Leveraging Large Data Sets In Continuous Chromatography Applications: Monitoring Critical Process Parameters Using Mvda, Engin Ayturk Nov 2015

Leveraging Large Data Sets In Continuous Chromatography Applications: Monitoring Critical Process Parameters Using Mvda, Engin Ayturk

Integrated Continuous Biomanufacturing II

Continuous multicolumn chromatography is gaining momentum in the industry as an enabling technology to establish continuous biomanufacturing platforms for monoclonal antibodies and recombinant proteins. Some companies are exploring continuous multicolumn chromatography processes as a stand-alone unit operation to improve specific productivity and/or reduce buffer consumption, while other companies are considering an integrated continuous downstream process based on continuous multicolumn chromatography.

One of the key features of continuous multicolumn chromatography is the amount of data generated during a single manufacturing batch or campaign. Traditional chromatography processes typically yield one (or a few) elution peaks per batch. Continuous chromatography processes, on the …

Understanding Acceptable Level Of Risk: Incorporating The Economic Cost Of Under-Managing Invasive Species, Alisha D. Davidson, Chad L. Hewitt, Donna R. Kashian

Environmental Science and Geology Faculty Research Publications

Management of nonindigenous species includes prevention, early detection and rapid response and control. Early detection and rapid response depend on prioritizing and monitoring sites at risk for arrival or secondary spread of nonindigenous species. Such monitoring efforts require sufficient biosecurity budgets to be effective and meet management or policy directives for reduced risk of introduction. Such consideration of risk reduction is rarely considered, however. Here, we review the concepts of acceptable level of risk (ALOR) and associated costs with respect to nonindigenous species and present a framework for aligning risk reduction priorities with available biosecurity resources. We conclude that available …

Monitoring And Control Of Reproducibility In Quasi-Continuous Integrated Production Processes Of Active Pharmaceutical Ingredients, Reiner Luttman, Sven Oliver Borchert, Jan Patrick Voss, Gesine Cornelissen

Integrated Continuous Biomanufacturing II

The development of integrated production processes include the combination and transformation of current batch oriented unit operations into linked sequential/parallel production strategies.

The presented process starts with a two-stage upstream consisting of cell cultivation and subsequent protein production, which in turn results in a five step downstream process, consisting of cell clarification via a separator, retention of cellular debris using microfiltration, concentration of the secreted product by ultrafiltration with subsequent buffer exchange through diafiltration, followed by a final purification using column chromatography.

The three main operations cell breeding, protein production and the complete downstream line are running in series, but …

Advanced Computational Tools To Enhance Continuous Monoclonal Antibody Production, Maria M. Papathanasiou, Ana Quiroga, Athanasios Mantalaris, Efstratios Pistikopoulos

Integrated Continuous Biomanufacturing II

Leading pharmaceutical companies invest high percentage of their revenue in the improvement of existing technologies used for the production of monoclonal antibodies (mAbs). Recently, there has been a paradigm shift towards the development of continuous/quasi-continuous purification operations, aiming to reduce capital and operational costs [1]. At the moment, however, there are no standardized methods and/or tools that can be used for global control and monitoring of integrated processes.

Mathematical models and advanced computational tools can be the key for the development of robust, integrated processes, as they can provide valuable insight in the process dynamics and ensure optimal operation [2]. …

Molecular Network Control Through Boolean Canalization, David Murrugarra, Elena S. Dimitrova Nov 2015

Molecular Network Control Through Boolean Canalization, David Murrugarra, Elena S. Dimitrova

Mathematics Faculty Publications

Boolean networks are an important class of computational models for molecular interaction networks. Boolean canalization, a type of hierarchical clustering of the inputs of a Boolean function, has been extensively studied in the context of network modeling where each layer of canalization adds a degree of stability in the dynamics of the network. Recently, dynamic network control approaches have been used for the design of new therapeutic interventions and for other applications such as stem cell reprogramming. This work studies the role of canalization in the control of Boolean molecular networks. It provides a method for identifying the potential edges …

Design And Synthesis Of Novel Chloroquine-Based Antimalarials, Kevin Vincent Murphy Nov 2015

Design And Synthesis Of Novel Chloroquine-Based Antimalarials, Kevin Vincent Murphy

