Bone development and length relies on the growth plate formation, which is dependent on degradative enzymes such as MMPs. Indeed, deletion of specific members of this enzyme family in mice results in important joint and bone abnormalities, suggesting a role in skeletal development. As such, the control of MMP activity is vital in the complex process of bone formation and growth. We generated a transgenic mouse line to overexpress TIMP3 in mouse chondrocytes using the Col2a1-chondrocyte promoter. This overexpression in cartilage resulted in a transient shortening of growth plate in homozygote mice but bone length was restored at eight weeks …

Recollection and familiarity are independent processes that contribute to memory performance. Recollection is dependent on attentional control, which breaks down in early-stage Alzheimer disease (AD), whereas familiarity is independent of attention. The present study examines the sensitivity of recollection estimates based on Jacoby’s (1991) process dissociation procedure to AD-related biomarkers in a large sample of well-characterized cognitively normal older adults (N = 519) and the extent to which recollection discriminates these individuals from individuals with very mild symptomatic AD (N = 64). Motor systems must adapt to perturbations and changing conditions both within and outside the body. We refer to the ability of a system to maintain performance despite perturbations as “robustness,” and the ability of a system to deploy alternative strategies that improve fitness as “flexibility.” Different classes of pattern-generating circuits yield dynamics with differential sensitivities to perturbations and parameter variation. Depending on the task and the type of perturbation, high sensitivity can either facilitate or hinder robustness and flexibility. The Alaska Native Claims Settlement Act (ANCSA) was passed in 1971 to extinguish aboriginal rights of Alaska Natives and provide compensation for those rights extinguished. Instead of vesting assets (land and money) in tribal governments, Congress required the formation of Alaska Native corporations to receive and hold these assets. A major flaw in the settlement was the failure to provide statutory protections for the aboriginal hunting, fishing, and gathering rights extinguished by ANCSA. Moreover, while ANCSA did not directly address Alaska Native tribal status or jurisdiction, the Supreme Court interpreted the Act to terminate the Indian country status of ANCSA …

This paper analyzes the degree to which the administration of Title VIII of the Alaska National Interest Lands Conservation Act (ANILCA) of 1980 protects customary and traditional hunting and fishing by Alaska Natives and their tribal communities. A recent Memorandum of Understanding (MOU) entered into by the United States Fish and Wildlife Service (USFWS) and the Kuskokwim Inter-Tribal Fish Commission (KRITFC) for co-management of subsistence fisheries will be used as a means to analyze the issue. This paper concludes with suggestions for improving the administration of Title VIII to better secure Alaska Native and Tribal rights for self-determination.

Rift Valley fever (RVF) outbreaks are recurrent, occurring at irregular intervals of up to 15 years at least in East Africa. Between outbreaks disease inter-epidemic activities exist and occur at low levels and are maintained by female Aedes mcintoshi mosquitoes which transmit the virus to their eggs leading to disease persistence during unfavourable seasons. Here we formulate and analyse a full stochastic host-vector model with two routes of transmission: vertical and horizontal. By applying branching process theory we establish novel relationships between the basic reproduction number, R0, vertical transmission and the invasion and extinction probabilities. China is identified as posing a key challenge to US national security interests in cyberspace. These threats are incurred across the spectrum of conflict, ranging from low-level crime, to network penetration, to cyberattacks that have the potential to cause major physical destruction. Thus far, the majority of strategic assessments of China’s cyber capabilities have focused on the role of the People’s Liberation Army (PLA), which is officially tasked with undertaking offensive operations in cyberspace.[1] However, China does not employ its cyber capabilities in isolation. In English You Read With A Stopwatch: A Journey Towards Biliteracy In Two Older Adopted Salvadoran Children, Mary A. Petron, Barbara J. Greybeck

