Ratiometric approaches to quantifying molecular concentrations have been used for decades in microscopy, but have rarely been exploited in vivo until recently. One dual-tracer approach can utilize an untargeted reference tracer to account for non-specific uptake of a receptor-targeted tracer, and ultimately estimate receptor binding potential quantitatively. However, interpretation of the relative dynamic distribution kinetics is confounded by differences in local tissue absorption at the wavelengths used for each tracer. Microalgae are gaining attention as a potential feedstock for the production of biodiesel, mainly derived from triacylglycerols (TAG). In many algae, TAG synthesis increases dramatically upon certain stresses but this is often accompanied by growth retardation. Rational improvements to strain productivity are limited by the scant knowledge on algal lipid metabolism and gene regulatory mechanisms. In this context, systems-level approaches aimed at understanding and modeling metabolic and regulatory networks may enable hypothesis-driven genetic engineering strategies. The green microalga Chlamydomonas reinhardtii accumulates significant amounts of TAGs under nutrient starvation and provides a genetically tractable model for manipulating biosynthetic pathways. Chemotherapy either before or after surgery is a common breast cancer treatment. Long-term, high dose treatments with chemotherapeutic drugs often result in undesirable side effects, frequent recurrences and resistances to therapy. The anti-cancer drug, gemcitabine (GEM) was used in combination with pulse power technology with nanosecond pulsed electric fields (nsPEFs) for treatment of human breast cancer cells in vitro. eNOS (endothelial nitric oxide synthase) contains a MAPK (mitogen-activated protein kinase)-binding site associated with a major eNOS control element. Purified ERK (extracellular-signal-regulated kinase) phosphorylates eNOS with a stoichiometry of 2–3 phosphates per eNOS monomer. Phosphorylation decreases NO synthesis and cytochrome c reductase activity. Three sites of phosphorylation were detected by MS. All sites matched the SP and TP MAPK (mitogen-activated protein kinase) phosphorylation motif. Ser602 lies at the N-terminal edge of the 42-residue eNOS AI (autoinhibitory) element. The pentabasic MAPK-binding site lies at the opposite end of the AI, and other critical regulatory features are between them. Thr46 and Ser58 …

Online social network participation is widespread among American adolescents. Prolific creators, consumers and curators of content, they write themselves into being (boyd, 2007) on social network sites like Facebook. Drawing on Erving Goffman's study of symbolic interaction in the form of dramaturgical perspective and The Third Person Effect, this research explores how young women ages 14-17 craft their self-presentations, engage in impression management, and experience aggression and bullying on Facebook. The work presented here is an exploration of the poetry and life of Jean Sénac, and through Sénac, of the larger role of poetry in the political and social movements of the 50s, 60s, and early 70s, mainly in Algeria and America. While Sénac was part of the European community in Algeria, his position regarding French rule changed dramatically over the course of the Algerian War, (between 1954 and 1962) and upon independence, he became one the rare French to return to his adopted homeland. Bottlenose dolphins are neuroanatomically different and evolutionarily divergent from primates yet they exhibit mirror self-recognition (MSR), a rare cognitive ability in non-human animals. This research investigated the developmental and age-related aspects of MSR in this species. During a longitudinal study, a social group of bottlenose dolphins at the National Aquarium, Baltimore, MD were exposed to a mirror and their behavioral responses were recorded to: 1) further confirm the presence of MSR in this species, 2) determine the age of emergence of MSR and 3) draw comparisons with data documenting the emergence of this ability in humans and great ape species. …

Work-family (or work-life) studies aim to measure interactions between the realms of work and home. It is necessary to examine these interactions within a broad context to understand external sources of tension on the work-life dynamic, including environmental, economic, and political factors. Exploratory interviews were conducted with participants of working age with a mobility disability, and when applicable, their significant others. Questions focused on work, home and transportation environments. Sex offender risk instruments provide empirically based outlooks on recidivism risk and often serve as a critical part of sex offender management. If applied to unrepresented offender groups, these instruments may offer inaccurate pictures of risk and hinder efforts to reduce sexual violence. A component tree is a topological and geometric image descriptor that captures information regarding the structure of an image based on the connected components determined by different grayness thresholds. Classical ecological theory predicts that generally similar species ought to partition resources in order to minimize competition amongst themselves. This basic idea becomes complex when one is dealing with species that migrate over thousands of miles and forage in a broad diversity of habitats and geographical locations. I studied a suite of migratory sandpipers, and asked whether they partitioned niches at a major migratory stopover in Delaware Bay. During migration, shorebirds form large, usually mixed-species flocks, which forage on marshes, mudflats, beaches or similar two-dimensional habitats where all individuals are distributed on the same horizontal plane. Highly oxygenated cyclohexanes comprise the structures of a number of pharmacologically interesting molecules, including potently bioactive natural products and carbapyranosides. The latter, which are unnatural analogues of carbohydrates in which the ring oxygen of the parent sugar is replaced with a methylene group, have attracted interest as hydrolytically stable mimetics of their parent O-glycosides. This research reports the development of two general methodologies for the synthesis of highly oxygenated cyclohexanes: (1) Oxocarbenium Ion Cyclization (OCC) and (2) Ring Closing Metathasis (RCM).

