The self-assembly and aggregation of amyloid protein are associated with several neurodegenerative diseases. The evidence indicates that the oligomeric intermediates, formed prior to the final fibrillary product, are the primary culprits of neurotoxicity. Although tremendous efforts have been dedicated for the characterization of structures, dynamics and toxic-related hallmarks of the oligomers, to date, yet the mechanism of such assembly from disordered monomers and their structure remain elusive.
In this dissertation, I focused on understanding the dimerization process of amyloid proteins and peptides of different sizes and I combined experimental studies with high-power computer simulations. The AFM force spectroscopy experiments showed …

The purpose of this historical case study was to understand and describe rural community experiences during the 1918 influenza pandemic in Nebraska.
Examining the rural experience in Nebraska during the 1918 influenza pandemic provided a new level of insight into the differences and similarities between the urban and rural experience. As related by a detailed study of Omaha during the 1918 pandemic, the community was devastated by disease. Despite public ordinances and health department warnings, streetcars ran at capacity, parades were held to raise money for war bonds, and the annual Aksarben Coronation took place. Background: When determining eligibility for kidney transplantation, body mass index (BMI) is often a debatable criterion for transplant due to the clinical indication, an elevated BMI has adverse effects on kidney transplant outcomes. There is a current gap in research on the effect BMI has on kidney transplant outcomes.

The present study provides a systematic replication of the Jennett, Harris, and Delmolino (2008) study comparing discrete trial instruction (DTI) and mand training on the acquisition of mands for children with autism spectrum disorder (ASD). An adapted alternating treatment design was implemented across three participants. Independent mands, variation in requested items, and duration of sessions were assessed across conditions. Generalization probes were conducted to assess generalization across communication partners (e.g., novel research assistants), along with a maintenance probe one-week post-training. Furthermore, a concurrent-chains arrangement was implemented to assess participants’ preference for teaching strategies. All participants reached mastery criterion quickest with …

Combination chemotherapy is commonly used to treat cancer, because such a therapy regimens usually involve sequential administration of multiple drugs and allow targeting different cell signaling pathway. The co-delivery of drug combination at a controlled ratio via the same vehicle is offering the advantages such as spatial-temporal synchronization of drug exposure, synergistic therapeutic effects and suppression of drug resistance. Parkinson’s disease (PD) is the most common neurodegenerative movement disorder and second most common neurodegenerative disorder. PD is characterized by the selective loss of dopaminergic neurons and dopamine neurotransmitter within the substantia nigra and termini in the striatum. Progressive loss of dopaminergic neurons occurs over many years in PD, and by the time movement disorder symptoms manifest, up to 50-70% of dopaminergic neurons have been lost. Several aspects of PD pathology have been described in detail, but a better understanding of PD progression is needed to develop more efficient treatments.

Purpose: The purpose of this study was to evaluate the changes in volume, cross-sectional area and depth of the upper airway following the surgical removal of the adenoids.
Materials and Methods: 16 patients were diagnosed with hypertrophic adenoids and referred for surgical removal. Pre-surgical and post-surgical CBCT scans were taken on each patient. Volume measurements of the total airway, oropharynx and nasopharynx were recorded. In addition, cross-sectional areas and airway depths at the posterior nasal spine (PNS) and cervical vertebrae 2 were recorded. 15 patients diagnosed with no or mild adenoid hypertrophy were treated as the control group. Purpose: The purpose of this investigation was to determine the volumetric and cross-sectional changes that may occur within the posterior pharyngeal airway space after positioning the mandibular condyles in centric relation.
Materials and Methods: All subjects evaluated presented signs and symptoms of upper airway obstruction with significant CR-CO discrepancies. Stabilization maxillary splint therapy was employed to seat mandibular condyles in CR. Pre- and post-treatment CBCT scans were taken to compare airway changes. The effects of methamphetamine (Meth) in the periphery are not well studied and a comprehensive investigation on the effects and molecular mechanism will give insight into why Meth users are at an increased risk of infections. There are nearly 26 million people with diabetes mellitus (DM) in the US, and half of chronic DMs develop somatosensory deficits due to diabetic polyneuropathy or diabetic peripheral neuropathy (DPN). The absence or impaired somatosensory feedback (e.g. touch sensation or joint proprioception) resulted from the damage of large nerve fiber, and motor deficits such as attenuated muscle strength and abnormal plantar pressure of lower extremity have been identified in DPN, and these sensorimotor impairments lead to an increased number of falls. The current study tested the hypothesis that the effects of THC on affect are mediated by the ability of THC to alter attentional bias to positive and negative stimuli. The study was completed by 16 participants, all of whom were consistent marijuana users. During sessions, participants used a machine called a Quantified Smoke Delivery System, which delivers a controlled amount of smoke to the user, to smoke either a cigarette that contained THC or a placebo cigarette. In 2002, John Judis and Ruy Teixeira published The Emerging Democratic Majority, a book that postulated that the United States was in the beginning of a political realignment that would spell the end of the Reagan-era coalition that gave Republicans an electoral advantage on the presidency. Abstract Microsoft’s Windows Operating System provides a logging service that collects, filters and stores event messages from the kernel and applications into log files (.evt and .evtx). Volatility, the leading open source advanced memory forensic suite, currently allows users to extract these events from memory dumps of Windows XP and Windows 2003 machines. Currently there is no support for users to extract the event logs (.evtx) from Windows Vista, Win7 or Win8 memory dumps, and Volatility users have to rely on outside software in order to do this. Plants respond to environmental stress by altering their gene expression. Under stress conditions some genes are activated and some genes are repressed. Even though a lot of work has been done to understand mechanisms of gene activation under abiotic stress very little information is available on how stress responsive genes are kept repressed under normal growth conditions. Recent work has revealed that plants use transcriptional repression as common mechanism of gene repression. Transcriptional repression is achieved by recruitment co-repressor complexes to the target genes. The development of a numeric simulation for predicting the performance of an Ocean Current Energy Conversion System is presented in this thesis along with a control system development using a PID controller for the achievement of specified rotational velocity set-points. Nearly all discussions of Hemingway and his work touch on the theme of masculinity, a recurrent theme in all of his works. Examinations of Hemingway and his relationship to masculinity have almost unanimously treated the author as a misogynist and a champion of violent masculinity. However, since the posthumous publication of The Garden of Eden in 1986, there has been much discussion of Hemingway’s uncharacteristic use of androgynous characters in the novel. Critics have taken this as a clue that Hemingway possessed a complex attitude regarding gender fluidity, but have failed to examine the constructions of gender and identity in …

