Idiopathic scoliosis (IS) is the most common form of spinal deformity with unclear pathogenesis. In this study, we first reanalyzed the loci associated with IS, drawing upon previous studies. Subsequently, we mapped these loci to candidate genes using either location-based or function-based strategies. To further substantiate our findings, we verified the enrichment of variants within these candidate genes across several large IS cohorts encompassing Chinese, East Asian, and European populations. Consequently, we identified variants in the EPHA4 gene as compelling candidates for IS. To confirm their pathogenicity, we generated zebrafish mutants of epha4a. This dissertation grounded in a dual-theoretical framework of Bandura's Self-Efficacy Theory and the Black Panther Party’s Revolutionary Theory of CommunityEmpowerment presents a systematic review of the literature exploring how high school students impacted by trauma, poverty, and systemic inequities utilize self-efficacy to remain engaged in learning, especially when immediate access to school-based mental health support is limited. 150 research articles across educational, psychological and sociopolitical domains were screened and 28 articles were analyzed to answer the three guided research questions:

There is growing recognition that tropical forest restoration is key for sequestering carbon and enhancing ecosystem resilience. Soils, roots, and soil biota are central to ecosystem function and services, but belowground recovery is largely overlooked in restoration monitoring frameworks. The apicomplexan parasite Cryptosporidium parvum (Cp) is a major global contributor to infectious diarrhea in children and poses a serious threat to immunocompromised adults. This dissertation investigates Cp's unique life cycle, which alternates between sexual and asexual reproduction to achieve three critical objectives: producing oocysts capable of transmission, enhancing genetic variation through recombination, and generating sufficient progeny for survival. Sexual reproduction is essential for forming infectious oocysts that facilitate disease transmission. Yet, the process of meiosis in Cp has been historically understudied due to limitations of traditional in vitro culture methods. Using stakeholder theory, this article introduces a framework to inform decision making with respect to the regulation of on-premise signs. Because signage resides in a broader, shared environment and its regulation largely takes place at the local level of government, it can be considered unique among most other marketing-related communications. On-premise signs are important to individual businesses and the communities in which they are used, but at the same time, they are “out in the world,” cannot be avoided, and may not be relevant to or welcomed by many consumers who are exposed to them. Staphylococcus aureus (S. aureus) is an opportunistic pathogen that produces numerous immune evasion molecules that target various components of the host immune defense to cause infection. One of these is a family of virulence factors, Staphylococcal superantigen-like proteins (SSLs) with 14 members that exhibit host immune-evasion functions. SSL11 has been reported to inhibit neutrophil migration, preventing its recruitment to sites of infection. However, its role in vivo and effect on neutrophil phagocytosis have not been known. In its earliest form, the memory of the American Revolution was far from pure, riddled with partisan jabs and purposeful currents of present events. This, however, has received little attention even with the rise of memory studies within professional history. Scholars have addressed the popular memory of a variety of other pivotal moments from the Civil War to both World Wars. Revolutionary memory has been studied within the contexts of later eras, but almost never in the era immediately following the end of the war. Despite states having scored successes at the operational level, organized crime has proven too adaptable and too resilient to be seriously affected. Instead, ground has been lost politically, societally, and even territorially to armed gangs, cartels, and other illicit structures. The result is a steady erosion of the rule of law, of norms of sovereignty, of governance, and of human security.

The need for robotic systems capable of operating in confined, hazardous, or complex environments has driven interest in soft and legged platforms for industrial inspection and structural integrity assessment. This thesis presents the design and fabrication of a tentacle-like, soft-legged hexapod robot, which uses six cable-stiffened, pre-curved, tendon-driven continuum robot (TDCR) legs. These biologically inspired legs are designed to offer a balance between compliance and control, enabling the robot to navigate unstructured terrains and potentially grasp cylindrical structures such as pipes. In this study, core-shell microparticles were synthesized using coaxial electrospray techniques, with Zeolitic Imidazolate Framework-8 encapsulating either Rhodamine B or Doxorubicin in the core. ZIF-8 is a material well regarded for its effectiveness in drug delivery systems. Phase change materials are composed of a 4:1 mixture of lauric acid and stearic acid in the shell. The PCM chosen has a melting point close to physiological body temperature. The structural morphology and thermal stability of the microparticles were confirmed using Scanning Electron Microscopy, X-ray Diffraction, Differential Scanning Calorimetry, Thermogravimetric Analysis, and Infrared Spectroscopy. Photoplethysmography (PPG) is widely used to measure heart rate, blood oxygenation, and more recently, blood pressure. Implanted PPG systems offer the possibility to measure similar real-time measures of cardiovascular health, however, the detection method may vary due to a lack of capillary vessels for PPG sensors to observe in muscle tissue. To improve volumetric blood detection in large muscles, without relying on the capillary density of skin, a flexible PPG sensor was developed. Background/Objectives: Neuronal oscillations play a key role in the symptoms of Parkinson’s disease (PD). This study investigates the effects of random synaptic inputs, their correlations, and the interaction with synaptic dynamics and spike timing-dependent plasticity (STDP) on the membrane potential and firing patterns of subthalamic nucleus (STN) neurons, both in healthy and PD-affected states. Methods: We used a modified Hodgkin–Huxley model with a Langevin stochastic framework to study how synaptic conductance, random input fluctuations, and STDP affect STN neuron firing and membrane potential, including sensitivity to refractory period and synaptic depression variability. Though results are published and the component data is archived and made publicly available, distrust in and misunderstanding of science by the lay populace remains common. This work focuses on bolstering the outreach component of the International Tundra Experiment (ITEX)—Arctic Observing Network (AON) to increase public awareness, interactivity, and ultimately, the understanding of how the science of ITEX-AON takes place. Subunits of the SWI/SNF chromatin remodeling complex are altered in ~20% of human cancers. Exemplifying the alterations is the ARID1A mutation that occurs in ~50% ovarian clear cell carcinoma (OCCC), a disease with limited therapeutic options. Here, we showed that ARID1A mutations create a dependence on alanine by regulating alanine transporters to increase intracellular alanine levels. ARID1A directly repressed alanine importer SLC38A2 and simultaneously promoted alanine exporter SLC7A8. ARID1A inactivation increased alanine utilization predominantly in protein synthesis and passively through the tricarboxylic acid cycle. Indeed, ARID1A-mutant OCCCs were hyper-sensitive to inhibition of SLC38A2. In addition, SLC38A2 inhibition enhanced chimeric antigen …