Dissertations and Theses

Malaria is an infectious, often fatal disease that afflicts nearly 200 million people every year. The disease, characterized by recurring and extreme flu-like symptoms, is caused by the protozoan parasite Plasmodium falciparum. Victims usually contract the disease through a mosquito vector. Chloroquine is a chemotherapeutic that was introduced in the 1940s. For many years the drug was the foremost treatment of malaria, being effective and producing few side effects. Unfortunately, tolerance to chloroquine developed when the parasite evolved a resistance mechanism. Newer drugs have been developed and implemented, but these medicines also show a decreasing effect with continued administration. …

What Is The Valence Of Mn In Ga1-Xmnx N ?, Ryky Nelson, Tom Berlijn, Juana Moreno, Mark Jarrell, Wei Ku

Faculty Publications

We investigate the current debate on the Mn valence in Ga1-xMnxN, a diluted magnetic semiconductor (DMS) with a potentially high Curie temperature. From a first-principles Wannier-function analysis, we unambiguously find the Mn valence to be close to 2+ (d5), but in a mixed spin configuration with average magnetic moments of 4μB. By integrating out high-energy degrees of freedom differently, we further derive for the first time from first-principles two low-energy pictures that reflect the intrinsic dual nature of the doped holes in the DMS: (1) an effective d4 picture ideal for local physics, and (2) an effective d5 picture suitable …

Attractor-Based Obstructions To Growth In Homogeneous Cyclic Boolean Automata, Bilal Khan, Yuri Cantor, Kirk Dombrowski Nov 2015

Attractor-Based Obstructions To Growth In Homogeneous Cyclic Boolean Automata, Bilal Khan, Yuri Cantor, Kirk Dombrowski

Department of Sociology: Faculty Publications

We consider a synchronous Boolean organism consisting of N cells arranged in a circle, where each cell initially takes on an independently chosen Boolean value. During the lifetime of the organism, each cell updates its own value by responding to the presence (or absence) of diversity amongst its two neighbours’ values. We show that if all cells eventually take a value of 0 (irrespective of their initial values) then the organism necessarily has a cell count that is a power of 2. In addition, the converse is also proved: if the number of cells in the organism is a proper …

Settlement-Size Scaling Among Prehistoric Hunter-Gatherer Settlement Systems In The New World, W. Randall Haas, Cynthia J. Klink, Greg J. Maggard, Mark S. Aldenderfer Nov 2015

Settlement-Size Scaling Among Prehistoric Hunter-Gatherer Settlement Systems In The New World, W. Randall Haas, Cynthia J. Klink, Greg J. Maggard, Mark S. Aldenderfer

Anthropology Faculty Publications

Settlement size predicts extreme variation in the rates and magnitudes of many social and ecological processes in human societies. Yet, the factors that drive human settlement-size variation remain poorly understood. Size variation among economically integrated settlements tends to be heavy tailed such that the smallest settlements are extremely common and the largest settlements extremely large and rare. The upper tail of this size distribution is often formalized mathematically as a power-law function. Explanations for this scaling structure in human settlement systems tend to emphasize complex socioeconomic processes including agriculture, manufacturing, and warfare-behaviors that tend to differentially nucleate and disperse populations …

Optical Studies Of 13 Hard X-Ray Selected Cataclysmic Binaries From The Swift-Bat Survey, Jules P. Halpern, John R. Thorstensen

Dartmouth Scholarship

From a set of 13 cataclysmic binaries that were discovered in the Swift Burst Alert Telescope (BAT) survey, we conducted time-resolved optical spectroscopy and/or time-series photometry of 11, with the goal of measuring their orbital periods and searching for spin periods. Seven of the objects in this study are new optical identifications. Orbital periods are found for seven targets, ranging from 81 minutes to 20.4 hr. PBC J0706.7+0327 is an AM Herculis star (polar) based on its emission-line variations and large amplitude photometric modulation on the same period. Swift J2341.0+7645 may be a polar, although the evidence here is less …

Evaluating Corrosion Resistance Of Reinforcing Steel In A Novel Green Concrete, Andrea Carolina Ramirez Nov 2015

Evaluating Corrosion Resistance Of Reinforcing Steel In A Novel Green Concrete, Andrea Carolina Ramirez

USF Tampa Graduate Theses and Dissertations

Reinforced concrete structures are expected to have a long service life with minimal maintenance. Corrosion of reinforcing steel is a major factor in reducing concrete structure lifespan, as corrosion products occupy a larger volume than that of the consumed steel and generate tensile stresses that crack the concrete cover. Procedures to control corrosion in traditional concrete, which is made with Portland-cement (PC), have been well established. However, in recent years novel concrete materials based on alternatives to normally cured PC have been developed in response to global needs to reduce greenhouse gases emissions. In particular, a promising new cement has …

