A method of dissolved methane analysis was developed utilizing cavity ring-down spectroscopy and headspace equilibration. Samples of 70 mL were collected in 140 mL plastic syringes and equilibrated with a methane free headspace. Reproducibility was high (i.e. 4% typical RSD), and samples were successfully measured in the low nanomolar to high micromolar range. During method development, multiple research cruises were undertaken in the northern Gulf of Mexico. Stations included the Orca Basin, the Deepwater Horizon site, and the surrounding area. The Deepwater Horizon site showed no continuing leakage from October 2010 to June 2013. Samples collected from the northern Gulf …

The manner in which dolphin calves acquire their whistle repertoire is largely unknown. This paper focuses on whistle development in four bottlenose dolphin calves during the first thirty days of life in order to increase our understanding of the early emergence of whistles and whistle-like vocalizations. The acoustic parameters of whistle-type vocalizations (i.e., whistles and whistlesquawks) that coincided with a bubblestream emission from the focal calf and/or its mother were analyzed, as were the behavioral states of the mother-calf pair during the emission of such vocals. Live benthic foraminiferal assemblages were studied at contaminated and uncontaminated bathyal sites around the wellhead of the Deepwater Horizon oil spill. Samples collected in October of 2010 and 2011 and summer of 2012 were divided into uncontaminated (GIP 12, 21, K, and 25 and Obs0), moderately oiled (GIP 16 and 17) and heavily oiled groups (GIP 15), in which the TPAH concentrations ranged from 29 to 7,553 ng/g in 2010. Metric multidimensional scaling (MDS) and cluster analyses were conducted to compare all surface samples. Karenia brevis is a mixotrophic, marine dinoflagellate found in the Gulf of Mexico that generates periodic, if not annual, harmful algal blooms (also known as “red tides”) in certain coastal areas. In an effort to better understand the biology of this organism, a functional genomics project has been initiated. As part of that project, it has been determined that a significant number of natural antisense transcripts (NATs) as well as double-stranded RNA (dsRNA) molecules exist within the transcriptome of K. brevis. Several studies indicate that seasonal applications of road deicers can lead to saline runoff entering surface water and groundwater, which can in turn change the chemistry, density, and physical mixing patterns of urban lakes. An investigation at Asylum Lake, an urban lake located near three major roadways, was conducted to determine the degree of road salt contamination in the lake, to determine if salt loading disrupts seasonal mixing patterns, and to determine the extent of redox stratification present due to eutrophic conditions. This thesis is a case study of a family of farmers in Mississippi’s Piney Woods who use horse-drawn equipment to grow crops. This practice is notable in the context of a larger agricultural system that prioritizes mechanization (particularly the use of tractors). The production style and their use of local direct selling via farmers markets allowed them to thrive economically because they were able to tap into a niche market of consumers desiring an alternative to the modern, conventional agricultural system.

Through layer-by-layer method, the authors have constructed three Au nanoplexes: AuPEI/RNA/PEI, AuPEI/RNA/PEI-mPEG, and AuPEI/NA/PEI-PEG-FA. All the nanoplexes are characterized by UV-vis spectrometry, DLS, and zeta potential. The surface density of the first layer PEI and second layer RNA were also determined. Moreover, the Au nanoplexes can protect siRNA from RNase degradation and are stable in cell culture medium.

Contextual (e.g. job fit, job involvement), individual (e.g. job satisfaction, need for achievement), and demographic (e.g. gender, educational level) factors have been related to forms of career commitment (i.e. affective, continuance, and normative commitment), highlighting that the commitment one feels toward his or her career is a complex variable. Furthermore, commitment has been associated with intent to remain within a profession or organization (Bowling, Beehr, & Lepisto, 2006; Den Hartog & Belschak, 2007; Duffy, Dik, & Steger, 2011; Goulet & Singh, 2002), suggesting that commitment is an important component of retention within a career. Dolphin communication is multimodal and incorporates physical behaviors and vocalizations. Dolphins often exchange information with conspecifics using different types of vocalizations, and these vocalizations are sometimes associated with specific behaviors. However, the relationship of vocalization type and mouthing behavior type has not been investigated. This thesis examines simultaneous acoustic and visual recordings of bottlenose dolphins (Tursiops truncatus) to determine the relationship between type of mouthing behavior and type of vocalization (whistle, whistle-squawk, chirp, moan, burst-pulse type A, burst-pulse type B, and click trains). The current study examined associations between insecure adult romantic attachment and symptoms of psychopathology, disordered personality, and problematic sexual behaviors in a clinical sample of 402 men in treatment for sexual addiction. Anxious and avoidant attachment were hypothesized to correlate with certain constructs of psychopathology, disordered personality, and dimensions of problematic sexual behaviors. Multiple regression analyses were conducted to examine the hypothesized relationships using data from scales of the Minnesota Multiphasic Personality Inventory- 2-Revised Form (MMPI-2-RF), the Sexual Dependence Inventory – 4.0 (SDI – 4.0), and the Experiences in Close Relationships-Revised (ECR-R). Urbanization in coastal watersheds is becoming an increasingly important issue in the world. Increased impervious surface cover, a factor of urbanization, has drawn more attention to stormwater runoff as a source of contamination to receiving waters. In this study, nitrogen species from rain and stormwater were analyzed across three different landscape types along the Mississippi Sound (hardened, residential, and pristine), as well as from drainage pipes that flow directly into to the Mississippi Sound. Nitrogen stable isotopes were used to trace the stormwater nitrogen to the surface waters and biota within the Sound.

