Integrated Continuous Biomanufacturing II
Mammalian cell cultures are used to produce a range of recombinant therapeutic proteins where post-translational modifications, such as glycosylation, are of key importance. The demand for proteins expressed in mammalian cells has increased rapidly over the last decade, primarily driven by the success of monoclonal antibodies (mAbs) that are required at large doses for therapeutic use. Several host cell lines are employed for manufacturing (e.g. NS0, hybridomas, PER.C6, BHK, etc.) but the primary workhorses are Chinese Hamster Ovary (CHO)-derived cell lines accounting for over 70% of the recombinant therapeutics. CHO cells have a proven track record as a safe host …

Here, we present a fast and convenient strategy for developing a high-cell density perfusion process for antibody-producing Chinese hamster ovary (CHO) cells based on the commercially available ActiCHO™ Media System. ActiCHO P base medium was used as a starting point and ActiCHO Feed-A and Feed-B were added in various concentrations as supplements. The resulting perfusion medium prototypes were first evaluated in batch cultures, applying a design of experiment (DoE) strategy (Figure 1), and then tested in small-scale perfusion cultures in rocking single-use WAVE bioreactor™ systems (Figure 2). The medium optimization resulted in a final process with a cell-specific perfusion rate …

Integrated Continuous Biomanufacturing II
There is increasing interest in perfusion processing as a means for increasing overall yield of the upstream process, facilitating a reduction of manufacturing costs. However, as in many companies the focus was on fed-batch process development for decades, hands-on knowledge about medium development for perfusion processes is considered a key gap for the wider adoption of perfusion processing in biologics manufacturing today.
Compared to batch-based cultivation, perfusion processes have fundamentally different requirements and challenges in terms of medium composition. The continuous nature of perfusion processes allows to achieve steady-state, although not all practitioners consider this desirable. The main challenge in …

Carotenes such as β-carotene have a positive impact in human health as a precursor of vitamin A. Structural complexity of these compounds makes their chemical synthesis a difficult endeavor, facilitating the need for their biological production. Continuous operation can be used in these processes to allow for increased cell viability through the mitigation of substrate and product inhibition, thereby increasing the productivity of the carotenoid product. This work looks to examine the operating conditions necessary to achieve the maximum production of the desired carotenoid products when produced via continuous operation. First, a two-level parameter estimation method is utilized to develop …

Biosimilar drugs are emerging very fast as the patents of innovator’s drugs are expiring. Media formulation development has to be carried out for each bio similar, which takes up a significant amount of time and generally, a random approach is taken to meet comparability requirement and improve the product titer. A targeted approach with genome information of host cell as a function of media composition can provide useful information to explain the product titer variability and comparability. A 2-step correlation model explaining relationship between cell-culture productivity and quality, and media compositions taking gene expression as intermediate attributes can provide a …

Multicolumn chromatography (MCC) is a proven technique to significantly improve performance of bio-processes by increasing specific productivity, reducing volume of resin used and enabling disposable, continuous and integrated processes. But the transfer of process parameters from batch to continuous mode is perceived as complex and could limit adoption at clinical and manufacturing scales.
We have developed and tested a new method to translate data from a minimal number of single column runs into a multicolumn process with predictable performance at large scale. Depending on the titre of the product and the processing time required, the method predicts the optimal number …

At an early stage of bioprocess science, continuous operations were the workhorse in the industry. Therefore, EMD-Serono has legacy with continuous upstream processes as many of its molecules such as Rebif are being produced in perfusion mode. These processes from the 90’s consume large volumes of commercial media and demonstrate low productivities. Cells are mainly attached because no performant cell retention device existed for cells in suspension at that time.
Around 10-15 years ago, the industry decided to move on fed-batch operations. EMD-Serono also re-oriented towards fed-batch operations. Molecules such as Erbitux are now being produced in 15,000 L stainless …

Process intensification leveraging perfusion offers tremendous potential for yield improvement over fed-batch processes for the production of monoclonal antibodies. In the context of continuous processing, the goal is to achieve highly intensified perfusion processes that allow substantial footprint reduction and enable flexible adaptation in new facilities. However developing a perfusion process and medium without prior technology requires leveraging the existing fed-batch platform knowledge. Evolving a medium for perfusion relies on designing suitable mixtures of basal and feed media that serve as adequate starting points for development. Focus on optimization of the medium to decrease byproduct waste, reduce unnecessary cell growth …

