Thyroid Autoimmunity As A Window To Autoimmunity: An Explanation For Sex Differences In The Prevalence Of Thyroid Autoimmunity, Stephen Merrill, Ying Mu
Thyroid Autoimmunity As A Window To Autoimmunity: An Explanation For Sex Differences In The Prevalence Of Thyroid Autoimmunity, Stephen Merrill, Ying Mu
Mathematics, Statistics and Computer Science Faculty Research and Publications
Autoimmune thyroid diseases (AITDs), predominately Graves׳ disease and Hashimoto׳s thyroiditis, comprise the most common autoimmune diseases in humans. Both have the production of anti-thyroid antibody as an important aspect and both are much more prevalent in females, being at least 10 times more common than in males. Using these two clues, a hypothesis for the initiation of thyroid autoimmunity is proposed that helps to make the case that the thyroid is one of the most sensitive sites for autoimmunity and helps account for the prevalence and the observed sex differences in AITDs and associated diseases, such as type 1 diabetes …

The “Stratospheric Cricket Keeper” – Developing A Simple“Life-Support” Payload For High-Altitude Balloon Missions, Lucas Kramer, Chad Serba, James Flaten
2017 Academic High Altitude Conference
Exposure to the environmental conditions of “near-space” (AKA the stratosphere) is quickly fatal to nearly all forms of animal life. It is even challenging to build a sealable enclosure that can keep insects (crickets) alive through the dramatic and simultaneous pressure and temperature drops experienced during a high-altitude balloon mission. This poster describes the development of a rugged “cricket keeper” in which we were able to fly crickets to the stratosphere and, quoting the words of JFK, “return (them) safely to the earth!” This “life-support” payload had large windows (for the view!) and included Arduino-logged temperature and pressure sensors, an …

Systemic siRNA administration to target and treat glioblastoma, one of the most deadly cancers, requires robust and efficient delivery platform without immunogenicity. Here we report newly emerged multivalent naked RNA nanoparticle (RNP) based on pRNA 3-way-junction (3WJ) from bacteriophage phi29 to target glioblastoma cells with folate (FA) ligand and deliver siRNA for gene silencing. Systemically injected FA-pRNA-3WJ RNPs successfully targeted and delivered siRNA into brain tumor cells in mice, and efficiently reduced luciferase reporter gene expression (4-fold lower than control). The FA-pRNA-3WJ RNP also can target human patient-derived glioblastoma stem cells, thought to be responsible for tumor initiation and deadly …

The human head louse is a cosmopolitan ectoparasite and frequently infests many people, particularly school-age children. Due to widespread pyrethroid resistance and the lack of efficient resistance management, there has been a considerable interest in the protection of uninfested people and prevention of reinfestation by disrupting lice transfer. In this study, two nonclinical model systems (in vitro and in vivo) were used to determine the efficacy of the infestation deterrents, Elimax lotion and Elimax shampoo, against human head lice or poultry chewing lice, respectively. With in vitro assessments, female head lice exhibited significantly higher avoidance responses to hair tufts treated …

Slavic Village is a revitalizing urban neighborhood in the southeast area of the City of Cleveland. A historic neighborhood that was once home to 75,000 immigrants of Polish and Slavic descent, it has seen steady decline since the 1960’s, along with other Cleveland neighborhoods, and was especially hard hit during the housing and foreclosure crisis of 2007-2010. However, in the present day, it has many assets and opportunities that make it poised to become one of the vibrant Cleveland neighborhoods of the future.
This study was done as part of a student project to look at the potential to re-invigorate …

Xylooligosaccharide Production From Tobacco Stalk Xylan Using Xylanase Streptomyces Sp. Bo 3.2, Muhammad Nur Kholis, Yopi, Anja Meryandini
Makara Journal of Science
Tobacco stalk (TS), which is one type of lignocellulosic material, has a xylan content of up to 21.9%. Lignocellulose can be used to produce xylooligosaccharides (XOs). XOs are dietary fibers that have prebiotic activity. This study aimed to produce XOs from tobacco stalk xylan using xylanase from Streptomyces sp. BO 3.2. After the TS was delignified, the xylan was extracted using the alkali method. The delignification process, which used 1% natrium hypoclorite (NaOCl), decreased the lignins from 32.93% to 18.15%. Xylan extraction was conducted using 10% natrium hydoroxide (NaOH); this extraction produced xylan of 15.53% (w/w). The xylanase produced by …

Activation And Inhibition Of Pannexin 1 Channels, David George Jackson
Activation And Inhibition Of Pannexin 1 Channels, David George Jackson
Open Access Dissertations
Pannexin 1 is one of three pannexins, a vertebrate protein family related to the invertebrate innexins. Unlike its innexin relatives, Pannexin 1 does not form gap junctions in vivo, but it does form plasma membrane channels that act as ATP release channels. ATP release through Pannexin 1 serves multiple physiological roles throughout the body, including in the central nervous system, or CNS. Pannexin 1 also plays a pathophysiological role in CNS injury as part of the cell death-inducing P2X7-Pannexin 1 complex. During CNS injury, increased extracellular K+ activates Pannexin 1 by an unknown mechanism. Since Pannexin 1 block might ameliorate …

Non-Operative Management Of Hip Osteoarthritis, Ray Marks
Publications and Research
This paper reviews several non-operative and non-pharmacologic management strategies advocated for alleviating the pain and disability experienced by people with hip osteoarthritis. It analyzes whether painful debilitating hip osteoarthritis, which has no effective cure and is often progressive, may be affected positively by non-operative interventions designed to control osteoar- thritis pain. Finally, it provides an integrated plan of management for ameliorating hip osteoar- thritis pain and disability in light of this knowledge.

