Rational Design Of A Chalcogenopyrylium-Based Surface-Enhanced Resonance Raman Scattering-Nanoprobe With Attomolar Sensitivity, Stefan Harmsen, Matthew A. Bedics, Matthew A. Wall, Ruimin Huang, Michael R. Detty, Moritz F. Kircher

Publications and Research

High sensitivity and specificity are two desirable features in biomedical imaging. Raman imaging has surfaced as a promising optical modality that offers both. Here, we report the design and synthesis of a group of near infrared absorbing 2-thienyl-substituted chalcogenopyrylium dyes tailored to have high affinity for gold. When adsorbed onto gold nanoparticles, these dyes produce biocompatible SERRS-nanoprobes with attomolar limits of detection amenable to ultrasensitive in vivo multiplexed tumor and disease marker detection.

© 2015 by the authors; licensee MDPI, Basel, Switzerland. Oncostatin M (OSM) is a cytokine belonging to the gp130 family, whose members serve pleiotropic functions. However, several actions of OSM are unique from those of other gp130 cytokines, and these actions may have critical roles in inflammatory mechanisms influencing several metabolic and biological functions of insulin-sensitive tissues. In this review, the actions of OSM in adipose tissue and liver are discussed, with an emphasis on lipid metabolism.

Long-Term Forest Soil Warming Alters Microbial Communities In Temperate Forest Soils, Kristen M. Deangelis, Grace Pold, Begum D. Topcuoglu, Linda T. A. Van Diepen, Rebecca M. Varney, Jeffrey L. Blanchard, Jerry Melillo, Serita D. Frey Feb 2015

Long-Term Forest Soil Warming Alters Microbial Communities In Temperate Forest Soils, Kristen M. Deangelis, Grace Pold, Begum D. Topcuoglu, Linda T. A. Van Diepen, Rebecca M. Varney, Jeffrey L. Blanchard, Jerry Melillo, Serita D. Frey

Faculty Publications

Soil microbes are major drivers of soil carbon cycling, yet we lack an understanding of how climate warming will affect microbial communities. Three ongoing field studies at the Harvard Forest Long-term Ecological Research (LTER) site (Petersham, MA) have warmed soils 5°C above ambient temperatures for 5, 8, and 20 years. We used this chronosequence to test the hypothesis that soil microbial communities have changed in response to chronic warming. Bacterial community composition was studied using Illumina sequencing of the 16S ribosomal RNA gene, and bacterial and fungal abundance were assessed using quantitative PCR. Only the 20-year warmed site exhibited significant …

Matural Exams In Kosovo: An Analysis On 2014 Test Results, Genta Agaj Feb 2015

Matural Exams In Kosovo: An Analysis On 2014 Test Results, Genta Agaj

Theses

The focus of this research project was to address some of the criticisms associated with Kosovo’s standardized tests - Matura Exams. The main goal of the project was to statistically analyze students’ test results using three statistical techniques. This problem was tackled by obtaining the 2014 results for the Municipality of Prishtina.

First, Prishtina’s Matura Exam test results were checked for normality. The results from this test suggested that even though Matura Exam is supposed to have normally-distributed results, it failed to do so. Its implications include students and universities not being able to compare individual results with the population …

Flipping The Classroom And Student Performance In Advanced Statistics: Evidence From A Quasi-Experiment, Michael Touchton Feb 2015

Flipping The Classroom And Student Performance In Advanced Statistics: Evidence From A Quasi-Experiment, Michael Touchton

Political Science Faculty Publications and Presentations

I administer a quasi-experiment using undergraduate political science majors in statistics classes to evaluate whether “flipping the classroom” (the treatment) alters students’ applied problem-solving performance and satisfaction relative to students in a traditional classroom environment (the control). I also assess whether general student characteristics such as when and where students took the prerequisite course, grade point average (GPA), and gender influence performance. I find flipping the classroom gives students statistically significant advantages in difficult, applied areas emphasized in class. Furthermore, students in the flipped classroom feel they learned more and enjoyed the course more than those in a traditional classroom. …

