Cpg Improves Influenza Vaccine Efficacy In Young Adult But Not Aged Mice, Alejandro Ramirez, Mary Co, Anuja Matthew

Institute for Immunology and Informatics Faculty Publications

Several studies have shown a reduced efficacy of influenza vaccines in the elderly compared to young adults. In this study, we evaluated the immunogenicity and protective efficacy of a commercially available inactivated influenza vaccine (Fluzone®) in young adult and aged mice. C57/BL6 mice were administered a single or double immunization of Fluzone® with or without CpG and challenged intranasally with H1N1 A/California/09 virus. A double immunization of Fluzone® adjuvanted with CpG elicited the highest level of protection in young adult mice which was associated with increases in influenza specific IgG, elevated HAI titres, reduced viral titres …

Tick‐, Mosquito‐, And Rodent‐Borne Parasite Sampling Designs For The National Ecological Observatory Network, Yuri P. Springer, Howard S. Ginsberg, Et Al

Plant Sciences and Entomology Faculty Publications

Parasites and pathogens are increasingly recognized as significant drivers of ecological and evolutionary change in natural ecosystems. Concurrently, transmission of infectious agents among human, livestock, and wildlife populations represents a growing threat to veterinary and human health. In light of these trends and the scarcity of long‐term time series data on infection rates among vectors and reservoirs, the National Ecological Observatory Network (NEON) will collect measurements and samples of a suite of tick‐, mosquito‐, and rodent‐borne parasites through a continental‐scale surveillance program. Here, we describe the sampling designs for these efforts, highlighting sampling priorities, field and analytical methods, and the …

The Reliability And Validity Of Functional Brain Connectivity Compared To A Self-Reported Measure Of Pain, Ali Alsouhibani Jan 2016

The Reliability And Validity Of Functional Brain Connectivity Compared To A Self-Reported Measure Of Pain, Ali Alsouhibani

Maria Dittman Library Research Competition: Student Award Winners

Pain is a multidimensional perception that is complex in nature. It is a unitary construct that includes overlapping domains such as intensity, affect, quality, and frequency. These domains do not reflect the amount of tissue damage. It reflects the end result of the perception of pain in which multiple biopsychosocial factors are involved (Gatchel et al., 2007). Multiple self-reported measures have been used in an attempt to capture most factors that may influence pain such as psychological factors. However, there is no one scale that can be used to characterize pain as a whole with all its factors. Furthermore, physical …

Decreased Interleukin-4 Release From The Neurons Of The Locus Coeruleus In Response To Immobilization Stress, Hyun-Ju Lee, Hyun-Jung Park, Angela Starkweather, Kyungeh An, Insop Shim Jan 2016

Decreased Interleukin-4 Release From The Neurons Of The Locus Coeruleus In Response To Immobilization Stress, Hyun-Ju Lee, Hyun-Jung Park, Angela Starkweather, Kyungeh An, Insop Shim

School of Nursing Publications

It has been demonstrated that immobilization (IMO) stress affects neuroimmune systems followed by alterations of physiology and behavior. Interleukin-4 (IL-4), an anti-inflammatory cytokine, is known to regulate inflammation caused by immune challenge but the effect of IMO on modulation of IL-4 expression in the brain has not been assessed yet. Here, it was demonstrated that IL-4 was produced by noradrenergic neurons in the locus coeruleus (LC) of the brain and release of IL-4 was reduced in response to IMO. It was observed that IMO groups were more anxious than nontreated groups. Acute IMO (2 h/day, once) stimulated secretion of plasma …

Role Of Tissue And Systemic Hypoxia In Obesity And Type 2 Diabetes, Lei Xi, Chin-Moi Chow, Xingxing Kong Jan 2016

Role Of Tissue And Systemic Hypoxia In Obesity And Type 2 Diabetes, Lei Xi, Chin-Moi Chow, Xingxing Kong

Internal Medicine Publications

Human lifestyle in most modern and developing societies has dramatically changed over past decades. Physical inactivity along with unrestricted access to calorie dense foods has established an “obesogenic” environment and contributed to a serious epidemic of obesity and type 2 diabetes (T2D), associated with increased morbidity and mortality. In 2005 a population-based study conducted by Reichmuth et al. of University of Wisconsin with a cross-sectional and longitudinal analysis identified that among 1387 participants the odds ratio for T2D with an apnea-hypopnea index (AHI) > 15 versus an AHI < 5 was 2.30 (1.28–4.11; p < 0.01) after adjustment for age, sex, and body habitus. Therefore it has been assumed that intermittent hypoxic periods associated with obstructive sleep apnea (OSA) may play a pathogenic role in inducing insulin resistance and T2D. At organ/tissue levels, in 2007–2009 Ye and colleagues first proposed a central role played by adipose tissue hypoxia resulting from adipocyte expansion in promoting chronic inflammation, adiponectin reduction, adipocyte dysfunction, and death in obese individuals. This group of researchers later identified the mediator roles played by hypoxia inducible factor 1α (HIF-1α) and other hypoxia-triggered signaling mechanisms that may promote free fatty acid release and inhibit glucose uptake in adipocytes by inhibition of the insulin-signaling pathway and induction of cell death.

