C. albicans is a polymorphic fungal pathogen which has the ability to shift from yeast to hyphae. C. albicans cell wall is composed of glucan, chitin, mannoprotein and mannan. It is not possible, using standard extraction methods, to isolate mannan from C. albicans hyphae. To isolate hyphal mannan, we developed a simplified alkali extraction method. Using this method it was determined that hyphal mannan has a much lower molecular weight, a smaller polymer distribution and altered conformation structure when compared to yeast mannan. The hyphal mannan was found to contain little to no acid-labile portion with only α-Man-PO4 groups …

The specific goal of this study was to determine the use of the physical classroom environment as a teaching and learning tool in an era of the implementation of the Common Core State Standards Initiative (CCSSI). This qualitative multi-case study focused on the learning principles and epistemological beliefs of primary teachers with reference to the physical classroom environment and the teaching process in regard to meeting the expectations set forth by the CCSSI. Pseudomonas aeruginosa is a bacterial pathogen that can infect any human tissue. The lungs of cystic fibrosis patients become chronically infected with Pseudomonas aeruginosa. Virulence factor gene expression is under elaborate regulatory control that remains poorly characterized. Understanding the regulatory hierarchy involved during infection is essential for identifying novel drug targets. RsmA is a post-transcriptional regulatory protein that controls expression of several virulence factors. This investigation was an analysis of the interaction of adolescent exposure to methylphenidate (MPH; trade name: Ritalin) on nicotine sensitization and conditioned place preference (CPP) in a rodent model and underlying mechanisms of this effect. Animals were treated IP with 1 mg/kg MPH or saline using a ―school day‖ regimen of five days on, two days off, from postnatal day (P) 28-50. During the final two weeks of MPH treatment, animals were either behaviorally sensitized to nicotine (0.5 mg/kg free base) or saline for 10 days, or conditioned to nicotine or saline using the CPP behavioral paradigm. The internalization of mental illness stigma is associated with an array of negative consequences; however, not all individuals experience the deleterious effects of internalized mental illness stigma. The present dissertation focuses on factors associated with internalized stigma, and will be the first to examine simultaneously both risk (i.e., shame, shame proneness and shame aversion, insight, and centrality and valence) and protective factors (social support and self-compassion) of internalized mental illness stigma. Objectives: This study investigated the correlation between Tannerella forsythia, Porphyromonas gingivalis, Fusobacterium nucleatum, and Aggregatibacter actinomycetemcomitans at dual sites in concurrent endodonticperiodontal diseases.
Material and methods: Samples were collected from endodontium and periodontium in cases of concurrent endodontic-periodontal diseases from thirty participants. The sensitivity and specificity of SYBR Green real-time PCR was used to identify the targeted species. Absolute number of targeted genome copies in tested samples were extrapolated from respective calibration curve.
Results: No statistical difference was found in the number of detected endodontic-periodontal pathogens between the endodontium and periodontium. The Pearson test …

Whole animal physiological performance is highly polygenic and highly plastic, and the same is generally true for the many subordinate traits that underlie performance capacities. Quantitative genetics, therefore, provides an appropriate framework for the analysis of physiological phenotypes and can be used to infer the microevolutionary processes that have shaped patterns of trait variation within and among species. In cases where specific genes are known to contribute to variation in physiological traits, analyses of intraspecific polymorphism and interspecific divergence can reveal molecular mechanisms of functional evolution and can provide insights into the possible adaptive significance of observed sequence changes. Objectives: To determine the prevalence of habitual snoring among a sample of middle-aged Saudi adults, and its potential predictors. Methods: A cross-sectional study was conducted from March 2013 until June 2013 in randomly selected Saudi Schools in Jeddah, Kingdom of Saudi Arabia. The enrolled subjects were 2682 school employees (aged 30-60 years, 52.1% females) who were randomly selected and interviewed. The questionnaire used for the interview included: the Wisconsin Sleep Questionnaire to assess for snoring, medical history, and socio-demographic data. Objectives: To estimate normal linear dimensions and volume of spleen in Jordanians using ultrasonography, and to correlate splenic volume with age and body parameters: height, weight, body surface area (BSA), and body mass index (BMI). A Novel Missense Mutation In The Tbx5 Gene In A Saudi Infant With Holt-Oram Syndrome, Mohammad M. Al-Qattan, Hussam Abou Al-Shaar

We report on a Saudi infant with Holt-Oram syndrome caused by a de novo missense mutation of the TBX5 gene. The mutation (Thr72Lys) is novel and has not been previously reported. The cardiac and limb defects in our patient were both severe, and the infant also had micrognathia and cleft palate. Previously reported cases of the Holt-Oram syndrome caused by missense mutations were reviewed and their phenotypes were compared with the phenotype of our patient.

Existing measures of emotion dysregulation typically assess dispositional tendencies and are therefore not well suited for study designs that require repeated assessments over brief intervals. The aim of this study was to develop and validate a state-based multidimensional measure of emotion dysregulation. Psychometric properties of the State Difficulties in Emotion Regulation Scale (S-DERS) were examined in a large representative community sample of young adult women drawn from four sites (N = 484). During the last four decades, since the completion of the GEOSECS program, there has been a growing interest in the biogeochemical cycle of barium (Ba) due to its potential as a proxy for interpreting the geologic record. The typical barium distribution exhibits removal in the upper water column and regeneration at depth, consistent with its classification as a bio-intermediate element and similar to the nutrient-like profiles of silicic acid and alkalinity. To better constrain the uses of Ba, trace element clean samples were collected during the North Atlantic US GEOTRACES cruises.

