Ecologists currently cannot test concentrations of carbohydrates in sap in vivo. Testing carbohydrates with current technology would require destructive tissue sampling. The tissue sampling involves large amounts of time and money to collect and test. Aphids are an insect that can bypass a tree’s passive immune system and feed off a phloem region for weeks. A series of enzymatic biosensors could be used to detect the concentration changes of specific carbohydrates. A calcium chelant can be added to defeat a tree’s immune system like an aphid. Nitrous acid (HONO) is an important reactive nitrogen species and a precursor to the hydroxyl radical. It plays important roles in reactive nitrogen recycling and in photo-oxidant formation in the troposphere. However, much is still unknown about its formation mechanisms and distributions in the troposphere. This research is part of NOMADSS (Nitrogen, Oxidants, Mercury, and Aerosol Distributions, Sources and Sinks) field study contributed by over 10 research groups in the United States to investigate the tropospheric chemistry of reactive nitrogen, mercury, oxidant and aerosols. Engineered nanomaterials (ENMs) play an increasing role in manufacturing and consumer products. Currently, there is no standard approach to studying ENM toxicity, and a growing body of literature suggests that ENMs may have toxicity differing from similar compounds in bulk or dissolved form. I examined ENMs used in the semiconductor manufacturing process called chemical-mechanical planarization (CMP) for their properties, removal in the wastewater treatment system (WWT), in-vitro toxicity, and location post-inhalation in-vivo. It was found that ENMs in CMP slurries have morphology determined by their elemental composition, but assessment of size and concentration can differ substantially between accepted techniques. Simulations of biochemical systems are being increasingly used to further our understanding of those systems. In broad strokes, chemical simulations can be broken into two categories, those that are rooted in a quantum mechanical approach and those that are classically rooted. Molecular dynamics is one such classical method, which propagates motion of a chemical system via repeatedly solving Newton’s Laws of motion. This approach is less computationally intensive than the quantum mechanical methods and allows for the simulation of systems of tens of thousands of atoms, if not more.

In Melendez-Diaz v. Massachusetts (2009), the United States Supreme Court held that the Sixth Amendment right to confront adverse witnesses includes the right to confront forensic analysts who produce affidavits certifying the results of forensic examinations used against defendants at trial. The decision thus invalidated the practice of substituting forensic laboratory reports for live testimony in criminal trials. Melendez-Diaz was a divided decision in which four dissenting Justices predicted onerous practical consequences for the practice of forensic science in the United States. Conventional x-ray imaging is a widely used imaging technique in medical diagnostics as well as material analysis. It utilizes only absorption-contrast imaging, which records intensity attenuation due to absorption. A major limitation of this technique for medical imaging is the poor contrast of weakly absorbing details in the soft tissue. Mammography and angiography are two examples where improved contrast is required. Prevalence of breastfeeding initiation in New York State (NYS) nearly meets Healthy People 2020 goals. Improvements in breastfeeding duration and exclusive breastfeeding (EBF) have lagged behind those achieved for initiation. Implementation of Ten Steps to Successful Breastfeeding in hospital maternity care has improved breastfeeding initiation, duration and exclusivity; but data on the independent association with specific Steps have been equivocal. Utilizing NYS-linked data from the Maternity Practices in Infant Nutrition and Care (mPINC) and the Pregnancy Risk Assessment Monitoring System (PRAMS), the associations of individual maternity care practices/Steps with breastfeeding behaviors were evaluated. This inquiry seeks to understand how preservice teachers interact with White privilege during a teacher education course dedicated to diversity and teaching. By pairing grounded theory with phenomenology, insights into participants’ pre-existing beliefs around race and Whiteness are examined. Given the cultural mismatch between an increasingly diverse public school student population and the historically stable White preservice teacher population, a closer look at persistent and resistant linguistic phenomena undergirding White dominance in the educational setting is useful. Axon outgrowth requires coordinated expression of critical cytoskeletal genes in response to extracellular cues. The spatial and temporal expression of these genes is regulated transcriptionally and posttranscriptionally. Such regulation is largely mediated through the activity of cis-regulatory elements within the cytoskeletal DNAs and RNAs, which recruit specific sets of trans-factors such as DNA- and RNA-binding proteins, as well as microRNAs. Amyloid aggregation is a specific form of protein misfolding and self-assembly related to several degenerative pathologies like Alzheimer's and Parkinson's diseases, type-II diabetes, etc. This association with deadly diseases has made amyloid fibrils the focus of intensive research for many decades. Most recently, a significant interest to protein fibrils was also due to the application of the remarkable self-assembling capabilities to form well-structured nano-biomaterials. A strong correlation between vitamin D deficiency and increased breast cancer incidence and mortality has been well established, leading researchers to further examine the possibility of using vitamin D in treatment and prevention strategies. Our long term goal is to elucidate the mechanisms by which vitamin D pathway signaling impacts on breast cancer. Generation of the high-affinity vitamin D receptor (VDR) ligand 1,25-dihydroxyvitamin D (1,25-D) from 25-hydroxyvitamin D (25-D) is mediated by CYP27B1 which is present and developmentally regulated in multiple tissues including the mammary gland. This study explores the multiplicity of challenging issues that lesbian clergy face today from a social work perspective, utilizing the intersecting concepts found in social work and theology as an interpretive framework. From the social work perspective, the theoretical constructs of social identity theory, ecological theory, and anti-oppressive theory inform the qualitative questions of this study. Among ocular pathologies, glaucoma remains the second leading cause of blindness. The molecular mechanisms that lead to glaucoma have been attributed to damage of the conventional outflow tract. Conventional outflow tissues, a composite of the trabecular meshwork and the Schlemm's canal, regulate and maintain homeostatic responses of aqueous humor outflow. In glaucoma, drainage of aqueous humor into the Schlemm's canal is hindered, leading to an increase in intraocular pressure (IOP). That increase in IOP is directly correlated with retinal ganglion cell death, eliminating the relay of visual information to the thalamus and visual cortex, leading to blindness. The interaction of X rays in diagnostic energy range with soft tissues can be described by Compton scattering and by the complex refractive index, which together characterize the attenuation properties of the tissue and the phase imparted to X rays passing through it. Many soft tissues exhibit extremely similar attenuation, so that their discrimination using conventional radiography, which generates contrast in an image through differential attenuation, is challenging. However, these tissues will impart phase differences significantly greater than attenuation differences to the X rays passing through them, so that phase-contrast imaging techniques can enable their discrimination.