Studies in human populations have shown a significant correlation between procollagen C-endopeptidase enhancer protein 2 (PCPE2) single nucleotide polymorphisms and plasma HDL cholesterol concentrations. PCPE2, a 52-kDa glycoprotein located in the extracellular matrix, enhances the cleavage of C-terminal procollagen by bone morphogenetic protein 1 (BMP1). Our studies here focused on investigating the basis for the elevated concentration of enlarged plasma HDL in PCPE2-deficient mice to determine whether they protected against diet-induced atherosclerosis. PCPE2-deficient mice were crossed with LDL receptor-deficient mice to obtain LDLr-/-, PCPE2-/- mice, which had elevated HDL levels compared with LDLr-/- mice with similar …
Estimation Of Optimal Productivity In Labor-Intensive Construction Operations, Krishna Prasad Kisi
Estimation Of Optimal Productivity In Labor-Intensive Construction Operations, Krishna Prasad Kisi
Department of Construction Engineering and Management: Dissertations, Theses, and Student Research
In an attempt to evaluate the efficiency of labor-intensive construction operations, project managers typically compare actual with historical productivity for equivalent operations. However, this approach toward examining productivity only provides a relative benchmark for efficiency and may lead to the characterization of operations as objectively efficient when in reality such operations might simply be comparably efficient.
Optimal productivity is the highest sustainable productivity achievable under good management and typical field conditions. Optimal productivity is useful in the determination of the absolute efficiency of construction operations because an accurate estimate of optimal labor productivity allows for the comparison of actual vs. …

The Integration Of Play Into The First-Grade Language Arts Curriculum For English Language Learners, Cheryl Jean Sanderson
School of Education and Leadership Student Capstone Theses and Dissertations
The research question addressed in this project was, can the integration of play and games with rules into the first-grade Language Arts curriculum improve the oral speaking and reading skills for English language learners? To investigate the answer to that question, I studied how a child’s first language develops. For children who learn a second language, Krashen’s models were utilized in learning how a second language is acquired. Play and games with rules are defined. Looking primarily to Piaget and Vigotsky, I learned about the development of the child, play, games with rules and how a child learns through play. …

A potential mechanism by which highly pathogenic avian Influenza A virus subtype H5N1 could more readily infect human beings is through the infection of and adaptation in pigs. To detect the occurrence of such infection, monitoring of pig populations through serological screening would be highly desirable. In the current study, hemagglutination inhibition assays were able to detect antibodies against H5N1 developed in pigs, but because of antigenic variation between clades, the use of multiple virus strains were required. Whole recombinant virus and recombinant hemagglutinin antigen enzymelinked immunosorbent assays (ELISAs) were generated that could detect antibody against multiple H5N1 strains, but …

Translational Neuroscience
The existence of α7β2 nicotinic acetylcholine receptors (nAChRs) has recently been demonstrated in both the rodent and human brain. Since α7-containing nAChRs are promising drug targets for schizophrenia and Alzheimer's disease, it is critical to determine whether α7β2 nAChRs are present in the human brain, in which brain areas, and whether they differ functionally from α7 nAChR homomers. We used α-bungarotoxin to affinity purify α7-containing nAChRs from surgically excised human temporal cortex, and found that α7 subunits co-purify with β2 subunits, indicating the presence of α7β2 nAChRs in the human brain. We validated these results by demonstrating co-purification of β2 …

Loris Malaguzzi And The Teachers: Dialogues On Collaboration And Conflict Among Children, Reggio Emilia 1990, Carolyn Edwards, Lella Gandini, John Nimmo
Zea E-Books Collection
In 1990, three American scholars participated in an extraordinary research experience with Loris Malaguzzi and the educators of the Diana School in Reggio Emilia, Italy. They were studying “cooperation”— how preschool educators promoted collaboration and community in their classrooms and schools—and they used videotapes of classroom episodes to provoke teachers to reflect on the meanings suggested by the actions of themselves and others. In October 1990 the three traveled to Reggio Emilia and spent several days with the Italian educators.
The Diana School faculty viewed these encounters as powerful opportunities for their own professional development through the documentation process, rather …

Chromatin To Clinic: The Molecular Rationale For Parp1 Inhibitor Function., Felix Y. Feng, Johann S. De Bono, Mark A. Rubin, Karen E Knudsen
Department of Cancer Biology Faculty Papers
Poly(ADP-ribose) polymerase 1 (PARP1) inhibitors were recently shown to have potential clinical impact in a number of disease settings, particularly as related to cancer therapy, treatment for cardiovascular dysfunction, and suppression of inflammation. The molecular basis for PARP1 inhibitor function is complex, and appears to depend on the dual roles of PARP1 in DNA damage repair and transcriptional regulation. Here, the mechanisms by which PARP-1 inhibitors elicit clinical response are discussed, and strategies for translating the preclinical elucidation of PARP-1 function into advances in disease management are reviewed.