Quantifying the impacts of restoration on coastal waterways is crucial to understanding their effectiveness. Here, I look at the impacts of multiple restoration projects on urban waterways within the city limits of New Orleans, LA, with an emphasis on the response of fishes. First I report the effects of two projects designed to improve exchange down estuary on the hydrologic characteristics of Bayou St. John (BSJ). Within BSJ, flow is dominated by subtidal wind driven processes. Removal of an outdated flood control structure did not appear to alter exchange in BSJ, but removal combined with sector gate openings did. I …

Huntington’s disease (HD) is a heritable, neurodegenerative disorder characterized by motor, cognitive, and psychiatric disturbances. The progressive disease is caused by an unstable CAG expansion within the gene that normally encodes for the huntingtin protein (Htt). The expanded mutant form of Htt (mHtt) is expressed ubiquitously throughout patients’ bodies; however, neuronal degeneration is prominent only in the corpus striatum and, to a lesser extent, the cortex. The Ras homolog Rhes is also preferentially localized to the striatum. The putative co-factor Rhes has been shown to act with mHtt to cause neuronal death. The generation and subsequent measurement of far-infrared radiation has found numerous applications in high-resolution spectroscopy, radioastronomy, and Terahertz imaging. For about 45 years, the generation of coherent, far-infrared radiation has been accomplished using the optically pumped molecular laser. Once far-infrared laser radiation is detected, the frequencies of these laser emissions are measured using a three-laser heterodyne technique. With this technique, the unknown frequency from the optically pumped molecular laser is mixed with the difference frequency between two stabilized, infrared reference frequencies. These reference frequencies are generated by independent carbon dioxide lasers, each stabilized using the fluorescence signal from an external, …

We classify n-point extensions of ergodic Zᵈ-actions up to relative orbit equivalence and establish criteria under which one n-point extension of an ergodic Zᵈ-action can be sped up to be relatively isomorphic to an n-point extension of another ergodic Zᵈ-action. Both results are characterized in terms of an algebraic object associated to each n-point extension which is a conjugacy class of subgroups of the symmetric group on n elements.

The high degree of heterogeneity observed in breast cancers makes it very difficult to classify cancer patients into distinct clinical subgroups and consequently limits the ability to devise effective therapeutic strategies. In this study, we explore the use of gene mutation profiles to classify, characterize and predict the subgroups of breast cancers. We analyzed the whole exome sequencing data from 358 ethnically similar breast cancer patients in The Cancer Genome Atlas (TCGA) project. Identified somatic and non-synonymous single nucleotide variants were assigned a quantitative score (C-score) that represents the extent of negative impact on the function of the gene. Using …

The Patient Protection and Affordable Care Act of 2010 ushered in a new era of fiscal accountability for healthcare organizations. Healthcare organizations and providers are now jointly held responsible for the improved quality of patient care and sustained reductions in patient care events termed healthcare-acquired conditions. To ensure compliance with this newly enacted legislation, the Centers for Medicare and Medicaid Services (CMS) began penalizing hospitals for targeted conditions leading to 30-day readmissions beginning in October 2012.
