Hybrid materials based on tetraethyl orthosilicate (TEOS), hydroxypropyl cellulose (HPC), or hydroxypropyl methyl cellulose (HPMC) with silver nanoparticles were synthesized. They were analyzed and characterized using differential thermal analysis and thermogravimetry, atomic force microscopy, and static contact angle measurements. It was experimentally demonstrated that the silver-doped hybrid materials have pronounced antibacterial behavior by studying the reduction of Pseudomonas aeruginosa PAO1 biofilm formation on the tested materials. The results revealed biofilm reduction of 35.7% and 30% by SiO2/HPC/2.5% Ag and SiO2/HPMC/2.5% Ag hybrid materials, respectively, compared to the control. Cytotoxicity of examined materials and actin cytoskeleton organization of fibroblasts seeded on …

Turkish Journal of Biology
This study investigates whether the reciprocal changes in transient receptor potential canonical (TRPC)1 and TRPC6 expressions, which have previously been observed in aging rat aorta, are functional in store-operated calcium (SOC) entry and proliferation in human vascular smooth muscle cells. TRPC1 levels were modulated via silencing and overexpression vectors in human primary aortic smooth muscle cells. Following TRPC1 gene modulation, TRPC1 and TRPC6 expression levels were measured using quantitative real-time RT-PCR. In functional analyses, real-time changes in intracellular calcium levels and cell proliferation were determined. Microarray analysis was performed to identify genes associated with functional alterations following TRPC1 silencing. TRPC1 …

To create new varieties resistant to biotic and abiotic stress factors that possess high productivity and improved fruit quality, breeders require modern breeding methods and techniques. Double haploid (DH) lines are an invaluable breeding element due to their guaranteed, completely homozygous nature. One of the fastest methods for obtaining DH lines is anther culture. Microspore embryogenesis provides considerable opportunities in the areas of breeding and biotechnology and represents a model system for fundamental biological research. The effectiveness of pepper anther culture is still low, which affects the application of DH lines in breeding programs. A detailed study of the anther …

Although interspecific chimeras can be identified using laborious techniques, more accurate and rapid detectable methods need to be established. The present study develops a nested polymerase chain reaction (nPCR) assay to detect human-mouse chimeras. A set of previously validated outer primers, specific to the human SRY gene, was used for conventional one-step PCR, while the inner primers for nPCR were designed. The specificities of PCR and nPCR assays were examined; both primer sets yielded PCR amplification products from male human epidermis-derived mesenchymal stem cell-like pluripotent cell DNA but no amplification products from negative control DNA. The sensitivity of this nPCR …

Turkish Journal of Biology
Electroporation is a safe and efficient method to transfect human tooth germ stem cells (hTGSCs), which are mainly mesenchymal stem cells (MSCs). In this study, pluronic F68 (PF68), a nonionic and low-foaming surfactant, was used for the first time to increase the transfection efficiency in electroporation of hTGSCs. Our results revealed that when PF68 was used in the preincubation, electroporation, and recovery medium, it dramatically increased transfection efficiency. Moreover, use of PF68 did not alter the differentiation capacity of hTGSCs, whereas cells electroporated without PF68 had significantly lower expression of Oct-4, pluripotency marker gene, and alkaline phosphatase (ALP) activity upon …

The Effects Of Dna Methyl Transferases On Antiaging Klotho Gene Expression, Eli̇f Çağlayan, Kadi̇r Turan
Turkish Journal of Biology
Several intracellular metabolic pathways in which multiple genes have a role affect the aging process. One important genetic factor associated with aging is klotho gene. In some recently published studies, klotho gene has also been associated with several types of cancer and identified as a tumor suppressor gene. Therefore, elucidation of the control of klotho gene expression has become more important. Results suggesting that the promoter region of human klotho gene could be epigenetically controlled by DNA methylation have been reported. In contrast, a study revealing whether a change in the expression level of DNMT enzymes in the cells affects …

