Ketone Body Metabolism Preserves Hepatic Function During Adaptation To Birth And In Overnutrition, David Graham Cotter

Mammalian ketone body metabolism partially oxidizes hepatic acyl-chains to ketone body intermediates, which can serve as alternative fuels in extrahapetic tissues during carbohydrate restricted states. Ketone body production (ketogenesis) occurs primarily in liver, due to hepatocyte-specific expression of the fate committing ketogenic enzyme, mitochondrial 3-hydroxymethylglutaryl-CoA synthase (HMGCS2). In contrast, the fate committing enzyme of ketone body oxidation, mitochondrial Succinyl-CoA:3-oxoacid CoA Transferase (SCOT), is expressed ubiquitously, except in liver. Here I demonstrate novel roles for ketone body metabolism during a classically ketogenic period, the transition to birth, and in a classically `non-ketogenic' state, overnutrition, using novel genetic mouse models, high-resolution measures …

Urinary tract infections (UTIs) are among the most common bacterial infections worldwide, costing greater than $2 billion in healthcare costs and lost wages yearly in America alone. The lifetime risk for a woman exceeds 50% with 25-40% of these suffering recurrent infections. Two of the most important risk factors for recurrent UTI are prior UTIs and sexual intercourse. Over 80% of UTIs are caused by uropathogenic Escherichia coli (UPEC), which binds and invades superficial facet cells lining the bladder surface. UPEC expresses extracellular fibers called type 1 pili with a terminal mannose-binding adhesin, FimH, which interacts with mannosylated uroplakin residues …

Background Ossabaw miniature swine when fed a diet high in fructose, saturated fat and cholesterol (NASH diet) develop metabolic syndrome and nonalcoholic steatohepatitis (NASH) characterized by liver injury and fibrosis. This study was conducted to further characterize the development of NASH in this large animal model. Methods Ossabaw swine were fed standard chow (control group; n = 6) or NASH diet (n = 6) for 24 weeks. Blood and liver tissue were collected and liver histology were characterized at 0, 8, 16 and 24 weeks of dietary intervention. Hepatic apoptosis and lipid levels were assessed at week 24. This thesis examines the intersections of bodybuilding and performance through a masculine lens. Much like theater, the body builder is activated by staging. In an investigation of its history and a sampling of its theories, we can challenge the hyper-masculine identity which is supported through the gym culture and gender constructs. The arena of the gym contains cues contributing to the artworks listed, and the icons of the gym itself are brought into the studio to transform not only the objects, but the actions themselves. This dissertation comprises three papers on spatial features of labor markets and links to the housing market. The first two papers look at how a local parental leave policy and the neighborhood in which one resides can influence women's decision to work. One paper shows that New Jersey's 2009 family leave insurance program induces women to remain employed following childbirth. The other reveals that, for women, having other women with similar aged children to yours among your closest neighbors makes you emulate their work behavior. Intimate partner violence (IPV) affects the health and mental health of many women in the United States, with some estimates projecting that one in three women will experience physical, sexual, and/or psychological abuse from a current or former partner during their lifetimes (National Intimate Partner and Sexual Violence Survey [NISVS], 2011). This dissertation investigates the relationship between mobility and urbanism in an Iron Age West African city. Isotopic and archaeological evidence reveal complex relationships among mobility, sedentism, and the livestock economy of Jenné-jeno, one of the earliest and best known urban systems in sub-Saharan Africa (occupied ca. 250 BC to AD 1400). Drawing on excavations at the ancient city of Jenné-jeno and those conducted at two neighboring sites (Tato à Sanouna and Thièl), and on serial, intra-tooth isotopic analysis (87Sr/86Sr, 13C, 18O) of cattle, sheep, and goat teeth, this analysis shows that multiple populations involved in animal husbandry, from seasonally mobile …

