Excess iron induces tissue damage and is implicated in age-related macular degeneration (AMD). Iron toxicity is widely attributed to hydroxyl radical formation through Fenton's reaction. We report that excess iron, but not other Fenton catalytic metals, induces activation of the NLRP3 inflammasome, a pathway also implicated in AMD. Additionally, iron-induced degeneration of the retinal pigmented epithelium (RPE) is suppressed in mice lacking inflammasome components caspase-1/11 or Nlrp3 or by inhibition of caspase-1. Iron overload increases abundance of RNAs transcribed from short interspersed nuclear elements (SINEs): Alu RNAs and the rodent equivalent B1 and B2 RNAs, which are inflammasome agonists. Targeting …

The phosphorylation status of the BAD protein is a determinant of ovarian cancer cell responsiveness to platinum chemotherapy. Indirect manipulation of BAD phosphorylation status influences cisplatin sensitivity. BAD phosphorylation represents a biomarker that predicts platinum sensitivity and is a therapeutic target to increase platinum sensitivity. The methods employ phospho-specific antibody against a particular amino acid residue or site. Phospho-specific protein characterization methods include immunohistochemical (IHC), flow cytometric, immunofluorescent, capture-and-detection, or reversed phase assay.

Heidegger's Gods: An Ecofeminist Perspective, Susanne Claxton Jun 2015

Heidegger's Gods: An Ecofeminist Perspective, Susanne Claxton

Philosophy ETDs

The aim of this dissertation is to address the importance and significance of Heidegger's engagement with the Greeks and the ways in which his views are commensurate with ecofeminism and the insights that a study of that intersection provides. I defend the thesis that a proper return to myth and art as a means by which the transcendental realities that constitute the phenomenology of our embodied existence may be better understood is what may allow us to truly dwell in the Heideggerian sense and live full lives rich in meaning and value. My methodology might best be described as a …

The Sox9 Upstream Region Prone To Chromosomal Aberrations Causing Campomelic Dysplasia Contains Multiple Cartilage Enhancers, Baojin Yao, Qiuqing Wang, Chia-Feng Liu, Pallavi Bhattaram, Wei Li, Timothy J. Mead, James F. Crish, Véronique Lefebvre Jun 2015

The Sox9 Upstream Region Prone To Chromosomal Aberrations Causing Campomelic Dysplasia Contains Multiple Cartilage Enhancers, Baojin Yao, Qiuqing Wang, Chia-Feng Liu, Pallavi Bhattaram, Wei Li, Timothy J. Mead, James F. Crish, Véronique Lefebvre

Biomedical Sciences

Two decades after the discovery that heterozygous mutations within and around SOX9 cause campomelic dysplasia, a generalized skeleton malformation syndrome, it is well established that SOX9 is a master transcription factor in chondrocytes. In contrast, the mechanisms whereby translocations in the –350/–50-kb region 5 of SOX9 cause severe disease and whereby SOX9 expression is specified in chondrocytes remain scarcely known. We here screen this upstream region and uncover multiple enhancers that activate Sox9-promoter transgenes in the SOX9 expression domain. Three of them are primarily active in chondrocytes. E250 (located at – 250 kb) confines its activity to condensed prechondrocytes, …

Nonparametric Bayes Approach For A Semi-Mechanistic Pharmacokinetic And Pharmacodynamic Model, Yan Dong Jun 2015

Nonparametric Bayes Approach For A Semi-Mechanistic Pharmacokinetic And Pharmacodynamic Model, Yan Dong

Mathematics & Statistics ETDs

Both frequentist and Bayesian approaches have been used to characterize population pharmacokinetics and pharmacodynamics(PK/PD) models. These methods focus on estimating the population parameters and assessing the association between the characteristics of PK/PD and the subject covariates. In this work, we propose a Dirichlet process mixture model to classify the patients based on their individualized pharmacokinetic and pharmacodynamic profiles. Then we can predict the new patients dose-response curves given their concentration-time profiles. Additionally, we implement a modern Markov Chain Monte Carlo algorithm for sampling inference of parameters. The detailed sampling procedures as well as the results are discussed in a simulation …

Background: Daily rhythms in mammals are programmed by a master clock in the suprachiasmatic nucleus (SCN). The SCN contains two main compartments (shell and core), but the role of each region in system-level coordination remains ill defined. Herein, we use a functional assay to investigate how downstream tissues interpret region-specific outputs by using in vivo exposure to long day photoperiods to temporally dissociate the SCN. We then analyze resulting changes in the rhythms of clocks located throughout the brain and body to examine whether they maintain phase synchrony with the SCN shell or core. Results: Nearly all of the 17 …

