Aging And Cancer: Molecular Facts And Awareness For Turkey, Gülgün Gündüz, Kayahan Fişkin
Turkish Journal of Biology
Two important subjects concerning extension of the human life-span are biological aging processes and their relationship to diseases, and especially to the formation of cancer. It is clear that an aging population poses some challenges for developing countries. We set out to show the demographic structure of Turkey and to discuss the feasibility of decreasing the prevalence of cancer. We overviewed the literature on the relationship between biological aging and cancer formation, the environmental causes of cancer, and age-related characteristics of cancer in the elderly. In addition, some statistical data on the changing demography of Turkey were reviewed. It is …

Autophagy And Cancer, Hacer Ezgi̇ Karakaş, Devri̇m Gözüaçik
Turkish Journal of Biology
Autophagy is an evolutionary conserved intracellular degradation and stress response mechanism that is mainly responsible for the breakdown and recycling of cytoplasmic materials, including long-lived proteins, protein aggregates, and damaged organelles. In this way, autophagy provides the cell with building blocks and allows the maintenance of homeostasis under stress conditions such as growth factor deficiency, nutrient deprivation, hypoxia, and toxins. Consequently, abnormalities of autophagy contribute to a number of pathologies ranging from neurodegenerative diseases to cancer. Autophagy was reported to have a dual role in cancer. Depending on cancer stage, autophagy seems to act as tumor suppressor or as a …

Invadopodia: Proteolytic Feet Of Cancer Cells, Gi̇zem Bati, Devri̇m Pesen Okvur
Turkish Journal of Biology
The leading cause of death in cancer patients is metastasis. Invasion is an integral part of metastasis and is carried out by proteolytic structures called invadopodia at the cellular level. In this introductory review, we start by evaluating the definition of invadopodia. While presenting the upstream signaling events involved, we integrate current models on invadopodia. In addition, we discuss the significance of invadopodia in 2D and 3D and in vivo. We finally point out technical challenges and conclude with open questions in the field.

It Takes 2 Antioxidants To Tango: The Interaction Between Manganese Superoxide Dismutase And Glutathione Peroxidase-1, Dede N. Ekoue, Alan M. Diamond
Turkish Journal of Biology
Reactive oxygen species (ROS) are generated during mitochondrial oxidative metabolism. Accumulation of ROS without an effective antioxidant response can lead to oxidative stress, resulting in macromolecular damage that is implicated in the etiology of various diseases including cancer. ROS detoxification is regulated by various antioxidant proteins, specifically manganese superoxide dismutase (MnSOD), which catalyzes the conversion of superoxide into H_{2}O_{2}, and the subsequent conversion of H_{2}O_{2} into water is catalyzed by glutathione peroxidase 1 (GPx-1). In vitro and in vivo evidence supports a conflicting role of MnSOD in tumor biology and indicates that an interaction between MnSOD and GPx-1 can modulate …

Failure Of Immunological Cells To Eradicate Tumor And Cancer Cells: An Overview, Rohit Sharma, Daizee Talukdar, Parth Malik, Tapan K. Mukherjee
Turkish Journal of Biology
Inflammation can be broadly understood as a successive immune response of an organism's immune system towards nonnative or foreign antigens. This is a protective mechanism of the immune system, mediated by diverse immunological cells, to ensure homeostasis of an individual. Once activated, these immunological cells release a number of cytokines, chemokines, reactive oxygen species, reactive nitrogen species, histamines, prostaglandins, and other materials leading to inflammation. Tumor cells express altered proteins due to mutations of their genes, DNA modifications such as histone modification, DNA methylation, or other mechanisms of altered protein expression. The body's immunological cells actively recognize these altered proteins, …

