Atmospheric vertical velocities and advective tendencies are essential as large-scale forcing data to drive single-column models (SCM), cloud-resolving models (CRM) and large-eddy simulations (LES). They cannot be directly measured or easily calculated with great accuracy from field measurements. In the Atmospheric Radiation Measurement (ARM) program, a constrained variational algorithm (1DCVA) has been used to derive large-scale forcing data over a sounding network domain with the aid of flux measurements at the surface and top of the atmosphere (TOA). We extend the 1DCVA algorithm into three dimensions (3DCVA) along with other improvements to calculate gridded large-scale forcing data. We also introduce …

Mechanical Heart Valves (MHV) provides life-saving solutions to patients suffering from cardiovascular diseases. But these devices are often plagued by non-physiological flow patterns resulting in increased shear stress conditions. This leads to platelet damage, a precursor of thromboembolism, thus impeding the device usability and demanding lifelong anticoagulation treatments. While the flow and stresses are in the micron level, the platelet behavior is at the nm level, thus presenting a major computational challenge for simulating this multiscale multi-physics modeling problem. Manufacturing industries have evolved tremendously in the past decade with the introduction of Additive Manufacturing (AM), also known as 3D Printing. The medical device industry has been a leader in adapting this new technology into research and development. 3D printing enables medical devices and implants to become more customizable, patient specific, and allows for low production numbers. This study compares the mechanical and thermal properties of traditionally manufactured parts versus parts manufactured through 3D printing before and after sterilization, and the ability of an FDM printer to produce reliable, identical samples. Chemotherapy is one of the most commonly used cancer treatments. However, many side effects could occur due to the non-specific bindings of extremely toxic anti-cancer drugs to healthy cells and organs. Targeted delivery has been developed to enhance the selectivity of drug delivery vehicle and minimize side effects of chemotherapy. In this study, the hydrophobically modified glycol chitosan (HGC) micelle was further modified with a biotin-avidin-biotin (B-A-B) strategy to improve the tumor-selective ability of HGC. Our breath is a very complex mixture of chemical compositions. Usually, the majority of our exhaled breath consists of nitrogen, oxygen, carbon dioxides, water and inert gases. The remaining small fraction of breath contains thousands of components in trace amount, including inorganic gas molecules like nitric monoxide and diverse kinds of volatile organic compounds (VOCs). The concentrations of our exhaled gases are at a relatively stable level, though minor fluctuations may occur at different time of the day and they are also linked to the body status. This dissertation focuses on the use of composite materials for environmental applications. For the first time, applications of both fresh and aged concrete as inexpensive adsorbents for nitrogen dioxide (NO2) removal is demonstrated. Concrete is the most widely used composite material of the modern era. Cement manufacturing (a major component of concrete) is considered to be one of the leading contributors to air pollution, resulting in 7% of the global carbon dioxide emissions along with a number of other harmful pollutants such as oxides, mercury and particulates. Thermal Spray (TS) coatings have seen extensive application as protective surfaces to enhance the service life of substrates prone to damage in their operating environment (wear, corrosion, heat etc.). Boron phosphide (BP), a light III-V indirect semiconductor compound with medium band gap of 2.0 eV, has emerged as a promising material with its applications in solid-state neutron technology and semiconductor devices in extreme conditions. Since bulk BP is unavailable, heteroepitaxial film growth is being attempted on various substrates including silicon, silicon carbide (4H, 6H, 3C polytypes), AlN and sapphire of different orientations to determine the best substrate for growing high quality thin films. Stress-assisted grain growth in nanocrystalline metals transpires collectively with a number of competing deformation mechanisms. In this study, molecular dynamics simulations of surface nanoindentation were performed to quantify the plastic strain distribution among competing mechanisms as a function of grain size and temperature during stress-assisted grain growth in nanocrystalline Ni and a Ni-1 at. % P alloy. Under identical conditions