From Fed-Batch To Perfusion: Productivity And Quality Considerations For A Late-Stage Program, Sen Xu, Hao Chen
Integrated Continuous Biomanufacturing II
In this poster, we will present the rapid development of a perfusion process with high productivity for late-stage manufacturing. The process uses a Chinese Hamster Ovary (CHO) cell line for monoclonal antibody production. Prior to the development, the clinical supply was manufactured by a fed-batch process with the same cell line.
To achieve desired productivity and comparable quality attributes, we developed a repeated batch shake flask model to rapidly screen media and process conditions. Shake flasks were inoculated at 10 × 106 cells/mL and maintained in a CO2 incubator with medium exchange for every 48 hours, repeated for …

Integrated Continuous Biomanufacturing II
Due to the limits of feed volume addition and the problem of amino acid counter ion, miscellaneous osmolyte, and cell growth inhibitor accumulation, the fed-batch mode of bioreactor operation for CHO cell production of protein therapeutics is inherently limited with respect to the cell densities and productivities that are achievable. Continuous or perfusion culture with cell retention can overcome some of these limitations, but suffers from the disadvantages of large volume media consumption, long times to reach peak cell densities, and complications with cell retention devices. We will describe hybrid versions of continuous culture that overcome many of these limitations …

Integrated Continuous Biomanufacturing II
Perfusion processes have traditionally been used for the generation of unstable proteins in cell culture systems. The use of perfusion for production of stable proteins has been limited by low product concentration, media costs, and system complexity. However, with the advent of new single-use technology, cell culture media specifically formulated to support high density perfusion, and high-producing cell lines, perfusion processes are gaining widespread industry attention. Additionally, perfusion processes are considered an integral part of the “Factory of the Future” vision through enabling continuous processing while delivering a product effluent with consistent product quality and concentration.
In this study, we …

Continuous Downstream Processing Of A Monoclonal Antibody Using Periodic Counter Current Chromatography (Pcc) And Straight Through Processing (Stp), Hans Blom, Christer Eriksson, Annika Forss, Helena Skoglar
Integrated Continuous Biomanufacturing II
There is an increased interest to perform process intensification in order to reduce costs and improve throughput in the development and production of monoclonal antibodies (MAbs). One solution to these demands can be to implement continuous or semi-continuous downstream processing. New emerging technologies such as periodic counter-current (PCC) chromatography and straight through processing (STP) are entering the market. Here, these two technologies were evaluated in a continuous three step chromatography MAb process. The capture step was performed with protein A media (resin) on a 3 column PCC chromatography system followed by two polishing steps which were connected in series with …

Optical Enzymatic Sensors For Continuous Monitoring Of Bioreactors, Kenneth Reardon, Brian Heinze, Trenton Danna
Integrated Continuous Biomanufacturing II
Continuous monitoring of biotechnological processes is important for control and optimization of quality and productivity. Typically, samples must be removed from the cultivation and analyzed in a laboratory to determine the concentrations of substrates and products. These time-delayed data cannot be used for real-time process control. The Process Analytical Technology initiative of the FDA supports the use of on-line measurement techniques for process development, production, and quality. An overview of sensors will be presented, focusing on in-situ sensors that contact the biological medium, and external sensors that contact the medium either optically (ex situ) or via a sterile (and disposable) …

Development of Protein A based purification platforms have simplified the downstream processing of monoclonal antibodies (mAb), the largest component of biopharmaceuticals. The ever increasing titer of the cell culture process is putting more pressure on the downstream process to further increase its productivity. The classical Protein A based mAb purification platform consists of two polishing steps, bind and elute cation-exchanger and flowthrough anion-exchanger. Cation-exchange chromatography is very efficient at the separation of HCP, leached Protein A and product-related impurities such as aggregates and fragments in bind and elute mode. However, anion-exchange chromatography is a proven technology to remove DNA, viruses, …

Exploring The Role Of Identification And Moral Disengagement In The Enjoyment Of An Antihero Television Series, Sophie Janicke, Arthur A. Raney
Communication Faculty Articles and Research
Affective disposition theory explains well the process of enjoying hero narratives but not the appeal of narratives featuring antiheroes. Recent antihero studies suggest that character identification and moral disengagement might be important factors in the enjoyment of such fare. The current study builds on this work. A sample of 101 self-identified fans and nonfans of the television series 24 viewed a condensed version of Season 1, providing evaluation of various protagonist perceptions, moral judgments, and emotional responses to the narrative, as well as overall enjoyment. As expected, fans reported greater liking of the protagonist and greater enjoyment. But more importantly, …