The mature eye lens contains a surface layer of epithelial cells called the lens epithelium that requires a functional mitochondrial population to maintain the homeostasis and transparency of the entire lens. The lens epithelium overlies a core of terminally differentiated fiber cells that must degrade their mitochondria to achieve lens transparency. These distinct mitochondrial populations make the lens a useful model system to identify those genes that regulate the balance between mitochondrial homeostasis and elimination. This study investigated the effect of perceived gender coaching style and gender stereotypes on the extrinsic motivation of female athletes. A t-test was utilized, with Masculine and feminine coaching styles as independent variables. The research question asked whether female athlete’s extrinsic motivation could be affect based on the type of gender coaching style used by female head coaches. H1 predicted that a masculine coaching style, respectively, would negatively influence the extrinsic motivation of female athletes. This work addresses the coordination issue in distributed optimization problem (DOP) where multiple distinct and time-critical tasks are performed to satisfy a global objective function. The performance of these tasks has to be coordinated due to the sharing of consumable resources and the dependency on non-consumable resources. Knowing that it can be sub-optimal to predefine the performance of the tasks for large DOPs, the multi-agent reinforcement learning (MARL) framework is adopted wherein an agent is used to learn the performance of each distinct task using reinforcement learning. Citizen science projects can serve as constructivist learning environments for programming focused on science, technology, engineering, and math (STEM) for youth. Attributes of rich environments for active learning (REALs) provide a framework for design of Extension STEM learning environments. Guiding principles and design strategies for the University of Minnesota Extension's Driven to Discover: Enabling Authentic Inquiry through Citizen Science project demonstrate how education and investigations grounded in real-world citizen science projects can capitalize on REAL environments to generate meaningful STEM learning. Positive evaluation results support the efficacy of the design for enhancing youth science identity and practice.

Recent work has demonstrated that competition and facilitation likely operate jointly in plant communities, but teasing out the relative role of each has proven difficult. Here we address how competition and facilitation vary with seasonal fluctuations in environmental conditions, and how the effects of these fluctuations change with plant ontogeny. We planted three sizes of pine seedlings (Pinus strobus) into an herbaceous diversity experiment and measured pine growth every two weeks for two growing seasons. Negative density dependence (NDD) and habitat specialization have received strong empirical support as mechanisms that explain tree species diversity maintenance and distribution in tropical forests. In contrast, disturbance appears to play only a minor role. Previous studies have rarely examined the relative strengths of these diversity maintenance mechanisms concurrently, and few studies have included plant groups other than trees. Here we used a large, spatially explicit data set from Barro Colorado Island, Panama (BCI) to test whether liana and tree species distribution patterns are most consistent with NDD, habitat specialization, or disturbance. Florida Tech’s campus-wide GTA professional development training, the GTA Seminar, has no clear theory on which it was designed, employs only measures of satisfaction and open-ended feedback, and performs limited follow-up on GTAs. Further, two pilot studies I conducted indicate that GTAs’ concerns may not be fully addressed by this seminar. This is notable because the GTA Seminar is often the only means of professional development available to more than half of the GTAs on campus. Birds are well-established in cycles of zoonotic diseases, generally as hosts of an infected arthropod vector such as a mosquito or tick. Since the 1980s, research in both the United States and Europe has focused on their role in the ecology of Lyme disease and its causative agent, Borrelia burgdorferi, since they are fed upon by larval and nymphal ticks and can transport them and their associated diseases geographically during migrations. Understanding the long-term effects and possible toxicity of nanoceria, a widely utilized commercial metal oxide, is of particular importance as it is poised for development as a therapeutic agent based on its autocatalytic redox behavior. We show here evidence of acute and subacute adverse hepatic responses, after a single infusion of an aqueous dispersion of 85 mg/kg, 30 nm nanoceria into Sprague Dawley rats. Light and electron microscopic evidence of avid uptake of nanoceria by Kupffer cells was detected as early as 1 hr after infusion. This paper explores new, game-based volunteering platforms in the sciences and discusses their viability for nonprofit organizations, which have long used gaming for fundraising but not typically in other aspects of their operations. The paper unfolds in two parts. Examples of game-based volunteering platforms in the sciences are examined in Part 1, and their broader significance discussed in regard to the history of science and the history of gaming. Currently all species of sea turtles are listed as threatened or endangered with extinction under the U.S. Endangered Species Act. In order to effectively construct management approaches we need as much information on various sea turtle populations as possible including demography, genetic origin, and critical habitat. One demographic piece of data that is lacking is the sex ratio of turtle populations in foraging habitats, as this information is integral in determining overall population abundance. Because secondary sex characteristics (i.e. males have longer tails) are not evident until turtles start to reach sexual maturity, the sex of juvenile turtles cannot be …

Insulin resistance which precedes Type II diabetes is a complex state of the body where the body fails to respond to insulin.
