Aminoacyl-tRNA synthetases (AARSs) are a superfamily of enzymes responsible for the faithful translation of the genetic code and have lately become a prominent target for synthetic biologists. Our large-scale analysis of >2500 prokaryotic genomes reveals the complex evolutionary history of these enzymes and their paralogs, in which horizontal gene transfer played an important role. These results show that a widespread belief in the evolutionary stability of this superfamily is misconceived. Although AlaRS, GlyRS, LeuRS, IleRS, ValRS are the most stable members of the family, GluRS, LysRS and CysRS often have paralogs, whereas AsnRS, GlnRS, PylRS and SepRS are often absent …

81 Pseudomonas aeruginosa from Guangdong Provincial Institute of Food Inspection were identified by 16S rRNA sequence analysis. The DNA was isolated and the sequences of 16S rRNA gene were amplified by PCR with the bacterium universal primers, and then the PCR products were sequenced after 2% agarose gel electrophoresis. Moreover, the corrected sequences were aligned with Clustal X and the phylogenetic tree was constructed by MEGA5.1. Consequently, the identified results of the 81 strains confirmed their original identification before. A method for determination of vanillin and ethyl-vanillin in milk powder was established by auto headspace solid-phase micro-extraction coupled with gas chromatography-mass spectrometry (GC—MS), and the influence factor were also studied, such as solid phase micro-extraction head, time and temperature of adsorption and desorption. As a result, the limits of determination (LOD) of vanillin and ethyl-vanillin were 0.5 mg/kg. The spiked recovery was between 90.0% and 100%, and the RSD ranged from 1.9%~4.1%(n=6). This method could eliminate the interference of complicated matrix, conveniently, quickly and exactly, which was applicable for quick determination of vanillin and ethyl-vanillin content in milk powder.

On-Line Determination Of Emergency Pollution Of 3-Nitropropionic Acid In Drinking Water By Flow Injection Chemiluminescence, Zhang Xuhui, Tang Shuze, Deng Xiaoling, Zhu Yao, Wu Shizheng

On the basis of inhibition of chemiluminescence reaction of luminol and potassium permanganate in sodium hydroxide medium by 3-nitropropionic acid, a novel flow-injection chemiluminescence method for determination of 3-nitropropionic acid in drinking water was developed. A linear range for 3-nitropropionic acid determination was 1~150 mg/L. The detection limit of 3-nitropropionic acid was 0.28 mg/L and relative standard deviation (RSD) was 1.9%, with the recovery of 76.5%~93.8%. This online determination system showed a good accuracy and selectivity and has great potential application in rapid detection and alarm of emergency pollution of 3-nitropropionic acid in drinking water.

Copyright © 2015 Wolters Kluwer Health, Inc. The Caenorhabditis elegans model is a rapid and inexpensive method to address pharmacologic questions. We describe the use of C. elegans to explore 2 pharmacologic questions concerning candidate antiobesity drugs and illustrate its potential usefulness in pharmacologic research: (1) to determine a ratio of betahistine-olanzapine that blocks the olanzapine-induced intestinal fat deposition (IFD) as detected by Nile red staining and (2) to identify the mechanism of action of a pharmaceutical candidate AB-101 that reduces IFD. Olanzapine (53 mg/mL) increased the IFD (12.1 6 0.1%, P < 0.02), which was blocked by betahistine (763 mg/mL, 39.3 6 0.01%, P < 0.05) in wild-type C. elegans (N2). AB-101 (1.0%) reduced the IFD in N2 (P < 0.05), increased the pharyngeal pumping rate (P < 0.05), and reversed the elevated IFD induced by protease inhibitors atazanavir and ritonavir (P < 0.05). AB-101 did not affect IFD in a ACS null mutant strain acs-4(ok2872) III/hT2[bli-4(e937) let-?(q782) qIs48](I;III) suggesting an involvement of the lipid oxidation pathway and an upregulation of CPT-1. Our studies suggest that C. elegans may be used as a resource in pharmacologic research. This article is intended to stimulate a greater appreciation of its value in the development of new pharmaceutical interventions.

I made numerous mistakes during my years in grade and middle school. These issues eventually led me to dropping-out in the ninth-grade. But my problems in the public school system weren’t the only area that I struggled in. I also made reckless choices in my social life. Linking Transcriptional And Physiological Responses To High Light Stress In A Wild-Type And A Photomorphogenic Tomato Mutant, Jordyn Radke

Tomato (Solanum lycopersicon) is the leading vegetable crop worldwide and an important part of a healthy human diet. In this study we used Next-Generation Sequencing, metabolomics and gene ontologies to study the effects of high light stress on the photomorphogenic hp-2dg tomato mutant. Wild-type (cv. ‘Manapal’) and mutant plants were grown for six weeks under low light and transferred to high light for one week. Illumina-based RNA Sequencing was performed and analysis of differential gene expression identified over 650 transcripts that were strongly regulated by light intensity (FC>4, p2,p<0,01). HPLC analysis of tomato leaves showed differential accumulation of phenylpropanoid metabolites such as chlorogenic acid, rutin, and caffeoylglucaric acid which was strongly dependent on genotype and light condition. Our findings show insightful differences between the wildtype and the photomorphogenic mutant as well as novel genes involved in high light adaptation in plants.