Many planning and land use decisions in New York State are controlled at the local (town or municipal) level, not an optimal scale for planning and implementing resource conservation management. Watershed boundaries provide a more ecologically meaningful scale for conservation, because they capture a full range of natural ecosystem processes that span political boundaries. However, defining an urban watershed is complicated by stormwater infrastructure, so standard topographic watershed boundaries may be inadequate for urban resource conservation even when applied at the watershed scale. The textbook model of bacterial transcription regulation posits that promoters occur immediately upstream of genes and that transcription factors (TFs) modulate transcription through promoter-proximal binding. However, the recent application of unbiased genome-wide approaches, such as ChIP-seq and RNA-seq, has revealed a much more complex picture, including TF binding and transcription initiation occurring in unexpected locations. This dissertation describes the use of deep sequencing-based approaches to evaluate the genome-wide binding of transcription-related proteins and identify locations of transcription initiation. Most precautionary principles discussed in the regulatory literature argue that one ought to be more risk averse when one is uncertain about the probability of some sig- nificant danger. I explore a new kind of precautionary principle, one that addresses avoiding dangers we did not explicitly consider at all. Since it is unclear how uncon- sidered dangers could be avoided, I sketch a methodology for identifying situations where unconsidered dangers seem especially likely, by drawing from the literature in psychology on heuristics and biases and evaluating our cognitive “blind spots.”

In my dissertation I examine two pieces of criminological knowledge in order to explore the point at which they converge. First, crime victims call the police for a variety of reasons, one of which is to stop future attacks; and second, research suggests that previous victimization is a significant predictor of future victimization. In my research I seek to expand current knowledge about repeat victimization using the National Crime Victimization Survey (NCVS). Glaucoma refers to a group of slowly progressing eye disorders that lead to damage to the optic nerve, resulting in irreversible vision loss. Recent statistics by the World Health Organization places glaucoma as a leading cause of blindness worldwide, affecting nearly 80 million people. Lowering intraocular pressure (IOP) is currently the only effective target for therapeutic intervention in glaucoma. IOP is mostly controlled by the outflow of the aqueous humor (AH) through the trabecular meshwork (TM). The TM and adjacent endothelium of Schlemm’s canal, known as the conventional outflow-tract, control AH outflow and thus determine IOP.

Exposure to 1,25-Dihydroxyvitamin D (1,25D) decreases proliferation and induces differentiation in telomerase-immortalized human mammary epithelial (hTERT-HME1) cells. The studies described here addressed the mechanisms by which these effects are exerted. Microarray experiments were used to identify a subset of metabolic genes and pathways that are altered by 1,25D. In particular, genes involved in glutamate and glutamine utilization, including SLC1A1 and GLUL, were studied. Interestingly, qPCR analysis in a panel of six cell lines, representing either normal epithelial tissue or breast cancer, demonstrated diverse gene expression responses to 1,25D. Follicular fluid (FF) is in direct contact with granulosa cells that surround a developing oocyte, and high density lipoprotein (HDL) is the sole lipoprotein found in FF. Therefore, we investigated 41 FF HDL particle components, including 26 HDL-cholesterol (HDL-C) subfractions measured for particle size, apolipoproteins, enzymes, and lipophilic micronutrients, in association with embryo quality on 180 women undergoing in vitro fertilization (IVF).

Foraminifera have unusually fast dynamic movements resulting in the reorganization of the microtubule cytoskeleton (Travis and Bowser, 1988; Travis et al., 1983). We are interested in using Reticulomyxa filosa as a model to investigate the mechanisms and factors that regulate such fast microtubule assembly/disassembly rates (Chen and Schliwa 1990; Travis and Bowser, 1991; Welnhofer and Travis, 1996). Compared to conventional microtubule assembly and disassembly models, foraminifera tubulin can exist in another assembly state termed helical filaments.
