Intestinal CD4+ T cell depletion is rapid and profound during early HIV-1 infection.This leads to a compromised mucosal barrier that prompts chronic systemic inflammation.The preferential loss of intestinal T helper 17 (Th17) cells in HIV-1 disease is a driver of the damage within the mucosal barrier and of disease progression.Thus, understanding the effects of new therapeutic strategies in the intestines has high priority. Histone deacetylase (HDAC) inhibitors (e.g., panobinostat) are actively under investigation as potential latency reversing agents in HIV eradication studies. These drugs have broad effects that go beyond reactivating virus, including modulation of immune pathways. We examined colonic …

Fibronectin (Fn1) is an evolutionarily conserved extracellular matrix glycoprotein essential for embryonic development. Global deletion of Fn1 leads to mid-gestation lethality from cardiovascular defects. However, severe morphogenetic defects that occur early in embryogenesis in these embryos precluded assigning a direct role for Fn1 in cardiovascular development. We noticed that Fn1 is expressed in strikingly non-uniform patterns during mouse embryogenesis, and that its expression is particularly enriched in the pharyngeal region corresponding with the pharyngeal arches 3, 4, and 6. This region bears a special importance for the developing cardiovascular system, and we hypothesized that the localized enrichment of Fn1 in …

This brief communication provides information to those developing monitoring plans for serious adverse events (SAE’s) following regulatory approval of a new drug. In addition, we (1) illustrate how many patients would need to be treated in order to have high confidence of seeing at least 1 pre-specified SAE, (2) show that absence of proof of a SAE is not proof of absence of that SAE, and (3) identify statistical methodology that could be used for formal statistical monitoring of SAE’s.

Inflammatory demyelination of the central nervous system (CNS) is the hallmark of multiple sclerosis (MS), a chronic debilitating disease that affects more than 2.5 million individuals worldwide. It has been widely accepted, although not proven, that the major pathogenic mechanism of MS involves myelin-reactive T cell activation in the periphery and migration into the CNS, which subsequently triggers an inflammatory cascade that leads to demyelination and axonal damage. Virtually all MS medications now in use target the immune system and prevent tissue damage by modulating neuroinflammatory processes. Although current therapies such as commonly prescribed disease-modifying medications decrease the relapse rate …

Hatching success in birds is influenced by the temperature and turning rate of the egg, but our understanding of the environmental factors that effect incubation temperatures and egg turning rates in birds is limited. Especially little is known of these effects for species that nest in burrows or crevices, such as the Cassin's auklet. On Southeast Farallon Island, California, USA, a subset of the Cassin's auklet (hereafter auklet) population nest in artificial nest boxes. The nest boxes are above ground and made out of a single layer of plywood. Temperatures in unshaded nest boxes can increase significantly with high ambient …

This dissertation explores the intersection of new media and narrative, as it is presented through a cinematic aesthetic. The narrative language of film is analyzed through the theoretical framework of Bakhtin’s concepts of Heteroglossia, Chronotope, and Dialogism. Bakhtin’s ideas of classifying language act as strong tools for demonstrating how cinematic narrative can inform and alter the perception of its spectators. Lev Manovich’s principles of New Media, specifically Variability, Modularity, and Automation are also utilized to demonstrate how cinema is a constantly evolving paradigm.

The present study reports on reactive oxygen species (ROS), antioxidant enzyme activities, electrolyte leakage and levels of abscisic acid (ABA) in healthy and phytoplasma infected (PI) tissues of Euphorbia coerulescens and Orbea gigantea. Histochemical staining for ROS indicated that PI tissues possess higher levels of hydrogen peroxide (H2O2) rather than superoxide (O2 ·-) as compared with healthy tissues in both plant species. The results indicated that superoxide dismutase (SOD) is not playing an important role in eliminating O2− in PI tissues. This was confirmed by a significantly decreased activity of SOD and a non-significant difference in O2 ·- content in …

Avian Haemosporida are common, vector-transmitted blood parasites of birds throughout the world. During my dissertation research, I explored how multiple host species respond immunologically to natural infections in the wild (Chapter 1) and to experimental infections in the laboratory (Chapter 2). Despite their tractability as a model host-parasite system and a burgeoning literature on avian Haemosporida, little is known about how their populations interact across large areas (hereafter “regions”). MicroRNAs have been identified as potential cancer biomarkers due to their presence and stability in many body fluids including urine and plasma, but the relationship of the pattern of expression of these messengers across various biological media has not been addressed and could provide important information in order to translate these biomarkers for epidemiologic or clinical use. We analyzed microRNA of matched FFPE-tumor tissue, plasma, urine exosomes (n = 16) and WBCs (n = 11) from patients with bladder cancer, using Nanostring miRNA assays and droplet digital PCR for validation. Water quality monitoring programs across multiple disciplines use total suspended solids (TSS), and volatile suspended solids (VSS), to assess potential impairments of surface water and groundwater. While previous methods for instream filtering have been developed, the need for rapid, cost-effective, high volume sampling has increased with the need to verify and supplement data produced by sondes and instantaneous data loggers. We present an efficient method to filter water instream with a portable drill pump that results in reduced sample processing time, and potentially reduced error associated with sample transportation, preservation, contamination, and homogenization. The main research question explored in this curriculum capstone is what are some simple things that children could know about where they live and what edible wilds grow around them that would enrich their sense of place? A goal of this project is to get children outside with an adult companion exploring natural settings by way of foraging for wild edibles. The purpose of this wild edibles guide is to give novice foragers the basic tools necessary to ensure wildcrafting success. Studies of divergence and speciation patterns in island systems have played an important role in the development and establishment of the allopatric speciation model. However, recent empirical support for divergence and speciation with gene flow means the importance of isolation for divergence in island systems needs to be re-examined. Here I explore the roles of geographic isolation and gene flow in the early stages of divergence of evolutionarily independent replicate populations of the Rufous Fantail Rhipidura rufifrons on satellite islands in southeastern Solomon Islands. Shiga toxin-producing Escherichia coli (STEC) strains are important food-borne pathogens that have been linked to human illness. In the United States, STEC O157:H7 strains have been attributed to the majority of disease and outbreaks; however, the frequency of disease associated with STEC non-O157 strains have also increased and is often under recognized due to diagnostic limitations. In 2012, the Food Safety and Inspection Service (FSIS) implemented mandatory testing for non-O157 STEC serogroups O26, O45, O103, O111, O121, and O145. Shiga toxin-producing Escherichia coli (STEC) have been identified by the USDA as a serious threat to the nations health stemming from contaminations in the food supply, specifically, the beef chain. I have developed an assay that is able to screen samples for STEC in a rapid and multiplexed format. Multiplex oligonucleotide ligation-PCR (MOL- PCR) is a nucleic acid based assay patented at Los Alamos National Laboratory (LANL) that uses flow cytometry and multiplex microsphere arrays for detection of nucleic acid based signatures.
