The Role Of The Unusual Threonine String In The Conversion Of Prion Protein, Romany Abskharon, Fei Wang, Kayla J. Vander Stel, Kumar Sinniah

The conversion of normal prion protein (PrP) into pathogenic PrP conformers is central to prion disease, but the mechanism remains unclear. The α-helix 2 of PrP contains a string of four threonines, which is unusual due to the high propensity of threonine to form β-sheets. This structural feature was proposed as the basis for initiating PrP conversion, but experimental results have been conflicting. We studied the role of the threonine string on PrP conversion by analyzing mouse Prnpa and Prnpb polymorphism that contains a polymorphic residue at the beginning of the threonine string, and PrP mutants in which threonine 191 …

Vocabulary Development In The Science Classroom: Using Hypermedia Authoring To Support English Learners, Robert Pritchard, Susan O'Hara

This study investigated the impact of authoring hypermedia projects on the academic vocabulary development of middle school, ESL students. Vocabulary definitions, inprocess verbalizations, observations and semi-structured interviews were the primary means of collecting data and assessing vocabulary growth. The results of this study indicate that hypermedia authoring had a positive impact on students' understanding of grade level, science concepts as well as on student engagement in and attitudes toward vocabulary building activities. Thus, the procedures implemented in this study provide a potential model for teachers to follow as they attempt to facilitate their students' vocabulary and concept development.

With an increasingly aging population, American society needs to more effectively address living options for the aging population in the present and future. Older adults have unique needs due to their age, physical and mental condition, varying needs for medical and health care and assistance with their daily activities. Freedom Village is an example of a very successful continuing care retirement community (CCRC) in Holland, Michigan. Using a case study approach and structured questions to ask two residents of Freedom Village, her aunt and uncle, over a period of several months, the author learned that they have lived very happily …

