Rats initially fear humans which can lead to negative affect, poor welfare, and difficult handling. Also, modeling and measuring positive affect states in rats can pose an additional challenge. Heterospecific play, or “tickling,” is a handling habituation technique that mimics rat rough-and-tumble play that is being used to study positive affect. It can also be used to reduce fear of human, improve welfare, and elicit a positive affect state. Bone morphogenetic proteins (BMPs) drive a range of cellular processes especially in the early stages of embryonic development. This family of proteins acts as one of the most important extracellular signals in development pattern formation across the animal kingdom. Cells in embryos differentiate into different cell types in response to the concentration level of BMP. Improper lawn fertilization with excess N or phosphorus has the potential to contaminate both surface and groundwater. Invasive Burmese pythons are established in the Everglades and are altering the ecology of southern Florida. Their distribution in Florida is expanding northward into more urbanized and fragmented habitats. An understanding of the suitability of habitat throughout southern Florida for Burmese pythons and their interaction with Florida’s landscapes through movement behavior is vital for predicting the python’s ability to persist in habitats outside of the Everglades. In this thesis, we use ecological modeling to predict habitat suitability and to investigate personality-dependent dispersal.

The dopamine transporter (DAT) is a monoamine transporter that regulates dopamine (DA) neurotransmission by clearing DA from the synapse. DAT is acted on by a number of psychostimulant drugs, which block reuptake and potentiate DA signaling. Several inhibitors of DAT, both amphetamines and cathinones, also cause reversal of transport. Previous research has shown the importance of a conserved salt bridge in another monoamine transporter, the serotonin transporter, in amphetamine-induced efflux. In our studies here, we engineered a mutant Drosophila melanogaster DAT (dDAT D475N) designed to interrupt this salt bridge and examine the effects on amphetamine- and cathinone-induced efflux in vitro. …

Non-typhoidal Salmonella is the leading cause of foodborne illness in the United States, resulting in about 20,000 hospitalizations and nearly 380 deaths annually. The meat processing industry has been especially plagued by Salmonella, from meat-inherent sources and more alarmingly, cross-contamination. For ready-to-eat (RTE) meat products specifically, this can cause significant problems in processing facilities ensuring safe product for consumption, resulting in foodborne illness.

