Kombucha(Khubdat Humza) is composed of yeast and acetic acid bacteria especially, Acetobacter xylinum which forms a cellulose pellicle on tea broth. Kombucha(Khubdat Humza) produces bacterial cellulose pellicles, with unique purity and fine structure. It can be used in many forms, such as an emulsifier, stabilizer, dispersing agent, thickener and gelling agent but these are generally subsidiary to its most important use of holding on to water. Recently, bacterial cellulose is used in many special applications such as a scaffold for tissue engineering of cartilages and blood vessels, also for artificial skin for temporary covering of wounds, as well as its …

No single analytical method can cover the whole metabolome and the choice of which platform to use may inadvertently introduce chemical selectivity. In order to investigate this we analysed a collection of uropathogenic Escherichia coli. The selected strains had previously undergone extensive characterisation using classical microbiological methods for a variety of metabolic tests and virulence factors. These bacteria were analysed using Fourier transform infrared (FT-IR) spectroscopy; gas chromatography mass spectrometry (GC-MS) after derivatisation of polar non-volatile analytes; as well as reversed-phase liquid chromatography mass spectrometry in both positive (LC-MS(+ve)) and negative (LC-MS(-ve)) electrospray ionisation modes. A comparison of the discriminatory …

Sphingosine 1-phosphate (S1P) is a bioactive lipid that is formed by the phosphorylation of sphingosine and catalysed by sphingosine kinase 1 (SK1) or sphingosine kinase 2 (SK2). Sphingosine kinases play a fundamental role in many signaling pathways associated with cancer, suggesting that proteins belonging to this signaling network represent potential therapeutic targets. Over the last years, many improvements have been made in the treatment of T-cell acute lymphoblastic leukemia (T-ALL); however, novel and less toxic therapies are still needed, especially for relapsing and chemo-resistant patients. Here, we analyzed the therapeutic potential of SKi and ROMe, a sphingosine kinase 1 and …

Understanding spatial patterns in freshwater fish communities is critical for the successful management of natural resources as well as a vital component for understanding aquatic ecosystems. Spatial patterns of species similarity of freshwater fish assemblages can be affected by dispersal processes and environmental conditions. We hypothesized that as distance increased between study systems, species similarity would decrease. We sampled 15 drowned river mouths (DRMs) connected to Lake Michigan by conducting 10-minute electrofishing transects (n = 5-6 per DRM) parallel to the shoreline in each DRM to characterize littoral fish assemblages. At each transect, we also characterized environmental conditions (e.g., specific …

ΔNp63α, a proto-oncogene, is up-regulated in non-melanoma skin cancers and directly regulates the expression of both Vitamin D receptor (VDR) and phosphatase and tensin homologue deleted on chromosome ten (PTEN). Since ΔNp63α has been shown to inhibit cell invasion via regulation of VDR, we wanted to determine whether dietary Vitamin D3 protected against UVB induced tumor formation in SKH-1 mice, a model for squamous cell carcinoma development. We examined whether there was a correlation between dietary Vitamin D3 and ΔNp63α, VDR or PTEN expression in vivo in SKH-1 mice chronically exposed to UVB radiation and fed chow containing …

We have recently reported that mouse embryonic stem cells (mESCs) are deficient in expressing type I interferons (IFNs) in response to viral infection and synthetic viral RNA analogs (Wang, R., Wang, J., Paul, A. M., Acharya, D., Bai, F., Huang, F., and Guo, Y. L. (2013) J. Biol. Chem. 288, 15926–15936). Here, we report that mESCs are able to respond to type I IFNs, express IFN-stimulated genes, and mediate the antiviral effect of type I IFNs against La Crosse virus and chikungunya virus. The major signaling components in the IFN pathway are expressed in mESCs. Therefore, the basic molecular mechanisms …

Development Of A Digital Noise Exposure System For Research On Noise Induced Hearing Loss, Jun Qin, Qing Wu, Jacob Walker, Kathleen Campbell, Daniel Fox
Conference Proceedings
Over thirty million Americans suffer from noise induced hearing loss (NIHL). Previous research demonstrated that different types of noises, even with equal sound energies, could produce different amounts of hearing loss. In this project, a novel digital noise exposure system has been developed for generating various noise signals (e.g. pure-tone, Gaussian, impulsive, and complex noise). The developed system can be used to study NIHL in animal models. The system could produce impulse noise with peak sound pressure level (SPL) up to 160 dB, which effectively mimics the noise generated by a military weapon (e.g., M-16 rifle). The preliminary results of …

Use Of Caenorhabditis Elegans As A Model To Study Alzheimer’S Disease And Other Neurodegenerative Diseases, Adanna G. Alexander, Vanessa Marfil, Chris Li
Publications and Research
Advances in research and technology has increased our quality of life, allowed us to combat diseases, and achieve increased longevity. Unfortunately, increased longevity is accompanied by a rise in the incidences of age-related diseases such as Alzheimer’s disease (AD). AD is the sixth leading cause of death, and one of the leading causes of dementia amongst the aged population in the USA. It is a progressive neurodegenerative disorder, characterized by the prevalence of extracellular Aβ plaques and intracellular neurofibrillary tangles, derived from the proteolysis of the amyloid precursor protein (APP) and the hyperphosphorylation of microtubule-associated protein tau, respectively. Despite years …

The intergeniculate leaflet (IGL) plays an important role in the entrainment of circadian rhythms and the mediation of acute behavioral responses to light (i.e., masking). Recently, we reported that IGL lesions in diurnal grass rats result in a reversal in masking responses to light as compared to controls. Here, we used Fos as a marker of neural activation to examine the mechanisms by which the IGL may influence this masking effect of light in grass rats. Specifically, we examined the patterns of Fos activation in retinorecipient areas and in brain regions that receive IGL inputs following 1-h light pulses given …

Background: An algebraic method for information fusion based on nonadditive set functions is used to assess the joint contribution of Boolean network attributes to the sensitivity of the network to individual node mutations. The node attributes or characteristics under consideration are: in-degree, out-degree, minimum and average path lengths, bias, average sensitivity of Boolean functions, and canalizing degrees. The impact of node mutations is assessed using as target measure the average Hamming distance between a non-mutated/wild-type network and a mutated network.
Results: We find that for a biochemical signal transduction network consisting of several main signaling pathways whose nodes represent signaling …

GDP-6-deoxy-D-talose and its 4-epimer GDP-D-rhamnose are unusual sugars found on the cell surface of certain Gram-negative bacteria. GDP-6-deoxy-talose and GDP-rhamnose are produced by enzymes called GTS and RMD, respectively. These sugars are produced from the same common intermediate and are 4'-epimers of each other, which provides an interesting paradigm to study the stereo-selective reduction of unusual sugars. We have determined a novel X-ray crystallographic structure of RMD from Pseudomonas aeruginosa that contains its cosubstrate NADPH within the active site. A superposition of our RMD structure with a previously-determined one gives clear insight into the mechanism of hydride transfer from the …
