Alpha-fetoprotein (AFP) represents a classical model system to study developmental gene regulation in mammalian cells. We previously reported that liver ZBTB20 is developmentally regulated and plays a central role in AFP postnatal repression. Here we show that ZBTB20 is a sequence-specific transcriptional repressor of AFP. By ELISA-based DNA-protein binding assay and conventional gel shift assay, we successfully identified a ZBTB20-binding site at -104/-86 of mouse AFP gene, flanked by two HNF1 sites and two C/EBP sites in the proximal promoter. Importantly, mutation of the core sequence in this site fully abolished its binding to ZBTB20 in vitro, as well …

B Lymphocytes Are Required During The Early Priming Of Cd4+ T Cells For Clearance Of Pneumocystis Infection In Mice, Michael M. Opata, Melissa L. Hollifield, Frances E. Lund, Troy D. Randall, Robert Dunn, Beth A. Garvy, David J. Feola

Microbiology, Immunology, and Molecular Genetics Faculty Publications

B cells play a critical role in the clearance of Pneumocystis. In addition to production of Pneumocystis-specific Abs, B cells are required during the priming phase for CD4+ T cells to expand normally and generate memory. Clearance of Pneumocystis was found to be dependent on Ag specific B cells and on the ability of B cells to secrete Pneumocystis-specific Ab, as mice with B cells defective in these functions or with a restricted BCR were unable to control Pneumocystis infection. Because Pneumocystis-specific antiserum was only able to partially protect B cell–deficient mice from infection, we …

Corrosion Of Phosphor Bronze In Different Biodiesel Blends / Rathinee Balakumar, Balakumar Rathinee

Student Works (2010-2019)

Biodiesel fuel is gaining rapid recognition around the globe as a reliable alternative to the use of petroleum fuel in the automotive industry. This safer, greener, renewable energy source originates from naturally forming feedstocks such as animal fats and vegetable oils. Biodiesel blends consist of mixtures of biodiesel with diesel at different composition. Biodiesel fuel is much more corrosive when compared to petroleum fuel, thus giving rise to many compatibility issues with the material construction of a typical combustion ignition engine. This research studies the effects of Tert-butylamine and Benzotriazole additive on the corrosion behaviour of phosphor bronze in different …

It has been known for several decades that serotonergic neurotransmission is a key regulator of cognitive function, mood, and sleep. Yet with the relatively recent discoveries of novel serotonin (5-HT) receptor subtypes, as well as an expanding knowledge of their expression level in certain brain regions and localization on certain cell types, their involvement in cognitive processes is still emerging. Of particular interest are cognitive processes impacted in neuropsychiatric and neurodegenerative disorders. The prefrontal cortex (PFC) is critical to normal cognitive processes, including attention, impulsivity, planning, decision-making, working memory, and learning or recall of learned memories. Furthermore, serotonergic dysregulation within …

Integrin-mediated cell adhesion to the ECM regulates many physiological processes in part by controlling cell proliferation. It is well established that many normal cells require integrin-mediated adhesion to enter S phase of the cell cycle. Recent evidence indicates that integrins also regulate cytokinesis. Mechanical properties of the ECM can dictate entry into S phase; however, it is not known whether they also can affect the successful completion of cell division. To address this issue, we modulated substrate compliance using fibronectin-coated acrylamide-based hydrogels. Soft and hard substrates were generated with approximate elastic moduli of 1600 and 34,000 Pascals (Pa) respectively. Our …

Reel Women: Gender Stereotypes In Film, Kayla Marie Krahn Jul 2015

Reel Women: Gender Stereotypes In Film, Kayla Marie Krahn

Master's Theses and Doctoral Dissertations

Horror films portray female characters as sexual, helpless victims. When horror films attempt to portray female characters as the hero, this sends a message to audiences that women can be strong and independent. An experiment was conducted to test an audience's perceptions gender stereotypes of women in the horror film, Scream. Results indicate that stereotypes present in the horror film effected the participants' perception of women as competent heroes. Female participants were also more likely to perceive women as more competent than male participants. Then, from a qualitative standpoint, five individuals were interviewed about their perceptions of characters in their …