of rate and temperature in nominally the same grain size structure, stress-assisted grain growth found to be prevalent in pure nanocrystalline Ni was virtually absent in the Ni-P alloy with P enriched grain boundaries. Interest in nonlinear sigma models with target space a symmetric space of positive curvature comes from both mathematics and physics. In this dissertation we focus on the two-dimensional nonlinear sigma model with target space the k-dimensional projective space P^k. We study the properties of a finite-dimensional approximation, which we denote by L_N(P^k), to the loop space L(P^k), and construct resolutions of singularities for these truncated loop spaces. We also look into the geometries of such spaces and propose a conjecture about the mass gap of the Schrodinger operator H on L_N(P^k) with analytic formula. | 55 pages

The first story begins with a question of Steenrod. He asked if the product in the cohomology of a triangulated space, which is associative and graded commutative, can be induced from a cochain level product satisfying the same two properties. He answered it in the negative after identifying homological obstructions among a collection of chain maps he constructed. Using later language, his construction could be said to endow the simplicial chains with an $E_\infty$-coalgebra structure. A generic smooth immersed closed curve in the 2-sphere has a finite number of circles tangent to it in three points. We present a classification of these tritangent circles and study how each class changes during deformations of the curve. We relate these classes to finite type invariants of the curve and derive a relation among the numbers of tritangent circles in the classes and finite type invariants of the curve. | 39 pages

In this dissertation we prove certain deformation behaviors of Kahler-Einstein metrics with singularities. Based on smooth approximation and a priori estimates, we first give a new proof to Donaldson's Openness Theorem of deforming the cone angle of conical Kahler-Einstein metric on smooth Fano manifold, and prove a direct generalization to conical Kahler-Einstein metrics along simple normal crossing pluri-anti-canonical divisors on smooth Fano manifold. Then we use continuity method to study the deformation of weak conical Kahler-Einstein metric on Q-Fano variety. How do contemporary artworks address and give visibility to the wounded body without reinvigorating trauma, without repeating the spectacular and homogenizing strategies used by the media? A model's validity, or its goodness-of-fit to the real world system, is commonly assessed by quantifying the level of agreement between the observed data and their corresponding model-simulated outputs. However, the observed data could be uncertain given inaccuracies in the observational tools and techniques while the model-simulated outputs may be incomparable since models are simplified versions, and not exact replicas, of the real world system. Westvaco Inspirations was a promotional magazine first published in 1925 by the West Virginia Pulp and Paper Company. The main purpose of the magazine was to promote paper and to show an intricate relationship between print, paper and design. Bradbury Thompson was the designer for the magazine from 1939 to its final year of publication in 1962. Protein O-fucosyltransferase 2 (POFUT2) elongates Thrombospondin Type 1 Repeats (TSR) with an O-linked fucose. Forty-nine potential proteins are modified by POFUT2 in the endoplasmic reticulum. Nearly half the POFUT2 targets belong to the A Disintegrin and Metalloprotease with ThromboSpondin type-1 motifs (ADAMTS) or ADAMTS-like family of proteins. One of POFUT2’s target proteins, ADAMTS9, contains 15 TSRs and is reported to be embryonic lethal around the same time as our Pofut2 LoxP mutants. In this study, we compared the Pofut2 and Adamts9 knockout phenotypes and used tissue specific deletion of Pofut2 to define the role of Pofut2 during gastrulation. Knockout of …

The use of cyclodextrins have shown promise to exchange lipids of model membranes in various conditions. Several types of cyclodextrins have been shown to exchange lipids in different conditions. In this study, we used Methyl-alpha cyclodextrin (MαCD) to facilitate lipid exchange between donor multilamellar vesicles (MLVs) containing brain SM and large unilamellar vesicle acceptors (LUVs) composed of POPE, POPS and cholesterol. Vps34 is the sole member of the Class III phosphoinositide 3-kinases (PI3Ks) identified in mammals thus far. It phosphorylates phosphatidylinositol (PI) to generate PI(3)P on intracellular membranes. Previous work has suggested that Vps34 is important for the protein degradation pathways of autophagy and