The purpose of this culminating comprehensive project is to advise adult empowerment focused NGOs on the importance of identifying and leveraging the strengths of the individuals they serve. Given the evidence that all individuals innately have strengths, it is important to be aware of them in order to properly engage such assets. Character strengths are studied in this paper as one of the main pillars in the field of positive psychology. The N-terminal 25 amino acids (AAs) of turnip crinkle virus (TCV) capsid protein (CP) are recognized by the resistance protein HRT to trigger a hypersensitive response (HR) and systemic resistance to TCV infection. This same region of TCV CP also contains a motif that interacts with the transcription factor TIP, as well as a nuclear localization signal (NLS). However, it is not yet known whether nuclear localization of TCV CP is needed for the induction of HRT-mediated HR and resistance. Here we present new evidence suggesting a tight correlation between nuclear inclusions formed by CP and the manifestation of HR. …

Simian immunodeficiency virus (SIV) is a robust pathogen used in non-human primates to model HIV vaccines. SIV encodes a number of potential vaccine targets. By far the largest and most conserved protein target in SIV is its gag-pol protein that bears many epitopes to drive multivalent immune T cell responses. While gag-pol is an attractive antigen, it is only translated after a frame shift between gag and pol with the effect that gag and pol are expressed at an approximate 10/1 ratio. The codon bias of native lentiviral genes are also mismatched with the abundance of tRNAs in mammalian cells …

The purpose of this study was to determine whether a relationship exists between Indiana End-of-Course Assessments (ECA) in English 10 and Algebra I and the following indicators at the high school level: (a) socioeconomic status, (b) instructional expenditures per student, (c) attendance rate, (d) school size, (e) percentage of ELLs, (f) mobility rate, (g) minority rate, and (h) percentage of specialEducation students. This study examined which indicators had the greatest effect on English 10 ECA scores and Algebra I ECA scores in Indiana. In this thesis, I analyze Reducing the Risk: Building the Skills to Prevent Pregnancy, STD & HIV (RTR). As one of the first evidence-based curricula in the U.S., RTR is now one of the numerous programs funded by the U.S. government. While this curriculum has been evaluated twice for its ability to influence student sexual behavior, messages found in the inclusion of abstinence and protection has not. Using an open coding method, I delve into this curriculum and find a disconnect between the stated goal of RTR—to provide students with the skills to abstain and use protection—and the material presented. …

This thesis explores adaptation through translation in Luis Riaza’s dramatic text, Antígona...¡Cerda!. I have translated this play from Spanish to English and staged it at New College of Florida. The first chapter describes the sociohistorical and cultural context in which Riaza wrote the play as well as an explanation and analysis of his style of work. My translation of the play is the second chapter, which includes notes on directorial and translation choices. The final chapter is a reflection on the performance and the rehearsal process including an explanation on specific actor technique. A copy of the performance is attached …

Bats are highly social, but the study of bat social behavior was limited until recently due to technological limitations. Most of bat behavior is imperceptible to our senses, including both their use of ultrasound and their nocturnal activities. During the maternity seasons (May-August) of 2013 and 2014 near Plainfield, Indiana, we recorded Indiana bat (Myotis sodalis) roost site behaviors with emergence count observation, video (Sony Nightshot HandyCams and IR lights), and acoustic detectors (Pettersson D500X ultrasonic detectors). Since the birth of modern artificial intelligence (AI) at the 1956 Dartmouth Conference, the AI community has pursued modeling and coding of human intelligence into AI reasoning processes (HI Þ MI). The Dartmouth Conference's fundamental assertion was that every aspect of human learning and intelligence could be so precisely described that it could be simulated in AI. With the exception of knowledge specific areas (such as IBM's Big Blue and a few others), sixty years later the AI community is not close to coding global human intelligence into AI. Intracytoplasmic membranes are structures that form within cells which help facilitate a variety of different metabolic processes. This feature of intracellular membranes makes them particularly valuable for studying compartmentalization and cell dynamics in bacteria. In the past, transmission electron microscopy has been the primary method for imaging bacteria with intracytoplasmic membranes. Because transmission electron microscopy takes images of a cell in fixed slices, it’s impossible to follow a cell’s growth and development over time. Fluorescence microscopy is a particularly effective method of measurement that can combat these issues when evaluating live bacterial cells. Maybe the most important lesson that can be learned from cases like Myriad (ones in which the legal problems are complex) is a subtle one: the big picture is complicated. After all, if every case were easy to resolve on the merits, all lawyers and judges would be out of jobs quickly. Technology is complex, also. Given its importance to the law, it is regrettable that judges and lawyers do not fully understand how the DSM is constructed, and the bedrock of values on which it rests. As evidence of this, lawyers and judges often refer to the DSM as the “psychiatric bible.” This language is both fascinating and perplexing. This Article will attempt to correct the notion that the DSM is a legal “psychiatric bible” by explaining how it is created and used by the medical field. In defending Queen Caroline in the House of Lords, Henry Brougham declared, “[a]n advocate, by the sacred duty of his connection with his client, knows, in the discharge of that office, but one person in the world, that client and none other.” Brougham’s ethic of advocacy has been cited repeatedly as stating the American lawyer’s duty of zealous representation of a client. It has often been called the “classic statement” of zealous representation and representing the “traditional view of the lawyer’s role.”

