Biomedical and Pharmaceutical Sciences Faculty Publications

Objective: To evaluate whether Nrf2 deficiency impacts insulin resistance and lipid accumulation in liver and white adipose tissue.

Methods: Lepob/ob mice (OB) with targeted Nrf2 deletion (OB‐Nrf2KO) were generated. Pathogenesis of obesity and type 2 diabetes was measured in C57BL/6J, Nrf2KO, OB, and OB‐Nrf2KO mice. Hepatic lipid content, lipid clearance, and very low‐density lipoprotein (VLDL) secretion were determined between OB and OB‐Nrf2KO mice.

Results: OB‐Nrf2KO mice exhibited decreased white adipose tissue mass and decreased adipogenic and lipogenic gene expression compared with OB mice. Nrf2 deficiency prolonged hyperglycemia in response to glucose challenge, which was paralleled by reduced insulin‐stimulated Akt …

The Brighton Collaboration Viral Vector Vaccines Safety Working Group (V3SWG) was formed to evaluate the safety of live, recombinant viral vaccines incorporating genes from heterologous viruses inserted into the backbone of another virus (so-called "chimeric virus vaccines"). Many viral vector vaccines are in advanced clinical trials. The first such vaccine to be approved for marketing (to date in Australia, Thailand, Malaysia, and the Philippines) is a vaccine against the flavivirus, Japanese encephalitis (JE), which employs a licensed vaccine (yellow fever 17D) as a vector. In this vaccine, two envelope proteins (prM-E) of YF 17D virus were exchanged for the corresponding …

Distinct Signaling Mechanisms Of Mtorc1 And Mtorc2 In Glioblastoma Multiforme: A Tale Of Two Complexes, Meena Jhanwar-Uniyal, John L. Gillick, Jayson Neil, Michael Tobias, Zachary E. Thwing, Raj Murali

NYMC Faculty Publications

Mechanistic target of rapamycin (mTOR) is a serine-threonine kinase that functions via two multiprotein complexes, namely mTORC1 and mTORC2, each characterized by different binding partners that confer separate functions. mTORC1 function is tightly regulated by PI3-K/Akt and is sensitive to rapamycin. mTORC2 is sensitive to growth factors, not nutrients, and is associated with rapamycin-insensitivity. mTORC1 regulates protein synthesis and cell growth through downstream molecules: 4E-BP1 (also called EIF4E-BP1) and S6K. Also, mTORC2 is thought to modulate growth factor signaling by phosphorylating the C-terminal hydrophobic motif of some AGC kinases such as Akt and SGK. Recent evidence has suggested that mTORC2 …

I first met the amazing HeroRATs in 2008 when I was an APOPO deminer detecting landmines in Mozambique. I had been invited to APOPO’s HQ and Training & Research Center in Tanzania and was about to embark on my new career as a Mine Detection Rat Handler. Although I had worked with trained dogs before I was still amazed at the rat’s abilities; their speed and accuracy. The Tanzania team taught me about scent detection theory and technology, as well as animal welfare and veterinary skills. In return I told them about the suffering in my country. At that time, …

Marginalized Fathers And Demonized Mothers: A Feminist Look At The Reproductive Freedom Of Unmarried Men, Michael J. Higdon Jan 2015

Marginalized Fathers And Demonized Mothers: A Feminist Look At The Reproductive Freedom Of Unmarried Men, Michael J. Higdon

Scholarly Works

Just last month, in the state of Utah, twelve biological fathers filed suit, challenging the state’s adoption laws — laws the fathers allege permit “legalized fraud and kidnapping.” Specifically, these laws require nonmarital fathers to promptly take legal action in Utah to preserve their paternal rights. A problem arises, however, as mothers from other states have started traveling to Utah specifically to surrender newborn children for adoption. The fathers, unaware that their children are being placed for adoption in another state, fail to take action in Utah and, as a result, are permanently deprived of all parental rights. In that …

The Use Of Commercial Bacterial Soil Inoculant Regime In An Urban Prairie Restoration, W. J. Leonard, Kelly G. Lyons Jan 2015