Laboratory mice are nocturnal, spending most of their daylight hours asleep. But they live in the diurnal world of human investigators and husbandry staff, who primarily work during this rest period. In humans, lack of sleep or sleep that occurs outside the normal circadian sleep period (as in shift work) has adverse effects. These include increased risk of cardiovascular disease, cancer, metabolic disorder, mood disorders, type II diabetes, and obesity. Listeria monocytogenes (Lm) is a foodborne pathogen, found ubiquitously in nature, and has a high morbidity rate among immunocompromised individuals, the elderly, and especially pregnant women and their fetuses resulting in abortion, stillbirth, and neonatal infection. There are currently no preventative medical interventions against Lm infection. The Listeria adhesion protein (LAP) is present in both pathogenic and non-pathogenic Listeria (i.e., L. innocua) and has shown to interact with host epithelial proteins causing tight junction dysregulation aiding in pathogen attachment and paracellular translocation across the host intestinal epithelium. High performance computing (HPC) systems frequently suffer errors and failures from hardware components that negatively impact the performance of jobs run on these systems. We analyzed system logs from two HPC systems at Purdue University and created statistical models for memory and hard disk errors. We created a small-scale error injection testbed—using a customized QEMU build, libvirt, and Python—for HPC application programmers to test and debug their programs in a faulty environment so that programmers can write more robust and resilient programs before deploying them on an actual HPC system. The hardness and reduced modulus of aspirin, RDX, HMX, TATB, FOX-7, ADAAF, and TNT/CL-20 were experimentally measured with nanoindentation. These values are reported for the first time using as-received micron sized crystals of energetic materials with no additional mechanical processing. The results for TATB, ADAAF, and TNT/CL-20 are the first of their kind, while comparisons to previous nanoindentation studies on large, carefully grown single crystals of the other energetic materials show that mechanical properties of the larger crystals are comparable to crystals in the condition they are practically used. Hypoxia inducible factors (HIFs) are central mediators of cellular responses to fluctuations of oxygen, an environmental regulator of stem cell activity. Muscle satellite cells are myogenic stem cells whose quiescence, activation, self-renewal and differentiation are influenced by microenvironment oxygen levels. However, the in vivo roles of HIFs in quiescent satellite cells and activated satellite cells (myoblasts) are poorly understood. Expression analyses indicate that HIF1α and HIF2α are preferentially expressed in pre- and post-differentiation myoblasts, respectively. Aflatoxin contamination in feed and feed ingredients is of high worldwide prevalence, posing serious risks to the livestock industry. A series of studies were conducted to better understand the comprehensive impact of aflatoxicosis and its interrelationships with dietary modifications in broiler chicks and ducks. Results of these studies emphasize the many factors that the severity of aflatoxicosis depends on, including animal species, age, presence of mycotoxin adsorbent, and concentration of major dietary nutrients. Proxy variables from palaeolimnological studies of lakes in the Prairie Pothole Region of North America have been used to infer large oscillations during the late Holocene between longer periods of high-salinity–dry conditions and shorter periods of low-salinity–wet conditions producing a normative pattern marked by the absence of hydrological stability. Studies of the historical rise in lake level at Devils Lake have identified 1980 as a transition point between two such hydroclimatic modes. The temperature sensor presented in this paper is based on a microwave dielectric resonator, which uses alumina ceramic as a substrate to survive in harsh environments. The resonant frequency of the resonator is determined by the relative permittivity of the alumina ceramic, which monotonically changes with temperature. A rectangular aperture etched on the surface of the resonator works as both an incentive and a coupling device. A broadband slot antenna fed by a coplanar waveguide is utilized as an interrogation antenna to wirelessly detect the sensor signal using a radio-frequency backscattering technique. Siderophores are low-molecular weight species utilized by bacteria for the sequestration of iron, an essential nutrient. Siderophores and their cognate receptors are considered to be virulence factors, due to their prominent role in pathogenicity. The work presented here focuses on ferrioxamine (FOx) as an “immutable” ligand for pathogen detection. A number of bacterial strains expressing high-affinity FOx receptors were identified by a proteomic BLAST search, and screened against microarrays patterned with FOx conjugates for detection using label-free optical imaging. These experiences give students the ability to engage in undergraduate research at an early point in their academic career by replacing traditional laboratory activities with semester-long research projects. Nanoparticles (NPs) are widely used in a myriad of commercial and industrial products making their entry to the environment a likely event. NPs have unique physical-chemical properties that result from their small size and high surface area to volume ratio, making them highly reactive and potentially toxic. SYK is a protein tyrosine kinase that plays an essential role in the development and activation of immune cells. Its expression, however, is not limited to immune cells. SYK is expressed in a variety of epithelial cell types and epithelial-derived tumors. Reports regarding the role of SYK expression in these diverse cell types and tumors have been opposing. In breast cancer, SYK expression has been overwhelmingly associated with tumor suppression. Predictive models in microbiology are used for estimating the growth or survival of microorganism in a set of environmental conditions. A validated predictive model provides an alternative to extensive survival and shelf life studies. In this study, a predictive inactivation model for non-O157 shiga toxin producing Escherichia coli (STEC) in ground beef was developed. Six strains of non-O157 STEC; E. coli O26:H1, E. coli O45:H2, E. coli O103:H2, E. coli O111:H8, E. coli O121:H9, and E. coli O145: nonmotile, has similar pathogenicity as E. coli O157:H7 and can cause serious food borne illnesses. Since the 1970’s over 1,880 incidents have been documented in agricultural confined spaces of which 65% of all recorded incidents were grain entrapments and engulfments. There have been several studies conducted on the contributing factors behind these incidents; however, there have been very few attempts to understand the environmental, physiological or psychological factors the victims experience while entrapped, engulfed, or extricated.
