of IDO1. Structure-activity studies of …