A short, provocative review of Rutger Bregman’s Moral Ambition. I sketch his technocratic utopia—“Rutger-land”—a colder, centralized world where fossil fuels are banned, richer people are taxed into moral compliance, UBI prevails, and the landscape is “tobacco-free, malaria-free, poverty-free, and meat-free… just fields of kale and solar panels.” I argue Bregman’s approach is resolutely top-down—activism, lobbying, and policy over enterprise, churches, and grassroots civic life—misunderstanding how wealth is created and how real change happens. Against this centralized moralism, I make the case for decentralization, subsidiarity, and the quiet, dignified work of civil society.

Immune dysfunction and late mortality from multiorgan failure are hallmarks of severe sepsis. Arginine, a semi-essential amino acid important for protein synthesis, immune response, and circulatory regulation, is deficient in sepsis. However, arginine supplementation in sepsis remains controversial due to the potential to upregulate inducible nitric oxide synthase (iNOS)-mediated excessive nitric oxide (NO) generation in macrophages, leading to vasodilation and hemodynamic catastrophe. Citrulline supplementation has been considered an alternative to replenishing arginine via de novo synthesis, orchestrated by argininosuccinate synthase 1 (ASS1) and argininosuccinate lyase (ASL). Purpose
Body armor is used to protect the wearer from penetrating injuries. However, when the armor defeats the projectile, it deforms at high rates into the wearer, referred to as back face deformation (BFD). This deformation can cause a variety of superficial and internal injuries and consequently should be considered when designing body armor. However, current design standards do not adequately consider the effects of BFD, and new data are necessary to inform future design thresholds for BFD. Osteoarthritis is a degenerative joint disease that significantly impacts dog health, often leading to chronic pain, reduced mobility, and behavioral changes that may predispose dogs to secondary conditions such as urolithiasis. This study aimed to investigate the association between osteoarthritis and urolithiasis in dogs using radiographic imaging data collected from the Small Animal Teaching Hospital, Chulalongkorn University, Thailand, over a three-year period (January 2018 to December 2020). A total of 16,377 dogs were included, and cases of osteoarthritis, urolithiasis, or the concurrent occurrence of both conditions were identified through a systematic database review. Most studies on the character of Théoden in The Lord of the Rings center on Germanic heroic themes like courage and oath-keeping, often emphasizing his renewal by Gandalf at Meduseld and his death at the Battle of Pelennor Fields. This perspective overlooks Théoden's growth in wisdom and hope between these events. During this period, Théoden learns about the various forces in Middle-earth and contemplates his role in the broader world, leading to a profound sense of humility. Assessing Natural Resources Conservation Service (Nrcs) Impacts On Rice And Soybean Efficiency In The Mid-South, Ayodeji Samuel Ayiti

The study investigates how the Mid-South region of the U.S. rice and irrigated soybean production efficiency is influenced by (NRCS) incentives. To evaluate the effect of crop insurance, subsidies, and conservation incentives on lowering inefficiency, this study used a stochastic frontier model (SFA). The dataset covered the years 2010 – 2023, which allowed for a thorough evaluation of efficiency trends over time. While crop insurance programs reduce inefficiency in rice production, it shockingly increases inefficiency in soybean production, according to the study, which highlights the disparate effects of net support (insurance-subsidy).

Lack of access to sanitation is a challenge that persists globally, with low sewerage connection rates in many low- and lower-middle-income countries. Engineered nonsewered sanitation (NSS) technologies can meet treatment requirements without sewers, but their relative sustainability varies across potential deployment sites. Here, we characterize the costs and carbon intensity (CI) of three emerging NSS technologies, two community reinvented toilets (CRTs) and one Omni Processor (OP), across 77 countries, identify sustainability performance typologies, and map typology prevalence in countries across the globe. The Cre/loxP system is widely used for site-specific genetic manipulation in mice. The PgrCre mouse model, where Cre recombinase is driven by the progesterone receptor promoter, is commonly used for gene ablation in Pgr-positive uterine cells. However, the PgrCre is active in the neonatal uterus and does not allow temporal control. To enhance the functionality of the PgrCre mouse, we generated and characterized an inducible PgriCreERT2 mouse, in which iCreERT2 is inserted downstream of the endogenous Pgr promoter. PgriCreERT2 mice crossed with Rosa26-CAG-LSL-Sun1-sfGFP-myc reporter mice demonstrated tamoxifen-dependent recombination in uterine stromal fibroblasts and a subset of epithelial cells.
