They are submitting this brief because they believe that the U.S. Court of Appeals for the District of Columbia Circuit made serious errors when it held that the Federal Energy Regulatory Commission (FERC) lacked authority to regulate operators’ rules for demand response (DR) in the wholesale electricity markets.

Assembly of polymerase chain reactions at room temperature can sometimes lead to low yields or unintentional products due to mispriming. Mutation of isoleucine 707 to leucine in DNA polymerase I from Thermus aquaticus substantially decreases its activity at room temperature without compromising its ability to amplify DNA. To understand why a conservative change to the enzyme over 20 Å from the active site can have a large impact on its activity at low temperature, we solved the X-ray crystal structure of the large (5′-to-3′ exonuclease-deleted) fragment of Taq DNA polymerase containing the cold-sensitive mutation in the ternary (E–DNA–ddNTP) and binary …

In War and Responsibility, John Hart Ely argues that Congress has willingly and cravenly surrendered its rightful role as the branch responsible for determining when and the extent to which the United States engages in armed conflicts. Since the publication of this seminal work on the war power, presidents have continued to commit troops to hostilities absent or outside of explicit congressional authorizations-and the legislature and the judiciary rarely challenge such actions.

Interstate coordination presents one of the most difficult challenges for American federalism as well as for energy markets and policy. Existing laws vest the approval of large-scale energy infrastructure projects such as interstate oil pipelines and high-voltage, interstate electric transmission lines with state and local levels of government. At the same time, state siting and eminent domain regimes routinely enable and even encourage state regulators to hold out from approving interstate infrastructure projects, hobbling any hope for interstate coordination. Objectives: The purpose of this study was to investigate the effect of the canonical Wnt/β-catenin signaling pathway on the proliferation and osteogenic differentiation of stem cells from human exfoliated deciduous teeth in vitro. Methods: Stem cells from human exfoliated deciduous teeth (SHED) were stimulated with human recombinant Wnt3A at various concentrations (50, 100 and 200ng/ml) in basic growth medium and osteogenic medium. Modern postural yoga is an amalgamation of practices that continues to defy traditional explanatory models that understand religious, spiritual, and secular to be mutually exclusive categories. In this project, I conduct in-depth interviews with yoga instructors in the Ozarks. In these interviews, I asked my informants to explain how they understand religious, spiritual, and secular. I then asked if and how they would use those words to describe yoga. These interviews revealed that religious, spiritual, and secular are not mutually exclusive, but are instead related, blended, and connected, especially when used to describe yoga. The organizational constructs that we examined were distributive justice, procedural justice and interactional justice. The research led to us proposing 12 hypotheses using correlations, t-tests, and logistic regressions. This thesis investigated whether Social Stories used in conjunction with perspective- taking questions would lead to an improved Theory of Mind (ToM) in three participants diagnosed with Autism Spectrum Disorder (ASD). We assessed participants’ perspective taking in a baseline condition, implemented a Social Story condition, and tested participants’ perspective taking again in a post-intervention condition. ToM was assessed using two variations of the false belief measures, namely the change-location and unexpected-contents paradigms. Each Social Story included three perspective-taking questions that required participants to infer a character’s emotion or perspective. This study examines the relationship between auditor partner rotation and fraudulent financial reporting in the post Sarbanes-Oxley conservative environment. Audit firm tenure impacts the quality of the audit and the likelihood of fraudulent financial reporting. Data were retrieved from Accounting and Auditing Enforcement Releases and Computstat spanning the years 1998 through 2008. A logistic regression model similar to one used by Carcello and Nagy (2004) was utilized for the study. Fraudulent financial reporting served as a proxy for audit quality. Threshold concepts are increasingly inescapable at library conferences and in general information literacy discourse, and this visibility will likely only increase as they figure so prominently in the Association of College and Research Libraries inchoate Framework for Information Literacy for Higher Education. Nevertheless, very little has been done to critically consider the wider intellectual ramifications of certain assumptions fundamental