Collagen/Peo/Gold Nanofibrous Matrices For Skin Tissue Engineering, Ömer Aktürk, Di̇lek Keski̇n

Turkish Journal of Biology

As a novel approach in skin tissue engineering, gold nanoparticles (AuNPs) were synthesized and incorporated at different concentrations into collagen/PEO nanofibrous matrices in this study. The group containing 14.27 ppm AuNPs (CM-Au) had the best nanofibrous morphology. CM-Au was cross-linked with glutaraldehyde vapor (CM-AuX). All groups were disrupted in collagenase in 2 h, but cross-linked groups and Matriderm1397904493 resisted hydrolytic degradation for 7 and 14 days, respectively. Due to its small pores and dense structure, lower water swelling results (7.26 ± 2.62 g/g) were obtained for CM-AuX than Matriderm (17.51 ± 1.97 g/g). CM-Au and Matriderm had statistically similar tensile …

The Sca-1+ Mesenchymal Stromal Cells Modulate Macrophage Commitment And Function, Yaozhen Chen, Yan Chen, Dandan Yin, Yazhou Wang, Zheng Liu, Ning An, Fan Wen, Na Li, Jiajia Xin, Xingbin Hu, Huijie Zhang, Wen Yin

Turkish Journal of Biology

Mesenchymal stromal cells (MSCs) have been characterized as an important component of the hematopoietic niche that plays a role in regulating hematopoietic stem cell (HSC) quiescence, self-renewal, and lineage commitment. Macrophages, one kind of innate immune cells, are mainly developed from HSCs in niche. However, how MSCs and their subpopulations regulate macrophage commitment is still unknown. The current study compared the contribution of MSCs and Sca-1+MSCs to macrophage commitment and further modulation on the function of macrophages. We found that Sca-1+MSCs could promote macrophage commitment through the M-CSFR gene, and Sca-1+MSCs led macrophage polarization towards the M2 phenotype. Further functional …

Simvastatin Combined With Bone Marrow Stromal Cells Treatment Activatesastrocytes To Ameliorate Neurological Function After Ischemic Stroke In Rats, Yonghua Zhao, Qian Zhang, Zhenwei Chen, Naiwei Liu, Chienchih Ke, Youhua Xu, Weikang Wu

Turkish Journal of Biology

Evidence indicates that reactive astrocytes play an important role in neurological functional amelioration after stroke. Bone marrow stromal cell (BMSC) transplantation-promoted astrocyte-derived neurotrophic factors' expressions resulted in decreasing the number of apoptotic neurons and improving neurogenesis and angiogenesis in the ischemic boundary zone. Recent studies found that simvastatin had neuroprotective activity in acute stroke. However, it is still ambiguous as to whether simvastatin combined with BMSCs can further activate astrocytes, which contribute to neurological functional recovery after ischemic stroke. In our present study, we established a middle cerebral artery occlusion model in rats and found that combination treatment of simvastatin …

Production Of Recombinant Human Dipeptidyl Peptidase Iv From Sf9cells In Microbial Fermenters, Özlem Ayteki̇n, Sai̇me İsmet Gürhan, Kayoko Ohura, Teruko Imai, Gaye Öngen

Turkish Journal of Biology

The human dipeptidyl peptidase IV (hDPPIV/CD26) is expressed as an immune response in some cancer cells as well as intestine and incretin metabolism, and deficiency of the enzyme leads to metabolic disorders. In the present study, recombinant hDPPIV/CD26 genes were expressed in baculovirus-insect cell systems in a 5-L stirred-tank fermenter. Because of the shear sensitivity of the insect cell line, production from insect cells should be performed in new-generation type bioreactors, which are commonly more expensive than microbial fermenters. The worldwide decline in amphibian populations has made heavy metal pollution a subject of interest. The aim of this work was to analyze the effect of sublethal doses of cadmium (Cd) on the liver, kidney, and blood of Rhinella arenarum specimens. Serum markers indicative of hepatic injury (gamma glutamyltransferase, alkaline phosphatase, pseudo cholinesterase, and total cholesterol) and markers of renal dysfunction (blood urea nitrogen, serum creatinine, calcium, and glucose) showed variation in animals treated with a 0.5 mg/kg dose compared to controls. Histopathological images revealed alterations in the liver (hepatocyte ballooning and hyperplasia of Kupffer cells) and kidney (renal tubular …

Oxytropis and Astragalus represent one of the largest angiosperm genera complexes. Although phylogenetic studies of this complex exist, the evolutionary relationships among Astragalus and Oxytropis species sharing similar habitats in the Old World have not been studied in detail. The phylogenetic relationships among 13 Oxytropis and 56 Astragalus species native to Turkey were inferred from nucleotide sequence variations in the nuclear ribosomal internal transcribed spacer (ITS) and chloroplast maturase-like protein (matK) gene regions. In addition to our samples, 36 Oxytropis ITS and 6 Oxytropis matK sequences were retrieved from GenBank and included in the analysis. Phylogenies derived from a maximum …