Phylogenetic studies in the Melastomataceae have demonstrated the need of taxonomic rearrangements in the current classification. Melastomes are among the most diverse groups of plants and several cases of known artificial taxa are observed and awaiting further resolution. One example is the Leandra s.str. clade, which includes the majority of the taxa traditionally treated in the genus Leandra. This research project considered two questions regarding forensic interviewers: Do forensic interviewers hold implicit biases toward people with disabilities? If so, could this influence whether a forensic interviewer finds a child with a disability believable? To examine these questions, a quantitative exploratory study was conducted. Using an online survey, participants were randomly assigned to read a scenario about a child's disclosure of sexual abuse (children with and without a disability), and respond to questions about the believability of the child. Participants then completed an adapted version of the Disability Attitude Implicit Association Test (DA-IAT). The results yielded four significant findings. …

The work presents the results of optical studies of Xe-related defect in diamond. This defect is one of a few having narrow zero-phonon line in the near-infrared part of the photo-luminescence spectra. It appears in diamond after Xe+ ion implantation followed by thermal annealing. Given unique physical properties of diamond (hardness, optical transparency in wide spectral range, chemical inertness, high thermal conductivity, low thermal expansion coefficient) and stability of Xe-related center it can be viewed as an potential candidate for the source of single-photons, or as optically manipulated qubit, not unlike nitrogen-vacancy center. This dissertation focused on the methodological and theoretical improvement of correlative ecological niche models (ENMs) to better understand the processes governing species distributions and associated evolutionary divergence in rodents inhabiting mesic conditions in the Neotropics. Focusing on a widespread rodent from northern South America (Heteromys anomalus), in the first chapter I proposed and tested a methodological approach to surmount the challenge of incorporating environmental information from the margins of species geographic ranges into ENMs. In so doing, I argue how populations that exist on the borders of species' local ranges (spatial margins) can lead to exaggerated estimates of their niches …

Pavlovian incentive motivation provides a theoretical framework on the basis of which experiments can be designed to examine the effects of motivational variables on timing behavior. The Pavlovian-Instrumental transfer (PIT) paradigm is a prototype for studying Pavlovian incentive motivation. The current study examined the effect of a Pavlovian appetitive conditioned stimulus (CS) on timing performance, based on the PIT paradigm. Nine pigeons were exposed to pairings of a 120-s conditioned stimulus (flashing or steady houselight) and unconditioned stimulus (food). The pigeons were then trained on the peak procedure (FI 30 s). The plasma membrane consists of lipids and proteins. Among the integral membrane proteins are P-type ATPases, widely expressed in both prokaryotes and eukaryotes, which use ATP to translocate ions across a plasma membrane. Type IV enzymes are putative aminophospholipid translocases (APLTs) and catalyze phosphatidylserine (PS) transfer into the cytosolic leaflet of a lipid bilayer. Previously, our group showed pronounced PS externalization in Atp8a1 (-/-) mice but not wild type (WT). Candida albicans adhesins have amyloid-forming sequences (Ramsook et al. 2010, Otoo et al. 2008). Similarly, Tango and Waltz predicted that amyloid-forming sequences are also present in Saccharomyces cerevisiae flocculins, Flo1p and Flo11p. The cell surface of Flo1p- and Flo11p-expressing cells have ordered domains that are thioflavin T fluorescent and Congo red birefringent, two hallmarks of amyloids. Flo1p and Flo11p amyloids were important for activities of the flocculins including cell-to-cell adhesion, cell-to-substrate adhesion, and agar invasion. In addition, amyloid-perturbing dyes thioflavin S and Congo red inhibited aggregation, biofilm formation and agar invasion.

In recent decades music psychologists have explained the functioning of tonal music in terms of the tonal hierarchy, a stable schema of relative structural importance that helps us interpret the events in a passage of tonal music. This idea has been most influentially disseminated by Carol Krumhansl in her 1990 monograph Cognitive Foundations of Musical Pitch. The nasopharynx is a centrally located region of the upper respiratory tract (URT) integral to several physiological functions. However, few have focused on this area within the context of human evolution. This study investigated osseous morphology, soft tissue histology, development, and evolutionary change of the nasopharynx. Multimodal analyses were performed:

Analysis 1: This study tested hypotheses on the morphological relationships of the osseous nasopharyngeal boundaries with the splanchnocranium and basicranium among dry crania representing humans and non-human primates using 3D geometric morphometrics (3D-GM). The formation of struvite, MgNH4PO4*6H2O (MAP), from wastes is one of the methods that can be used to recover P from wastes efficiently. However, since there are usually toxic components in the wastes, like arsenic (As), the possibility of having toxic contaminants in MAP is a big concern. So, the interaction between As and MAP during coprecipitation (CPT) and adsorption (ADS) processes were studied at pH 8-11. MAP precipitated without As at pH 8-11 was also characterized.

Cocaine addiction is a serious public health hazard, and contributes to disastrous outcomes for individuals who suffer from it. Addiction is accompanied by an inability to control one's own behavior, and a preoccupation with cocaine at the expense of other rewarding pursuits.
