Purification Of 26s Proteasomes And Their Subcomplexes From Plants, Richard S. Marshall, David C. Gemperline, Richard D. Vierstra Oct 2016

Purification Of 26s Proteasomes And Their Subcomplexes From Plants, Richard S. Marshall, David C. Gemperline, Richard D. Vierstra

Biology Faculty Research

The 26S proteasome is a highly dynamic, multisubunit, ATP-dependent protease that plays a central role in cellular housekeeping and many aspects of plant growth and development by degrading aberrant polypeptides and key cellular regulators that are first modified by ubiquitin. Although the 26S proteasome was originally enriched from plants over 30 years ago, only recently have significant advances been made in our ability to isolate and study the plant particle. Here, we describe two robust methods for purifying the 26S proteasome and its subcomplexes from Arabidopsis thaliana; one that involves conventional chromatography techniques to isolate the complex from wild-type …

Co-Operative Collective Motion Of Benzoquinone Particles At The Air-Water Interface And Assessment Of Cd62l As A Protein Biomarker For Metastatic Bladder Cancer, Jennifer E. Satterwhite 4469944

Doctoral Dissertations

Active matter is an entity observed in both animate and inanimate systems and exhibits spontaneous movement. An internal mechanism generates the self-driven unit to convert potential energy into mechanical movement. Biomotors and organisms are biologically well-known active systems; however, their movements are quite a complex phenomenon and understanding their driving motive forces and thermodynamic principles are essential. Therefore, the first portion of this thesis focused on the dynamics of an active multiple particle system at the air-water interface.

Benzoquinone active particles were studied and remarkable locomotion phenomena were observed at the air-water interface. Self-motion was evaluated with multiple symmetric circular …

Forgetting And The Value Of Social Information, Benjamin James Abts Oct 2016

Forgetting And The Value Of Social Information, Benjamin James Abts

Theses

Information is everywhere in nature, however it can be deceitful or incorrect, so not all information should be used. Foraging pollinators utilize variable and ephemeral resources so learning about patch quality and nectar replenishment rates are essential to success and survival. However, remembering information after it is no longer relevant is not advantageous. It has been theorized that a pollinator’s memory should reflect their environment. Bumblebees are known to use both personal information (information gathered through trial and error) and social information (information gained through observations of or interactions with other animals or their products) in foraging decisions; however, it …

Mitochondrial Superoxide Signaling Contributes To Norepinephrine-Mediated T-Lymphocyte Cytokine Profiles., Adam J. Case, Colton T. Roessner, Jun Tian, Matthew C. Zimmerman

Journal Articles: Cellular & Integrative Physiology

Norepinephrine (NE) produces multifaceted regulatory patterns in T-lymphocytes. Recently, we have shown that NE utilizes redox signaling as evidenced by increased superoxide (O2●-) causally linked to the observed changes in these cells; however, the source of this reactive oxygen species (ROS) remains elusive. Herein, we hypothesized that the source of increased O2●- in NE-stimulated T-lymphocytes is due to disruption of mitochondrial bioenergetics. To address this hypothesis, we utilized purified mouse splenic CD4+ and CD8+ T-lymphocytes stimulated with NE and assessed O2●- levels, mitochondrial metabolism, cellular proliferation, and cytokine profiles. We demonstrate that the increase in O2●- levels in response to …

Effects Of Elaborative Interrogation After Reading Belief-Inconsistent Arguments And Need For Cognition On Argumentation And Topic Beliefs, Ruomeng Zhao

College of Education and Human Sciences: Dissertations, Theses, and Student Research

Constructing quality argumentation to justify one’s own beliefs on a topic is important both for a thorough topic understanding and the development of argumentation writing skills. Also, one’s change or retention of topic beliefs should be based on quality argumentation, such that the belief can be considered rational. The purpose of this study was to test whether a cognitive strategy, elaborative interrogation, can improve the understanding of belief-inconsistent arguments on a controversial topic and then improve argumentation quality, as well as result in reflective belief change. Elaborative interrogation is a cognitive strategy which prompts individuals to answer “why” questions on …

Using Lidar To Determine Early Successional And Shrubland Structure In Eastern Connecticut, Jacob P. Conshick Oct 2016

Using Lidar To Determine Early Successional And Shrubland Structure In Eastern Connecticut, Jacob P. Conshick

Master's Theses

Since the decline of agriculture in the state of Connecticut due to the opening of the Erie Canal, the land originally used for cropland reverted back to forest. During this time dense shrubland shade intolerant plants sprouted in the abandoned fields. The dense cover is known as early successional habitat and is utilized by many different species such as the New England cottontail, bobcat, and woodcock. These species use early successional habitat for a source of food, cover and shelter to survive. Currently disturbance events such as clearing land for agriculture, hurricanes, clear cuts, and fires are on a decline, …