The prevalence of allergic airway diseases such as asthma and rhinitis has increased dramatically to epidemic proportions worldwide. Besides air pollution from industry derived emissions and motor vehicles, the rising trend can only be explained by gross changes in the environments where we live. The world economy has been transformed over the last 25 years with developing countries being at the core of these changes. Around the planet, in both developed and developing countries, environments are undergoing profound changes. Many of these changes are considered to have negative effects on respiratory health and to enhance the frequency and severity of …

Daubert And Judicial Review: How Does An Administrative Agency Distinguish Valid Science From Junk Science?, D. Hiep Truong

Akron Law Review

This broad authority to assess risk, however, leaves too much discretion to administrative agencies. Even more disturbing is the fact that different agencies assess the same risks differently, which leads to inconsistent results. The Environmental Protection Agency (EPA), for example, in determining the cancer risks from pesticides on food, produced an estimated risk of cancer mortality ten times greater than the Food and Drug Administration (FDA). To use a law and economics model, valuing equivalent (or identical) risks differently leaves open the possibility of economic misallocation. For example, if one agency has determined the proper level of risk, and assuming …

Emerging evidence demonstrates that the DNA repair kinase DNA-PKcs exerts divergent roles in transcriptional regulation of unsolved consequence. Here, in vitro and in vivo interrogation demonstrate that DNA-PKcs functions as a selective modulator of transcriptional networks that induce cell migration, invasion, and metastasis. Accordingly, suppression of DNA-PKcs inhibits tumor metastases. Clinical assessment revealed that DNA-PKcs is significantly elevated in advanced disease and independently predicts for metastases, recurrence, and reduced overall survival. Further investigation demonstrated that DNA-PKcs in advanced tumors is highly activated, independent of DNA damage indicators. Combined, these findings reveal unexpected DNA-PKcs functions, identify DNA-PKcs as a potent driver …

Earthquake Stories From Minto And Nenana, Alaska, Carl Tape

Faculty

On Wednesday October 15, 1947 at 4:10pm local time, a magnitude 7.2 earthquake struck Interior Alaska, near Healy. This is a collection of stories of six life-long Alaskan elders who felt this earthquake and shared their recollections in fall of 2014, sixty-seven years after the earthquake. Geraldine Charlie had recently turned 18 years old and worked in the village store in Minto when the earthquake hit. Geraldine was crouching down to weigh a bag of potatoes at the moment the earthquake hit. She felt dizzy and noticed Coleman lanterns swaying from side to side, and items shook off the shelves. …

The Role Of Philornis Downsi In Avian Disease Transmission In The Galápagos Islands, Courtney Lauren Pike Jul 2015

The Role Of Philornis Downsi In Avian Disease Transmission In The Galápagos Islands, Courtney Lauren Pike

Theses

Philornis downsi is a dipteran fly species known to parasitize passerine birds in the Neotropics. P. downsi is speculated to be a recently introduced species in Galápagos Islands, so it is important to understand its effects on the archipelago’s native and endemic avian species. Avian parasites and pathogens have been previously reported in the Galápagos, leading the Galápagos National Park and the Charles Darwin Foundation to focus efforts on the study and management of potential avian disease vectors. The main goal of this study was to determine whether P. downsi is a host for avian parasites and pathogens found in …

Dumbbell-Pcr: A Method To Quantify Specific Small Rna Variants With A Single Nucleotide Resolution At Terminal Sequences., Shozo Honda, Yohei Kirino

Computational Medicine Center Faculty Papers

Recent advances in next-generation sequencing technologies have revealed that cellular functional RNAs are not always expressed as single entities with fixed terminal sequences but as multiple isoforms bearing complex heterogeneity in both length and terminal sequences, such as isomiRs, the isoforms of microRNAs. Unraveling the biogenesis and biological significance of heterogenetic RNA expression requires distinctive analysis of each RNA variant. Here, we report the development of dumbbell PCR (Db-PCR), an efficient and convenient method to distinctively quantify a specific individual small RNA variant. In Db-PCR, 5'- and 3'-stem-loop adapters are specifically hybridized and ligated to the 5'- and 3'-ends of …