This longitudinal case study provides an in-depth exploration of the journey toward bilingualism and biliteracy of two older adopted Salvadoran siblings in U.S. schools. Data sources include observations in the home and school, interviews, written artifacts, field notes, and various reading test scores. Analysis suggests that literacy instruction in English tended to focus more on phonics and fluency than comprehension and vocabulary, and it assumed a level of oral proficiency in English that neither of the children had. Spanish literacy instruction was aimed toward children of Mexican origin that was neither culturally relevant nor geared toward their specific language needs. …

Home To School Transitions: A Guatemalan Family Portrait, Lisa Crayton

This study applies a sociocultural perspective to examine the home to school transitions in literacy achievement of three low-income children from Guatemala. Through participant observation and informal conversations with the family during home visits, two factors appeared to influence the literacy development of the family's young children: bilingualism and cultural assimilation. Investigating the home literacy environment of an immigrant family provides an insider's perspective of the life experiences of children from non-mainstream homes. Understanding their home reading and writing experiences, and their transition to school literacy, is valuable for reading teachers facing increasingly diverse students from multicultural backgrounds.

Taking the highway along the California coast and swinging inland into one of the state's agricultural belts, the hills appear golden in the distance, spotted with gnarled oak trees. Vineyards rise up on either side of the highway, and occasionally cowboys may be seen in the distance herding grazing cattle. Yet as clouds of dust rise from the fields in this agricultural community, the idyllic scene fades dramatically in the town of Rancho Benito, a community wearing the signs of the hard economic times. Noise induced hearing loss (NIHL) remains a severe health problem worldwide. Current noise metrics and models have assessment limitations on gradually developing NIHL (GDNIHL). In this study, we apply a complex velocity level (CVL) auditory fatigue model to quantitatively assess the impact of basilar membrane (BM) movement on GDNIHL. The transfer functions of chinchilla and human auditory systems, including the triple-path nonlinear (TRNL) filters to simulate the inner ear responses, are applied to obtain BM velocity distribution. I characterize the entire class of consumption rules for finite-horizon models in which consumption is proportional to lifetime wealth. Any such rule can be obtained from a preference model with CRRA period utility. In a steady state with constant interest rates, a proportional consumption rule can be derived from a model with time-consistent preferences or from a model with possibly time-inconsistent preferences in which a household continually reoptimizes future utility discounted relative to the present instant. These two preference models will only coincide for the special case when the discount function is exponential. More generally, there will be two distinct …

Quantum biology is the study of quantum effects on biochemical mechanisms and biological function. We show that the biological production of reactive oxygen species (ROS) in live cells can be influenced by coherent electron spin dynamics, providing a new example of quantum biology in cellular regulation. ROS partitioning appears to be mediated during the activation of molecular oxygen (O2) by reduced flavoenzymes, forming spin-correlated radical pairs (RPs). We find that oscillating magnetic fields at Zeeman resonance alter relative yields of cellular superoxide (O2•−) and hydrogen peroxide (H2O2) ROS products, indicating coherent singlet-triplet mixing at the point of ROS formation. This research uses an experimental design to study if the pattern and positioning of rewards influence the amount of effort participants put in. The three key hypotheses tested here are: 1) are people more likely to complete a task if the incentives are given in more regular intervals, 2) do uncertainty of reward timing hurt or help in maintaining motivation, 3) is intrinsic motivation more influential than the patterns in which incentives are structured? Enlightenment emphasis on rationalism in philosophy and the arts prefigures Samuel Taylor Coleridge’s and William Wordsworth’s Romantic recovery of a subject’s empirical relationship to nature and the phenomenal world. Coleridge and Wordsworth respond to philosophical precedents that emphasize rationalism and the autonomy of a subject while introducing empiricism and sensation as primary components of the speaker’s experience. The poets delineate a fluid shift from the Enlightenment to Romanticism through an interchangeable reliance on Kantian and Burkean philosophical methods. The Biopharmaceutics Classification System (BCS) classifies pharmaceutical compounds based on their aqueous solubility and intestinal permeability. The BCS Class III compounds are hydrophilic molecules (high aqueous solubility) with low permeability across the biological membranes. While these compounds are pharmacologically effective, poor absorption due to low permeability becomes the rate-limiting step in achieving adequate bioavailability. Several approaches have been explored and utilized for improving the permeability profiles of these compounds. The approaches include traditional methods such as prodrugs, permeation enhancers, ion-pairing, etc., as well as relatively modern approaches such as nanoencapsulation and nanosizing. The purpose of this curriculum development Capstone was to create a unit of lessons to address how we educate children that live in a water rich environment about the need for water conservation. The curriculum contains lessons in a kit that educate children about water cycles, water sheds, water run-off, water pollution, and water conservation. Methods of teaching include hands-on lessons, small and large group activities that incorporate literature. The overall goal is to create curriculum that would foster a sense of water conservation stewardship in students. Included with this curriculum are copies of handouts, assessments, and literature.

