Phosphorus Sorption Dynamics In Shallow Groundwater, Coastal Everglades, Florida, Usa, Hilary Flower

USF Tampa Graduate Theses and Dissertations

For this dissertation I studied phosphorus (P) sorption dynamics in the shallow groundwater of the southern Everglades. In particular, I examined how the ambient water type governs soluble reactive P (SRP) availability through adsorption/desorption reactions with the aquifer matrix. Chapter 2 investigated how P sorption dynamics of the mangrove root zone sediment are affected by high bicarbonate brackish groundwater compared to both fresh groundwater and saltwater. The results from chapter 2 show that the sediment exhibited exceptionally low sorption efficiency in the high bicarbonate brackish water, which would allow ambient water SRP concentration to be maintained at a higher level. …

Water in the World: Interdisciplinary Approaches to Access and Sustainability

Every odd-year in March, from 2005-2015, the Río Lagarto, Costa Rica has been surveyed by students in Tropical Watersheds and Coastal Ecology. The Río Lagarto originates at approximately 1,500 m in the Monteverde cloudforest region of the Cordillera de Tilarán and steeply descends toward the Pacific coast discharging into the mangrove forests bordering the Gulf of Nicoya. This small Pacific slope river is the western-most of the three rivers, along with their watersheds, that define the Bellbird Biological Corridor. The Bellbird Biological Corridor is a long-term community-based effort to restore natural forest cover to a largely deforested terrain. This reforestation …

Background

Ad5-GUCY2C-PADRE is a replication-deficient human type 5 recombinant adenovirus (Ad5) vaccine encoding guanylyl cyclase C (GUCY2C) fused to the PAn DR Epitope (PADRE). GUCY2C, a paracrine hormone receptor producing the second messenger cyclic GMP (cGMP), is selectively expressed by intestinal epithelial cells and a subset of hypothalamic neurons, but not other tissues. Importantly, GUCY2C is over-expressed in nearly all primary and metastatic human colorectal tumors. Preclinical studies in mice demonstrated selective tolerance of GUCY2C-specific CD4+ T cells, but not CD8+ T or B cells, necessitating inclusion of the exogenous CD4+ T helper cell epitope PADRE to maximize GUCY2C-specific CD8+ …

The pomegranate fruit moth, Apomyelois ceratoniae (Zeller), is the most important pest of pomegranate orchards in Iran, where infestations lead to 20%–80% fruit loss. A. ceratoniae overwinters as larvae in several instars. The success in overwintering determines the fruit loss in the following season, thus overwintering physiology of A. ceratoniae could provide insights into population prediction. To this end, overwintering strategy and some seasonal physiological and biochemical changes were investigated in the field-collected larvae of A. ceratoniae. Arsenic (As3+) is a carcinogen with considerable environmental and occupational relevancy. The present study shows that As3+-transformed human lung bronchial epithelial BEAS-2B cells (AsT cells) exhibit the property of apoptosis resistance. The level of basal reactive oxygen species (ROS) is very low in AsT cells in correlation with elevated expressions of both antioxidant enzymes and antiapoptotic proteins. Nuclear factor erythroid 2-related factor (Nrf2) and p62 are constitutively expressed. These two proteins up-regulate antioxidant enzymes and antiapoptotic proteins. The knockdown of Nrf2 or p62 by small interfering RNA (siRNA) enhanced both ROS levels and As3+-induced …

Mario Sprouse, born October 10th, 1948 in Spanish Harlem, is a well-known musician,arranger, composer, and musical director. His parents immigrated to the United States from the Dominican Republic in 1929, and would meet in Harlem and begin their family after the war.

His parents worked hard to provide for their children, but always had a passion for music, which they would instill upon Mario and his siblings. Background: A priority for conservation is the identification of endemic populations. We developed microsatellite markers for common raven (Corvus corax), a bird species with a Holarctic distribution, to identify and assess endemic populations in Alaska. Results: From a total of 50 microsatellite loci, we isolated and characterized 15 loci. Eight of these loci were polymorphic and readily scoreable. Eighteen to 20 common ravens from Fairbanks, Alaska were genotyped showing the following variability: 3-8 alleles per locus, 0.25-0.80 observed heterozygosity (Ho), and 0.30-0.80 expected heterozygosity (He). RNA interference, or RNAi, refers to a set of biological processes that make use of conserved cellular machinery to silence genes. Although there are several variations in the source and mechanism, they are all triggered by double stranded RNA (dsRNA) which is processed by a protein complex into small, single stranded RNA, referred to as small interfering RNAs (siRNA) with complementarity to sequences in genes targeted for silencing. As revealed in a previous microarray study to identify genes regulated by 20-hydroxyecdysone (20E) and juvenile hormone (JH) in the silkworm, Bombyx mori, E93 expression in the fat body was markedly low prior to the wandering stage but abundant during larval-pupal metamorphosis. Induced by 20E and suppressed by JH, E93 expression follows this developmental profile in multiple silkworm alleles. The reduction of E93 expression by RNAi disrupted 20E signaling and the 20E-induced autophagy, caspase activity, and cell dissociation in the fat body. Reducing E93 expression also decreased the expression of the 20E-induced pupal-specific cuticle protein genes and prevented growth …

The impact of consuming biologically active compounds is often dose-dependent, where small quantities can be medicinal while larger doses are toxic. The consumption of plant secondary compounds can be toxic to herbivores in large doses, but can also improve survival in parasitized herbivores. In addition, recent studies have found that consuming nectar secondary compounds may decrease parasite loads in pollinators. However, the effect of compound dose on bee survival and parasite loads has not been assessed. Integrin α6β4 is up-regulated in pancreatic adenocarcinomas where it contributes to carcinoma cell invasion by altering the transcriptome. In this study, we found that integrin α6β4 up-regulates several genes in the epidermal growth factor receptor (EGFR) pathway, including amphiregulin (AREG), epiregulin (EREG), and ectodomain cleavage protease MMP1, which is mediated by promoter demethylation and NFAT5. The correlation of these genes with integrin α6β4 was confirmed in The Cancer Genome Atlas Pancreatic Cancer Database. Population structure analyses and genome-wide association studies (GWAS) conducted on crop germplasm collections provide valuable information on the frequency and distribution of alleles governing economically important traits. The value of these analyses is substantially enhanced when the accession numbers can be increased from ~1,000 to ~10,000 or more. In this research, we conducted the first comprehensive analysis of population structure on the collection of 14,000 soybean accessions [Glycine max (L.) AIM: To investigate the ability of a Prunella vulgaris (P. vulgaris) ethanolic extract to attenuate spontaneous typhlocolitis in mdr1a-/- mice.

METHODS: Vehicle (5% ethanol) or P. vulgaris ethanolic extract (2.4 mg/d) were administered daily by oral gavage to mdr1a-/- or wild type FVBWT mice from 6 wk of age up to 20 wk of age. Clinical signs of disease were noted by monitoring weight loss. Mice experiencing weight loss in excess of 15% were removed from the study.