The organism is one of the fundamental concepts of biology and has been at the center of many discussions about biological individuality, yet what exactly it is can be confusing. The definition that we find generally useful is that an organism is a unit in which all the subunits have evolved to be highly cooperative, with very little conflict. We focus on how often organisms evolve from two or more formerly independent organisms. Two canonical transitions of this type—replicators clustered in cells and endosymbiotic organelles within host cells—demonstrate the reality of this kind of evolutionary transition and suggest conditions that …

Southern cooking is fluid–ever-changing, influential, and influenced by both Southerners and non-Southerners alike. In this thesis, I explore what constitutes Southern cooking. The foods, cooking methods, and acquisition of foodstuffs and ingredients influence Southerners as cooks and the communities we live in. In the following pages, I’ve connected primary research via interviews with existing literature about Southern foods, cooking, techniques, lifestyles, and cultures. My primary research is derived from authentic interviews with real Southerners. The results from my research are not groundbreaking, but they do serve to help Southerners understand their relationship to and with Southern food and Southern cooking.

River otters (Lontra Canadensis) were extirpated in Nebraska by the early 1900’s, but in 1986, the Nebraska Game and Parks Commission (NGPC) began reintroduction efforts. Following reintroductions, the otter was listed as a tier-1 at-risk species in Nebraska. With increasing otter populations, NGPC is evaluating a de-listing plan. In order to inform de-listing efforts, I surveyed Nebraska’s rivers documenting otter sign and used modeling techniques to estimate otter distribution and habitat suitability.
Otter sign surveys were conducted on the navigable rivers of Nebraska. Occupancy modeling techniques were used to examine patterns in otter detections. The best model incorporated distance …

Summary: 1. Warming temperatures cause changes in growing seasons and prey that drive earlier breeding by birds, especially dietary specialists within homogenous habitat. Less is known about how generalists respond to climate-associated shifts in growing seasons or prey, which may be occurring at different rates across land cover types used by predators. 2. We studied whether breeding phenology of a generalist predator, the American kestrel (Falco sparverius), in a mosaic of non-irrigated shrub/grasslands and irrigated crops and pastures, was associated with changes in growing seasons and, presumably, prey abundance. A myoelectric-controlled prosthesis holds the promise of renewed quality of life for the amputee population. When it comes to prosthesis control, muscle fatigue presents a significant challenge for all task discrimination algorithms due to changed muscle activation patterns and significant shifts in the spectral contents of the surface electromyogram (sEMG) signals. The effect of muscle fatigue on the classification accuracy of two supervised machine-learning algorithms, i.e. the linear discriminant analysis (LDA), and the muscle synergy based task discrimination (MSD) was investigated. Objective—Breastfeeding is linked to lower rates of childhood obesity. Human milk contains cortisol, known to regulate glucose storage and metabolism. We aimed to test the hypothesis that early exposure to cortisol in human breast milk helps to modulate infant BMI trajectories over the first two years of life.
Methods—Growth curve modeling was used to examine whether infant exposure to cortisol in human milk at 3 months predicted changes in child body mass index percentile (BMIP) at 6, 12, and 24 months of age in 51 breastfeeding mother-child pairs.

In order to investigate the beef quality changes under handling cattle in different platform slope, Twenty simon tal beef cattle were randomly divided into 4 groups, the cow is respectively loading and unloading through (30,40,50 °) slope, then cattle are slaughtered. The pH value, flesh-colored, drip loss percentage, cooking loss percentage and shear force of postmortem beef were measured respectively. Results showed, with the increase of loading platform slope, pH values, L* value and a* value of postmortem beef significantly decreased (P<0.05), while shear force, b* value, drip loss and cooking loss of postmortem beef significantly increased (P<0.05). This study indicated that beef quality was better through handling cattle in 30 degrees platform slope, and it should be reduced platform slope in cattle actual production thereby improve beef quality.

Bright Along The Body, Ashley Michelle Roach

Bright Along the Body is a collection of poetry that explores the nature of desire within the liminal space of marriage. The poems fuse domestic, garden, and urban wildlife imagery to explore the speaker's conflicting physical, spiritual, and emotional desires. The speaker subverts her own longings by cultivating a psychological space that is beautiful and lonely, representing transcendence that may be read as dissociation. Feminist themes of safety and independence inform the collection, which is sectioned in three parts to represent conflict, interiority, and resolution. The poems are stylistically diverse and music-rich, showing affinity for both contemporary poetry and tradition.