Born Naked By Farley Mowat, Ted Geier Feb 2015

Born Naked By Farley Mowat, Ted Geier

The Goose

Farley Mowat's charming childhood memoir covers the usual Mowat terrain: riotous humour, humble reverence, and a meticulous accounting of the little things that make life--all life--dear to us. Mowat's work deserves regular attention in animal studies and environmental literary studies. His autobiographical techniques, as this review suggests, can be both assets and occasional impediments. But there is no replacing or replicating a Farley Mowat, and Born Naked deserves to be read immediately and repeatedly as one of the lasting legacies of a long life, well-lived.

© 2015 Australasian Society for Immunology Inc. Respiratory syncytial virus (RSV) infection remains a significant global health burden disproportionately affecting infants and leading to long-term lung disease. Interleukin (IL)-17A has been shown to be involved in regulating viral and allergic lung inflammatory responses, which has led to a more recent interest in its role in RSV infection. Using a neonatal mouse model of RSV, we demonstrate that neonates fail to develop IL-17A responses compared with adult mice; the main immediate IL-17A contributor in adults were γδ T cells. Antibody neutralization of IL-17A in adult mice caused increased lung inflammation and …

Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, we conducted genome-wide association meta-analyses of waist and hip circumference-related traits in up to 224,459 individuals. We identified 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (WHRadjBMI) and an additional 19 loci newly associated with related waist and hip circumference measures (P<5×10-8). Twenty of the 49 WHRadjBMI loci showed significant sexual dimorphism, 19 of which displayed a stronger effect in women. The identified loci were enriched for genes expressed in adipose tissue and for putative regulatory elements in adipocytes. Pathway analyses implicated adipogenesis, angiogenesis, transcriptional regulation, and insulin resistance as processes affecting fat distribution, providing insight into potential pathophysiological mechanisms.

Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10-8), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20 % of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

Background

Peripheral artery disease (PAD), which affects an estimated 27 million people in Europe and North America, is caused by atherosclerotic plaques that limit blood flow to the legs. Chronic, repeated ischemia in the lower leg muscles of PAD patients is associated with loss of normal myofiber morphology and myofiber degradation. In this study, we tested the hypothesis that myofiber morphometrics of PAD calf muscle are significantly different from normal calf muscle and correlate with reduced calf muscle strength and walking performance.

Methods

Gastrocnemius biopsies were collected from 154 PAD patients (Fontaine stage II) and 85 control subjects. Morphometric parameters …

We use fluctuational electrodynamics to determine spectral emissivity and van der Waals contribution to surface energy for various spherical shapes, such as a sphere, a bubble, a spherical shell, and a coated sphere, in a homogeneous and isotropic medium. The dyadic Green's function formalism of radiative energy and fluctuation-induced van der Waals stress for different spherical configurations has been developed. We show (1) emission spectra of micro- and nano-sized single and coated spheres display several emissivity sharp peaks as the size of object reduces and (2) surface energy becomes size dependent due to van der Waals phenomena when size of …

Elimination Of Hydrogenase Active Site Assembly Blocks H2 Production And Increases Ethanol Yield In Clostridium Thermocellum, Ranjita Biswas, Tianyong Zheng, Daniel G. Olson, Lee R. Lynd, Adam M. Guss

Dartmouth Scholarship

Background: The native ability of Clostridium thermocellum to rapidly consume cellulose and produce ethanol makes it a leading candidate for a consolidated bioprocessing (CBP) biofuel production strategy. C. thermocellum also synthesizes lactate, formate, acetate, H2 , and amino acids that compete with ethanol production for carbon and electrons. Elimination of H2 production could redirect carbon flux towards ethanol production by making more electrons available for acetyl coenzyme A reduction to ethanol. Results: H2 production in C. thermocellum is encoded by four hydrogenases. Rather than delete each individually, we targeted hydrogenase maturase gene hydG, involved in converting the …
