The neurotransmitter acetylcholine (Ach) is arguably the most important in the CNS. Two classes of receptors are activated by Ach: nicotinic (ionotropic) and muscarinic (metabotropic). Muscarinic receptor pharmacology is well-defined, but still lacks receptor sub-type specificity seen in other GPCR families like adrenergic receptors. We have found that Gβ5-RGS7, a unique G-protein obligate heterodimer complex, selectively attenuates Ca2+ signaling through the muscarinic M3 receptor (M3R). The G protein beta subunit Gβ5 uniquely forms heterodimers with R7 family regulators of G protein signaling (RGS) proteins (RGS6, RGS7, RGS9, and RGS11) instead of Gγ. While the Gβ5-RGS7 complex attenuates Ca2+ signaling mediated …

Background
In order for Informed Consent to be ethical and valid each clinical trial participant must be able to make a voluntary decision to participate, free from pressure or coercion. Nonetheless, many factors may influence the decision reached, and such influences may be different for male and female volunteers. Being aware of these differences may help researches develop better processes for obtaining consent that safeguard the right of autonomy for all participants. The goal of this study was to evaluate potential gender-based differences in the factors influencing clinical trial participation.

Background: The Lung Open Ventilation Study (LOV Study) compared a low tidal volume strategy with an experimental strategy combining low tidal volume, lung recruitment maneuvers, and higher plateau and positive end-expiratory pressures (PEEP) in adults with acute respiratory distress syndrome (ARDS). Herein, we compared sedative, opioid, and neuromuscular blocker (NMB) use among patients managed with the intervention and control strategies and clinicians' assessment of comfort in both groups. Methods: This was an observational substudy of the LOV Study, a randomized trial conducted in 30 intensive care units in Canada, Australia, and Saudi Arabia. In 16 centers, we recorded daily doses …
The Development Of A Valid And Reliable Biogeochemistry Concept Inventory, Chris Mead
The Development Of A Valid And Reliable Biogeochemistry Concept Inventory, Chris Mead
Discipline-Based Education Research Group Speaker Series
Concept inventories are a commonly used tool to measure conceptual understanding. To date, concept inventories have been published for geology, chemistry, and biology, but no instrument has been designed to measure conceptual understanding at the intersection of those fields. To fill that gap, we constructed a 32‐item biogeochemistry concept inventory (BGC‐CI). Item response theory analysis, using the Rasch model, shows the BGC‐CI is a reliable and valid tool to measure the biogeochemistry knowledge of science majors. Because biogeochemistry is an interdisciplinary field, we were concerned about the unidimensionality of the instrument. However, our analysis showed the BGC‐CI to be acceptably …

The Use Of Artificial Neural Networks To Describe And Predict The Presence Of Harmful Algae In The Indian River Lagoon, Florida, Erin L. Faltin
The Use Of Artificial Neural Networks To Describe And Predict The Presence Of Harmful Algae In The Indian River Lagoon, Florida, Erin L. Faltin
USF St. Petersburg campus Master's Theses (Graduate)
Harmful algal blooms are a natural phenomenon of growing global concern. Dense blooms of single celled phytoplankton can have wide reaching effects on both the aquatic ecosystem and surrounding economies. This study constructed artificial neural network models of the northern Indian River Lagoon, Florida, using an existing dataset. Models attempted to both describe and predict chlorophyll a, as an indicator of total algal biomass, or Pyrodinium bahamense, a dinoflagellate known to bloom and produce the paralytic shellfish toxin saxitoxin in the lagoon. Descriptive models used current data while predictive models used time-lagged data as input. Further analyses were conducted on …

Latino Men Managing Hiv: An Appraisal Analysis Of Intersubjective Relations In The Discourse Of Five Research Interviews, Will Caston
Dissertations and Theses
Latino men, particularly those who have sex with other men, have been disproportionately affected by HIV/AIDS. Scholars have sought for nearly two decades to understand how various social and cultural factors in the Latino community exacerbate HIV risk among these men. Although following the advent of life-sustaining medications in 1996, HIV is often regarded as a manageable chronic illness, as opposed to a death sentence, scant attention has been devoted to how HIV-positive Latino men experience managing the illness. Among studies that have focused on HIV-positive persons' illness management, few Latino men have participated.

The Stories We Tell: A Qualitative Inquiry To Multiracial Family Storytelling, Mariko O. Thomas
The Stories We Tell: A Qualitative Inquiry To Multiracial Family Storytelling, Mariko O. Thomas
Dissertations and Theses
A narrative inheritance is comprised of the stories told by family members that are received by a younger generation and used to help construct identity. According to the communication theory of identity, identity is formed through communication. Additionally, the storied resource perspective looks at narratives as a major method of creating and maintaining identity. This study looks at the kinds of narrative inheritance concerning race that people in multiracial families receive and possible ways it affects racial identity formation. Findings from 12 semi-structured interviews indicate that narratives of racism, cultural pride, and hardship are prevalent in multiracial families. Additionally, findings …

Genome-wide association studies (GWAS) of colorectal cancer (CRC) have led to the identification of a number of common variants associated with modest risk. Several risk variants map within the vicinity of TGFβ/BMP signaling pathway genes, including rs4939827 within an intron of SMAD7 at 18q21.1. A previous study implicated a novel SNP (novel 1 or rs58920878) as a functional variant within an enhancer element in SMAD7 intron 4. In this study, we show that four SNPs including novel 1 (rs6507874, rs6507875, rs8085824, and rs58920878) in linkage disequilibrium (LD) with the index SNP rs4939827 demonstrate allele-specific enhancer effects in a large, multi-component …

Chip-Seq And In Vivo Transcriptome Analyses Of The Aspergillus Fumigatus Srebp Srba Reveals A New Regulator Of The Fungal Hypoxia Response And Virulence, Dawoon Chung, Bridget M. Barker, Charles C. Carey, Brittney Merriman
Dartmouth Scholarship
The Aspergillus fumigatus sterol regulatory element binding protein (SREBP) SrbA belongs to the basic Helix-Loop-Helix (bHLH) family of transcription factors and is crucial for antifungal drug resistance and virulence. The latter phenotype is especially striking, as loss of SrbA results in complete loss of virulence in murine models of invasive pulmonary aspergillosis (IPA). How fungal SREBPs mediate fungal virulence is unknown, though it has been suggested that lack of growth in hypoxic conditions accounts for the attenuated virulence. To further understand the role of SrbA in fungal infection site pathobiology, chromatin immunoprecipitation followed by massively parallel DNA sequencing (ChIP-seq) was …

Examining Activity Organization In Plazas Through Geochemical Analysis At Tlalancaleca, Puebla, Mexico (800 Bc-Ad 100), Paige Gale Phillips
USF Tampa Graduate Theses and Dissertations
This research aims to understand the organization of activities across a prehispanic urban center at the Formative period site of Tlalancaleca (800 BC- AD 100), located in Puebla, Mexico. This study analyzes soil samples at the central civic-ceremonial complex of Cerro Grande in an attempt to understand the use of space. This work is a part of the larger Proyecto Arqueológico Tlalancaleca, Puebla (PATP), which is focused on understanding the socio-political organization at Tlalancaleca that led to this site of early urbanism. Soil samples from Tlalancaleca are analyzed using three chemical methods to perform a cross-comparison of analytical methods. These …