Begone, Euclid!: Leasing Custom And Zoning Provision Engaging Retail Consumer Tastes And Technologies In Thriving Urban Centers, Michael N. Widener

Pace Law Review

Is urban center retailing in a death spiral? Competition for consumers with Internet vendors is afoot; winners and losers shall be anointed. The threats to physical retailing in an era of the “Internet of Goods” initially are described below. Adaptations by tenants, landlords, and stakeholders in urban centers will be required quickly, and new perspectives and partnerships, including those among local and regional governments, are instrumental if physical retail operations in municipal cores are to survive. The balance of this article describes these needs from the vantage point of each stakeholder; but this article argues that integrating information and communication …

Understanding The Sympathetic Nervous System In Response To Food In Children With Autism And Typically Developing Children, Jonathon D. Haskell

Occupational Therapy Graduate Student Evidenced-Based Research Reviews

Previous research behind sympathetic nervous system functioning in children with Autism Spectrum Disorder has shown disturbances in their ability to regulate their arousal level (Miller et al., 2003; Tomcheck and Dunn, 2000). Depicting this notion in previous research, children diagnosed with autism are likely to be comorbid with Sensory Over-responsitivity (SOR) or Sensory under-responsitivity (SUR), and could ultimately elicit variable sympathetic nervous system functioning in response to their sensory environment-whether it be taste, smell, auditory, tactile, or visual stimuli. Although previous research depicts inconsistent measures involving sympathetic responses to stimuli in children with autism, research specifically involving food selectivity provided …

Effect Of Social Skill Training Programs On Bullying, Kaycee Johns, Megan Kloska Jun 2015

Effect Of Social Skill Training Programs On Bullying, Kaycee Johns, Megan Kloska

Occupational Therapy Graduate Student Evidenced-Based Research Reviews

Bullying is a problem that is often found in schools during the adolescence age. Although this is not a new concept, bullying continues to lead to the victimization of children and adolescents all over the world. Research has played a critical role in understanding bullying, victimization, and prevention programs. However, bullying and victimization still continue to be problems within schools and peer relationships.

The primary purpose of this systematic review is to assist practitioners by investigating evidence-based studies related to school social skill programs, and their effectiveness in reducing victimization of bullying throughout adolescence.

Effectiveness Of Executive Function Performance Test (Efpt) In Stroke Population, Faryal Shaheen Jun 2015

Effectiveness Of Executive Function Performance Test (Efpt) In Stroke Population, Faryal Shaheen

Occupational Therapy Graduate Student Evidenced-Based Research Reviews

Stroke is the leading cause of long term disability in the U.S, causing 15,000 deaths each year (Hall et al, 2015). It has been estimated that 71% of clients are discharged with minimal to no services from acute/subacute after stroke because of deficits that go undetected (2015).

One of the most common deficits in stroke is loss in executive functioning (EF). Occupational therapists (OTs) have typically used a combination of tools to measure EF and most of these assessments did not involve observations in the everyday environment (Morrison et al, 2015). A new measure, known as the Executive Function Performance …

Effects Of Reducing Fatigue In Muscular Dystrophy Through Physical Activity, Callie Toaso, Alea Brown Jun 2015

Effects Of Reducing Fatigue In Muscular Dystrophy Through Physical Activity, Callie Toaso, Alea Brown

Occupational Therapy Graduate Student Evidenced-Based Research Reviews

Myopathy is a functional impairment of muscle tissue; muscular dystrophy is a type of myopathy (Rakowicz & Lane, 2004). Muscular dystrophy (MD) consists of progressive muscle weakness and affects limbs and facial muscles. The most common types include Duchenne muscular dystrophy, Fascioscapulohumeral muscular dystrophy (FSHD), and Limb-girdle muscular dystrophy (Mercuri & Muntoni, 2013). One common complaint found with patients with muscular dystrophy is fatigue. It is noted that fatigue affects daily activities, but it is believed that physical activity can help reduce this. The purpose of this systematic review is to research how effective physical activity is at reducing fatigue …

Estrogen receptor–α (ERα) activity in the brain prevents obesity in both males and females. However, the ERα-expressing neural populations that regulate body weight remain to be fully elucidated. Here we showed that single-minded–1 (SIM1) neurons in the medial amygdala (MeA) express abundant levels of ERα. Specific deletion of the gene encoding ERα (Esr1) from SIM1 neurons, which are mostly within the MeA, caused hypoactivity and obesity in both male and female mice fed with regular chow, increased susceptibility to diet-induced obesity (DIO) in males but not in females, and blunted the body weight–lowering effects of a glucagon-like peptide-1–estrogen …

BACKGROUND. TGF-β has potent profibrotic activity in vitro and has long been implicated in systemic sclerosis (SSc), as expression of TGF-β–regulated genes is increased in the skin and lungs of patients with SSc. Therefore, inhibition of TGF-β may benefit these patients.