Temperature is one of the most pervasive and influential abiotic factors affecting the life histories of fish, regulating a host of physiological processes and influencing behavioral thermoregulation. Fundulus notatus, the blackstripe topminnow, and F. olivaceus, the blackspotted topminnow, are widely distributed species located in drainages along the northern part of Gulf of Mexico (30°N latitude) and as far north as southern Illinois (39°N latitude). The purpose of this project was to provide insights about the short- and long-term patterns of habitat selection of Gulf-strain Striped Bass, Morone saxatilis, based on spatially and seasonally variable abiotic environmental characteristics (water temperature, dissolved oxygen (DO), salinity, specific conductivity, and pH) in the Biloxi River, MS system. Bycatch in the shrimp fishery became of particular concern in the 1980s during the peak of shrimp harvest in the northern Gulf of Mexico. Shrimp bycatch is of major importance because the majority of catch in the fishery is not intended target species of commercial shrimp, and most of the additional bycatch is returned to the sea dead or dying, inducing higher rates of mortality. This thesis research used techniques of dendrochronology to investigate the efficacy of using multiple co-occurring species (MCOS) in a climate reconstruction model compared to a single species (SS) in four old-growth forests in Indiana: Pioneer Mothers Memorial Forest (PM), Donaldson Woods (DW), Hoot Woods (HW), and Lilly Dickey Woods (LD). The objective is to evaluate the ability of aminoacyl-tRNA synthetases (aaRS) to recognize the non-standard nucleic acid, PNA (peptide nucleic acid). PNA has immense potential in biomedical applications due to its increased thermostability and nuclease resistance over natural nucleic acids. PNA represents a superior alternative to natural nucleic acids in many biomedical applications due to its specificity, strong binding, and nuclease resistance. This study is the initial data set that indicates PNA are recognized by translation enzymes.

David Lehman’s poem “The Breeders’ Cup” uses cross-generational coterie and ekphrasis to create a poetic poioumenon. When read in terms of art criticism, Lehman’s “The Breeders’ Cup” models creative processes from the past and calls for a rehabilitative ethic in postmodern poetics. Lehman follows the ekphrastic form, which associates a poem with a work of visual art, from his New York School predecessor Frank O’Hara. “The Breeders’ Cup” addresses Édouard Manet’s 1865 painting Olympia through ekphrasis, and the painting of a prostitute becomes a patron saint of parody for postmodern poetics. An understanding of regional orogenic, climatic, and eustatic processes is critical to the interbasinal correlation of Paleozoic strata in eastern North America. Tectonic activity associated with the culmination of Appalachian Orogenic events has been shown to have regional influence on paleostructure and sediment dispersal in the Appalachian foreland basin and adjacent intracratonic Illinois and Michigan basins. The culmination of the Acadian Orogeny at the end of the Devonian represents the beginning of a period of general tectonic quiescence extending throughout the early and middle Mississippian in eastern North America. Bronson Methodist Hospital in Kalamazoo is home to one of only 15 certified breast milk banks in the United States. Women have shared breast milk for centuries through wet nurses, but this institutionalized and regulated version of sharing is distinct from previous forms. Breastfeeding has become a symbol of successful motherhood; donor milk adds a new dimension to this aspect of idealized motherhood. This study explores how the milk bank works: its organizational structure within a hospital, how donors are selected, and how recipients qualify for donor milk. Ichthyoplankton in the Gulf of Mexico (GOM) aggregate in two different habitats that serve to provide refuge and concentrate prey in an otherwise oligotrophic environment: the Loop Current (LC) a mesoscale oceanographic feature characterized by the transition of environmental conditions of the water mass between the GOM and Caribbean; and Sargassum, a two species assemblage of floating brown algae that is habitat for many larval and juvenile obligate and associated fishes. The objectives of this study were to characterize the density and community composition of ichthyoplankton within these habitats using multivariate analysis. This thesis studies the craft beer culture in Hattiesburg in order to answer the research question: Is taste what draws people to this culture? Beyond conducting classic participant-observation, I deployed the method of sensory activated participant-observation in my fieldwork. In so doing, this research centers the perspective and the data collected on the senses, especially the taste of craft beer. The purpose of this research is to add to our knowledge of the anthropology of the senses and contribute to the literature on the anthropology of taste.

In order for them to succeed, they need access to reference materials as well as up-to-date information (Mankin, Warner, & Anderson, 1999). Incorporating mobile technology (i.e. tablets) into naturalists’ endeavors in natural history and environmental education can be used as supportive and educational tools. Ticks are ectoparasites that attach to their host for many days to weeks, feeding to engorgement and continuing their species life cycle. The mechanisms behind the successful feeding and manipulation of host immune responses have been associated with the secretion of their specialized saliva. Tick saliva includes a variety of modulatory molecules that must disrupt and counteract defense mechanisms, including a variety of proteases. Reprolysin Metalloproteases, in the Zn2+ dependent family, is secreted as a proenzyme and is shown to have proteolytic activity, degrading fibrinogen and gelatinase activity, as well as disturbing homeostasis12.