We investigate the effectiveness of a Hidden Markov Model (HMM) with random restarts as a mean of breaking a homophonic substitution cipher. Based on extensive experiments, we find that such an HMM-based attack outperforms a previously de- veloped nested hill climb approach, particularly when the ciphertext message is short. We then consider a combination cipher, consisting of a homophonic substitution and a column transposition. We develop and analyze an attack on such a cipher. This attack employs an HMM (with random restarts), together with a hill climb to recover the column permutation. We show that this attack can succeed on …

The development of a novel tuberculosis vaccine is a leading global health priority. SRL172, an inactivated, whole-cell mycobacterial vaccine, was safe, immunogenic and reduced the incidence of culture-confirmed tuberculosis in a phase III trial in HIV-infected and BCG immunized adults in Tanzania. Here we describe the immunogenicity and protective efficacy of DAR-901, a booster vaccine against tuberculosis manufactured from the same seed strain using a new scalable method.

Generalized arterial calcification of infancy (GACI) is an autosomal recessive disorder characterized by early onset of extensive mineralization of the cardiovascular system. The classical forms of GACI are caused by mutations in the ENPP1 gene, encoding a membrane-bound pyrophosphatase/phosphodiesterase that hydrolyzes ATP to AMP and inorganic pyrophosphate. The asj-2J mouse harboring a spontaneous mutation in the Enpp1 gene has been characterized as a model for GACI. These mutant mice develop ectopic mineralization in skin and vascular connective tissues as well as in cartilage and collagen-rich tendons and ligaments. The comprehensive understanding of cellular signaling pathways remains a challenge due to multiple layers of regulation that may become evident only when the pathway is probed at different levels or critical nodes are eliminated. To discover regulatory mechanisms in canonical WNT signaling, we conducted a systematic forward genetic analysis through reporter-based screens in haploid human cells. Comparison of screens for negative, attenuating and positive regulators of WNT signaling, mediators of R-spondin-dependent signaling and suppressors of constitutive signaling induced by loss of the tumor suppressor adenomatous polyposis coli or casein kinase 1α uncovered new regulatory features at most levels of the …

This study was part of a larger research intervention that uses motivational interviewing (MI) as part of an in-school substance abuse intervention in local high schools in the greater Seattle area. Our aim was to test hypothesized relationships between marijuana use, other delinquent behavior, and neutralization techniques used by participants and determine their impact on the effectiveness of an MI-based intervention. Hypotheses were that neutralization technique use would decrease the effectiveness of an MI intervention due to the conflicting cognitive processes of justification and developing discrepancy. This paper examines the effect of message characteristics on donation behavior using an economic model of giving. The utility of giving can come from one’s own contribution and possibly from the combined contributions of others. Donors are assumed to be constrained utility maximizers, and the message attributes affect the degree to which they react altruistically or egoistically. The model is estimated with data from an incentive-aligned study of South Korean consumers, and implications for message optimization and donor targeting are explored.
