In her book How We Became Posthuman (1999), Katherine Hayles analysed the process through which the conception of the liberal humanist subject led the way to the posthuman subject, a subject who lives in complete entwinement with the digital. This process, however, was not innocuous: it made the (fallacious) perception that information could do without material instantiation pervasive within many fields of knowledge, a process that Hayles contends originates in the Macy Conferences and the evolution of cybernetic theory. This dissertation claims that the reciprocal exchange of voice—an element for constructing community and strengthening political recognition—may be fostered in small-scale farming communities by (1) the appropriation and transformation of information and communications technologies, (2) artistic intervention, and (3) cross-community research. My aim for That Dark Remembered Day was to create a work of fiction with a strong sense of the literary, one whose themes of violence, landscape and survivor guilt shone a light on the human condition, specifically the effects of post-war trauma on one family. The novel’s structure would be crucial in achieving its emotional heft, as each of the central characters is allocated at least one section, the reader experiencing events from disparate and increasingly illuminating perspectives. The main aim of this thesis was to determine how marine metazoans might persist as ocean acidification (OA) conditions intensify. This was done using a combination of field surveys, field transplants and laboratory experiments with polychaetes from a site where volcanically-derived CO2 gas bubbles through the seafloor and drives the seawater pH down, resulting in a marine ecosystem representative of global OA projections for, or before, the year 2100. My first objective was to identify phenotypes, or traits, associated with OA tolerance (Chapter 2 and 5). This thesis takes a theoretical framework constructed for transdisciplinary research within different natural science disciplines and investigates what kind of new knowledge is produced when this framework is applied to projects at the interface of art and natural science. The main case study is “Sauti ya Wakulima – The Voice of the Farmers”, which involves collaboration with another intervention artist, and with natural scientists and farmers. This research centres on the Balinese performance tradition known as topeng which translates as Balinese masked dance-drama. In Bali this genre is performed traditionally in spaces reserved for religious ceremonies. The research questions the extent to which, and how, it may be possible for a non-Balinese person to embody a culturally coded, sacred object (the mask) and how a woman is able to make meaning and express herself within a genre which is traditionally the preserve of men. More than sixty-five percent of people in the U.S. are considered overweight or obese. African-Americans in the U.S. have a higher risk of obesity than any other racial group. One way to reduce this statistic is physical activity. Recreational green spaces (parks) can serve as an avenue to complete the 150 min/wk of recommended physical activity for adults by the ACSM. Data from SISTAS and HEALS interventions, that recruited overweight/obese African-Americans from Columbia and Florence, SC, was used to assess the association of recreational green space (parks) around a residence and physical activity. Maternal smoke exposure produces long-term adverse cognitive and behavioral outcomes in offspring, including an increased likelihood of attention problems (e.g., attention deficit hyperactivity disorder; ADHD) and drug abuse. Preclinical research shows that gestational exposure to nicotine, the primary psychoactive compound in tobacco smoke, influences the neurodevelopment of attention and reward neuronal circuits. This study investigated hypotheses about five brain regions, to determine if prenatal nicotine (PN) exposure altered expression of nicotinic acetylcholine receptors (nAChRs). Failure of atherosclerotic plaques can lead to potentially life threatening clinical events such as myocardial infarction (MI), stroke, or transient ischemic attack (TIA). The most frequently described plaque failure mechanism is tensile rupture of the fibrous cap; however, often during angioplasty another plaque failure mechanism occurs in which the atherosclerotic plaque separates from the internal elastic lamina (IEL). This study aims assess the material strength of atherosclerotic lesions using mechanical concepts.

An important feature of such data is that the survival time of interest is usually not completely known but is censored due to the design of the study or an early dropout. I demonstrate that a member of the protein family A Disintegrin and Metalloprotease (ADAM), ADAM23, is expressed in the olfactory epithelium (OE) and may inhibit neural progenitor cell (NPC) proliferation. I generated a list of ten gene candidates and selected ADAM23 for further study, based on its conserved protein structure and accumulating evidence for its role in cell cycle progression of proliferative cells. I next determined ADAM23 protein localization using immunohistochemistry and measured both mRNA and protein expression post-lesion with RT-qPCR and Western blot. Our knowledge of avian Haemosporidian parasites has increased dramatically with the advent and increased used of molecular technologies. Yet Leucocytozoon, a common avian parasite, remains relatively understudied. Haemosporidian parasite diversity and prevalence has been shown to be dependent on several factors, including host family and habitat, on a microecological scale. In this study I screened 3,375 birds from sub-Saharan Africa for the presence of Leucocytozoon and demonstrate that the patterns of prevalence and diversity continue at a macroecological scale. Using molecular data, I found that the prevalence of Leucocytozoon varied between sampling areas from 3.9% to 62.64% and the diversity …

