Most philosophical debate over mental causation has been concerned with reconciling commonsense intuitions that there are causal interactions between the mental and the physical with philosophical theories of the nature of the mental that seem to suggest otherwise. My concern is with a different and more practical problem. We often confront some cognitive, affective, or bodily phenomenon, and wonder about its source – its etiology or its underlying causal basis. For much of North Carolina’s history its General Assembly sought to strike a balance between the undeniable utility of black people’s armed labor and the threat that gun-toting black people were thought to pose. Masters equipped their slaves with firearms much like many other tools and many citizens turned to the Assembly to undertake measures to ensure that this armed labor did not compromise white people’s safety or property. The state’s legislature dictated the terms under which masters could arm their slaves and while some slaveholders defiantly used armed African-descended laborers as they wished most white people believed that armed …

Although global history has been a part of the discipline of social studies in New York State for a number of years, the inclusion of global education and skills has been less prevalent in the discipline. This lack of global perspectives in education has proven to be detrimental to the global awareness of American students. Seagrasses are a diverse group of clonal marine macrophytes. Their decline in recent decades has been an alarming component of estuarine urbanization, with large portions of historical coverage lost or degraded. Understanding patterns of dispersal and recruitment within and among the remaining populations is now critical to predicting the form and pace of recovery. Working in a developing eelgrass (Zostera marina) meadow in Shinnecock Bay, we combined field manipulations, genotypic surveys and correlative analyses to understand how vegetative growth and seedling recruitment contribute to space acquisition. Mechanical Heart Valves (MHV) provides life-saving solutions to patients suffering from cardiovascular diseases. But these devices are often plagued by non-physiological flow patterns resulting in increased shear stress conditions. This leads to platelet damage, a precursor of thromboembolism, thus impeding the device usability and demanding lifelong anticoagulation treatments. While the flow and stresses are in the micron level, the platelet behavior is at the nm level, thus presenting a major computational challenge for simulating this multiscale multi-physics modeling problem. Further, the current methods of tackling this problem involve using chemicals such as disinfectants which are harmful to us humans. Chemotherapy is one of the most commonly used cancer treatments. However, many side effects could occur due to the non-specific bindings of extremely toxic anti-cancer drugs to healthy cells and organs. Targeted delivery has been developed to enhance the selectivity of drug delivery vehicle and minimize side effects of chemotherapy. In this study, the hydrophobically modified glycol chitosan (HGC) micelle was further modified with a biotin-avidin-biotin (B-A-B) strategy to improve the tumor-selective ability of HGC. How do contemporary artworks address and give visibility to the wounded body without reinvigorating trauma, without repeating the spectacular and homogenizing strategies used by the media? How do artworks move beyond denunciation to aesthetically and historically name and rearticulate bodily violence? And fundamentally, how do artworks offer an historical understanding of the political historicity of contemporary violence without uncritically resorting to myth and structural trauma? How do Bradbury Thompson’s designs in Westvaco Inspirations work as a guide for design and paper? Westvaco Inspirations was a promotional magazine first published in 1925 by the West Virginia Pulp and Paper Company. The main purpose of the magazine was to promote paper and to show an intricate relationship between print, paper and design. Bradbury Thompson was the designer for the magazine from 1939 to its final year of publication in 1962. Protein O-fucosyltransferase 2 (POFUT2) elongates Thrombospondin Type 1 Repeats (TSR) with an O-linked fucose. Forty-nine potential proteins are modified by POFUT2 in the endoplasmic reticulum. Nearly half the POFUT2 targets belong to the A Disintegrin and Metalloprotease with ThromboSpondin type-1 motifs (ADAMTS) or ADAMTS-like family of proteins. One of POFUT2’s target proteins, ADAMTS9, contains 15 TSRs and is reported to be embryonic lethal around the same time as our Pofut2 LoxP mutants. In this study, we compared the Pofut2 and Adamts9 knockout phenotypes and used tissue specific deletion of Pofut2 to define the role of Pofut2 during gastrulation. Knockout of …