The Deepwater Horizon (DWH) blowout of 2010 exposed many coastal and marine ecosystems in the Gulf of Mexico (GoM) to the acute and chronic effects of crude oil. To date, no studies have investigated the toxicity of DWH oil on shallow reef-building corals despite the relative proximity of some reef sites to the DWH (Flower Garden Banks, TX) and the potential for downstream oil transport to other more distant sites (Pulley Ridge and Florida Keys, FL). Calcified marine organisms typically experience increased oxidative stress and changes in mineralization in response to ocean acidification and warming conditions. These effects could hinder the potency of animal weapons, such as the mantis shrimp’s raptorial appendage. The mechanical properties of this calcified weapon enable extremely powerful punches to be delivered to prey and aggressors. We examined oxidative stress and exoskeleton structure, mineral content, and mechanical properties of the raptorial appendage and the carapace under long-term ocean acidification and warming conditions. Many college students find themselves struggling with mental health problems (ACHA, 2015). Individuals with mental illness not only battle with the disease, but with the stigma society has developed toward them, leading to a diminished quality of life. Education and contact are two antistigma approaches utilized in this study (Corrigan & Watson, 2002). This thesis aims to understand players’ involvement in the video game narrative (i.e., narrative engagement and transportation) and its relationship with empathy and attitudes toward people with mental illness. Coastal zones located in populated and rapidly developing areas face high risks of natural and anthropogenic disasters. In this study, a framework was developed to determine the indicators of vulnerability to natural and anthropogenic disasters in Chuwei and Tamsui, two northern Taiwanese fishing villages in the coastal zone of the Tamsui River. The analytical hierarchy process (AHP) was used to determine the vulnerability indices of the locations, with experts evaluating the weights assigned to a range of criteria, namely hydrological data (such as sea surface temperature and sea level), stakeholder perceptions, and fishery data. Landscape features, interspecific introgression, and adaptation work in concert to shape the evolutionary history of a clade. Understanding the independent and cumulative consequences of these evolutionary processes on diversification is critical to revealing the origins of extant biodiversity. Studying these processes within rapid radiations, a significant contributor to global biodiversity, can provide powerful insight into the process of diversification. To assess how diversification is shaped by these evolutionary forces, I examined the biogeographic history, patterns of interspecific introgression and adaptation to high elevation in a recent, rapid radiation of finches, the South American siskins (Fringillidae: Spinus). Global climate change and anthropogenic activity have facilitated the movement and invasive potential of nonnative plants in native environments. These invasions can have negative effects on ecosystem diversity and function. The nonnative and invasive plant, Chinese Tallow (Triadica sebifera), has already invaded much of the south eastern US where it is outcompeting native tree species and changing ecosystem diversity in a variety of habitats. Leaf litter from the Chinese tallow has been shown cause changes in dissolved oxygen and pH in the aquatic environment. Turbidity is also affected when Chinese tallow litter is present in water. Segmentation of 3D/4D biological images is a critical step for a wide range of applications such as treatment planning, quantitative analysis, virtual simulations, and rendering visualizations. Automatic segmentation methods are becoming more reliable, but many experts still rely on manual intervention which makes segmentation a time and resource intensive bottleneck. Marking boundary contours in 3D images can be difficult when images are often noisy or the delineation of biological tissue is unclear. Non-parallel contours can be more accurate and reduce the amount of marking necessary, but require extra effort to ensure boundary consistency and maintain spatial orientation. Implementation of daily functions in humans crucially relies on both the bottom-up moment-to- moment processing of relevant input and output information as well as the top-down controls that instantiate and regulate goal-directed strategies. The current dissertation focuses on different systems of brain regions related to task control. Cortical oscillations are electrical activities with rhythmic and/or repetitive nature generated spontaneously and in response to stimuli. Study of cortical oscillations has become an area of converging interests since the last two decades and has deepened our understanding of its physiological basis across different behavioral states. Collective migration is the process by which cells organize individual motions to productively migrate as a group and plays a fundamental role in organism development, tissue regeneration, and cancer invasion. In development, coordinated migration facilitates the formation of complex organ structures and is required for proper dissemination of neural crest cells throughout an organism. After injury, this process allows breaches in epithelial layers to be repaired while maintaining tissue integrity, and in cancer, collective behavior enhances invasion of tumor cells into the surrounding tissue. One of the most successful and diverse systems involved in the maintenance of behavioral barriers between closely related animal species is pheromonal communication. In the fruit fly, contact chemosensation input is especially important during sexual decision-making as it allows for the sensing of sex and species-specific non-volatile cuticular hydrocarbons (CHCs), which function as insect pheromones. However, how pheromonal systems support the maintenance of mating barriers is puzzling since any change in either pheromone ligands or their cognate receptors would carry a fitness cost, which should be eliminated by stabilizing selection. Uterine contractions are tightly regulated throughout pregnancy, remaining relatively quiescent for the majority of pregnancy to promote fetal development, while becoming highly activated at term to effectively expel the fetus. Contractions are coordinated through electrical and mechanical coupling of myometrial smooth muscle cells (MSMCs). Ion channel activity, which alters the electrical activity of MSMCs, can directly affect the contraction profile. Cancers, including hematologic malignancies, arise as a result of the stepwise accumulation of mutations. Some early mutations that potentially initiate clonal expansion might exist in patients many years before they develop obvious disease symptoms. Therefore, identifying and characterizing these early mutations are critical to understanding the genetic basis of tumorigenesis. Here, we analyzed blood-derived DNA sequencing data from 2,728 individuals without apparent hematologic malignancies and identified 77 blood-specific mutations in 31 cancer-associated genes. Importantly, 83% of all mutations occurred in 19 genes that have been previously linked to hematological malignancies, such as DNMT3A, TET2, JAK2, and ASXL1. By investigating these …

This project focuses on structural and prosodic effects during reading, examining their influence on agreement processing and comprehension in native English (L1) and Spanish–English bilingual (L2) speakers. We consolidate research from several distinct areas of inquiry—cognitive processing, reading fluency, and L1/L2 processing—in order to support the integration of prosody with a cue-based retrieval mechanism for subject-verb agreement. To explore this proposal, the experimental design manipulated text presentation to influence implicit prosody, using sentences designed to induce subject-verb agreement attraction errors. Materials included simple and complex relative clauses with head nouns and verbs that were either matched or mismatched for number. …

Background: Hyperphosphorylation and aggregation of tau protein are the pathological hallmarks of Alzheimer’s disease and related tauopathies. We previously demonstrated that the microglial activation induces tau hyperphosphorylation and cognitive impairment via activation of p38 mitogen-activated protein kinase (p38 MAPK) in the hTau mouse model of tauopathy that was deficient for microglial fractalkine receptor CX3CR1.
Method: We report an isoform-selective, brain-permeable, and orally bioavailable small molecule inhibitor of p38α MAPK (MW181) and its effects on tau phosphorylation in vitro and in hTau mice.