Overexpression of 5-hydroxytryptamine (5-HT) in human cancer contributes to tumor metastasis, but the role of 5-HT receptor family in cancer has not been thoroughly explored. Here, we report overexpression of 5-HT1D receptor (5-HT1DR) was associated with Wnt signaling pathway and advanced tumor stage. The underlying mechanism of 5-HT1DR-promoted tumor invasion was through its activation on the Axin1/β-catenin/MMP-7 pathway. In an orthotopic colorectal cancer mouse model, we demonstrated that a 5-HT1DR antagonist (GR127935) effectively inhibited tumor metastasis through targeting Axin1. Epidermal growth factor receptor (EGFR) mutants drive lung tumorigenesis and are targeted for therapy. However, resistance to EGFR inhibitors has been observed, in which the mutant EGFR remains active. Thus, it is important to uncover mediators of EGFR mutant-driven lung tumors to develop new treatment strategies. The protein tyrosine phosphatase (PTP) Shp2 mediates EGF signaling. Nevertheless, it is unclear if Shp2 is activated by oncogenic EGFR mutants in lung carcinoma or if inhibiting the Shp2 PTP activity can suppress EGFR mutant-induced lung adenocarcinoma. All Staphylococcus aureus genomes contain a genomic island, which is termed νSaα and characterized by two clusters of tandem repeat sequences, i.e. the exotoxin (set) and 'lipoprotein-like' genes (lpl). Based on their structural similarities the νSaα islands have been classified as type I to IV. The genomes of highly pathogenic and particularly epidemic S. aureus strains (USA300, N315, Mu50, NCTC8325, Newman, COL, JH1 or JH9) belonging to the clonal complexes CC5 and CC8 bear a type I νSaα island. Recent observations have identified a new species of leucothoid amphipod, Leucothoe “sp. F,” associated with the sponge Cliona varians. This project examined the relationship between this amphipod and its sponge host at three sites in the Florida Keys with differing hydrodynamic regimes. Ninety-eight sponge samples with a total of 2,030 amphipods were collected between December 2011 and September 2012. Leucothoe “sp. F” is currently a common species in the Florida Keys strongly associated with C. varians; its distribution strongly coincides with open tidal currents from the Gulf of Mexico. Caffeine is one of the most popular psychostimulant drugs worldwide. Its effects are exerted through a variety of complex mechanisms, apparently primarily via interactions with adenosine A1 and A2A receptors. This drug has also been proven to elicit neuroprotective responses in a number of different brain disorders of the Central Nervous System (CNS), as well as provide enhancement of cognitive abilities. In 2014, a major epidemic of human Ebola virus disease emerged in West Africa, where human-to-human transmission has now been sustained for greater than 12 months. In the summer of 2014, there was great uncertainty about the answers to several key policy questions concerning the path to containment. What is the relative importance of nosocomial transmission compared with community-acquired infection? How much must hospital capacity increase to provide care for the anticipated patient burden? Undergraduate research experiences have become a widely accepted goal of colleges and universities for enhancing student development and success. Texas Southern University (TSU) is an Historically Black College and University (HBCU) with approximately 6,000 undergraduate students and as with most HBCUs, retention and graduation rates are extremely low. This study addresses the relationship between undergraduate research at TSU and progression, graduation rates and GPA. The experimental population includes 34 students that participated at a 10-week undergraduate research program (URP) during the summers of 2012 or 2013. Engineered metal oxide nanoparticles (MO NPs) are finding increasing utility in the medical fields as anticancer agents. Before validation of in vivo anticancer efficacy can occur, a better understanding of whole-animal toxicity is required. We compared the toxicity of seven widely used semiconductor MO NPs made