Background: The regulation of specific target genes by transcription factors is central to our understanding of gene network control in developmental and physiological processes yet how target specificity is achieved is still poorly understood. This is well illustrated by the Hox family of transcription factors as their limited in vitro DNA-binding specificity contrasts with their clear in vivo functional specificity. Results: We generated genome-wide binding profiles for three Hox proteins, Ubx, Abd-A and Abd-B, following transient expression in Drosophila Kc167 cells, revealing clear target specificity and a striking influence of chromatin accessibility. In the absence of the TALE class homeodomain …

Cree/Métis filmmaker Danis Goulet’s science fiction short Wakening (2013) is set in Canada’s near future, yet the film reveals a slipstream of time where viewers are invited to contemplate the horrors of ecosocial crises—future, past, and present. I argue Wakening, as futuristic ecohorror, produces horrific feelings in the moment of its viewing that are inevitably entangled with the past, inviting its audiences to experience the monstrous contexts of Indigenous lives across time. To articulate this temporal dynamism, I overlay two key conceptual understandings: Walter Benjamin’s critiques of Western progress and historicism, and Indigenous notions of a Native slipstream. When brought …

Nanomedicine is becoming very popular over conventional methods due to the ability to tune physico-chemical properties of nanovectors, which are used for encapsulation of therapeutic and diagnostic agents. However, the success of nanomedicine primarily relies on how specifically and efficiently nanocarriers can target pathological sites to minimize undesirable side effects and enhance therapeutic efficacy. Here, we introduce a novel class of targeted nano drug delivery system, which can be used as an effective nano-theranostic for cancer. We formulated pH-sensitive niosomes (80–90 nm in diameter) using non-ionic surfactants Span20 (43–45 mol%), cholesterol (50 mol%) and 5 mol% of pH (Low) Insertion …

A series of cyclic peptides containing a number of tryptophan (W) and glutamic acid (E) residues were synthesized and evaluated as pH-sensitive agents for targeting of acidic tissue and pH-dependent cytoplasmic delivery of molecules. Biophysical studies revealed the molecular mechanism of peptides action and localization within the lipid bilayer of the membrane at high and low pHs. The symmetric, c[(WE)4WC], and asymmetric, c[E4W5C], cyclic peptides translocated amanitin, a polar cargo molecule of similar size, across the lipid bilayer and induced cell death in a pH- and concentration-dependent manner. Fluorescently-labelled peptides were evaluated for targeting …

We have developed a way to measure cell surface pH by positioning a pH-sensitive fluorescent dye, seminaphtharhodafluor (SNARF), conjugated to the pH low insertion peptide (pHLIP). It has been observed that many diseased tissues are acidic and that tumors are especially so. A combination of effects acidifies tumor cell interiors, and cells pump out lactic acid and protons to maintain intracellular pH, acidifying the extracellular space. Overexpression of carbonic anhydrases on cell surfaces further contributes to acidification. Thus, the pH near tumor cell surfaces is expected to be low and to increase with distance from the membrane, so bulk pH …

pH (low) insertion peptides (pHLIP peptides) target acidic extracellular environments in vivo due to pH-dependent cellular membrane insertion. Two variants (Var3 and Var7) and wild-type (WT) pHLIP peptides have shown promise for in vivo imaging of breast cancer. Two positron emitting radionuclides (64Cu and 18F) were used to label the NOTA- and NO2A-derivatized Var3, Var7, and WT peptides for in vivo biodistribution studies in 4T1 orthotopic tumor-bearing BALB/c mice. All of the constructs were radiolabeled with 64Cu or [18F]-AlF in good yield. The in vivo biodistribution of the 12 constructs in 4T1 orthotopic allografted …