Regulation Of Reactive Oxygen Species And The Antioxidant Protein Dj-1 In Mastocytosis, Do-Kyun Kim, Michael A. Beaven, Joseph M. Kulinski, Avanti Desai, Geethani Bandara, Yun Bai, Calman Prussin, Lawrence B. Schwartz, Hirsh Komarow, Dean D. Metcalfe, Ana Olivera

Internal Medicine Publications

Neoplastic accumulation of mast cells in systemic mastocytosis (SM) associates with activating mutations in the receptor tyrosine kinase KIT. Constitutive activation of tyrosine kinase oncogenes has been linked to imbalances in oxidant/antioxidant mechanisms in other myeloproliferative disorders. However, the impact of KIT mutations on the redox status in SM and the potential therapeutic implications are not well understood. Here, we examined the regulation of reactive oxygen species (ROS) and of the antioxidant protein DJ-1 (PARK-7), which increases with cancer progression and acts to lessen oxidative damage to malignant cells, in relationship with SM severity. ROS levels were increased in both …

Analyses were conducted on a sample of 5,532 adolescents from 37 schools at 4 sites. About half (49%) of participants were male; 48% self-identified as Black non-Hispanic; 21% as Hispanic, 18% as White non-Hispanic. Adolescents completed the PBFS and measures of beliefs and values related to aggression, and delinquent peer associations at the start of the 6th grade and over 2 years later. Ratings of participants’ behavior were also obtained from teachers on the Behavioral Assessment …

Fe/Male Mother Of Two: Gender And Motherhood In Lionel Shriver's We Need To Talk About Kevin, Amy B. Smialek Jan 2016

Fe/Male Mother Of Two: Gender And Motherhood In Lionel Shriver's We Need To Talk About Kevin, Amy B. Smialek

ETD Archive

There are critical reviews regarding Lionel Shriver’s We Need to Talk About Kevin that discuss many controversial topics in the novel. Of these reviews, most critics limit their arguments to the taboo topics of American school shootings and Eva’s character as an ostensibly ambivalent mother. Unfortunately, there is little academic criticism on Shriver’s most recognized novel and, among such analyses, two of Shriver’s most crucial depictions are overlooked. Firstly, readers must acknowledge the impact that contemporary American society has on females and mothers. This novel shows how much a culture relies on societal “rules” that govern human expectations. Secondly, Shriver’s …

The Regulatory Role Of Interleukin-1 Receptor Associated Kinase M In Toll-Like Receptor Signaling, Hao Zhou Jan 2016

The Regulatory Role Of Interleukin-1 Receptor Associated Kinase M In Toll-Like Receptor Signaling, Hao Zhou

ETD Archive

Toll-like receptors (TLRs) are one of the major groups of pattern recognition receptors. TLRs are able to recognize the pathogen-associated molecular patterns and transduce their signals through the adaptor molecule MyD88 and members of the IL-1R-associated kinase family (IRAK1, 2, M and 4). IRAKM was previously known to function as a negative regulator that prevents the dissociation of IRAKs from MyD88, thereby inhibiting downstream signaling. However, we now found that IRAKM was also able to interact with MyD88-IRAK4 to form IRAKM Myddosome to mediate TLR7-induced MEKK3-dependent second wave NF¿B activation. As a result, the IRAKM-dependent pathway only induced expression of …

The Synthesis And Characterization Of Multifunctional Nanoparticles Of Elastin-Like Polypeptides For Theranostic Applications, James T. Cole Jan 2016

The Synthesis And Characterization Of Multifunctional Nanoparticles Of Elastin-Like Polypeptides For Theranostic Applications, James T. Cole

ETD Archive

Theranostics is a promising field that aims to combine therapeutics and diagnostics into single multifunctional formulations. This field is driven by advancements in nanotechnology and specifically in the creation of multifunctional nanoparticles capable of providing the necessary functionalities. Elastin-like polypeptides (ELPs) are a class of environmentally responsive biopolymers that are known to undergo a transition in response to various stimuli. The organic nature of ELPs along with the ability to control their design at the gene level and the aforementioned responsive behavior make them a promising candidate for use in theranostic systems. The system presented here is one of the …