As many as 1 in 2 women will have at least one urinary tract infection (UTI) in their lifetime. UTIs can cause complications in pregnancy and decrease quality of life, and their treatment and prevention are expensive. Uropathogenic E. coli (UPEC) is the primary cause of UTI. The probiotic and bactericidal capacities of gut and vaginal Lactobacillus isolates have been studied, but the same attention has not been paid to urinary strains. Hepatocellular Carcinoma is the second most lethal cancer after pancreatic cancer. Unresectable HCC tumors carry a poor prognosis and few treatment options are available. The dismal prognosis is mainly due to limited therapy options and molecularly targeted therapy is deemed as solution. Here, we report a novel role of c-Abl in HCC development. We provide evidence of c-Abl activation in human HCC samples compared to normal liver. Using genetic and pharmacological tools, we show that c-Abl plays a vital role in HCC progression in vitro and in vivo. We have identified Axl as an effector in processes mediated by c-Abl. …

Misfolded, toxic forms of amyloid alpha synuclein (α-syn) have been implicated in Parkinson’s disease (PD) pathology and have been shown to spread throughout the brain in a prion-like manner, explaining the progressive loss of neurons in PD. The mechanisms of spread and cell death are unknown, however, evidence from previous studies suggests that neurons may undergo apoptosis due to the uptake of exogenous α-syn aggregate through the endocytic pathway and subsequent lysosomal rupture.

Estrogens are an important class of steroid hormones involved in many cellular signaling processes. Levels of estrogens fluctuate across the lifespan suggesting that they target intracellular mediators, such as microRNAs, to fine-tune homeostatic cellular processes. Previous work in our lab demonstrated that 17β-estradiol (E2), the predominant circulating estrogen, stabilized miR-181a in a neuronal cell line derived from rat hypothalamus. These results lead to the hypothesis that E2 regulated miR-181a stability by altering the pool of endogenous RNA and by recruiting PNRC2 away from the miRNA degradation machinery. The overall objective of this thesis work is to determine the mechanism by which celecoxib modulates Kv7 potassium channels in vascular smooth muscle cells. The central hypothesis of the project is that celecoxib modulates Kv7 potassium channels in vascular smooth muscle cells through inhibition of phosphodiesterase 4 (PDE4, which catalyzes the breakdown of cyclic adenosine monophosphate (cAMP)), thereby activating a cAMP/protein kinase A (PKA) signaling pathway. The aims of this thesis targeted different points in the cAMP/PKA pathway with pharmacological interventions using whole cell perforated patch clamp electrophysiology and biochemical methods.

The innate immune system is many organisms first line of defense against pathogenic insult or tissue damage. This defense strategy is intent on restoring homeostasis upon perturbation. Upon activation of the innate immune system in humans, an oligomeric protein complex termed the “Inflammasome” forms in myeloid cells. The canonical output of activation of any subset of inflammasome is Caspase-1-mediated secretion of pro-inflammatory cytokines IL1β and IL18. In this study, I argue that the appearance of anti-miscegenation writings in Ohio spiked during periods that saw massive threats to the notion of white male supremacy, such as the months just prior to the onset of the Civil War, several especially tense points during the Civil War and the Reconstruction period, and the early to middle years of the 1880s. During these times, Ohioans used at least one of three major rhetorical strategies—each of which coincided with a major trend in national events and politics—to justify and explain their anti-miscegenation attitudes.

This project concerns itself with the theoretical framework and application of ethnoarchaeological research methods in the Maya region. Following an in-depth discussion of ethnoarchaeology and its theoretical locus within archaeology as well as the transformations it has seen in recent year, the current work focuses on the following source- and subject-side cultural groups and phenomena: cave use at La Ventana and La Ventana Campana by Maya peoples from the Suchitepéquez and Sololá Department of Guatemala in comparison with Problematical Deposit 21 at Tikal, Petén, Guatemala. Ion channels play an important role in the swimming behavior of Paramecium tetraurelia. Electrophysiological experiments indicate that a calcium-dependent sodium channel exists in Paramecium, but a gene has not been identified for this channel. It is hypothesized that this channel evolved from a voltage-dependent sodium channel after a whole genome duplication event. In this paper, 13 candidate genes were identified from the Paramecium genome based on their similarity to a reference human voltage-dependent sodium channel. Bioinformatics analysis of the selectivity filter of these putative ion channel genes suggested that at least 8 of the candidate genes were sodium channels …

The BlackLivesMatter Movement has caught the media’s eye as it addresses racism in our criminal justice system. However, there seems to be a racial divide in support for this race-based movement, namely, black Americans tend to be supportive of the cause, and white Americans appear to be less so. Previous literature suggests that an emotional reaction to injustice, specifically moral shock, may trigger cross-racial support for race-based movements. In addition, racial attitudes can also be influential on cross-racial social movement support.