Premorbid Personality Indicators: A Tool To Identify Those Who Have Liability For Schizophrenia, Daniel V. Hudak Jr.
All-Inclusive List of Electronic Theses and Dissertations
Schizophrenia is an often-debilitating mental illness whose etiology remains unknown, although a number of conceptualizations have been suggested. Among the most prominent of these conceptualizations is the diathesis-stress model, which posits that liability to the disorder is genetically mediated, although ultimate expression is dependent upon exposure to various environmental stressors. Meehl (1962) is arguably most responsible for this conceptualization of schizophrenia. Identifying those who are liable for schizophrenia has the potential to inform treatment which may lead to better outcomes. In an effort to better understand the developmental process of schizophrenia and to identify those who may be at increased …

The Clinical Utility Of Standardized Test Observation In Diagnosis Of Adhd, Sarah K. Thomas
All-Inclusive List of Electronic Theses and Dissertations
The purpose of this study was to investigate the clinical utility of a standardized observation measure in the diagnostic assessment of ADHD in children. Evidenced-based assessment of ADHD recommends the use of multiple informants and multiple methods, and ADHD diagnostic criteria requires impairment in at least two settings. Commonly, parent and teacher reports of child behavior differ and blur the diagnostic picture, so standardized observations during testing may provide additional useful diagnostic information. Archival data from 140 children (age 5 – 12) referred to a university-based ADHD evaluation clinic was utilized in the present study. Testing observations measured by the …

Attitudes Towards Tax Evasion In Turkey And Australia: A Comparative Study, Robert W. Mcgee, Ken Devos, Serkan Benk
College of Business and Economics
The authors conducted a survey of 502 Turkish and Australian undergraduate and graduate business and economic students to determine their views regarding the ethics of tax evasion. These two groups were selected on the premise that their views represented the perceptions of two very different cultures, which has not been investigated in previous studies. The survey instrument required students to indicate their level of agreeableness to 18 general statements representing various scenarios in the socio-economic environment. The statements in the survey reflected the three main viewpoints regarding the ethics of tax evasion which have emerged from the literature to date. …

Potential Population And Assemblage Influences Of Non-Native Trout On Native Nongame Fish In Nebraska Headwater Streams, Kelly C. Turek, Mark A. Pegg, Kevin L. Pope, Steve Schainost
Nebraska Cooperative Fish and Wildlife Research Unit: Staff Publications
Non-native trout are currently stocked to support recreational fisheries in headwater streams throughout Nebraska. The influence of non-native trout introductions on native fish populations and their role in structuring fish assemblages in these systems is unknown. The objectives of this study were to determine (i) if the size structure or relative abundance of native fish differs in the presence and absence of non-native trout, (ii) if native fish-assemblage structure differs in the presence and absence of non-native trout and (iii) if native fish-assemblage structure differs across a gradient in abundances of non-native trout. Longnose dace Rhinichthys cataractae were larger in …

Influence Of Anglers’ Specializations On Catch, Harvest, And Bycatch Of Targeted Taxa, Kevin L. Pope, Christopher J. Chizinski, Christopher L. Wiley, Dustin R. Martin
Nebraska Cooperative Fish and Wildlife Research Unit: Staff Publications
Fishery managers often use catch per unit effort (CPUE) of a given taxon derived from a group of anglers, those that sought said taxon, to evaluate fishery objectives because managers assume CPUE for this group of anglers is most sensitive to changes in fish taxon density. Further, likelihood of harvest may differ for sought and non-sought taxa if taxon sought is a defining characteristic of anglers’ attitude toward harvest. We predicted that taxon-specific catch across parties and reservoirs would be influenced by targeted taxon after controlling for number of anglers in a party and time spent fishing (combine to quantify fishing effort …

Erforschung biologischer Ressourcen der Mongolei / Exploration into the Biological Resources of Mongolia, ISSN 0440-1298
A revised annotated checklist of Mongolian mammals is presented, supplemented with comments on modifications in species- and genus-level taxonomy introduced over the last 30 years. The checklist includes 140 species compared to 124 species recognized three decades ago by SOKOLOV & ORLOV (1980). Since then, four species were newly described; seven species were registered for the first time on Mongolian territory. Names for more than 20 taxa have changed, on most occasions this change occurred not for purely nomenclatural reasons but rather as a result of taxonomic revisions at the species level which have elevated the rank of many nominal …