Respiratory Sinus Arrhythmia (Rsa) In Adults With Possible Autism Spectrum Disorder (Asd) Symptoms, Arielle Bowers Jan 2016

Respiratory Sinus Arrhythmia (Rsa) In Adults With Possible Autism Spectrum Disorder (Asd) Symptoms, Arielle Bowers

ETD Archive

Autism Spectrum Disorders (ASD) are a group of developmental disorders, which are becoming a major health concern within the US. According to The Diagnostic and Statistical Manual of Mental Disorders (5th ed.; DSM–5; American Psychiatric Association, 2013) an ASD includes the following features: continuous impairments associated with social communicative and interactive skills, replication of specific, patterned behaviors and/or attentiveness to specific interests. Recent psychophysiological research has looked at how high and low respiratory sinus arrhythmia (RSA) activity may be related to a lack of social communicative skills relating to social cognition, which are hallmark features of individuals who have been …

The Black Death And Giovanni Bocaccio's The Decameron's Portrayal Of Merchant Mentality, Rachel D. Rickel Jan 2016

The Black Death And Giovanni Bocaccio's The Decameron's Portrayal Of Merchant Mentality, Rachel D. Rickel

ETD Archive

Giovanni Boccaccio was a contemporary witness to the effects of the Black Death pandemic, the Yersinia pestis bacterial pandemic in Europe between the years 1346-53, causing 75 million to 200 million deaths across the continent alone. In The Decameron, Boccaccio depicts the outbreak’s high-mortality rates and how that was a catalyst for many social and cultural changes within fourteenth-century Europe. He also goes on to portray the devastating effects of death on, not only the physical bodies of people and animals, but also on their mental, emotional, and spiritual states, and how this accelerated their acceptance of the rising …

Green Buildings: Lease Structure, Productivity, And Regional Economic Impacts, Eun Kyu Lee Jan 2016

Green Buildings: Lease Structure, Productivity, And Regional Economic Impacts, Eun Kyu Lee

ETD Archive

In an effort to apply the sustainability movement to the built environment, sustainable construction has gained substantial strength and momentum in global society. A growing body of literature reveals the positive effects of sustainable, or green, buildings in terms of real estate market premiums, energy cost savings, employee productivity, and regional economic growth. In this context, my dissertation examines three research topics pertaining to sustainable buildings, and does so in three separate chapters.

The first study discusses the effect of lease structure on the tenant’s willingness to pay for energy-efficient building features, and compares the tenant’s stated willingness to pay …

Sex-Bias In Lupus-Prone (Nzbxnzw)F1 Mice: The Interplay Of Gr1+Cd11b+ Cells, Testosterone And Genetics, Abhishek Trigunaite

ETD Archive

Systemic Lupus Erythematosus (SLE) is a prototypic systemic autoimmune disorder predominantly (9:1) targeting females. Currently, there is no cure for SLE and also there is a lack of disease predicting biomarkers. The major aim of my research is to determine the cellular basis of the sex bias in SLE and subsequently facilitate the development of better therapeutics and biomarkers for the disease. In (NZBxNZW) F1 mice a similar female bias is observed as in humans and testosterone is shown to be protective against lupus-like disease development, but the mechanism is not well understood.

We found higher levels of Gr1hiCD11b+ myeloid …

Development And Validation Of Uplc/Ms/Ms Methods For Quantification Of Gangliosides In The Clinical Study Of Ganglioside Gm3 Synthase Deficiency, Qianyang Huang Jan 2016

Development And Validation Of Uplc/Ms/Ms Methods For Quantification Of Gangliosides In The Clinical Study Of Ganglioside Gm3 Synthase Deficiency, Qianyang Huang

ETD Archive

Gangliosides are a large subclass of glycosphingolipids that structurally characterized by the addition of mono- or poly-sialyated carbohydrate moieties onto the ceramide scaffold. They are presented at significant abundance in the central nervous system of vertebrates. Their interactions with trans-membrane receptors, especially receptor tyrosine kinase, are believed to play critical roles in modulating signal transduction events during cell proliferation, differentiation, migration, and adhesion. Furthermore, the disruption and dysfunction on their metabolism have been found to be associated with various neurodegenerative disorders, such as Alzheimer disease, Parkinson’s disease, and GM3 synthase deficiency. GM3 synthase deficiency (GSD) is a newly identified neurological …