endocytosis, as well as nutrient-induced mTOR signaling. However, due to the pluripotent nature of available PI3K inhibitors, a clear understanding of the functions of mammalian Vps34 remains to be illustrated. Although they predominantly function in the animal germline, increasing evidence has pointed to their involvement in somatic cells. Some flatworms possess a unique somatic stem cell system named neoblasts. Mitochondria are dual membrane-bound organelles found in the cytoplasm of all eukaryotic cells. Human mitochondria contain a 16.5 kb double-stranded DNA genome that encodes thirteen essential protein subunits of the oxidative phosphorylation system, as well as the two ribosomal RNAs and 22 transfer RNAs needed to translate the thirteen messenger RNAs. Oxidative phosphorylation produces the majority of energy needed for cells to sustain life, and this process is strictly dependent on expression of the mitochondrial genome. During neuronal development, the neuronal cell polarizes to specify the structures of axons and dendrites. This polarized architecture regulates the input/output functions of the neuronal cell. In my thesis, I studied the molecular mechanisms of dendrite formation. My main hypothesis was that dendrite specification results from local elevation of the cGMP signal. Soluble guanylyl cyclase (sGC) is the major enzyme for the synthesis of cGMP in the embryonic brain. DNA Polymer dynamics are fundamental to the function of biological systems. Examples include gene regulation, cell division, threading and transport through pores. We address the limitations of previous experimental studies of polymer dynamics by designing, building, and characterizing a novel, custom Fluorescence Cross-Correlation Spectroscopy (FCCS) system. We detail the optical considerations for quantitatively describing and shaping the confocal volume. The hardware is supplemented by a robust theoretical foundation of correlation analysis and how it applies to polymer dynamics. Assembly of carbon nanomaterials into three-dimensional (3D) architectures is necessary to harness their unique physiochemical properties for tissue engineering and regenerative medicine applications. In this dissertation, fabrication and characterization of 3D chemically crosslinked macro-sized (5-8 mm height and 4-6 mm diameter) porous carbon nanotube (CNT) scaffolds, their in vitro cytocompatibility and interactions with human mesenchymal stem cells (hMSCs) are reported. Single- or multi-walled carbon nanotubes (SWCNTs or MWCNTs) scaffolds were fabricated using a novel radical initiated thermal crosslinking and annealing method using SWCNTs or MWCNTs as nanoscale building blocks. Despite numerous advances in our understanding of the mechanisms that underlie Alzheimer’s disease (AD), there is still no cure, and the disease continues to pose a severe public health problem. It has become clear that many factors, including certain lifestyle practices, may contribute to the development of the disease; however, much remains to be learned about how lifestyle factors may prevent or delay cognitive decline. Unlike human epidemiological studies, murine models can provide clearer inferences about causality. Tuberculosis (TB) is one of the major causes of mortality in the world, causing approximately 1.5 million deaths every year, among which children take a great part. However, due to the perception that children are rarely infectious or they rarely develop severe disease, children with TB have always been neglected. What is worse, diagnosis of childhood TB is difficult, making it harder to control the disease. PET has played a key role in the field of molecular imaging and nuclear medicine over the past decades. PET represents a non-invasive molecular imaging technique to study and visualize human physiology by detecting positron-emitting radiopharmaceuticals labeled with short half-live radioisotopes (i.e. | 11C, 13N, 15O, and 18F) that visualize in vivo biochemical processes and metabolic pathways (i.e. | glycolysis and protein/DNA synthesis). The world’s dependence on the limited supply of fossil fuels has provided the motivation for research into alternative renewable fuel sources. Ethanol is a promising alternative due to its low toxicity, high energy density, and compatibility with the current fuel distribution system. An attractive route to ethanol production would be through the conversion of synthesis gas or ‘syngas’ (CO + H2), which is currently used to produce methanol, hydrogen, and synthetic fuels (diesel and gasoline). However, there are currently no commercially used catalysts for efficient ethanol production from syngas. In general, catalysts for syngas conversion to ethanol should promote C‒C …