The Use Of Commercial Bacterial Soil Inoculant Regime In An Urban Prairie Restoration, W. J. Leonard, Kelly G. Lyons

Biology Faculty Research

For successful grassland restoration, commercial soil inoculants are often recommended to increase establishment success. In spring 2009, a 0.94-ha tract was targeted for restoration at Phil Hardberger Park, a 126-ha park in the heart of San Antonio, Texas. Woody species, mainly Texas persimmon (Diospyros texana Scheele), and Ashe juniper (Juniperus asheii Buchholz), were removed and the area was divided into 10 subplots measuring 911 m2 on average. In September 2009, over 40,000 plugs of seven native grass species were planted. In addition, native prairie seed mixes, including various grass and forb seeds, were sown into the site …

Convergent Evolution Of Brain Morphology And Communication Modalities In Lizards, Christopher D. Robinson, Michael S. Patton, Brittany M. Andre, Michele A. Johnson Jan 2015

Convergent Evolution Of Brain Morphology And Communication Modalities In Lizards, Christopher D. Robinson, Michael S. Patton, Brittany M. Andre, Michele A. Johnson

Biology Faculty Research

Animals communicate information within their environments via visual, chemical, auditory, and/or tactile modalities. The use of each modality is generally linked to particular brain regions, but it is not yet known whether the cellular morphology of neurons in these regions has evolved in association with the relative use of a modality. We investigated relationships between the behavioral use of communication modalities and neural morphologies in six lizard species. Two of these species (Anolis carolinensis and Leiocephalus carinatus) primarily use visual signals to communicate with conspecifics and detect potential prey, and two (Aspidoscelis gularis and Scincella lateralis) …

Evolutionary History Influences The Salinity Preference Of Bacterial Taxa In Wetland Soils, Ember M. Morrisey, Rima B. Franklin Jan 2015

Evolutionary History Influences The Salinity Preference Of Bacterial Taxa In Wetland Soils, Ember M. Morrisey, Rima B. Franklin

Biology Publications

Salinity is a major driver of bacterial community composition across the globe. Despite growing recognition that different bacterial species are present or active at different salinities, the mechanisms by which salinity structures community composition remain unclear. We tested the hypothesis that these patterns reflect ecological coherence in the salinity preferences of phylogenetic groups using a reciprocal transplant experiment of fresh- and saltwater wetland soils. The salinity of both the origin and host environments affected community composition (16S rRNA gene sequences) and activity (CO2 and CH4 production, and extracellular enzyme activity). These changes in community composition and activity rates were …

Risk Assessment Based On Indirect Predation Cues: Revisiting Fine-Grained Variation, Michael W. Mccoy, Stefan K. Wheat, Karen M. Warkentin, James R. Vonesh Jan 2015

Risk Assessment Based On Indirect Predation Cues: Revisiting Fine-Grained Variation, Michael W. Mccoy, Stefan K. Wheat, Karen M. Warkentin, James R. Vonesh

Biology Publications

To adaptively express inducible defenses, prey must gauge risk based on indirect cues of predation. However, the information contained in indirect cues that enable prey to fine-tune their phenotypes to variation in risk is still unclear. In aquatic systems, research has focused on cue concentration as the key variable driving threat-sensitive responses to risk. However, while risk is measured as individuals killed per time, cue concentration may vary with either the number or biomass killed. Alternatively, fine-grained variation in cue, that is, frequency of cue pulses irrespective of concentration, may provide a more reliable signal of risk. Here, we present …

Knowledge of the diversity and ecological function of the microbial consortia of James River in Virginia, USA, is essential to developing a more complete understanding of the ecology of this model river system. Metagenomic analysis of James River's planktonic microbial community was performed for the first time using an unamplified genomic library and a 16S rDNA amplicon library prepared and sequenced by Ion PGM and MiSeq, respectively. From the 0.46-Gb WGS library (GenBank:SRR1146621; MG-RAST:4532156.3), 4 × 106 reads revealed >3 × 106 genes, 240 families of prokaryotes, and 155 families of eukaryotes. From the 0.68-Gb 16S library (GenBank:SRR2124995; MG-RAST:4631271.3; EMB:2184), …