Current Paradigms Of Cancer Chemoprevention, Rajendra G. Mehta
Turkish Journal of Biology
Cancer chemoprevention has been continually evolving ever since the term was coined in the 70s. From the original approaches of identifying chemopreventive agents from the dietary constituents based on the epidemiological data to the current status of adapting them from the molecular targets, significant understanding of cancer as a disease and its possible prevention has been achieved. The identification of the chemopreventive agent from a complex mixture of natural product extracts involves numerous steps including bioassay guided fractionation; evaluation of the extracts, fractions and isolated new chemicals selectively using in vitro and in vivo experimental models; and understanding the mechanism …

Turkish Journal of Biology
Roscovitine and purvalanol are specific cyclin-dependent kinase (CDK) inhibitors, which induce apoptosis by triggering cell cycle arrest in various cancer cells such as colon, prostate, and breast cancer cells. Although the apoptotic action of roscovitine was clarified at the molecular level, the exact mechanism of purvalanol-induced apoptosis is still under investigation. The mitogen-activated protein kinase (MAPK) signaling cascade is activated by different inducers related to growth, proliferation, differentiation processes, or environmental stress factors. Recent reports showed that modulation of MAPKs might lead to regulation of c-Myc, which is a transcription factor for the polyamine (PA) biosynthesis enzyme, ornithine decarboxylase (ODC). …

Cytotoxic And Apoptotic Effects Of Ceranib-2 Offering Potential For A New Antineoplastic Agent In The Treatment Of Cancer Cells, Djanan Vejselova, Hati̇ce Mehtap Kutlu, Gökhan Kuş, Selda Kabadere, Ruhi̇ Uyar
Turkish Journal of Biology
Fibrocarcinomas are malignant tumours that originate in mesenchymal cells. The tumours typically form in the presence of connective tissue and are of key consideration when assessing research priorities for future healthcare management. Ceramidase inhibitors, such as ceranib-2, have demonstrated capacity to interfere with cellular DNA functionality, initiating apoptosis in many cancer cell lines. The enzyme ceramidase can regulate cellular levels of sphingosine and sphingosine-1-phosphate by controlling hydrolysis of ceramide. The present study investigates the antigrowth effects of ceranib-2 on mouse embryonic fibroblast cells (NIH/3T3 normal cell line) and rat embryonic fibroblast cells (5RP7 cancer cell line). Research was conducted using …

The Choice Of Auxin Analogue For In Vitro Root Induction Influences Post-Induction Root Development In Eucalyptus Grandis, Muhammad Nakhooda, Maria Paula Watt, David Mycock
Turkish Journal of Agriculture and Forestry
Previous studies on in vitro rooting for improved micropropagation of eucalypts indicated that root graviperception and post-acclimatisation architecture are determined by the relative exogenous auxin analogue and its stability, supplied during the pre-rooting culture stages. The specific roles of indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) in the rooting medium on the in vitro root morphological processes were explored using a good-rooting clone. In vitro rooting percentage was significantly reduced when either of the auxin inhibitors 2,3,5-triiodobenzoic acid (TIBA) and ρ-chlorophenoxyisobutyric acid (PCIB) or the auxin antagonist kinetin was supplied at rooting, with or without exogenous auxin. For all treatments, …

Genetic Comparison Between Natural And Planted Populations Of Pinus Brutia And Cupressus Sempervirens In Syria, Batoul N. Al-Hawija, Viktoria Wagner, Isabell Hensen
Turkish Journal of Agriculture and Forestry
There is wide consensus that ongoing deforestation contributes to global warming and poses a threat to species diversity. Less understood is whether the practice of creating plantations might also erode genetic diversity and undermine the genetic structure of tree populations. We tested these hypotheses in natural and planted populations of Pinus brutia Ten. subsp. brutia and Cupressus sempervirens L. var. horizontalis (Mill.), 2 important forestry species in the Mediterranean region. We used plant material from 3 different bioclimatic regions in Syria. Using RAPD markers, we evaluated the genetic diversity and structure of 12 populations of P. brutia (6 natural, 6 …