to their manifestation in library/information literacy instruction. In order to be acknowledged, chronic pain must be voiced yet disclosing of chronic pain is fraught with social and professional repercussions. Moreover, there is a perceived disinterest in hearing about, and a stigma associated with the experience of chronic pain. This research explores the therapeutic value of communicating about pain. Nineteen individuals with chronic pain participated in a six-week online writing workshop to describe the way chronic pain impacts daily activities. These qualitative responses were analyzed using discourse analysis and four interpretive repertoires emerged which convey the multi-faceted impacts of living with chronic pain. The decision by an animal near a road to either cross or not cross may be influenced by characteristics of the road, environmental conditions, traits of the individual animal, and other aspects of the context within which the decision is made. Canis latrans (Coyotes) are a management concern in the southeastern US because of their potential impacts on agriculture, other wildlife species, and human health and safety. This region is part of a recent range expansion by Coyotes, and information about their population structure in the southeastern US is lacking. In this study, we used microsatellite DNA to assess genetic diversity and population structure among Coyotes in east-central Alabama. We detected high genetic diversity (H_E = 0.78) and no population structure across the total sampling area. Additionally, we investigated population structure between urban and rural groups. While genetic diversity is hypothesized to be an important factor explaining invasion success, there is no consensus yet on how variation in source populations or demographic processes affects invasiveness. We used mitochondrial DNA haplotypic and microsatellite genotypic data to investigate levels of genetic variation and reconstruct the history of replicate invasions on three continents in a globally invasive bird, the monk parakeet (Myiopsitta monachus). Invasive vertebrates are a leading cause of extinction on islands and rats (Rattus spp.) are one of the most damaging to island ecosystems. Methods to eradicate rats from islands are well established and there have been over 580 successful eradications to date. Increasingly, rat eradications are being implemented on tropical islands, a reflection of the need to protect the threatened biodiversity in the tropics. Beginning at 1838 h on 30 May 2014 at the Caves Branch Jungle Lodge in Belize, we observed an encounter between a Leptodeira septentrionalis and an Incilius valliceps that appeared to have initiated as a predation attempt by the toad on the snake. Given their free-ranging habits, feral swine could serve as reservoirs or spatially dynamic ‘mixing vessels’ for influenza A virus (IAV). To better understand virus shedding patterns and antibody response dynamics in the context of IAV surveillance amongst feral swine, we used IAV of feral swine origin to perform infection experiments. The virus was highly infectious and transmissible in feral swine, and virus shedding patterns and antibody response dynamics were similar to those in domestic swine. In the virus-inoculated and sentinel groups, virus shedding lasted ≤6 and ≤9 days, respectively. Antibody titres in inoculated swine peaked at 1 : 840 on …

This study examined the relationship between personality and the preferences expressed by latent entrepreneurs among different ways of creating value. Latent entrepreneurs are individuals who would prefer to be an entrepreneur rather than a salaried employee. Latent entrepreneurs take all the first time decisions on entering entrepreneurship and their entry choices have been linked in the literature to their expressed preferences. This thesis is an exploration of relationships and issues of professional boundaries at Welcome Inn Community Centre, a faith-based community centre offering programs and services to address issues of poverty in Hamilton, Ontario. Data was gathered through semi-structured qualitative interviews with fifteen staff members, volunteers, and participants at Welcome Inn. A strengths-based perspective combined with mutual relationships and flexible boundaries were found to foster inclusion, acceptance, community building, and personal transformation at Welcome Inn Community Centre; Welcome Inn staff, volunteers, and program participants described these qualities positively. The role of investor countries in large-scale land acquisitions is poorly understood in the contemporary “land grab” literature. Orthodox explanations largely build on deductive analyses that deviate from the emerging empirical evidence, and/or face analytical difficulties when trying to capture why large-scale land acquisitions happen. This thesis investigates the global phenomenon of “land grabbing” from the comparative perspective of two major investor countries: the UK and China.