from zinc oxide (ZnO), titanium dioxide, cerium dioxide and tin dioxide prepared in pure water and in synthetic seawater using a five-day embryonic zebrafish assay. Opinion is hardly unanimous, but many authors endorse the idea that Chaco Canyon is and was a marginal place for growing corn (Zea mays), a chief source of food energy for Puebloan groups in the Southwest. Poor soils with “toxic” levels of salts, inadequate and unpredictable precipitation, and a short growing season have all been identified as contributing to the agricultural marginality of the place (Benson 2011a; Bryan 1954; Force et al. 2002; Judd 1954:59–61). Enteric methane production from cattle and its effect on climate change has been a topic of debate. Multiple studies have explored methods to reduce cattle enteric methane production while simultaneously improving performance. However, most strategies developed have not been widely implemented by cattle producers. Numerous advancements in machinery performance on row-crop agricultural tractors have been made in recent years. Currently, the Organisation for Economic Co-operation and Development (OECD) only tests predetermined points (E.g., max. power, max. torque) for drawbar and hydraulic power. This method of testing has been used for many years with the tractor at a steady state which is usually not typical of row-crop operations. An integral part of functional genomics studies is to assess the enrichment of specific biological terms in lists of genes found to be playing an important role in biological phenomena. Contrasting the observed frequency of annotated terms with those of the background is at the core of overrepresentation analysis (ORA). Gene Ontology (GO) is a means to consistently classify and annotate gene products and has become a mainstay in ORA. Alternatively, Medical Subject Headings (MeSH) offers a comprehensive life science vocabulary including additional categories that are not covered by GO. We have characterized the molecular and genomic diversity of the microbiota of the starlet sea anemone Nematostella vectensis, a cnidarian model for comparative developmental and functional biology and a year-round inhabitant of temperate salt marshes. Molecular phylogenetic analysis of 16S rRNA gene clone libraries revealed four ribotypes associated with N. vectensis at multiple locations and times. These associates include two novel ribotypes within the ε-Proteobacterial order Campylobacterales and the Spirochetes, respectively, each sharing 99% 16S rRNA identity with Endozoicomonas elysicola and Pseudomonas oleovorans, respectively. La Reserva de la Biosfera Barranca de Metztitlán es un área natural protegida por la gran diversidad de flora y fauna existente en la misma. La zona es atravesada por un río de gran longitud que se extiende desde el municipio de Tulancingo hasta su desembocadura en la Laguna de Metztitlán, en la zona núcleo de la reserva. En el presente trabajo se realizó un análisis sobre la percepción de calidad de vida, contaminación y riesgo en nueve comunidades rurales del Municipio de Tecoanapa, Guerrero. Para ello, se seleccionaron nueve comunidades que cumplieran con tres aspectos: rezago social (muy alto, medio y muy bajo); ubicación (cerca de ríos); y vivencias en conflictos y embates ambientales. Se levantó una encuesta a jefes de familia en febrero de 2014. Registrando que el 70% de los habitantes afirmaron tener una buena calidad de vida. Existen muy pocos trabajos etnobotánicos realizados en la Reserva de la Biosfera Barranca de Metztitlán, por lo que el objetivo de este estudio fue generar un listado preliminar de plantas útiles de la Reserva. Se aplicaron entrevistas a habitantes de localidades ubicadas en la zona, para colectar las plantas útiles mencionadas se hicieron recorridos de campo en compañía de informantes, posteriormente las especies fueron identificadas con el uso de claves dicotómicas. Se registraron 83 especies de plantas útiles, distribuidas en 39 familias y 20 categorías de uso, siendo la medicinal la mejor representada por número de especies. Conocer el uso …