Turkish Journal of Biology
Glucocorticoids (GCs) induce cell cycle arrest and apoptosis in lymphoid cells and constitute a central component in the treatment of lymphoid malignancies. The molecular basis of this clinically important phenomenon remains, however, poorly understood. Using whole genome expression profiling we have previously identified glucocorticoid response genes in children with acute lymphoblastic leukemia (ALL). The promyelocytic leukemia zinc finger (PLZF) appeared as one of the most promising candidate genes, which has been implicated in the pathogenesis of several leukemia types. We have already established that transgenic PLZF reduced the sensitivity to GC-induced apoptosis in the CEM-C7H2-2C8 leukemic cell line and knockdown …

Turkish Journal of Agriculture and Forestry
Irrigation water can be polluted by the tobacco etch virus (TEV), which causes serious economic loss in tobacco. However, it is difficult to monitor the sanitary status of irrigation water because TEV presents at extremely low concentrations. This study designed a procedure for concentrating TEV from a standard water sample using polyethylene glycol and detecting it using SYBR Green-based quantitative real-time polymerase chain reaction (qRT-PCR). The concentration factors were optimized through orthogonal tests and the highest recovery efficiency of the TEV genome from a standard water sample was 92.05%. The sensitivity was evaluated using nine 10-fold dilution series of TEV …

Chloroplast Dna Variation And Pollen Contamination In A Pinus Brutia Ten. Clonal Seed Orchard: Implication For Progeny Performance In Plantations, Behi̇ye Banu Bi̇lgen, Nuray Kaya
Turkish Journal of Agriculture and Forestry
Pollen contamination is one of the important factors affecting the yield, adaptability, and genetic quality of the seed produced from seed orchards in forest tree breeding programs. Incoming pollen from the forests surrounding the seed orchard is a major concern in tree breeding because it contributes to losses in the expected genetic gains from seed orchard crops. The genetic variation and the level of pollen contamination in a 16-year-old Pinus brutia Ten. first-generation clonal seed orchard was studied using chloroplast microsatellite markers (cpSSRs). In total, 23 alleles and 36 unique allelic combinations (haplotypes) were detected based on the 6 cpSSR …

Exhaustive exercise is a strong stress factor and can impact cytokine production in the brain. Interleukin-6 (IL-6) is produced in greater amounts than any other cytokine in response to exercise, and its effects are closely related to both exercise duration and intensity. In this study, we measured the differences between the amount of IL-6 reactivity of the hippocampus after an exhaustive session of running in long-term exercise-trained and untrained rats. Materials and methods: The exercise-trained group ran on a treadmill for 12 weeks. Both groups were forced to run until exhaustion. Each group of rats was sacrificed immediately, 1 day, …

To determine the roles of hepcidin and its related genes in a renal ischemia/reperfusion model. Materials and methods: A total of 20 Wistar albino rats were equally divided into 2 groups: Group I was the control group and Group II was the ischemia and reperfusion (I/R) group (60 min of ischemia + 48 h of reperfusion). I/R was performed on the left kidneys of these rats and then the I/R-treated kidneys were removed. The levels of serum biochemical markers were evaluated after renal I/R. The expression levels of hepcidin-linked genes [growth differentiation factor 15 (GDF-15), bone morphogenetic protein 6 (BMP6), …

To investigate serum asymmetric dimethyl arginine (ADMA) levels in maintenance hemodialysis (HD) patients and to assess their potential correlations with C-reactive protein (CRP), albumin, and cholesterol levels as the established cardiovascular and nutritional parameters. Materials and methods: Forty-nine patients on maintenance HD treatment and 22 healthy volunteers with similar age and sex characteristics were recruited into the study. Serum albumin, CRP, creatinine, calcium, phosphate, total cholesterol, low-density lipoprotein (LDL) cholesterol, high-density lipoprotein (HDL) cholesterol, hemoglobin, white blood cell counts, and serum ADMA levels were measured. Results: HD patients had significantly higher ADMA levels compared with healthy controls (0.51 ± 0.25 …