Evaluation Of Low-Cost Low Impact Development Practices In Southwest Florida For The Control Of Urban Runoff, Laura Kathren Rankin Nov 2015

Evaluation Of Low-Cost Low Impact Development Practices In Southwest Florida For The Control Of Urban Runoff, Laura Kathren Rankin

USF Tampa Graduate Theses and Dissertations

Stormwater management is required due to development and alteration of the natural environment. It is heavily regulated in Florida and at the national level. Over the last two decades, Low Impact Development (LID) has been promoted as a sustainable and environmentally friendly method of controlling urban runoff. Case studies, provided in Chapter 2, show that LIDs can restore watershed hydrology by balancing the water budget. The difference in runoff between pre-development and post-development appears to increase with soil perviousness. However, the potential for mitigating the impacts of urbanization through runoff reduction is also greater for pervious, sandy soils that dominate …

Linking Insomnia And Suicide Ideation: The Role Of Socio-Cognitive Mechanisms In Suicide Risk, Melanie Lauren Bozzay Nov 2015

Linking Insomnia And Suicide Ideation: The Role Of Socio-Cognitive Mechanisms In Suicide Risk, Melanie Lauren Bozzay

USF Tampa Graduate Theses and Dissertations

Despite what is known about predictors of suicide risk and consequences of insomnia, research has yet to delineate mechanisms that may explain the known relationship between insomnia symptoms and suicide risk. There is some disagreement in the literature regarding whether this relationship could be primarily explained by recent depressive symptoms, or whether there may be other explanatory factors related to sleep deficits. The present study addressed this contention in the literature by examining 1) whether socio-cognitive variables (e.g. fatigue, appraised social problem-solving ability, and hopelessness) explained this insomnia-ideation relationship, and 2) whether these variables contributed some explanatory variance in suicide …

Shopminer: Mining Customer Shopping Behavior In Physical Clothing Stores With Passive Rfids, Longfei Shangguan, Zimu Zhou, Xiaolong Zheng, Lei Yang, Yunhao Liu, Jinsong Han

Research Collection School Of Computing and Information Systems

Shopping behavior data are of great importance to understand the effectiveness of marketing and merchandising efforts. Online clothing stores are capable capturing customer shopping behavior by analyzing the click stream and customer shopping carts. Retailers with physical clothing stores, however, still lack effective methods to identify comprehensive shopping behaviors. In this paper, we show that backscatter signals of passive RFID tags can be exploited to detect and record how customers browse stores, which items of clothes they pay attention to, and which items of clothes they usually match with. The intuition is that the phase readings of tags attached on …

Overview Of Estimation Methods For Industrial Dynamic Systems, John Hedengren, Ammon Eaton Nov 2015

Overview Of Estimation Methods For Industrial Dynamic Systems, John Hedengren, Ammon Eaton

Faculty Publications

Measurement technology is advancing in the oil and gas industry. Factors such as wireless transmitters, reduced cost of measurement technology, and increased regulations that require active monitoring tend to increase the number of available measurements. There is a clear opportunity to distill the recent flood of measurements into relevant and actionable information. Common methods to do this include a filtered bias update, implicit dynamic feedback, Kalman filtering, and moving horizon estimation. The purpose of these techniques is to validate measurements and align imperfect mathematical models to the actual process. Additionally, they can determine a best-estimate of the current state of …

Retention specialists come from many areas of higher education. Some, such as a First Year Programs Director, seem like a very natural fit. Others come from departments not traditionally associated with retention. At Embry-Riddle Aeronautical University, which offers degree programs primarily in STEM fields, the Director of First Year Programs (FYP) had been considered the informal retention specialist of record until late 2013 when the Library Director was recruited to take on the newly created position of Dean of Retention and Student Success. This paper chronicles the ways in which the two colleagues learned about the state of retention at …

Earth Sciences

Remote sensing observations suggest Greenland ice sheet (GrIS) albedo has declined since 2001, even in the dry snow zone. We seek to explain the apparent dry snow albedo decline. We analyze samples representing 2012–2014 snowfall across NW Greenland for black carbon and dust light-absorbing impurities (LAI) and model their impacts on snow albedo. Albedo reductions due to LAI are small, averaging 0.003, with episodic enhancements resulting in reductions of 0.01–0.02. No significant increase in black carbon or dust concentrations relative to recent decades is found. Enhanced deposition of LAI is not, therefore, causing significant dry snow albedo reduction or driving …