Game meats represent a multibillion dollar industry in the United States with high economic incentives associated with species substitution and mislabeling. However, there is currently a lack of information regarding the prevalence of mislabeled game meat on the U.S commercial market. In the pursuit of green chemistry, adequate substitutes for lead acetate and hydrogen sulfide were investigated for use in the ACS test procedure for stabilizer in meta-phosphoric acid. In the current ACS procedure lead acetate is used to form insoluble lead phosphate. In order to remove lead from the solution lanthanum chloride was investigated with positive results. Hydrogen Sulfide is currently used to precipitate lead by forming insoluble lead sulfide. Several substitutes including oxalic acid and sulfuric acid were investigated. Preliminary testing showed that lanthanum chloride and oxalic acid are sufficient in precipitating phosphate and lead out of the sample …

The purpose of this study was to determine the existence of sex differences following a 40% caloric restrictive diet and its impact on tibial bone mineral density (BMD) and bone strength between male and female rats. Thirty-two, six-week old Sprague Dawley rats (16 males and 16 females) were randomly divided into an ad libitum fed control group (MC, n=8 and FC, n=8) and a pair fed diet group placed on the 40% caloric restriction (MD, n=8 and FD, n=8) for a 6 week period. Hypoxia, or oxygen depletion, is a process that occurs in conjunction with eutrophication. In hypoxic conditions the dissolved oxygen levels in the water column sink to unlivable conditions for the marine organisms causing them to flee or die. Despite efforts of improvement, the annual summer Gulf of Mexico dead zone continues to be a serious threat to aquatic ecosystems. The 2012 smaller than usual dead zone was a false indicator of future improvement. In this study, we evaluated the chemical and physiological effect of low-dose gamma irradiation on the post-harvest quality of Chandler Pummelos (Citrus maxima (Burm.) Merr.), an emerging crop of interest in the U.S. Chandler pummelos from a local grower in California were irradiated at target doses of 150 Gy and 1000 Gy. Irradiated and untreated pummelos were stored at 12 °C for 3 weeks and at 20 °C for the 4th week to reflect three weeks of sea shipment at ideal temperature for storage of pummelos and an additional week of retail under ambient conditions. The risk of poor food safety is a major focus for managers in the food manufacturing industry. Despite industry-led and regulatory efforts to improve the overall food safety of US packaged consumer foods, product recalls and market withdrawals are increasing. This is especially true for the most frequent cause for food recall: the undeclared allergen. Elder abuse is a serious issue that negatively affects physical and mental health in a significant portion of the older population. Abuse of older adults is not adequately detected or reported. Nurses have frequent contact with older victims of abuse, providing them the opportunity to play a significant role in detecting, reporting, and intervening in such cases. Introduction: Parkinson’s disease (PD) is a chronic progressive movement disorder, affecting primarily people over sixty years of age. With aging populations, some estimates indicate as many as nine million people will be impacted by this disease by 2030. Thus it is imperative to identify means of prevention, delay of onset, and management of symptoms. Many elements of diet and nutrition have been investigated to accomplish these goals. This review will examine recent studies of potential diet and nutrition factors and PD incidence.

Insects are increasingly being recognized not only as a source of food to feed the ever growing world population but also as potential sources of new products and therapeutic agents, among which are sterols. In this study, we sought to profile sterols and their derivatives present in the desert locust, Schistocerca gregaria, focusing on those with potential importance as dietary and therapeutic components for humans. Using coupled gas chromatography-mass spectrometry (GC-MS), we analyzed and compared the quantities of sterols in the different sections of the gut and tissues of the locust. Social Communication Of Predator-Induced Changes In Drosophila Behavior And Germ Line Physiology, Balint Z. Kacsoh, Julianna Bozler, Mani Ramaswami, Giovanni Bosco

Behavioral adaptation to environmental threats and subsequent social transmission of adaptive behavior has evolutionary implications. In Drosophila, exposure to parasitoid wasps leads to a sharp decline in oviposition. We show that exposure to predator elicits both an acute and learned oviposition depression, mediated through the visual system. However, long-term persistence of oviposition depression after predator removal requires neuronal signaling functions, a functional mushroom body, and neurally driven apoptosis of oocytes through effector caspases. Strikingly, wasp-exposed flies (teachers) can transmit egg-retention behavior and trigger ovarian apoptosis in naive, unexposed flies (students).