METHODS. Patients with early, diffuse cutaneous SSc were enrolled in an open-label trial of fresolimumab, a high-affinity neutralizing antibody that targets all 3 TGF-β isoforms. Seven patients received two 1 mg/kg doses of fresolimumab, and eight patients received one 5 mg/kg dose of fresolimumab. Serial mid-forearm skin biopsies, performed before and after treatment, were analyzed for expression …

Recombinant subunit vaccines should contain minimal non-pathogen motifs to reduce potential off-target reactivity. We recently developed a vaccine antigen against respiratory syncytial virus (RSV), which comprised the fusion (F) glycoprotein stabilized in its pre-fusion trimeric conformation by “DS-Cav1” mutations and by an appended C-terminal trimerization motif or “foldon” from T4-bacteriophage fibritin. Here we investigate the creation of a cyste- ine zipper to allow for the removal of the phage foldon, while maintaining the immunogenic- ity of the parent DS-Cav1+foldon antigen. Constructs without foldon yielded RSV F monomers, and enzymatic removal of the phage foldon from pre-fusion F trimers resulted in …

Gonad: Genome-Editing Via Oviductal Nucleic Acids Delivery System: A Novel Microinjection Independent Genome Engineering Method In Mice., Gou Takahashi, Channabasavaiah B. Gurumurthy, Kenta Wada, Hiromi Miura, Masahiro Sato, Masato Ohtsuka

Journal Articles: Genetics, Cell Biology & Anatomy

Microinjection is considered the gold standard technique for delivery of nucleic acids (NAs; transgenes or genome editing tools such as CRISPR/Cas9 systems) into embryos, for creating genetically modified organisms. It requires sophisticated equipment as well as well-trained and highly skilled personnel to perform the micro-injection technique. Here, we describe a novel and simple microinjection-independent technique, called Genome-editing via Oviductal Nucleic Acids Delivery (GONAD). Using GONAD, we show that NAs (e.g., eGFP mRNA or Cas9 mRNA/sgRNAs) can be effectively delivered to pre-implantation embryos within the intact mouse oviduct by a simple electroporation method, and result in the desired genetic modification in …

The Qualitative Report

This inquiry was conducted to describe the experience of individuals with autism spectrum disorders who have experienced the higher education system. All participants have been diagnosed with an autism spectrum disorder and experienced some aspect of the higher education system. Data was collected using primarily face to face interviews. The data was then analyzed using the heuristic methodology of Dr. Clark Moustakas (1990). Ultimately, many patterns and themes emerged from this analysis, culminating in a creative synthesis which sums up the experience. The themes showed the highs and lows of being a college student coupled with the experience of having …

“What’S Race Got To Do With It?” An Investigation Into The Differences Between Self-Reported Television Exposure And Self-Reported Attitudes Towards Interracial Dating, Kimberly Reeb Jun 2015

“What’S Race Got To Do With It?” An Investigation Into The Differences Between Self-Reported Television Exposure And Self-Reported Attitudes Towards Interracial Dating, Kimberly Reeb

Theses

Television has the power to suggest what its viewers should think about and to impact individuals’ attitudes towards controversial topics. The present study examined the differences between individuals’ self-reported television exposure and self-reported interracial dating attitudes (IDA). Television viewing habits and demographics were also analyzed. A survey distributed via social media and email collected 216 responses. Cultivation theory suggests heavy viewers are likely to have more negative perceptions and attitudes. Analysis of differences for IDA between viewers was not statistically significant, but the IDA score was slightly more negative for heavy viewers. Overall, IDA scores were positive, however individuals’ attitude …

Dialogical Interspecies Ethics: Ataraxia, Desire And Hope In The Post-Human World Of Anne Carson's Pastoral, Thomas Bristow Dr

The Goose

This review essay implicitly revisits human and non-human power relations within a critical animal studies context that understands the affective conjunction between the manipulation of our worlds (action, partly through knowledge) and degrees of involvement with these others that live in our worlds (comportment via emotions). I take Louise Westling’s new study as the platform for an analysis of two book-length poems, The Autobiography of Red (1998) and red doc> (2013), which centre on the life of a shepherd, Geryon. Rather than revisit classical pastoral, these texts extract power-relations that classical myth and pastoral spatialise. In so doing, I argue, …