Vps34 is the sole member of the Class III phosphoinositide 3-kinases (PI3Ks) identified in mammals thus far. It phosphorylates phosphatidylinositol (PI) to generate PI(3)P on intracellular membranes. Previous work has suggested that Vps34 is important for the protein degradation pathways of autophagy and endocytosis, as well as nutrient-induced mTOR signaling. However, due to the pluripotent nature of available PI3K inhibitors, a clear understanding of the functions of mammalian Vps34 remains to be illustrated. piRNA pathway and its central components, PIWI proteins, are essential for transposon silencing and some aspects of gene regulation. Although they predominantly function in the animal germline, increasing evidence has pointed to their involvement in somatic cells. Some flatworms possess a unique somatic stem cell system named neoblasts. These totipotent cells are the only dividing cell population thus critical for postembryonic development, adult homeostasis and the remarkable regeneration ability. In the study led by Kaja A. During neuronal development, the neuronal cell polarizes to specify the structures of axons and dendrites. This polarized architecture regulates the input/output functions of the neuronal cell. In my thesis, I studied the molecular mechanisms of dendrite formation. My main hypothesis was that dendrite specification results from local elevation of the cGMP signal. Soluble guanylyl cyclase (sGC) is the major enzyme for the synthesis of cGMP in the embryonic brain. Assembly of carbon nanomaterials into three-dimensional (3D) architectures is necessary to harness their unique physiochemical properties for tissue engineering and regenerative medicine applications. In this dissertation, fabrication and characterization of 3D chemically crosslinked macro-sized (5-8 mm height and 4-6 mm diameter) porous carbon nanotube (CNT) scaffolds, their in vitro cytocompatibility and interactions with human mesenchymal stem cells (hMSCs) are reported. Single- or multi-walled carbon nanotubes (SWCNTs or MWCNTs) scaffolds were fabricated using a novel radical initiated thermal crosslinking and annealing method using SWCNTs or MWCNTs as nanoscale building blocks. Despite numerous advances in our understanding of the mechanisms that underlie Alzheimer’s disease (AD), there is still no cure, and the disease continues to pose a severe public health problem. It has become clear that many factors, including certain lifestyle practices, may contribute to the development of the disease; however, much remains to be learned about how lifestyle factors may prevent or delay cognitive decline. Actinic keratosis (AK), a cutaneous lesion resulting from the proliferation of atypical epidermal keratinocytes, has an incidence of 11%-26% and its prevalence is 58 million in the US alone. AK is a precursor to non-melanoma skin cancer (NMSC). NMSC, consisting of squamous cell and basal cell carcinomas, represents the most frequent human cancer. Tuberculosis (TB) infection is the single most lethal bacterial infections in the world. Isoniazid (INH), one of the most effective anti-TB drugs is usually administered to treat latent TB infection and is used in combination therapy to treat active TB. However resistance to INH is common and the emergence of multi-drug-resistant, extensively drug-resistant and now totally drug-resistant strains of TB, has resulted in an urgent need to develop novel anti-TB drugs. Previous research has identified puberty as a time of marked vulnerability to negative psychological outcomes, despite the fact that most individuals navigate this transition with little disturbance. The current study investigated neuroendocrine coupling, or the extent to which hormone reactivity is correlated, as a potential mechanism explaining the relationship between puberty and negative outcomes, as well as investigating early life stressors that may influence hormone coupling patterns. This thesis argues for the greater inclusion of fantasy literature in high school and middle school English classrooms. Through an analysis of the popularity of fantasy series, including recent movies and their widespread pop cultural appeal, this thesis illustrates that fantasy resonates with many readers. A close examination of various works of fantasy literature appropriate to be read in full or excerpted in the classroom demonstrates that fantasy usefully illustrates various literary concepts, such as symbolism, metaphor, allusion, allegory, and character foils; provides clear examples of the hero's journey, which appear in various canonical texts as well as other media; …

The pqbp1 (polyglutamine tract-binding protein 1) gene has been associated with a number of developmental and neurodegenerative disorders, such as Huntington’s disease, spinocerebellar ataxia type 1, and a variety of X-linked intellectual disabilities grouped together under the umbrella term Renpenning Syndrome. Renpenning Syndrome is an X-linked recessive disorder involving intellectual disability, as well as physical features such as microcephaly, micropthalmia, lean build, short stature, and small testes. Work in the Thomsen Lab previously determined that a disruption in cellular levels of PQBP1 can impact crucial developmental processes such as gastrulation and neural tube closure. The mechanisms by which transforming growth factor beta (TGF-beta) promotes lung adenocarcinoma (ADC) metastasis are largely unknown. Here, we report that in lung ADC cells TGF-beta potently induces expression of dedicator of cytokinesis 4 (DOCK4), but not other DOCK-family members, via the Smad pathway, and that DOCK4 induction mediates TGF-beta's pro-metastatic effects by enhancing tumor cell extravasation. TGF-beta-induced DOCK4 stimulates lung ADC cell protrusion, motility, and invasion, without affecting epithelial-to-mesenchymal transition (EMT). These processes, which are fundamental to tumor cell extravasation, are driven by DOCK4-mediated Rac1 activation, unveiling a novel link between TGF-beta and Rac1. Thus, our findings uncover the …

This dissertation addresses community belonging, community building, and community identity of women in the city of Granada during the early modern period, showing how women made use of their Granadan social context. It situates the women of Granada in the context of the economic and social turmoil throughout the early modern period, and in particular of the seventeenth century. Non-elite urban women in early modern Granada actively shaped the civic economy, culture, and religious life of the city and, in turn, their lives were shaped by the interplay of these elements. There is increasing recognition of the role of suspension-feeding bivalves in providing important ecosystem services (e.g. | water clarity, benthic-pelagic coupling, and habitat structure) within estuarine systems. Mercenaria mercenaria (aka hard clams, quahogs) are frequently the dominant suspension-feeder in estuarine systems across the US east coast and have had significant commercial fishery importance and a steadily growing mariculture industry. M. mercenaria have generally not had disease problems which contrasts the microbial infections that have plagued oyster fisheries. Adult born neurons in the hippocampal dentate gyrus start to assemble primary cilia at 14 days after their birth. At 21 days, all newborn neurons show a typical primary cilium. Although this small microtubule-based structure has been shown to be important for early neuronal development, its physiological/behavioral roles remain elusive. Therefore, I set out to investigate both behavioral and physiological functions of primary cilia in immature and mature dentate granule cells (DGCs) of the adult brain. After selectively knocking out IFT20 to ablate primary cilia from mature DGCs, I found defects of hippocampal spatial and cognitive memory, suggesting a crucial …