Silymarin (Sm) is widely used in treating diseases that affect organs such as the liver, kidney, and gallbladder thanks to its antioxidative, renoprotective, antihepatotoxic, and anticarcinogenic properties. However, this substance is poorly solved in water and tends to decompose in the intestine, its bioavailability decreasing before it can show real effect. With these limitations in mind, the present study aims to enhance the poor bioavailability of Sm by forming Sm-loaded solid lipid nanoparticles (Sm-SLNs) using the hot homogenization method. A characterization process was undertaken to determine possible impact of Sm on experimental kidney damage. Our biochemical and light microscopic results …

Physical exercise is beneficial for bone tissue health, yet its usage is limited for preventing osteoporosis. Even though natural for the bone tissue from development to homeostasis, mechanical loads present with a multitude of physical parameters, including amplitude, duration, frequency, and distribution. Utilizing the most beneficial parameters of mechanical loads may potentiate a nonpharmaceutical tool for biotechnology to prevent and treat bone loss related to aging, bedrest, sedentary lifestyles, weightlessness, and other diseases. Low intensity vibrations (LIVs) consist of mechanical loads with amplitudes smaller than loads prescribed by habitual activity, with a higher frequency. In this review, literature covering LIV …

Heart disease is one of the largest causes of death in humans throughout the developed world. Despite its critical role in maintaining physiologic homeostasis, the adult mammalian heart does not exhibit a significant capacity for regeneration. By contrast, several lower vertebrates exhibit a remarkable ability to regenerate the aging myocardium in response to severe injury. Additionally, recent evidence has shed light on a transient regenerative window in postnatal mammals. In this review, we discuss key findings that help to unravel the differences in cardiac regeneration across phylogeny and ontogeny. Furthermore, we highlight recent significant progress in the pursuit of adult …

Increased fragility of long bones and delay in fracture healing in adults as well as skeletal development disorders in children are thought to be associated with endochondral ossification problems. The extracellular matrix (ECM) of cartilage is absorbed during endochondral ossification. During the recovery process from damage, chondrocytes undergo hypertrophy and increase their cell volume; on the other hand, it is also necessary to digest the ECM elements around the cells. Recent research on the proteolytic enzymes in ECM assumed to be responsible for the digestion is connected to proteinases digesting type II collagen and aggrecan. ADAMTS1, 4, and 5 are …

Findings in the last decade suggest that there is a considerable amount of cardiomyocyte turnover in the human heart throughout life, albeit not sufficient for heart regeneration following myocardial infarctions. Only a few species are known to be remarkably efficient in cardiac regeneration. They restore lost cardiomyocytes via a process of cardiomyocyte dedifferentiation, which is followed by robust proliferation of cardiomyocytes and incorporation into the myocardium. Similarly, neonatal mice have been recently shown to regenerate their heart following myocardial injuries. Studies with a neonatal cardiac regeneration mouse model suggest that the major source of new cardiomyocytes is likely to be …

Cardiomyogenic differentiation from mesenchymal stem cells has emerged as a novel approach for repair of damaged myocardium. Cell transplantation through direct cell injection is not an optimal method due to the lack of cell-extracellular matrix interactions. In the present study, differentiation potential of human adipose-derived stem cells (ASCs) to cardiomyocytes has been investigated by growing them on hyaluronic acid/gelatin (HA/G) plasma gels and coverslips and supplementing the growth medium with chemical modifiers (activin-a, BMP-4, insulin, valproic acid, and 5-azacytidine) in various combinations. The HA/G plasma gels were produced from human blood plasma-derived fibrinogen, gelatin, and human umbilical cord-derived hyaluronic acid. …

Researchers have been focused on understanding the pathogenesis of human diseases. They have been working to find the roles of caspases in the balance between apoptosis, autophagy, pyroptosis, and necroptosis, and also in the regeneration of damaged tissue. At this point, besides their death-inducing roles, new findings indicate the role of caspases in proliferation for maintaining the viability of cells in response to signals from apoptotic cells. Recently, determining cell fate has also been identified among other functions of caspases. Findings indicate that caspases direct cellular pathways to cell differentiation by suppressing stem cell self-renewal. The common opinion about the …
